Species A rotaviruses (RVAs) replicate and assemble their immature particles within electron dense compartments known as viroplasms, where lipid droplets (LDs) interact with the viroplasm and facilitate viral replication. Despite the importance of LD formation in the life cycle of RVAs, the upstream molecules modulating LD formation remain unclear. This study aimed to find out the role of sterol regulatory element-binding proteins (SREBPs) in reprogramming of LD formation after RVA infection. Here, we demonstrate that RVA infection reprograms the sterol regulatory element-binding proteins (SREBPs)-dependent lipogenic pathways in virus-infected cells, and that both SREBP-1 and -2 transactivated genes, which are involved in fatty acid and cholesterol biosynthesis, are essential for LD formation. Our results showed that pharmacological inhibition of SREBPs using AM580 and betulin and inhibition of their downstream cholesterol biosynthesis (simvastatin for HMG-CoA reductase) and fatty acid enzymes (TOFA) negatively modulated the intracellular triacylglycerides and cholesterol levels and their resulting LD and viroplasm formations. Interestingly, pharmacological inhibition of SREBPs significantly reduced RVA protein synthesis, genome replication and progeny production. This study identified SREBPs-mediated lipogenic reprogramming in RVA-infected host cells, which facilitates virus replication through LD formation and its interaction with viroplasms, suggesting that SREBPs can be a potential target for the development of efficient and affordable therapeutics against RVA infection.
Objective: This study identifies single-nucleotide polymorphisms (SNP) or gene combinations that affect the flavor and quality of Korean cattle (Hanwoo) by using the SNP Harvester method. Methods: Four economic traits (oleic acid [C18:1], saturated fatty acids), monounsaturated fatty acids, and marbling score) were adjusted for environmental factors in order to focus solely on genetic effects. The SNP Harvester method was used to investigate gene combinations (two-way gene interactions) associated with these economic traits. Further, a multifactor dimensionality reduction method was used to identify superior genotypes in gene combinations. Results: Table 3 to 4 show the analysis results for differences between superior genotypes and others for selected major gene combinations using the multifactor dimensionality reduction method. Environmental factors were adjusted for in order to evaluate only the genetic effect. Table 5 shows the adjustment effect by comparing the accuracy before and after correction in two-way gene interactions. Conclusion: The g.3977-325 T>C and (g.2988 A>G, g.3977-325 T>C) combinations of fatty acid-binding protein4 were the superior gene, and the superior genotype combinations across all economic traits were the CC genotype at g.3977-325 T>C and the AACC, GACC, GGCC genotypes of (g.2988 A>G, g.3977-325 T>C).
Two SNPs, i.e. L127V and T172M, of bovine growth hormone (GH) causing the presence of GH gene haplotypes A, B, and C was previously shown to alter intramuscular fatty acid (FA) composition in Japanese Black (JB) heifers. To determine the SNP effect on somatotropic hormone concentration and lipogenesis, we measured plasma GH, insulin, and insulin-like growth factor-1 (IGF-1) concentrations. We also measured mRNA levels of fatty acid synthase (FASN), stearoyl-coA desaturase (SCD), and sterol regulatory element binding proteins-1 (SREBP-1) and FA composition in diaphragm tissues. Heifers with genotype CC had the lowest plasma insulin concentration and FASN and SCD mRNA levels among genotypes. FASN mRNA levels in haplotype A tended to positively correlate with saturated FA (SFA) content and negatively correlated with C18:2 and unsaturated FA (USFA) contents. SCD mRNA levels in haplotype A positively correlated with monounsaturated FA (MUFA) contents and negatively correlated with C18:0 content. They also tended to positively correlate with C16:1, C18:1, and USFA contents and USFA/SFA ratio and negatively correlate with SFA content. Taken together, GH gene polymorphism affects the lipogenic genes expression levels and their relationships with fatty acid compositions in diaphragm tissues of JB heifers at 31 months of age.
폴리코사놀의 콜레스테롤 감소 효과에 대한 기전은 아직 명확하게 규명되지 못하고 있다. 최근 몇몇 연구들은 지방산 합성 경로를 통한 콜레스테롤 합성의 조절에 sterol regulatory element-binding proteins(SREBP-1c)의 역할을 제시하였다. 그러나 현재까지 SREBP-1c에 의해 조절되는 지방산 합성에서 폴리코사놀의 효과에 대한 연구는 전무하다. 그러므로 본 연구의 목표는 SREBP-1c에 의해 매개되는 지방산 합성이 폴리코사놀의 콜레스테롤 감소 효과와 관계하는지를 조사하는 것이다. 7주령의 C57BL/6 수컷 생쥐를 7개군(군당 7마리)으로 나누고 8주간 다음과 같이 처리하였다; 1) 정상식이군(정상 대조군), 2) 고지방식이군(high-fat diet, HFD, 음성대조군), 3) 고지방식이+에탄올처리군(Pol-0), 4) 고지방식이+1 mg/kg 폴리코사놀처리군(Pol-1), 5) 고지방식이+2 mg/kg 폴리코사놀처리군(Pol-2), 6) 고지방식이+4 mg/kg 폴리코사놀처리군(Pol-4),7) 고지방식이+simvastatin 50 ㎍/kg 처리군(양성 대조군). 폴리코사놀과 simvastatin은 고지방식이를 유지하는 동안 매일 동일 시간에 처리하였으며 체중과 음식섭취량은 8주 동안 매주 측정하였다. 8주 후, 혈중 콜레스테롤 수치를 측정하였으며, 간의 조직학적 분석과 SREBP-1c와 지방산의 발현을 조사하였다. 폴리코사놀은 농도-의존적으로 체중과 음식섭취량을 감소시켰다. 혈중 콜레스테롤 수치는 Pol-1과 Pol-4군에서 유의하게 감소하였으며, SREBP-1c와 FAS의 발현 역시 Pol-4 군에서 유의하게 감소하였다. 이러한 결과들은 폴리코사놀의 콜레스테롤 감소 효과가 SREBP-1c와 FAS의 발현 억제에 기인하여 일어날 수 있음을 시사한다.
적무(Rahphanus sativus L.) 새싹은 십자화과 식물이다. 본 연구에서는 적무새싹 물 추출물의 ${\alpha}$-amylase, ${\alpha}$-glucosidase, 췌장 리파아제 효소에 대한 활성 억제능과 3T3-L1 지방 전구세포를 이용하여 지방합성 억제 효능을 평가하였다. 적무새싹 추출물을 처리한 결과 ${\alpha}$-amylase, ${\alpha}$-glucosidase, 췌장 리파아제 효소 활성을 농도 의존적으로 억제하는 것을 확인하였다. 더욱이 적무새싹 추출물은 3T3-L1 지방 전구세포의 지방세포 분화, 지방 및 중성지방 축적을 억제하였으며 세포독성은 나타나지 않았다. 적무새싹 추출물은 peroxisome proliferator-activated receptor (PPAR)${\gamma}$, sterol regulatory element-binding protein 1 (SREBP-1) and CCAT/enhancer binding protein (C/EBP)${\alpha}$와 같은 지방합성 전사 인자의 발현 조절을 통하여 지방합성을 억제하였다. 또한, 적무새싹 추추물은 지방합성과 수송 저장에 관여하는 단백질인 adiponectin, fatty acid synthesis (FAS), perillipin, and fatty acid bind protein-4(FABP4)의 발현을 억제하였다. 이 연구는 적무새싹이 지방합성 전사인자는 물론 지방합성 단백질 발현의 제어를 통해 비만을 억제할 수 있는 가능성을 보여주었다.
Hypertriglyceridemia is one of the metabolic syndrome that is often observed as a result of lipid abnormalities. It is associated with other lipids, metabolic disorders, cardiovascular disease and liver disease. Korean red ginseng is known to affect obesity, dyslipidemia, liver disease and liver function, but the mechanism of its effect is not clear. This study examined the beneficial effects of hypertriglyceridemia and the mechanism of action of Jinan red ginseng extract (JRG) in hepatoma cells. To measure the levels of triglyceride accumulation, we studied the expression of proteins and mRNAs related to lipidogenesis in hepatoma cells (Huh7 and HepG2). JRG decreases the lipidogenic markers, peroxisome proliferator-activated receptor γ (PPARγ), CCAAT-enhancer-binding proteins α (C/EBPα) and C/EBPβ which are major regulators of triglyceride synthesis in hepatoma cells. We also found that JRG reduced sterol regulatory element binding proteins 1c (SREBP-1c), C/EBPα and C/EBPβ by regulating liver X receptor (LXR) and stearoyl CoA desaturase (SCD) expressions. In addition, the first-limited step of synthesis triglyceride (TG), glycerol-3-phosphate (G3P) is decreased by JRG. These results suggest that the anti-hypertriglyceride effect of JRG in hepatoma cells could be accompanied with the inhibition of lipidogenic transcription factors by regulating LXR and SCD expression.
간 소포체(ER) 스트레스는 비알콜성지방간과 인슐린 저항성의 발달에 기여하고, unfolded protein response(UPR)의 구성요소는 지질 대사를 조절한다. 최근 연구에 따르면 ER 스트레스와 비정상적인 세포 지질 대사 사이의 연관성이 보고되었으며, 이 과정에서 지질 대사의 중심 조절자인 sterol regulatory element binding proteins(SREBPs)의 관련성이 확인되었다. 그러나 ER 스트레스 동안 지질 대사를 조절하는 SREBP의 정확한 역할과 비알콜성지방간에 대한 기여는 아직 밝혀지지 않았다. 본 연구에서 SREBP-1c 결핍은 UPR, 염증 및 지방산 산화 조절을 통해 ER 스트레스에 의해 유도된 비알콜성지방간으로부터 생쥐를 보호한다는 것을 보여준다. SREBP-1c는 inositol requiring kinase 1α (IRE1α) 발현을 직접적으로 조절하고 ER 스트레스에 의해 유도된 tumor necrosis factor-α의 활성화를 매개하여 peroxisome proliferator-activated receptor γ coactivator 1-α (PGC1α)의 감소와 그에 따른 지방산 산화의 장애를 유발한다. 그러나, SREBP-1c의 유전적 결핍은 이러한 현상을 보호하여 간 염증과 지방 축적을 완화시킨다. SREBP-1c 결핍이 ER 스트레스에 의해 유도된 염증 신호를 방지하는 메커니즘은 아직 밝혀지지 않았지만, SREBP-1c가 결핍된 Kupffer 세포에서 IRE1α 신호의 변화가 염증 신호에 관여할 수 있을 것으로 생각된다. 본 연구결과는 SREBP-1c가 ER 스트레스에 의해 유도된 비알콜성지방간에서 UPR 및 염증의 조절에 중요한 역할을 함을 시사한다.

BACKGROUND/OBJECTIVES: Obesity is a major cause of metabolic disorders; to prevent obesity, research is ongoing to develop natural and safe ingredients with few adverse effects. In this study, we determined the anti-obesity effects of Rosa multiflora root extract (KWFD-H01) in 3T3-L1 adipocytes and Sprague-Dawley (SD) rats. MATERIALS/METHODS: The anti-obesity effects of KWFD-H01in 3T3-L1 adipocytes and SD rats were examined using various assays, including Oil Red O staining, gene expression analyses, protein expression analyses, and blood biochemical analyses. RESULTS: KWFD-H01 reduced intracellular lipid accumulation and inhibited the mRNA expression of peroxisome proliferator-activated receptor γ (PPARγ), cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding proteins (C/EBPα), sterol regulatory element-binding transcription factor 1 (SREBP-1c), acetyl-CoA carboxylase (ACC), and fatty acid synthase (FAS) in 3T3-L1 cells. KWFD-H01 also reduced body weight, weight gain, and the levels of triglycerides, total and LDL-cholesterol, glucose, and leptin, while increasing high-density lipoprotein-cholesterol and adiponectin in SD rats. PPARγ, C/EBPα, SREBP-1c, ACC, and FAS protein expression was inhibited in the epididymal fat of SD rats. CONCLUSION: Overall, these results confirm the anti-obesity effects of KWFD-H01 in 3T3-L1 adipocytes and SD rats, indicating their potential as baseline data for developing functional health foods or pharmaceuticals to control obesity.
Fumigaclavine C (FC), an active indole alkaloid, is obtained from endophytic Aspergillus terreus (strain No. FC118) by the root of Rhizophora stylosa (Rhizophoraceae). This study is designed to evaluate whether FC has anti-adipogenic effects in 3T3-L1 adipocytes and whether it ameliorates lipid accumulation in high-fat diet (HFD)-induced obese mice. FC notably increased the levels of glycerol in the culture supernatants and markedly reduced lipid accumulation in 3T3-L1 adipocytes. FC differentially inhibited the expressions of adipogenesis-related genes, including the peroxisome proliferator-activated receptor proteins, CCAAT/enhancer-binding proteins, and sterol regulatory element-binding proteins. FC markedly reduced the expressions of lipid synthesis-related genes, such as the fatty acid binding protein, lipoprotein lipase, and fatty acid synthase. Furthermore, FC significantly increased the expressions of lipolysis-related genes, such as the hormone-sensitive lipase, Aquaporin-7, and adipose triglyceride lipase. In HFD-induced obese mice, intraperitoneal injections of FC decreased both the body weight and visceral adipose tissue weight. FC administration significantly reduced lipid accumulation. Moreover, FC could dose-dependently and differentially regulate the expressions of lipid metabolism-related transcription factors. All these data indicated that FC exhibited anti-obesity effects through modulating adipogenesis and lipolysis.
본 연구는 감귤 가공공정 중에 발생하는 가공부산물인 감귤피의 산업적 이용성 및 유용성을 증대시키고자 효소적 추출법을 적용하여 추출물을 제조하고 항비만 활성을 측정하였다. 감귤피 효소적 추출물(MPCE)의 폴리페놀 함량은 이전의 연구에서 보고한 유기용매 추출물보다 높은 함량을 나타내었다. MPCE의 항비만 활성을 알아보기 위해 지방세포 분화억제에 미치는 영향을 확인한 결과 MPCE는 농도 의존적으로 지방세포의 분화 및 지방축적을 감소시켰으며 이는 세포독성에 의한 것이 아님을 세포생존율 측정을 통해 확인하였다. 또한 MPCE는 adipogenesis 관련 전사인자인 C/EBPα, SREBP-1, PPARγ, FABP4의 단백질 발현을 감소시켰다. 이러한 결과들을 통해 MPCE는 adipogenesis 관련 전사인자의 단백질 발현 억제하여 지방세포의 분화 및 지방축적을 억제하였으며, 이를 통해 항비만 효과를 나타낸다는 것을 확인하였다. 따라서 MPCE는 비만 예방 효능을 가진 기능성 식품 소재로서의 활용 가치가 있는 것으로 판단되며, 보다 명확한 항비만 효능을 확인하기 위해 추후 동물 실험 등에 대한 연구가 필요할 것으로 생각된다.
