Allelic diversity of 44 microsatellite marker loci originated from the coding regions of specific genes or the non-coding regions of barley genome was analyzed for 19 barley genotypes. Multi-allelic variation was observed at the most of marker loci except for HVM13, HVM15, HVM22, and HVM64. The number of different alleles ranged from 2 to 12 with a mean of 4.0 alleles per micro-satellite. Twenty-one alleles derived from 10 marker loci are specific for certain genotypes. The level of polymorphism (Polymorphic Information Content, PIC) based on the band pattern frequencies among genotypes was relatively high at the several loci such as HVM3, HVM5, HVM14, HVM36, HVM62 and HVM67. In the cluster analysis using genetic similarity matrix calculated from microsatellite-derived DNA profiles, two major groups were classified and the spike-row type was a major factor for clustering. Correlation between genetic similarity matrices based on microsatellite markers and pedigree data was highly significant ($r=0.57^{**}$), but these two parameters were moderately associated each other. On the other hand, RAPD-based genetic similarity matrix was more highly associated with microsatellite-based genetic similarity ($r=0.63^{**}$) than coefficient of parentage.

This experiment was conducted to evaluate genetic variation in 48 rice accessions (Oryza sativa L.) using AFLP and RAPD markers. For AFLP, a total of 928 bands were generated with 11 primer combinations and 327 bands (35.2%) of them were polymorphic among 48 accessions. In RAPD analyses using 22 random primers 145 bands were produced, and 121 (83.4%) were polymorphic among 48 accessions. Each accession revealed a distinct fingerprint by two DNA marker systems. Cluster analysis using AFLP-based genetic similarity tended to classify rice cultivars into different groups corresponding to their varietal types and breeding pedigrees, but not using RAPD-based genetic similarity. The AFLP marker system was more sensitive than RAPD in fingerprinting of rice cultivars with narrow genetic diversity.

The diesel-degrading activities of biofilms sampled from petroleum-contaminated groundwaters in urban subway drainage systems were examined in liquid cultures, and the microbial populations of the biofilms were characterized by denaturing gel gradient electrophoresis (DGGE) and 16S rDNA sequence analysis. Biofilm samples derived from two sites (19 K and 20 K) at subway Station N and Station I could degrade around $80\%$ of applied diesel within 20 and 40 days, respectively, at $15^{\circ}C$, and these results were strongly correlated with the growth patterns of the biofilms. The closest phylogenetic neighbor of a dominant component in the 19 K biofilm was Thiothrix fructosivorans strain Q ($100\%$ similarity). Four dominant strains in the 20 K biofilm were closely related to Thiothrix fructosivorans strain Q ($100\%$ similarity), Thiothrix sp. CC-5 ($100\%$ similarity), Sphaerotilus sp. IF14 ($99\%$ similarity), and Cytophaga-Flexibacter-Bacterioides (CFB) group bacterium RW262 ($98\%$ similarity). Three dominant members in the Station I biofilms were very similar to uncultured Cytophagales clone CRE-PA82 ($91\%$ similarity), Pseudomonas sp. WDL5 ($97\%$ similarity), and uncultured CFB group bacterium LCK-64 ($94\%$ similarity). The microbial components of the biofilms differed depending on the sampling site. This is the first report on the isolation of clones highly similar to Thiothrix fructosivorans and Thiothrix sp. from biofilms in petroleum-polluted groundwaters, and the first evidence that these organisms may play major roles in petroleum degradation and/or biofilm-development.
Randomly amplified polymorphic DNA(RAPD) analysis was applied to detect the molecular polymorphisms in Gentiana scabra var. buergeri. A high level of molecular variability was found among wild plants and cultivars. In genetic analysis, eight of twenty primers were selected and 54 amplification products ranged 2. 2 to 0.2 kb were compared. Twenty - nine amplified products showed polymorphic, while five were monomorphic. Twenty of line specific bands were found. In genetic similarity and cluster analysis using PCR products, three wild plants collected from Naejangsan, Daedunsan and Keojedo and one cultivar Seochunjaerae were grouped into one cluster, while cultivar Jinbujaerae and Japanese line separated into another clusters, respectively. The identification of DNA polymorphisms by the RAPD technique will facilitate the selection of the lines from different origin.

In this study, the smoke flows of the inner subway station were visualized through a numerical analysis and visualization experiment in the subway fire. A transparent acrylic model was designed and installed as 1:25th scale-down as the actual subway station by using geometrical similarity The properties of subway fire were reconstructed according to Densimetric Froude Similarity. The 47 to 53 ratio of the mixed air and Helium was inputted in the inner acrylic model to describe 1MW fire intensity with reference to the experiment paper. For the same time, the fire smoke from a smoke generator was inputted in the inner acrylic model with the mixture. At this time, the buoyancy effect of Helium gas went up the smoke to the acrylic model. When the sheet beam of Ar-lon laser was given out to the top and stair of subway model, the digital camcorder took the images of the scattered cluster of smoke particles when applying the smoke management system and PSD.
Genetic diversity of 45 tea accessions from Korea, Japan, China and Taiwan was investigated by using RAPD analysis. Out of the eighty primers screened, twenty primers generated 99 polymorphic bands with a polymorphic rate 87.0%. The size of the amplified fragments ranged from about 3,138 bp to 520 bp. By cluster analysis, all of the 45 accessions can be grouped into five groups. Over 90% of the 32 Korean accessions belonged to group II, III, IV and V. Moreover, newly developed Korean cultivars (accession no. 13, 14 and 15) belonged to very different group compared with any other Korean accessions. Among the Korean accessions, the minimum genetic similarity 0.500 was obtained between accession no. 17 and 37 and the largest genetic similarity 0.912 between no. 20 and 21.

To evaluate the intraspecific variations among the Kasiogalpi(Eleutherococcus senticosus Max.) collections. randomly amplified DNA polymorphisms were examined. Twenty primers from 90 primers applied were selected. The range of polymorphism was $7.1{\sim}90.9%$ in 113 randomly and specifically amplified DNA fragments. Collections were divided into two major groups at the similarity coefficient value of 0.65. A considerable degree of genetic diversity was also detected among plants within the same collections. Deokyu (1, 2, 3, 4, 6), Bukhaedo (7, 8) and Odae (9, 10) col1ections showed higher degree of genetic similarity with a value of $0.65{\sim}0.86$, while Deokyu 5 showed much lower genetic similarity than other col1ections.
Insight into behaviour of pedestrians as welt as tools to assess passenger flow condition is important in such instances as planning and geometric design of railway station under regular and safety-critical circumstances. Algorithm for passenger flow analysis based on DEM (Discrete Element Method) is newly developed. There are lots of similarity between particle-laden two phase flow and passenger flow. The velocity component of 1st phase corresponds to the unit vector of calculation cell, each particle to passenger, volume fraction to population density and the particle velocity to the walking velocity, etc. And, the walking velocity of passenger is also represented by the function of population density. Key algorithms are developed to determine the position of passenger, population density and numbering to each passenger. To verify the effectiveness of new algorithm, passenger flow analysis for the basic models of railway station is conducted.
Landslide, one of the serious natural disasters, has Incurred a large loss of human and material resources. Recently, many forecasting or alarm systems based on various kinds of measuring equipment have been developed to reduce the damage of landslide. However, only a few of these equipments are guaranteed to evaluate the safety of whole side of land slope with their accessibility to the slope. In this study, we performed some experiments to evaluate the applicability of a terrestrial LiDAR as a surveying tool to measure the displacement of a land slope surface far a slope collapsing protection system. In the experiments, we had applied a slope stability method to a land slope and then forced to this slope with a load increasing step by step. In each step, we measured the slope surface with both a total station and a terrestrial LiDAR simultaneously. As the result of Slope Fracturing analysis using all targets, the LiDAR system showed that three was 1cm RMSE on X-axis, irregularity errors on Y-axis and few errors on Z-axis compare with Total Station. As the result of Slope Fracturing analysis using continuous targets, the pattern of Slope Fracturing was different according to the location of continuous targets and we could detect a continuous change which couldn't be found using Total station. The accuracy of the LiDAR data was evaluated to be comparable to that of the total station data. We found that a LiDAR system was appropriate to measuring the behaviour of land slope. The LiDAR data can cover the whole surface of the land slope, whereas the total station data are available on a small number of targets. Moreover, we extracted more detail information about the behavior of land slope such as the volume and profile changes using the LiDAR data.
