Studies on the Effects of Some Chemicals on the Germinative Stimulatilon of Clostridiu chauvoei Spore (우기종저균(牛氣腫疽菌) 아포(芽胞)에 대(對)한 몇가지 화학제(化學劑)의 발아촉진(發芽促進) 시험(試驗))

  • The effects of some chemicals for the germinative stimulation of Clostridium chauvoei spore studied. The results obtained were as follows: 1. Cooked meat medium (CMM) was superior to Liver-liver bouillon (LLB) for cultivation of the organisms. 2. Heating the organisms at $70^{\circ}C$ for 30 minutes prior to cultivation in CMM stimulated the germination rate. 3. The addition of 0.5mM L-alanine to CMM was found to be most effective for the rapid germination of the spores. 4. The addition of 0.2 mM D-alanine to CMM inhibited the germination of the spores even if the spores were heat treated. 5. The addition of 0.1mM manganese retarded the germination time, but the final germination rate was greater with 0.1mM manganese than with 0.5mM L-alanine. 6. The addition of both 0.5mM L-alanine and 0.1mM manganese stimulated the germination rate to 100 per cent. 7. The germination rate was greater with the addition of germination stimulants than without any stimulants. The germination rate was approximately 6 per cent greater with prior heat treatment than without heat treatment. 8. The optimum time for the harvest of vegetative forms of the organisms was 15 hours after cultivation in the media which contain suitable germination stimulants.

Decrease of glycogen synthase kinase 3β phosphorylation in the rat nucleus accumbens shell is necessary for amphetamineinduced conditioned locomotor activity

  • Phosphorylation levels of glycogen synthase kinase 3β (GSK3β) negatively correlated with psychomotor stimulant-induced locomotor activity. Locomotor sensitization induced by psychomotor stimulants was previously shown to selectively accompany the decrease of GSK3β phosphorylation in the nucleus accumbens (NAcc) core, suggesting that intact GSK3β activity in this region is necessary for psychomotor stimulants to produce locomotor sensitization. Similarly, GSK3β in the NAcc was also implicated in mediating the conditioned effects formed by the associations of psychomotor stimulants. However, it remains undetermined whether GSK3β plays a differential role in the two sub-regions (core and shell) of the NAcc in the expression of drug-conditioned behaviors. In the present study, we found that GSK3β phosphorylation was significantly lower in the NAcc shell obtained from rats expressing amphetamine (AMPH)-induced conditioned locomotor activity. Further, we demonstrated that these effects were normalized by treatment with lithium chloride, a GSK3β inhibitor. These results suggest that the behavior produced by AMPH itself and a conditioned behavior formed by associations with AMPH are differentially mediated by the two sub-regions of the NAcc.

PMA Activates Stat3 in the Jak/Stat Pathway and Induces SOCS5 in Rat Brain Astrocytes

  • Suppressors of cytokine signaling (SOCS) family members are negative feedback regulators of the Jak/Stat pathway, which is an essential inflammatory signaling pathway. We investigated expression of eight members of the SOCS family in rat astrocytes, using two inflammatory stimulants, PMA and IFN-${\gamma}$. Only a few SOCS genes were induced by both stimulants, and we detected an increase in SOCS5 protein with PMA. PMA activated the Jnk, Erk, p38, and Jak/Stat signal pathways. In addition, it increased the level of activated-Stat3 resulting from tyrosine phosphorylation. A gel-shift assay showed that a protein in nuclear extracts from PMA-treated cells was able to bind to Stat binding elements. These results suggest that activated Stat3 binds to SOCS promoters and leads to their transcriptional induction.

GINSENG USE AMONG TWO GROUPS IN THE UNITED STATES

  • Tow groups of subjects were studied before, during, and after long-term use of commercial Panax ginseng preparations. Group A (n = 10) consisted of users who also used other psychomotor stimulants including caffeine, while subjects in Group B (n = 8) used no drugs other than ginseng. During 12 weeks of regular ginseng use, users in Group A showed signs of central nervous system stimulation and reported feelings of well-being. Two subjects manifested possible allergic reactions while one developed a ginseng abuse syndrome marked by hypertension, nervousness, sleeplessness, skin eruptions, edema, and morning diarrhea. This syndrome was associated with excessive dosages, concomitant intake of caffeine, and possible allergic reactions. Subjects in Group B reported stimulation and feelings of well-being while showing some signs of nervousness. One subject developed a possible contact urticarial reaction to a ginseng cosmetic. All adverse reaction were transient and disappeared when ginseng use was discontinued. It is suggested that adverse reactions can be prevented by moderate use of standardized ginseng preparations while restricting use of other psychomotor stimulants.

  • The effect of acetylcholine, oxytocin and prostaglandin $F_{2{\alpha}}$ ($PGF_{2{\alpha}}$) on cyclic nucleotide levels in estrogen-primed rabbit whole uterus were studied in the presence and absence of 1-methyl-3-isobutyl xanthine (MIX), a phosphodiestrase inhibitor, and indomethacin, a prostagandin inhibitor. In the absence of MIX, acetylcholine increased guanosine 3', 5'-cyclic monophosphate (cGMP), but had no effect on adenosine 3', 5'-cyclic monophosphate (cAMP) levels. In contrast, oxytocin had no influence on cGMP, but decreased cAMP levels. $PGF_{2{\alpha}}$ increased cGMP and decreased cAMP levels. MIX increased both cAMP and cGMP levels. Oxytocin and $PGF_{2{\alpha}}$ further increased cGMP levels, indicating activation of guanylate cyclase activity. The ratio of cAMP/cGMP was decreased by uterine stinulants both in presence and absence of MIX. Indomethacin elevated cAMP and cGMP revels. The effects of uterine stimulants in the presence of indomethacin on cyclic nucleotide levels were varied from tissue to tisse. In general, oxytocin decreased cGMP and $PGF_{2{\alpha}}$ increased cAMP/cGMP levels, but the effects were statisically nonsignicficant. The cAMP/cGMP ratio was increased by uterine stimulant in the presence of indomethacin. In conclusion, uterine stimulants eased cAMP/cGMP ratio which indicates that the uterine stimulants have opposing effects on adenylate cyclase and guanylate cyclase activities. The endometrium plays a role in the regulation of cyclic nucleotide levels and uterine contraction by means of PG synthesis. Indomethacin has an unknown activities besides both of PG synthetase and phosphodiesterase inhibitions.

Effects of Adenosine and CNS Stimulants on Motor Activity in Mice (중추신경흥분제 및 Adenosine이 마우스의 자발운동에 미치는 영향)

  • The behavioral pattern of an animal is influenced by endogenous and endogenous stimuli such as humoral secretion, neurohumoral transmitters, drugs, light and environmental change. It has teen known that adenosine is a normal constituent of brain, and has sedative or hypnotic effects and anticonvulsant effects, inhibiting the spontaneous firing of cells in the brain via membrane adenosine receptors. Recent studies suggest that the excitatory responses to xanthines in the CNS might be related to the competitive antagonism of xanthines to adenosine. This study was undertaken to Investigate the effects of adenosine and the CNS stimulants such as picrotoxin, strychnine and caffeine on the spontaneous activity of mire, and to examine the influence of adenosine on the seizures induced by large doses of CNS stimulants. Subjects were $20{\sim}30\;g$ adult mice, and the spontaneous activity was measured using the Selective Activity Meter after intraperitoneal injection of adenosine (10 mg/kg), caffeine (100 mg/kg), strychnine(0.2 mg/kg) or picrotoxin(0.5 mg/kg) with or without adenosine pretreatment. The seizures were induced with caffeine(200, 250 and 300 mg/kg), strychnine(1.25 and 1.5 mg/kg) or picrotoxin(10 and 15 mg/kg). The results are summarized as follows : 1) The spontaneous activity in mite was significantly inhibited between 10 and 20 minutes after adenosine treatment. 2) Caffeine and picrotoxin increased the motor activity significantly while strychnine had no effect on the activity. 3) The ambulatory activity in the caffeine, strychnine and picrotoxin treated groups was significantly inhibited by adenosine pretreatment. 4) The seizures were observed with caffeine(200, 250 and 300 m9/kg), strychnine(1.25 and 1.5 mg/kg) or picrotoxin(10 and 15 mg/kg). The caffeine induced seizures were inhibited by adenosine pretreatment, but the strychnine or picrotoxin induced seizures were not affected.

  • Blast furnace slag (BFS) have many advantages that are related to effective value improvement on applying to concrete while side effects of blast furnace slag also appear. Thus, research team conducted an experiment with high volume slag to see if the attribute of waste alkali accelerator for mixing rate, mixed use of NaOH and $Na_2SiO_3$, and early strength agent for mixing rate for replacement ratio and for the types of the stimulants in order to increase the use of blast furnace slag1s powder. As the result of the experiment, when it comes to compression strength, all of the alkali stimulants have been improved as the replacement rate increases except for sodium hydroxide. Among the alkali stimulants, sodium silicate was high on dynamic elastic modulus and absorption factor. In case of early strength agent, the mix of mixing 1.5% and blast furnace slag 75% have showed high strength enhancement. In event of Waste Alkali accelerator, it has showed different consequences for each experiment.

Influence of Panax Ginseng on the Hypothermia in Rats Elicited by Various Drugs (인삼(人蔘)의 각종약물투여(各種藥物投與)로 인(因)한 체온하강(體溫下降)에 미치는 영향(影響))

  • Attempts have been made upon the temperature response of the rat induced by some central nervous system depressants as well as stimulants, so as to secure some hidden facets of Panax Ginseng acting upon central nervous system. Although considerable works have been done with regard to Panax Ginseng, it is quite apparent that neither definite implication in terms of its effective chemical constituents is with us nor its pharmacological activity thus far. The author could, however, arrive at some results through procedures preceded by intraperitoneal administration of various drugs in combination with Panax Ginseng in albino rats, that is: (1) Nembutal and chlorpromazine displayed a highly inhibitory effect upon temperature response in the presence of Panax Ginseng, while meprobamate, reserpine, phenacetin and aspirin exerted potentiation actions upon hypothermia. Phenobarbital, serotonin and histamine, on the contrary, did not appear to produce any effect of significance. (2) Nembutal with Panax Ginseng caused prolongation of hypnosis in rat, whereas sodium phenobarbital did not have any effect on it. (3) $LD_{-50}$ in each experimental group of administration of central nervous system stimulants such as strychnine, picrotoxin with Panax Ginseng, necessitated marked increase in the lethal doses. The observations from this study seemed to imply that the complicated mechanism of action of Panax Ginseng might be referred to both central nervous depressive action and influence to basal metabolic rate of mammalian.

Simultaneous Determination of Amphetamine-Type Stimulants (ATS) in Human Hair by GC-MS (가스크로마토그래피-질량분석기를 이용한 모발에서 암페타민형 각성제 약물의 동시 분석)

  • Gas chromatography-mass spectrometric (GC-MS) procedure is presented for the simultaneous qualification and quantitation of methamphetamine (MA), amphetamine (AMP), 3,4-methylenedioxymethamphetamine (MDMA), 3,4-methylenedioxyamphetamine (MDA), and 3,4-methylenedioxyethylamphetamine (MDEA) in human hair. The method procedure involves decontamination of hair with distilled water and acetone, acidic hydrolysis, extraction in the presence of deuterated internal standards, and GC-MS analysis after derivatization with trifluoroacetic anhydride (TFAA) in ethylacetate. The limit of detection for 5 drugs were about 0.1∼0.15 ng/mg using 30 mg hair sample. Coefficient variations of correlation ranged from 0.9941 to 0.9993. The recoveries of these drugs were found to be 93.4∼104.4%. The concentrations of AMP, MA, MDA, and MDMA in abusers' hair samples were measured 0.17∼2.88, 2.09∼18.34, 0.24∼3.83, and 3.10∼22.81 ng/mg, respectively. The ratios of MA/AMP and MDMA/MDA ranged 5.67∼49.57 and 4.78∼54.31, respectively. This assay has been successfully utilized in the evaluation of the deposition of amphetamine-type stimulants (ATS) in human hair.

  • Methylphenidate is a widely used stimulant for treatment of attention-deficit hyperactivity disorder (ADHD). In addition to core symptoms of attention and behavioral symptoms, methylphenidate is even effective for executive functions. However, abuse and misuse of stimulants, including methylphenidate, for the purpose of cognitive enhancement is an issue of concern worldwide. Some prejudices and misunderstandings against methylphenidate are popular ; however, little attention has been given to the neuropsychiatric evidence of methylphenidate for enhancement of cognitive function among healthy populations. In this article, our aim was to conduct a review of previous studies investigating the effect of methylphenidate in healthy populations. Findings from several recent studies have demonstrated the effectiveness of methylphenidate for enhancement of cognitive function in healthy populations. The mechanisms of cognitive enhancement are thought to be associated with motivation and the reward circuit in the brain. However, when considering the risk to benefit, an official discussion of the use of methylphenidate among healthy members of the population would be premature. Instead, investigation of epidemiological studies of the prevalence of misuse of stimulants among healthy members of the population, particularly adolescents and college students, is needed. In addition, based on achievements reported in previous studies, investigation of the effect of an approach using non-pharmacological enhancing motivation, which will in turn result in increased cognitive function would be helpful.
