Stem-ML에 기반한 한국어 억양 생성 (Korean Prosody Generation Based on Stem-ML)

  • In this paper, we present a method of generating intonation contour for Korean text-to-speech (TTS) system and a method of synthesizing emotional speech, both based on Soft template mark-up language (Stem-ML), a novel prosody generation model combining mark-up tags and pitch generation in one. The evaluation shows that the intonation contour generated by Stem-ML is better than that by our previous work. It is also found that Stem-ML is a useful tool for generating emotional speech, by controling limited number of tags. Large-size emotional speech database is crucial for more extensive evaluation.

Selective in vitro cytotoxicity of Hypericum hookerianum towards cancer cell lines

  • The methanol extracts of the aerial parts, leaves and stem of Hypericum hookerianum were tested for in vitro cytotoxicity on selected normal and cancer cell lines and anti tumor activity using DLA cells. Cell viability and morphological changes were assessed. Among the three extracts tested, the stem extract of Hypericum hookerianum showed potent cytotoxicity against HEp-2 and RD cell lines. The $CTC_{50}$(concentration required to reduce viability by 50%) of this extract was found to be $2.02\;{\mu}g/ml$ for RD cell line, $10.25\;{\mu}g/ml$ for HEp-2 cell line and $100.06\;{\mu}g/ml$ for Vero cell line. In the clonogenic assay, no colony formation was observed up to a concentration of $100\;{\mu}g/ml$. In the short term cytotoxicity studies using DLA cells, 50% viability was observed in the concentration range of $50-100\;{\mu}g/ml$ for aerial parts, $100-200\;{\mu}g/ml$ for stem and more than $200\;{\mu}g/ml$ for leaf extracts of Hypericum hookerianum. In the long-term activity using HEp-2 cell line, no colony formation was observed over a concentration of 200 mg/ml for the stem extract. Hypericum hookerianum stem extract was fractionated into petroleum ether, chloroform, ethyl acetate and methanol soluble fractions. The petroleum ether and chloroform soluble fractions showed higher cytotoxic activity against HEp-2 cell line when compared to the other two fractions. The methanol stem extract of Hypericum hookerianum has the potential for further investigation in animal models to determine its anti-tumor activity and to identify its active principles.

BMP-2에 의한 협부 지방 성체 줄기세포의 골형성 (OSTEOGENESIS BY BMP-2 IN ADULT STEM CELL DERIVED FROM BUCCAL FAT PAD)

  • Bone morphogenetic proteins (BMPs) in combination with stem cells gain more significance for their use in bone tissue engineering. The mesenchymal stem cell can be differentiated into osteoblast by the treatment of BMP. The aim of this study is to characterize the osteogenic differentiation process of adult stem cells derived from buccal fat pad according to BMP-2 within culture media and decide the appropriate concentration of BMP-2 to facilitate osteogenesis. The authors procured the stem cell from buccal fat pad and analyzed for presence of stem cell by flow cytomety against CD-34, CD-105 and STRO-1. The buccal fat derived stem cells (BFDC) were treated by application of the different concentration with BMP-2 of 0, 10, 50, 100 and 200ng/ml, respectively. And their ability to differentiate into osteogenic pathway were checked by alkaline phosphatase(ALP) staining, Alizarin red staining and RT-PCR for osteocalcin(OC) gene expression at 7, 14 and 21day of culture. Flow cytometric analysis and biochemical assays demonstrated that BFDC might be a distinguished stem cells, and mineralization was accompanied in proportion to BMP-2 concentration. However, with 100ng/ml concentration of BMP-2, the BFDC demonstrated most efficient staining pattern of ALP and Alizarin red. The feasibility of the osteogenic differentiation in the group of both 50ng/ml and 200ng/ml of BMP-2 showed similar activity and relatively weaker than that of 100ng/ml. These results suggest that the BMP-2 stimulate osteogenesis by BFDC effectively and that bone induction might be controlled through negative regulatory feedback in higher concentration.

이화명나방 유충에 대한 곤충병원성 선충 Steinernema carpocapsae(Steinernematidiae)와 Heterorhabditis bacteriophora(Heterohabditidae)의 살충효과 (Evaluation of Entomopathogenic Nematodes, Steinernema carpocapsae (Steinernematidae) and Heterorhabditis bacteriophora (Hetrorhabditidae) against Rice Stem Borer Chilo suppressalis (Walker) (Lepidoptera: Pyralidae))

  • 이화명나방 유충을 방제할 목적으로 이화명충이 가해한 벼 줄기에 곤충병원성선충인 Steinernema carpocapsae와 Heterorhabditis bacteriophora를 살포한 결과 효과가 좋았다. 8cm 길이의 벼 줄기 2본에 S. carpocapsae 250, 500, 1,000, 2,000마리/ml를 살포한 결과 68.6$\pm$10.0~94.9$\pm$2.6%의 치사율을 얻었으며 H. bacteriophora 100, 200, 400, 800마리 ml살포구에서는 91.4$\pm$0.7~100%의 방제를 얻었다. 그러나 3본의 벼줄기를 H.bacteriophora 현탁액에 침적한 경우는 100마리/ml 구에서는 42.6$\pm$4.7%, 200마리/ml구에서는 63.1$\pm$4.7%로 선충의 현탁액을 살포하는 것보다는 치사율이 낮아 효과가 적었다. 벼 줄기는 선충의 생존과 기주 탐색에 좋은 환경을 유지해 주어 곤충병원 성선충은 이화명충의 방제에 유용한 생물적 인자로 확인 되었다.

Potential antifilarial activity of the fruit, leaf and stem extract of Melia azedarach Linn. on cattle filarial parasite Setaria cervi in vitro

  • The effect of aqueous and alcoholic extracts of the fruit, leaf and stem of Melia azedarach Linn. (Meliaceae) on the spontaneous movements of both the whole worm and the nerve-muscle (n.m.) preparation of Setaria (S.) cervi and on the survival of microfilariae in vitro was studied. Alcoholic extracts of fruit, leaf and stem caused inhibition of the spontaneous movements of the whole worm and the n.m. preparation of S. cervi, while only aqueous extract of fruit caused inhibition of the spontaneous movements of the whole worm and the n.m. preparation of S. cervi. The initial stimulatory effect was not observed by the aqueous and alcoholic extracts of fruit on n.m. preparation. The concentrations required to inhibit the movements of the whole worm and n.m. preparation for alcoholic extracts of fruit, leaf and stem were 250, $40\;{\mu}g/ml$; 280, $40\;{\mu}g/ml$ and 270, $25\;{\mu}g/ml$ respectively, whereas an aqueous extract of fruit caused inhibition of whole worm and n.m. preparation at $200\;{\mu}g/ml$ and $40\;{\mu}g/ml$ respectively. Alcoholic extracts of the fruit, leaf and stem and aqueous extract of the fruit of M. azedarach caused concentration related inhibition on the survival of microfilariae (m.f.) of S. cervi. The $LC_{50}$ and $LC_{90}$ as observed after 6 h were found to be 5, 15, 10, 20 ng/ml and 10, 25, 20 and 35 ng/ml, respectively. This work was conducted in view of the exploration of potential antifilarial herbal drug.

울릉도 마가목의 다양한 부위의 항혈전 활성 (In-vitro Antithrombosis Activity of Different Parts of Sorbus commixta from Ulleung Island)

  • 마가목은 울릉도를 대표하는 약용식물 중 하나이다. 본 연구에서는 마가목을 이용한 혈행개선 기능성을 가진 고부가가치 음료 제조를 위해 마가목의 미숙과, 성숙과, 줄기 및 잎의 열수 추출물을 제조하고, 이를 대상으로 기존에 보고된 바 없는 항혈전 활성 및 항산화 활성을 검토하였다. 마가목 줄기의 경우 추출효율은 3.99%, pH 5.5이었으며, 열수 추출물은 총 폴리페놀 89.2 mg/g, 총 플라보노이드 28.3 mg/g의 함량을 나타내었다. 다양한 부위의 마가목 추출물들은 0.5 mg/ml 농도까지 인간 적혈구 용혈활성을 나타나지 않았다. 혈액응고 저해활성 평가결과, 줄기 추출물에서만 강력한 트롬빈, 프로트롬빈, 혈액응고인자 저해가 확인되었으며, 인간 혈소판 응집저해활성은 인정되지 않았다. 한편 혈전 생성억제와 관련된 항산화 활성 평가 결과, 줄기 추출물에서 강력한 DPPH 음이온, ABTS 음이온 소거능, nitrite 소거능을 확인하였으며, 각각의 RC50는 각각 119.7, 53.3, 및 117.5 μg/ml으로 계산되었다. 항산화제인 vitamin C의 DPPH 음이온, ABTS 양이온 및 nitrite 소거능에 대한 RC50가, 각각 13.7, 5.2, 및 14.9 μg/ml임을 고려할 때, 마가목 줄기 추출물은 정제되지 않은 상태에서도 우수한 항산화 활성을 나타내었으며, 환원력 역시 다른 부위에 비해 강력하였다. 본 연구결과는 강력한 항응고 활성과 항산화 활성을 나타내는 마가목 줄기 추출물이 신규의 항혈전 천연물 소재로 이용가능 함을 제시하고 있다.

Nitric Oxide Inhibition and Procollagen Type I Peptide Synthesis Activities of a Phenolic Amide Identified from the Stem of Lycium chinense Miller

  • The bioactivities of boxthron fruits, a source of oriental medicine, are well known, whereas phytochemical studies of the boxthorn stem are rare. In this study, the stem extract of boxthorn (Lycium chinense Miller) and its subfractions were evaluated for their effects on nitric oxide (NO) inhibition and procollagen type I peptide (PIP) synthesis. A phenolic amide isolated from the stem extract was also assayed for these effects. The compound, N-trans-feruloyltyramine, was identified by $^1H$, $^{13}C$, and 2D-nuclear magnetic resonance analyses. In NO inhibition, the chloroform fraction (CF) exhibited the strongest inhibitory activity ($MIC_{50}=24.69{\mu}g/ml$) among the subfractions of the ethanol extract (EE). N-trans-feruloyltyramine isolated from the CF showed strong NO inhibitory activity, presenting with an $MIC_{50}$ of $31.36{\mu}g/ml$. The EE, CF, and N-trans-feruloyltyramine shown to have NO inhibition activity were assayed for the activity of PIP synthesis. The EE and CF showed relatively high PIP values of 38.8% and 24.21% at $100{\mu}g/ml$, respectively. The PIP value for $20{\mu}g/ml$ N-trans-feruloyltyramine showed a 36% increase compared with the non-treated control, whereas that treated with $20{\mu}g/ml$ ascorbic acid as a positive control showed a 13% increase. The results suggest that the proper stem extract of boxthorn stem could be efficiently used to produce good cosmetic effects.

Chemical Composition of Leaves, Stem bark and Fruit Essential Oil from Premna foetida Linn

  • Premna foetida is a woody plant with short and twisted trunk. P. foetida is a scandent, erect shrub or small tree, thorny on the trunk and large branches. Leaves are opposite or whorled and entire or serrate. Premna foetida is a wild plant locally known as "Daun Sebuas". P. foetida is used for it nutritive and as traditional treatment. The fruit and leaves of P. foetida are prepared for salad. The study aimed at the hydrodistillation and antioxidant activity of leaves, stem-bark and fruits essential oil from Premna foetida Linn, they were analysed by capillary GC and GC-MS. Ninety eight compounds representing 81.68±0.02, 37.31±0.05 and 93.45±0.03 of the isolates of leaves, stem-bark and fruits respectively were identified, the most abundant were α -Duprezianene (77.27±0.03, leaves, α-Gurjunene (36.06±0.05) fruits and Hinesol acetate (77.19±0.03) stem-bark. Components among which sesquiterpenoids dominated. The total volatiles were assayed for antioxidant potentials using 2, 2-diphenyl-1-picrylhydrazyl (DPPH). The total volatiles showed strong activity with IC50 of 11.74±0.82㎍/ml, 9.63±0.34 ㎍/ml and 49.73±1.12 ㎍/ml for leaves fruits and stem-bark respectively.

해당화 줄기 및 뿌리의 주성분 분리 및 항산화활성 (Isolation of main component and antioxidant activities on the stem and root of Rosa rugosa)

  • 1. 해당화의 총페놀화합물 함량은 줄기추출물이 평균 7.8(g/100g, D.W.)이고 뿌리추출물이 평균 10.5(g/100g, D.W.)이며, 모든 시료에서 뿌리추출물의 함량이 높게 나타났다. 2. K-1의 줄기와 뿌리 추출물(0.5g)에서 주성분의 하나로 추정되는 S-4(84.1mg)과 R-2(70.0mg)의 화합물을 단리했고, 그 구조를 catechin으로 결정했다. 3. UPLC 분석에 의해 줄기추출물의 탄닌함량은 평균 752.5(mg/100g, D. W.)로, gallic acid 6.3, epicatechin 61.5, catechin 684.8이고, 뿌리추출물의 탄닌함량은 줄기의 약 2배 이상 높은 1676.0(mg/100g, D.W.)으로 gallic acid 6.8, epicatechin 160.3, catechin 1508.5로 나타났다. 4. 해당화의 free radical 소거활성에서 추출물의 IC50는 뿌리가 $13.7{\mu}g/ml$, 줄기가 $14.5{\mu}g/ml$으로 높은 소거활성을 나타냈고, SOD 유사활성은 $50{\mu}g/ml$의 농도에서 뿌리추출물이 평균 75.1%, 줄기추출물이 71.9%로써 높은 활성을 나타냈다.

High Dose of FGF-2 Induced Growth Retardation via ERK1/2 De-phosphorylation in Bone Marrow-derived Mesenchymal Stem Cells

  • Fibroblast growth factor (FGF)-2 is one of the most effective growth factors to increase the growth rate of mesenchymal stem cells (MSCs). Previously, we reported that low dose of FGF-2 (1 ng/ml) induced proliferation of bone marrow-derived mesenchymal stem cells (BMSCs) through AKT and ERK activation resulting in reduction of autophagy and senescence, but not at a high dose. In this study, we investigated the effects of high dose FGF-2 (10 ng/ml) on proliferation, autophagy and senescence of BMSCs for long term cultures (i.e., 2 months). FGF-2 increased the growth rate of BMSCs in a dose dependent manner for a short term (3 days), while during long term cultures (2 months), population doubling time was increased and accumulated cell number was lower than control in BMSCs when cultured with 10 ng/ml of FGF-2. 10 ng/ml of FGF-2 induced immediate de-phosphorylation of ERK1/2, expression of LC3-II, and increase of senescence associated ${\beta}$-galactosidase (SA-${\beta}$-Gal, senescence marker) expression. In conclusion, we showed that 10 ng/ml of FGF-2 was inadequate for ex vivo expansion of BMSCs because 10 ng/ml of FGF-2 induced growth retardation via ERK1/2 de-phosphorylation and induction of autophagy and senescence in BMSCs.
