Effect of Gaseous Chlorine Dioxide on Sterilization in Industrial Food-holding Cabinets (이산화염소가스를 이용한 식품산업용 소독장에서의 살균효과)

  • The aim of this study was to investigate the effect of different concentrations of chlorine dioxide ($ClO_2$) on sterilization and deodorization of food-holding cabinets under different exposure times. For the measuring sterilization and deodorization, a 6.5 L chamber and a 625 L cabinet with circulation systems were used. Two bacteria (Staphylococcus aureus KCTC1916 and Escherichia coli KCTC 1682) that were artificially inoculated in the plate respectively were put into the 6.5 L chamber and the 625 L cabinet. The $ClO_2$ gas was produced by ampules. In the 6.5 L chamber, neither of the two bacteria was detected after 24 hours treatment by $ClO_2$ gas. Moreover, the deodorization rate against ammonia and phenol was 94% and 70%, respectively, but deodorization against formaldehyde was not effective. When the concentration reached maximum (6 ampule, 4.6 ppm) levels in the cabinet, it lasted for approximately 2 h and then decreased slowly. When a circulator was used, the gas concentration was very low (6 ampule, 0.8 ppm) and the antibacterial activity against S. aureus and E. coli was low. The level of reduction against S. aureus and E. coli was 2.98 log CFU/plate and 6.06 log CFU/plate, respectively, in the cabinet after 24 h without a circulator. The reduction against S. aureus KCTC1916 and E. coli KCTC1682 was 2.69 log CFU/plate and 4.41 log CFU/plate for 24 h, respectively.

Effects of Gamma-Irradiated Korean Ginseng on Fertility and General Reproductive Toxicity in Rats (방사선 조사 인삼이 랫드의 수태능 및 일반 생식독성에 미치는 영향에 관한 연구)

  • Korean ginseng products have been fumigated with ethylene oxide (EO) for sterilization and prolongation of storage periods. However, there had been controversies indicating that the consumption of food treated with EO might cause harmful effects in human. Since, in Korea the use of EO gas for food treatment was banned in 1991. Since then, irradiation technique has been developed as an alternative. This study was carried out to investigate the effects of irradiated ginseng on fertility, and reproductive and developmental toxicity. Either EO gas fumigated or gamma-irradiated ginseng was administered to male rats by oral gavage for 63 days during the premating period. Female rats were administered from 14 days before mating to day 20 of gestation or to day 21 of lactation. The exposure amount of irradiation used was 5, 10 and 30 kGy, respectively. There were no treatment related changes of darns in clinical signs, and parturition. No treatment related changes in food consumption, body/organ weights, male/female reproductive and fertility performances were observed. F1 fetuses showed no external abnormality. Reflex/sensory junctions, physical/behavioral development, and reproductive performance of F1 rats were not adversary affected. The results of this study show that gamma-irradiated ginseng, up to 30 kGy, has no adverse effects on the fertility, reproduction and development in Wistar rats.

  • Bioterrorism intends to cause mass casualties and social panic by means of malicious pathogenic microorganisms. Environmental decontamination becomes very important as a follow-up measure if that happens. Conventional methods for decontamination is that aqueous disinfectants are being sprayed for killing or not spreading microorganisms with the purpose of preventing infection. However, these procedures are not enough to perfectly sterilize space or surface inside of building, requiring additional measures such as surface disinfection or gas treatment methods. This article deals with the issues about the present decontamination procedures, global trends, in order to formulate suggestion for advanced environmental decontamination.

Embryo and Fetal Developmental Toxicity Study on Gamma-Irradiated Korean Ginseng in Rats (방사선 조사 인삼이 랫드의 기형유발에 미치는 영향에 관한 연구)

However, there had been controversies indicating that consumption of EO treated foods might cause harmful effects in human. In Korea, the use EO gas for food treatment was banned in 1991. Since then, irradiation technique has been developed as an alternative. This study was carried out to evaluate the safety of irradiated ginseng on embryo and fetal developmental toxicity effects in rats. Either EO gas fumigated or $\gamma$-irradiated ginseng was administered to pregnant Wistar rats by oral gavage from gestational day 7 to 17. The amount of irradiation used in this study was 5, 10 and 30 kGy, respectively. There were no treatment related changes of dams in deaths, clinical signs, food consumption and body/organ weight. No treatment related changes in implantation ratio, litter size, sex ratio and body/organ weight of fetuses were observed. Also, no F1fetuses with external, visceral, head and skeletal mal-formation were observed. The results of this study showed that $\gamma$-irradiated ginseng, up to 30 kGy, has no adverse effects on fetal development of rats.

  • For the ultimate manufacture of vinegar with watermelon juice, we examined the preliminarily alcoholic fermentation of watermelon juice using Saccharomyces cerevisiae, Saccharomyces cerevisiae var. ellipsoideus and Kluyveromyces fragilis. The juice contained 3.2% total reducing sugar by the Somogyi-Nelson method. Strains of yeast culture, pH, temperature and concentration of sugar in the juice were important factors affecting alcoholic fermentation. Saccharomyces cerevisiae var. ellipsoideus produced 5.3% alcohol from the juice fortified with 12% glucose under the conditions of pH 5.73 and $27^{\circ}C$ for 8 days. Generally, small quantities of various salts and N-sources affected little the alcoholic fermentation of the juice. The sterilization of the juice by autoclaving improved efficiency of fermentation than that by $SO_2$ sterilization. Determination of alcohol was carried out by the gas chromatographic method using Chromosorb W: Carbowax as packing material.

  • The microwave of 2450 MHz, generated by a household cooking oven, was evaluated for its applicability to melt various rehydrated media and to remove dissolved oxygen from tubed media for anaerobic culture. The effect on the sterilization of E. coli in selective media was also evaluated. The following results were obtained. 10 The microwave oven was useful in saving time for melting media and in eliminating heat and combustion gas from the laboratory, which were inevitable by-products in the conventional flame method. 2) Dissolved oxygen could be removed without boiling over by exposing the tubes of anaerobic culture medium after putting them in a wire basket in a beaker with water. 30 The count of E. coli during the melting of MacConkey and EMB agar were similar to those treated with open flame. The microwave treatment was not considered a possible mean to replace autoclaving even in these selective media.

Sterilizing Effect of Plant Pathogenic Fungi using Ozone Microbubble (오존마이크로버블을 이용한 식물병원균 살균효과)

  • Sterilization is an important factor in reprocessing of drained nutrient solution (RDN). To evaluate the suitability of ozone microbubble for RDN in protected horticulture and plant factory, strong fungicidal activity of ozone and microbubble were applied. This experiment was taken advantage of ozonated water (OW) and ozone gas (OG). The Fusarium oxysporum (FO), Phytophthora capsici (PC) and Colletotrichum gloeosporioides (CG) were treated with OW 0.5, 1.0 and 2.0 ppm and OG $3.0g{\cdot}h^{-1}$ for 0, 30, 60, 120 and 180 sec. Results of this experiment can be summed up as follows : In the OW, FO was sterilized by 0.5 ppm in 60 sec and PC was sterilized by 2.0 ppm in 30 sec. In the OG, FO and PC both of them were sterilized in 180 sec. However, CG was not sterilized using OW and OG. Overall, ozone microbubble showed possibility of sterilization in RDN. However, CG is required to more ozone concentration and processing time.

  • The effects of filling temperatures of broth and degassing method on the residual oxygen content and gas composition in the pouch and physical strength of packaging material for Samgyetang depending on the contamination of broth on the sealing layer and sterilization process were investigated. The residual oxygen content in the broth and the oxygen proportion in the headspace of package were decreased with the increase of broth temperature at filling into the pouch from 50 to 100. When the products were packaged as air-contained (Air), manually squeezed the upper side of package out to minimize the headspace (Degas) and flushed with nitrogen gas ($N_2$-Flushing) while maintaining the broth temperatures of Samgyetang at 50 or 85. The residual oxygen content and oxygen proportion were increased in the order of $N_2$-Flushing

Preparation and Characterization of ClO2 Self-Releasing Smart Sachet (이산화염소 자체 방출 스마트 샤쉐의 제조 및 특성 연구)

  • Chlorine dioxide (ClO2) is widely used for post-harvest sterilization treatment. However, there are limitations in the retail application of ClO2 due to difficulties in handling, expensive facilities, and safety concerns. Therefore, it is necessary to develop a ClO2 technology that can be easily applied and continuously released for a long period. In this study, a series of ClO2 self-releasing sachets were developed. First, poly(ether-block-amide) (PEBAX) and polyethylene-glycol (PEG) composite films containing different ratios of citric acid (CA) were prepared using the solution casting method. The as-prepared PEBAX/PEG-CA composite films were evaluated using FT-IR, DSC, and TGA to confirm chemical structure and thermal properties. Subsequently, PEBAX/PEG-CA composite films were designed in the form of a sachet and NaClO2 powder was transferred into the sachet to achieve a ClO2 self-releasing system. The ClO2-releasing behavior of the sachet was investigated by measuring the release amount of the gas using UV-vis. The release amount of ClO2 increased with increasing CA contents owing to the existence of higher protons (trigger) in the polymer matrix. Further, ClO2 gas was released for a longer time. Therefore, the as-prepared smart sachet can be tuned according to applications and packaging sizes to serve an optimal sterilization effect.

Microscopic Study of Decomposition-Inhibition in Stabilized $ClO_2$ Gas in Kidney of Rat with Passage of Time (시간경과에 따른 안정화 이산화염소(Stabilized $ClO_2$)의 콩팥조직 부패억제에 대한 현미경적 연구)

  • The stabilized $ClO_2$ gas has been used for many years by the food industry as a strong oxidizing and sanitizing agent that has broad and high biocidal effectiveness. Therefore, "stabilized $ClO_2$" gas may be used in fields of disinfectant and sterilization. But, there have been few studies on the decomposition-inhibition effect of stabilized $ClO_2$ gas with passage of time. The main purpose of this study was to examine the decomposition-inhibition effect of stabilized $ClO_2$ gas and the morphological change of kidney by measuring of the light and electron microscope. Sprague-Dawley (SD) rats weighting from 230 gm to 250 gm were used as experimental animals. Under ether anesthesia, the right kidney of rat was obtained. Put each sample in $37^{\circ}C$ and humidity $80{\pm}5%$ incubator, we obtained each sample after 0 day, 1 day, 2 days, 3 days, 4 days and 5 days. We proceeded the observation of light and electron microscope. The results obtained in this study reveal that stabilized $ClO_2$ gas is an effective decomposition inhibitor until 2 days that was conducted at $37^{\circ}C$ and humidity $80{\pm}5%$ conditions.
