한약재복합 추출물의 인간피부섬유아세포 HS68에 대한 항노화 효과 (Antiaging Activity of Mixed Extracts from Korean Medicinal Herbs on HS68 Skin Fibroblast)

  • Objectives : The purpose of this study was to investigate antiaging and antioxidant effects on cultured human skin fibroblast with 80% ethanol extracts of plants including of stem of Dendropanax morbifera, Corni fructus and Lycii Fructus. Methods : An ethanol extract of three medicinal plants including stem of Dendropanax morbifera, Corni fructus and Lycii Fructus. Extracts were assessed to determine the mechanism of antioxidant and antiaging activities. Antioxidant activity of extract was evaluated by two different assays as 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and super oxide dismutase (SOD) like activities. These extracts were tested for cell viability on HS68 skin fibroblast by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay. We investigated the effects of Ultraviolet-B irradiation on cytotoxicity, type 1 collagen, elastin level and oxidative damage in cultured human skin fibroblast (HS68). Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. Results : The extracts obtained dose-dependently increased the scavenging activity on DPPH radical scavenging activity and SOD like activity. The extracts of complex herbal medicine showed low cytotoxicity as more than 100% cell viability in 100ppm/ml concentration. HS68 fibroblasts were survived 70% at 120 $mJ/cm^2$ UVB irradiation and treated tumor necrosis factor (TNF)-alpha. The levels of aging factors and cytotoxicity were decreased by ethanol extract of complex herbal medicine. Conclusions : These results suggest that ethanol extracts of complex medicinal plants of including of stem of Dendropanax morbifera, Corni fructus and Lycii Fructus may have value as the potential antioxidant and antiaging medicinal plant.

Recombinant Human Bone Morphogenetic Protein-2 Priming of Mesenchymal Stem Cells Ameliorate Acute Lung Injury by Inducing Regulatory T Cells

  • Mesenchymal stromal/stem cells (MSCs) possess immunoregulatory properties and their regulatory functions represent a potential therapy for acute lung injury (ALI). However, uncertainties remain with respect to defining MSCs-derived immunomodulatory pathways. Therefore, this study aimed to investigate the mechanism underlying the enhanced effect of human recombinant bone morphogenic protein-2 (rhBMP-2) primed ES-MSCs (MSCBMP2) in promoting Tregs in ALI mice. MSC were preconditioned with 100 ng/ml rhBMP-2 for 24 h, and then administrated to mice by intravenous injection after intratracheal injection of 1 mg/kg LPS. Treating MSCs with rhBMP-2 significantly increased cellular proliferation and migration, and cytokines array reveled that cytokines release by MSCBMP2 were associated with migration and growth. MSCBMP2 ameliorated LPS induced lung injury and reduced myeloperoxidase activity and permeability in mice exposed to LPS. Levels of inducible nitric oxide synthase were decreased while levels of total glutathione and superoxide dismutase activity were further increased via inhibition of phosphorylated STAT1 in ALI mice treated with MSCBMP2. MSCBMP2 treatment increased the protein level of IDO1, indicating an increase in Treg cells, and Foxp3+CD25+ Treg of CD4+ cells were further increased in ALI mice treated with MSCBMP2. In co-culture assays with MSCs and RAW264.7 cells, the protein level of IDO1 was further induced in MSCBMP2. Additionally, cytokine release of IL-10 was enhanced while both IL-6 and TNF-α were further inhibited. In conclusion, these findings suggest that MSCBMP2 has therapeutic potential to reduce massive inflammation of respiratory diseases by promoting Treg cells.

감마선 조사에 따른 흰쥐의 혈장 및 좌골신경의 혈소판-유래성 성장인자(PDGF)와 PDGF ${\alpha}$ - 및 ${\beta}$ - 수용체의 시간 의존성 변화 (Time Dependent Changes in Platelet-Derived Growth Factor(PDGF) and PDGF ${\alpha}$ - and ${\beta}$ - Receptors Following Gamma-Irradiation of Rat Plasma and Sciatic Nerve)

  • 이 실험의 목적은 4.5 및 7.5 Gy의 단일 선량으로 감마선의 전신조사에 따른 혈장 및 좌골신경 혈소판-유래성 성장인자(platelet-derived growth factor, PDGF)의 농도와 좌골신경 PDGF ${\alpha}$ -및 ${\beta}$ -수용체 밀도에 대한 시간의존성 변화를 조사 추구하는데 있다. 웅성 흰쥐(Sprague-Dawley계, 체중 $270{\pm}15.4g$)를 사용하여 $^{60}Co$ 감마선을 전신 조사(선량률 1.065 Gy/min)하고, 조사 후 1, 2, 3, 5 및 10일구에서 피폭에 따른 급성반응을 조사 추구하였다. 실험의 결과는 다음과 같다. 1) 4.5 및 7.5 Gy 선량군에서는 혈장 PDGF의 농도가 전 일구에 걸쳐 대조군($2.26{\pm}0.31\;ng/ml$)에 비해서 감소경향이 나타났으며, 한편 4.5 및 7.5 Gy 선량군 사이에서는 전 일구에 걸쳐 혈장 PDGF의 농도차에 유의적인 차이가 없었다(P>0.05). 피폭된 흰쥐의 혈장 PDGF의 농도는 4.5 및 7.5Gy 선량군에서 모두 대조군의 흰쥐에 비하여 5일구($4.5Gy:1.35{\pm}0.23\;ng/ml$; 대조군의 백분율로서 59.7%, 그리고 $7.5\;Gy:1.24{\pm}0.13\;ng/ml;54.9%$) 및 10일구($4.5Gy:1.27{\pm}0.28ng/ml;56.2%$ 그리고 $7.5Gy:1.07{\pm}0.19\;ng/ml:47.3%$)에서 각각 유의성으로 낮았다(P<0.05). 2) 4.5 및 7.5Gy 선량군에서는 좌골신경 PDGF의 농도가 전 일구간에 걸쳐 대조군($3.68{\pm}0.12\;ng/ml$)에 대하여 대체로 약간의 증가가 나타났으나, 한편 4.5 및 7.5Gy 선량군 사이에서 전 일구에 걸쳐 좌골신경 PDGF의 농도차는 무의성이었다(P>0.05). 피폭된 흰쥐의 좌골신경 PDGF의 농도는 4.5 및 7.5Gy 선량군에서 모두 대조군에 비하여 1일구($4.5Gy:4.33{\pm}0.03\;ng/m1$;대조군의 백분율로서 118% 그리고 $7.5Gy:4.81{\pm}0.36\;ng/ml$; 130%)에서 각각 증가하였으나 무의성이었다(P>0.05). 3) 방사선의 조사는 혈장 및 좌골신경 PDGF의 농도의 변화를 초래할 뿐만아니라 좌골신경 PDGF ${\alpha}$ - 및 ${\beta}$ -수용체 밀도의 변화도 초래하였다. 수용체의 밀도단위를 단백질의 상대적 함량(relative level of protein)으로 표시하고 일구-필적의 대조군을 백분율로 표시하였다. 4.5 및 7.5Gy 선량군에서의 좌골신경 PDGF ${\alpha}$ -수용체의 밀도는 대조군에 비하여 초기에 감소현상으로 나타나, 1일구에서는 40%(4.5Gy)와 50%(7.5Gy)를, 그리고 2일구에서는 33%(4.5Gy)와 50%(7.5Gy)로 가장 낮은 값을 나타낸 후 점차 대조군에 근접하는 값으로 증가하는 경향이었으며, 한편 4.5 및 7.5Gy 선량군에서 좌골신경 PDGF ${\beta}$ -수용체의 밀도는 역시 대조군에 비하여 각각 26%(4.5 Gy)와 27%(7.5Gy)의 최소 값을 나타낸 후점차 대조군에 근접하는 값으로 증가하는 경향으로 나타났다. 이러한 실험의 결과를 미루어 보아서, 혈장 및 좌골신경의 PDGF의 농도와 좌골신경 PDGF ${\alpha}$ - 및 ${\beta}$ -수용체의 밀도에 대한 방사선-유발성 변경이 골수 간세포 및 말초뉴런의 장해의 병인론에 관련되어 있는 것으로 생각된다.

  • Forty herbal drugs which are described to have potential antitumor activity were solvent-fractionated with petroleum ether, ether and ethyl acetate in sequence. The cytotoxic activity was mostly shown in the ether fraction(40.54%) and petroleum ether fraction (35.15%), but scarcely in the water phase (10.8%), meaning that most of the active components had less polar property. Twenty-seven percent of the drugs tested were active, which is higher value than 10.4% of the random sampled drugs The drugs possessing the $ED_{50}$ values less than $10{mu}g/ml$ were the roots of Lithospermum erythrorhizon, Curcuma domestica, Salvia miltiorrhiza, Astragalus membraneceus and Scutellaria indica, the leaves of Panax ginseng, S. indica and Liriodendron tulipifera, the barks of Picrasma ailanthoides and Rhus vernifera, the herbs of Agrimonia pilosa and Siegesbeckia pubescens the seeds of Tricosanthes kirilowii, P. ailanthoides, and the stem of P. ginseng.

당근 절편을 이용한 Agrobacterium spp. 분리, 동정과 선발균주에 의한 연초의 형질전환 (Isolation and identification of Agrobacterium spp. using carrot disc and transformation of Nicotiana gluca by selected strains)

  • These studies were carred out to obtain the transformant from tobacco cells by Agrobacterium spp. from crown gall and soil at the natural field in Korea, and identified their virulence. Kodo's and Clark's selective media were used for isolation of Agrobacterium spp. In these media, total of 99 strains were characterized based on the morphological characteristics of colonies. Among them 34 strains were able to induce on carrot discs. And hypervirulent strains C23-1 and K29-1 were identified as Agrobacterium tumefaciens biotype 1 and biotype 2, respectively. These strains formed fast growing, larger gall as compared to those induced by other strains on the carrot discs. Transformed tobacco callus was initiated on the phytohormone free MS medium with 250$\mu\textrm{g}$/ml carbenicillin after co-cultivation of tobacco stem explants and Agrobacteria. On the phytohormone free media, shoot was rarely formed from transformed callus. However, these shoot were teratoma shoots which were not grown as normal shoot, and teratoma shoot from transformant by C23-1 was smaller than that of K29-1.

무균돼지뼈를 이용한 복합 골지지체의 제조와 생체적합성 평가 (Preparation and Biocompatibility of Composite Bone Scaffolds Using Gnotobiotic Pig Bones)

  • Highly porous composite bioceramic bone scaffolds were developed using sintered gnotobiotic pig bones. These scaffolds consisted of poly-D,L-lactic acid (P(D,L)LA) and bioceramic materials of pig bone powder. The bone scaffolds were able to promote biocompatibility and possess interconnected pores that would support cell adhesion and proliferation adequately. The composite scaffolds were tested with dental pulp stem cells for cytotoxicity test. Cells seeded on the composite scaffolds were readily attached, well proliferated, as confirmed by cytotoxicity test, and cell adhesion assessment. The composite bone scaffold had no toxicity in cytotoxicity test on the extract of 0.013 g scaffold to 2 ml culture medium. The cells on the composite bone scaffold proliferated better than cells on the P(D,L)LA scaffolds.

Antibiotic Effect of Leaf, Stem, and Root Extracts in Smallanthus sonchifolius H. Robinson

  • This study was conducted with the aim of evaluating the antibiotic effects of leaves, stems, and roots in yacon (Smallanthus sonchifolius). Antibacterial activity of leaf extract by disk diffusion method with Bacillus subtilis and Escherichia coli respectively showed 13.3 and 13.75mm diameters of clear zone. There was no significant difference between the stems and leaves. The minimum inhibitory concentration of leaves' heating and agitation extraction showed a restrain of strain at 1mg/ml, but the stems and root extract did not appear. Yacon is a functional antibacterial material, and methanol extraction is more effective than water. This research was to investigate the growth stage of collection has the most effective antibacterial effects. It has collected yacon's leaves from June to October, which is an appropriate time for collection right before reaping. Yacon leaf has antibacterial effects on Bacillus subtilis, Escherichia coli, Enterococcus faecium, and Salmonella enteritifis. There were no significant differences by the growth stage of collection. Leaves collected in July are high in phenol which helps in sulfating activity works well considering the high scavenging capability of DPPH. Leaves collected in September are high in total flavonoid.

  • The n-hexane and carbon tetrachloride soluble fractions of a methanolic extract of the stem bark of the Couroupita guianensis furnished three compounds, identified as $\beta$-amyrin (1), betulin-$3{\beta}$-caffeate (2) and lupeol-$3{\beta}$-caffeate (3). The structures of the isolated compounds were deduced by extensive spectroscopic analysis as well as by comparison with published values. Compounds 1-3 were subjected to antioxidant screening through free radical scavenging activity by DPPH (1,1-diphenyl-2-picrylhydrazyl), where compound 2 showed moderate antioxidant activity with $IC_{50}$ value $108.0{\mu}g/ml$.

First Report of Soft Rot by Pectobacterium carotovorum subsp. brasiliense on Amaranth in Korea

  • Amaranth has the potential for good materials related to nutrients and health benefits. There are several diseases of amaranth such as leaf blight, damping-off, and root rot. As a causal agent of soft rot disease, Pectobacterium spp. could infect various plant species. In this study, we isolated the bacterial pathogen causing soft rot of amaranth in South Korea. In Gangneung, Gangwon province during 2017, amaranth plants showed typical soft rot symptoms such as wilting, defoliation and odd smell. To isolate pathogen, the macerated tissues of contaminated amaranth were spread onto LB agar plates and purified by a single colony subculture. One ml bacterial suspension of a representative isolate was injected to the stem of five seedlings of 2-week-old amaranth with a needle. Ten mM magnesium sulfate solution was used as a negative control. 16S rDNA gene and recA gene were sequenced and compared with the reference sequences using the BLAST. In the phylogenetic tree based on 16S rDNA gene and recA gene, GSA1 strain was grouped in Pcb.

무 시들음병에 대한 간편한 대량 저항성 검정법 개발 (Development of an Efficient Mass-screening Method for Testing the Resistance of Radish to Fusarium Wilt)

  • 무 시들음병 저항성 검정에는 대부분 뿌리 침지(root-dipping) 접종 방법을 사용하고 있지만, 이 방법은 복잡한 접종 과정 때문에 많은 노동력과 시간이 소요된다. 본 연구는 F. oxysporum f. sp. raphani에 의해 발생하는 무 시들음병의 간편 대량 저항성 검정법을 확립하기 위하여 수행하였다. 간편 저항성 검정법을 개발하기 위해 scalpel 접종 방법, 접종 농도 및 생장 단계에 따른 무 감수성 및 저항성 품종의 시들음병 발생을 조사하였다. 그리고 그 결과로부터 무 시들음병 대량 검정을 위한 효율적인 간편 저항성 검정방법으로 14일 동안 재배된 유묘에 scalpel을 사용하여 지제부에서 0.5 cm 떨어진 곳에서 $90^{\circ}$ 각도 2 cm 깊이로 찔러서 뿌리에 상처를 주고 포자현탁액($1.0{\times}10^7conidia/ml$)을 10 ml씩 관주하여 접종하고 $25^{\circ}C$에서 하루 12시간씩 광을 조사하면서 약 4주간 재배하는 것이 효율적이라고 생각되었다. 본 연구를 통해 확립한 접종방법은 대량의 무 시료로부터 시들음병에 대한 저항성 개체를 효과적으로 선발하는 것을 가능하게 할 것이다.
