소나무재선충 인공접종에 의한 해송묘목의 병 발달 및 수분함량 변화 (Change of Water Content and Disease Development on Pinus thunbergii Seedlings Inoculated with Bursaphelenchus xylophilus)

  • 본 연구는 국내산 4년생 해송을 대상으로 소나무재선충을 인공접종하여 나타나는 병징의 발달과 묘목내 수분함량 및 선충밀도의 변화를 조사 및 분석하여 소나무재선충에 의한 생리적 변화과정 구명에 사용될 수 있는 기초 자료로 활용되고자 수행되었다. 소나무재선충 인공접종 후 20일을 기점으로 외부적으로 잎의 변색 그리고 내부적으로 접종 부위 위 아래의 목부 건조 및 수 갈변 현상이 나타났다. 그러나 접종 후 10일에 대조구 묘목과 접종목의 목부 및 피층의 건조상태는 차이를 나타내어 목부 건조는 접종 후 10일 경부터 진행이 되고 있음을 알 수 있었다. 소나무재선충 밀도는 접종 후 5일과 10일 사이에서 소폭의 증가를 보였으나 외부적 또는 내부적 피해증상이 나타난 10일과 20일 사이에서는 급격한 증가를 보였다. 소나무재선충 접종 후 시간이 경과함에 따라 감염목의 줄기수분함량 및 당년생 잎과 줄기의 상대수분함량은 꾸준히 감소하였다. 소나무재선충의 개체 수가 증가함에 따라 줄기수분함량, 당년생 가지 및 잎의 상대수분함량이 유의적으로 감소하여, 소나무재선충 증식은 병 발달 및 수체내 목부의 수분건조 현상과 밀접한 상관관계를 보이지만 묘목의 고사와 함께 재선충 번식이 현격히 줄어드는 것으로 나타났다.

잣나무 조림목(造林木)의 조직특성(組織特性)의 변동(變動)과 성열재(成熱材)·미성열재(未成熱材)의 재질(材質) (Changes in Cellular Characteristics and Qualities of Matured and Juvenile Wood from Reforested Tree of Pinus koraiensis)

  • Pinus koraiensis is one of the most important reforestation species which had widely been planted around the country. Therefore, there are great research needs on the fundamental properties of wood to extend the frequency of uses and adequate availabilities. Research results on the characteristics of anatomical wood properties and changes are summarized as follows: 1. At a horizontal direction of tree trunk, the tracheid length at outer wood gradually becomes shorter as it reaches to pith. The tracheid length having between 32 and 42 annual rings showed over 2.5 times longer than that of pith, and also its length increases from the bottom to the top of the stem. 2. The tracheid diameters in latewood showed a little fluctuations, and the shortest near to the pith in earlywood. The tracheid diameters in earlywood are more than 2 times those of latewood, and the differences between the mature and juvenile wood in earlywood are less than 10${\mu}m$. 3. The tracheid wall thickness in early wood showed a little fluctuations. On the contrary, tracheid wall thickness in latewood rapidly increases from the pith to the bark. The tracheid wall of mature wood showed 10 to 20% thicker than that of juvenile wood. 4. The fibril angle at the secondary walls of tracheid within an annual ring showed higher degree in earlywood than that of latewood. The highest tracheid fibril angle was observed at around the pith of both earlywood and latewood. Then, fibril angle slowly decreased toward outer wood, and was stabilized after 15 years. 5. Structural boundaries between mature and juvenile wood from Pinus koraiensis are divided by 13 to 19 annual ring and distance of 5 to 8cm from the pith.

Chemical Components, Antitermite and Antifungal Activities of Cinnamomum parthenoxylon Wood Vinegar

  • Termiticidal and fungicidal activities of wood vinegar from Cinnamomum parthenoxylon (CP) stem wood have been evaluated against Coptotermes curvignathus and wood rotting fungi (Schizophyllum commune and Fomitopsis palustris). The utilized CP wood vinegar was produced in the operating temperature range 250-300℃ pyrolysis. A no-choice test was applied for evaluating termiticidal activity with 33 active termites and antifungal activity using the agar media assay. The result showed that an increase in the concentrations of CP wood vinegar significantly raised the mortality of termite. CP wood vinegar showed high termiticidal activity, organic acids (acetic acid 42.91%, 3-butenoic acid 6.89%, butanoic acid, 2-propenyl ester 2.26%), and ketones (1-hydroxy-2-propanone 5.14%, 3-methylcyclopentane-1,2-dione 2.34%) might be largely contributed to termiticidal activity in addition to other minor components. Furthermore, CP wood vinegar exhibited significant inhibition of fungal growth. These data showed that CP wood vinegar was more toxic to white-rot fungi (S. commune) than brown-rot (F. palustris). The results suggested that phenolic compounds from lignin degradation were responsible for good antifungal activity.

  • In order ot obtain the information on wood quality of the Populus nigra $\times$ maximowiczii the distributions of crystalliferous substancess, ty loses and tension woods were examined through light microscopy or scanning electron microscopy. 1. The crystalliferous subtance, presumably calcium oxalate. partially occurred in vessel elements of heartwood, and sporadically in ray parenchyma, but not in the wood fibers The types of crystalliferous subtances were mainly crystal sand, $2-3{\mu}m$ in diameter. 2. The tyloses arranged in a ladder-like series were sporadically formed in the vessel elements of heartwood. 3. Although the woods were cut from the straightly grown stem without any evident disorientation with respect to gravitational axis, single or group of gelatinous fibers were irregularly distributed among normal wood fibers.

Anatomical Characteristics of Paulownia tomentosa Root Wood

  • This study investigated several anatomical characteristics of Paulownia tomentosa roots. The root wood was separated into three parts from stem base (top, middle, and base) at different positions below ground. Qualitative anatomical data suggested that the growth rings in earlywood and latewood were structurally different. Furthermore, the root wood vessels were found having 2 to 3 radial multiples and they were appeared in the form of clusters. In addition, some sheath cells and septate axial parenchyma were observed. Regarding the quantitative anatomical characteristics, vessel and ray numbers per $mm^2$, as well as ray width and height differed significantly among the top, middle, and base rood wood parts. However, there were no significant differences in vessel diameters, cell wall thickness, and width and length of wood fibers among those parts. The crystallinity of the root top part was slightly higher than that of the middle and base parts. Furthermore, the vessel numbers, ray numbers, and ray width and height in the near pith (NP) area were higher compared to those in the near bark (NB) area. However, the fiber width and fiber length at NP were lower than those at NB. Overall, this study demonstrated some significant differences in the anatomical characteristics of the top, middle, and base parts of root wood from Paulownia tomentosa.

음나무 추출물의 세포 내 산화 스트레스와 항산화 활성 (Oxidative Stress in the Cell and Antioxidant Activity of Kalopanax Pictus Extracts)

  • 음나무의 새로운 용도 개발을 위하여 다양한 항산화 측정법을 이용하여 생리활성을 측정하였다. 즉, 음나무 수피 및 근피의 온수 추출물과 메탄올 추출물에 대한 세포 내 활성산소 농도를 측정하여 세포수준에서의 산화 스트레스 억제효과를 측정하였으며, DPPH법 및 NBT법을 이용하여 각 추출물의 화학적인 측면에서의 항산화력을 측정하여 이들 간의 차이를 비교하였다. NR assay와 MTT assay에 의해 측정한 세포독성 실험결과, 음나무 근피의 메탄올 추출물의 세포독성이 가장 강했으며, 50%의 세포 생존율을 나타내는 NR50과 MTT50은 각각 0.0018%와 0.0029%였다. 산화 스트레스 억제효과는 근피의 불용성 온수 추출물이 15분 처리에서 57.9%로 가장 우수하였다. DPPH법과 NBT법을 이용한 음나무 추출물에 대한 항산화 활성 측정결과, 각각 수피의 메탄올 추출물이 96% (0.1% 농도)이고, 수피의 불용성 온수 추출물이 95% (0.5% 농도)를 나타내어 우수한 항산화 활성을 나타내었다.

농림부산물을 이용한 펠릿의 열적 특성 (Thermal Characteristics of Pellets made of Agricultural and Forest by-products)

  • Biomass is considered to be a major potential fuel and renewable resource for the future. In fact, there is high potential to produce the large amount of energy from biomass around the world. In this study, to obtain basic data for practical application of agricultural and forest by-products as fuel of heating system in agriculture, agricultural and forest biomass resources were surveyed, the pelletizer with capacity of $50\;kg{\cdot}h^{-1}$ was designed and manufactured and pellets were made by the pelletizer. High heating value, ash content, etc. of pellets made of agricultural and forest by-products were estimated. Straw of rice was the largest agricultural biomass in 2009 and the total amount of rice straw converted into energy of $299{\times}10^3$ TOE. And in 2009, amount of forest by-product converted into energy of $9,579{\times}10^3$ TOE. High heating values of pellets made of stem and seed of rape, stem of oat, rice straw and rice husk were 16,034, 16,026, 16,089, 15,650, $15,044\;kJ{\cdot}kg^{-1}$ respectively. High heating values of pellets made of agricultural by-products were average 83.6% compared to that of wood pellet. Average bulk density of pellets made of stem and seed of rape, stem of oat, rice straw and rice husk was $1,400\;kg{\cdot}m^{-3}$ ($1.4\;g{\cdot}cm^{-3}$). Ash contents of the pellets were 6.6, 7, 6.2, 5.5, 33% respectively. Rice husk pellet produced the largest ash content compared to other kinds of pellets.

Variation in Tree Growth Characteristics, Pilodyn Penetration, and Stress-wave Velocity in 65 Families of Acacia mangium Trees Planted in Indonesia

  • Growth characteristics [stem diameter (D), tree height (H)] and wood properties [Pilodyn penetration (P) and stress-wave velocity (SWV)] were measured for 65 families of 6-year-old Acacia mangium trees planted in Indonesia, in order to characterize their variation in D, H, P, and SWV. Therefore, the correlations between the measured characteristics were also determined, and their significant differences observed. Furthermore, their low to moderate values of narrow-sense heritability was obtained, and the results indicated the characteristics to be genetically controlled in A. mangium. In addition, highly significant positive correlations were observed among the growth characteristics, suggesting a close relationship, while there was no significant association between the growth characteristics and P, as well as SWV, indicating their independent. Therefore, these results demonstrate a potential for the improvement of both growth and wood properties of A. mangium trees, using the appropriate breeding programs. In addition, 18 families showed good performance in D and SWV, signifying their positive prospect of being considered as plus trees for the next generation breeding cycles.

Constituents of Lindera Erythrocarpa Stem Bark

  • Three chalcones and a stilbenoid have been isolated from the stem bark of Lindera erythrocarpa. They were elucidated as 2'-hydroxy-3',4',5',6'- tetramethoxychalcone, 2',4'-dihydroxy-3',6'-dimethoxy chalcone, 2',4',5',6'- tetrahydroxy-3'-methoxychalcone, and 5,6-dihydroxy-2,3,4-trimethoxystilbene. Chemical structures were elucidated on the basis of MS, $^1H,\;^{13}C$ and 2D-NMR spectroscopic data. This is the first report on the isolation of these compounds from the L. erythrocarpa.

Migrations and Multiplications of Bursaphelenchus xylophilus and B. mucronatus in Pinus thumbergii in Relation to Their Pathogenicity

  • To evaluate the mechanisms of pathogenicity and nonpathogenicity of Bursaphelenchus xylophilus and B. mucronatus isolated in South Korea, we used 4-year-old P. thunbergii seedlings and 20-cm long one-year-old stem cuttings of 5-year-old Pinus thunbergii, and studied distributions and multiplications of pine wood nematodes after inoculation. The distributions of B. xylophilus in the 20-cm pine stem cuttings were not significantly different from that of B. mucronatus. Conversely, the proliferation rate of B. xylophilus on mycelial mats of Botrytis cinerea was significantly different from that of B. mucronatus. The study using 4-year-old P. thunbergii seedlings also showed that B. mucronatus can migrate to distal portions of the pine seedlings the same as B. xylophilus, but the populations of B. xylophilus remaining in the pine seedlings were relatively larger than those of B. mucronatus. Therefore, we concluded that the pathogenicity of B. xylophilus could be strongly influenced by its ability to multiply.
