Sexual reproduction of paramecia have been accomplished through conjugation between individuals which have opposite mating type substances on their cilia when they were starved. Using selfing clone in which mating takes place, I examined whether a mating type change in indicidual cells required new protein and new mRNA synthesis or not and also shether there is a precursor relationship between both of the complementary mating type substances in their synthetic pathway. I found that 1. Mating type change needs new protein(s) and new mRNA synthesis. 2. Mating type substances are synthesized sequentially from mating type XIII to XIV 3. There might be a common precursor pool from which the mating type XIII substnace is synthesized and then complementary mating type XIV is fromed by addition of small group to the mating type XIII substance.

We attempted to control the maturation promoting factor (MPF) activity and investigated the subsequent reprogramming of bovine somatic cell nuclear transfer (SCNT) embryos. Serum-starved adult skin fibroblasts were fused to enucleated oocytes treated with 2.5 mM caffeine or $150\;{\mu}M$ roscovitine. The MPF activity, nuclear remodeling patterns, chromosome constitutions and development of SCNT embryos were evaluated. Methylated DNA of embryos was detected at various developmental stages. The MPF activity was increased by caffeine treatment or reduced by roscovitine treatment (p<0.05). Blastocyst development was higher in the caffeine-treated groups (27.6%) than that of the roscovitine-treated group (8.3%, p<0.05). There was no difference in the apoptotic cell index among the three groups. However, the mean cell number of blastocysts was increased in the caffeine-treated group (p<0.05). Higher methylation levels were observed in the Day 3 embryos of the roscovitine-treated group (50.8%), whereas lower methylation levels were noted at Day 5 in the caffeine-treated group (12.5%, p<0.05). These results reveal that the increase in MPF activity via a caffeine-treatment creates a more suitable condition for nuclear reprogramming after SCNT.
Effect of fasting and refeeding on growth and blood chemistry of juvenile olive flounder Paralichthys olivaceus L. was investigated when fish achieved compensatory growth. Fish were fed the experimental diet for 6 days a week. Five treatments in triplicate were prepared: C, S1, S2, S3 and S4. Fish in the control group (C) were hand-fed to apparent satiation twice a day. Fish in treatments of S1, S2, S3 and S4 experienced 1, 2, 3 and 4 weeks of starvation and were then hand-fed to satiation twice daily during the remaining 7, 6, 5 and 4 weeks of the experiment, respectively. Weight gain of fish in C, S1 and S2 were higher than those of fish in S3 and S4. A significant difference in plasma total protein, glucose, triglyceride, $T_3$ and $T_4$ was observed in between starved and refed fish for the rest periods of the feeding trial. Plasma total protein and $T_3$ of flounder decreased with week of fasting and following correlationships were obtained; Y (Total protein) = -0.13X (week of fasting) + 1.54, $R^2=0.9792$ and $Y(T_3)=-11.48X$ (week of fasting) + 79.57, $R^2=0.8822$, respectively.

The effects of dietary nutrient composition on compensatory growth of grower olive flounder (Paralichthys olivaceus) under different feeding regimes at suboptimal temperature were determined. Four hundred five fish weighing 271.2 g were distributed into 27 300 L flow-through tanks (15 fish per tank). Nine treatments were prepared in triplicate: fish were hand-fed with control (C) diet for 12 weeks (12WF-C); four groups of fish were starved for 1 week and then fed C, high-protein (HP), high-lipid (HL), or combined high-protein and high-lipid (HPL) diets for 11 weeks; these groups are referred to as 11WF-C, 11WF-HP, 11WF-HL, and 11WF-HPL, respectively. Four other groups of fish were starved for 2 weeks and then fed C, HP, HL, and HPL diets for 10 weeks; these groups are referred to as 10WF-C, 10WF-HP, 10WF-HL, and 10WF-HPL, respectively. Weight gain and specific growth rate of fish from 12WF-C group were greater than those of fish from 11WF-C, 11WF-HP, 11WF-HL, 10WF-C, 10WFHP, and 10WF-HL groups, but not different from those of fish from 11WF-HPL and 10WF-HPL groups. Feed-efficiency ratio of fish from 12WF-C, 11WF-HP, 11WF-HPL, 10WF-HL, and 10WF-HPL groups was higher than that of fish in 11WF-C, 11WF-HL, 10WF-C, and 10WF-HP groups. The results of this study demonstrated that grower olive flounder subjected to 1- or 2-week feed deprivation were able to achieve full compensatory growth at suboptimal temperature only when fed HPL diet.

The effect of delayed initial feeding (1, 3, 5, 7 days) and starvation on morphological change, mortality and cannibalism on larvae of Siniperca scherzeri was examined by laboratory rearing. The larvae of S. scherzeri began to feed on Artemia nauplii at 4 days after hatching. In case of unfed and 7-days delayed groups, all of the larvae died at 12 days after hatching. The larvae of 1 day delayed feeding survived and grew as almost same as the control group, and 3-days delayed groups showed $33\%$ survival rate at the end of experiment (12 days after hatching). In case of the unfed group, total length of the starved larvae showed lower growth rate than the control group, and they did not reached at the same size of the larvae of the control group. Cannibalism were more common in the unfed group and the delayed fed group than the control group. The highest rate of daily mortality caused by cannibalism in the delayed fed group was $23\%$ at 8 days after hatching.
Following the previous experiments, a starvation experiment was conducted to determine the influence of feeding and starvation on the histological and biochemical changes, the morphormetric changes in the sectioned body and the morphometric changes in Rhynchocypris oxycephalus (Sauvage and Dabry). The influence of starvation on nutritional conditions of the histological changes of hepatocyte and intestinal epithelium as hepatosmatic index (HSI), protein, RNA and DNA concentrations of liver in R. oxycephalus was tested. Although the starved group showed higher concentrations of protein, DNA and RNA than the fed group, food deprivation resulted in a decrease in the HSI, hepatocyte nucleus size and nuclear height of the intestinal epithelium. The RNA - DNA ratio appears to be a useful index of nutritional status in R. oxycephalus and may be useful for determining if R. oxycephalus is in a period of rapid or slow growth at the time of sampling. Additionally, the data have been interpreted in detail and some biologically important relationships discussed. The effects of starvation on the morphometrical changes in sectioned body traits, condition factor, viscera index and dressing percentage were determined for evaluating nutritional conditions of R. oxycephalus. Starvation for nine weeks resulted in a decrease in most sectioned traits as well as in condition factor and viscera index (P<0.05). These findings suggest that nutritional parameters used in this study appear to be a useful index for nutritional status in this species. The data has been interpreted in detail and some important body sectioned values of interest to commercial growers discussed. A 75-day study was conducted to determine the effect of starvation on classical and truss parameters in R. oxycephalus. Truss dimensions of almost the entire head and trunk region as well as the abdomen were increased significantly through feeding or starvation (P<0.05). Truss dimensions of the caudal region generally decreased through feeding or starvation, particularly those dimensions at the hind part of the trunk. There were some significant decreases in classical dimensions of the head region during feeding, in relation to body depth characteristics in the trunk and caudal region during starvation, whereas there was only one decreasing classical dimension in the caudal region during feeding. The results of this study indicate that application of the truss network as a character set enforces classical coverage across the body form, discrimination among experimental groups thus being enhanced. Considering that the dimension of the lower part of the head and some truss and classical dimensions were least affected by feeding and starvation, these dimensions may then be useful as a taxonomical indicator to discriminate the species of Rhynchocypris sp. The value of trunk region dimensions with a large component of body depth in R. oxycephalus is most likely to be compromised by variability related to differences in feeding regimes of fish in different habitats.
Objectives : We Investigated the effect of Dioscoreae Rhizoma(山藥) on the change of the corticosterone and the rectal temperature(直腸溫渡) of the mice induced by starvation stress(創戰 스트레스). Methods : After administration of Dioscoreae Rhizoma (0.25g/kg, 0.5g, 1.0g/kg, 3g/kg) three times, mice were starved. The corticosterone and rectal temperature were measured after 36.5 hours starvation stress. Results : The plasma cortiosterone levels in the S-2, S-3 and S-4 group were decreased significantly comparing with the control group (P<0.01) after 36.5 hours starvation stress. and rectal temperature was decreased in the control goup comparing with the normal group, but there is no significant change in the Dioscoreae Rhizoma treated group. Conclusion : it might be recognized that Dioscoreae Rhizoma has preventive-effect against starvation stress and also it might be needed further study in various viewpoints. Objectives : This study was disegned to elucidate the short term effect of Rossa rugosae Radix on proliferation. differentiation and maturation of 3T3-L1 Preadipocyte. Methods: 3T3-L1 preadipocytes obtained from Korean Cell Line Bank were cultured in a D ulbecco’ s modified eagle medium(MEM) culture solution containing 10% fetal bovine serum(FBS) and various concentrations of aqueous extract of Rossa rugosae Radix.. The short term effect of the extract of Rossa rugosae Radix on proliferation. differentiation and maturation of 3T3-L1 preadipocytes were investigate after treatment for 24 hours by measuring MTT. Oil Red 0 and latate dehydrogenase activity.. Results: The Rossa rugosae Radix extract inhibited significantly the proliferation of 3T3-L1 preadipocytes and tended to increase latate dehydrogenase activity in the media of differentiated 3T3-L1 preadipocytes & matured 3T3-L1 preadipocytes. the extract also inhibit the lipid accumulation of differentiated 3T3-L1 preadipocytes & matuered 3T3-L1 preadipocytes. Conclusions: These results demonstrated that the Rossa rugosae Radjx extract inhibited the proliferation. differentiation and maturation of 3T3-L1 preadipocytes. suggesting that Rossa rugosae Radix has anti-obesity effect: however further in vivo study is needed to demonstrate its pharmacological effects.

Two consecutive feeding trials investigated the effects of feeding rate and pellet expansion by water-soaking on the growth performance, blood components and histology of olive flounder Paralichthys olivaceus. The first two experiments were carried out to determine the effects of pellet expansion and feeding rate. In the first experiment, growth performance, feed utilization and survival of fish were not significantly affected by pellet expansion for six weeks. There were no significant differences in hematocrit, hemoglobin, aspartate aminotransferase, alanine aminotransferase, glucose and total protein of fish fed the expanded pellet. However, whole-body lipid content of fish in the non-expanded group was significantly higher than that in the expanded group. Histological analysis of the anterior intestine revealed that fish in the expanded group had shorter and smaller mucous folds. These results indicate that pellet expansion had no beneficial effect in terms of growth performance, feed utilization and fish health. In the second experiment, weight gain and feed efficiency were significantly increased as feeding rate increased from 0 to 2.5% body weight per day (BW/d), but there were no significant differences in weight gain or feeding efficiency in fish fed the expanded pellet at ratios of 2.5% BW/d and satiation for three weeks. Plasma aspartate aminotransferase activity of fish fed the expanded pellet at a ratio of 2.5% was significantly lower than that of starved fish. Histological analysis of the anterior intestine revealed that fish in the 0% group had shorter mucous folds. Broken-line regression analysis suggested that the optimum juvenile olive flounder feeding rate was 3.5% BW/d during the low temperature season ($16-17^{\circ}C$).
This study was conducted to determine the circadian rhythm and the effect of starvation for the health assessment of olive flounder Paralichthys olivaceus, reared under 12:12 LD (light:dark) cycle and fed 1% of their body weight once a day. The blood collection was implemented from 10 fish every 3 hours for a day, and analyzed to serval health parameters of the experimental fish. Plasma cortisol, glucose and total protein concentration reflected diel rhythms with high level of photophase and low level of scotophse despites of glutamic oxalacetic transaminase (GOT) and glutamic pyruvate transaminase (GPT) showing no rhythmicity, while electrolyte (sodium, potassium and chloride ion) concentrations arrived at the peak 18 hour and then slowly down. In the experiment of starvation, the result showed that there were no significant differences of the health parameters between the starved group and the fed. In short, our findings described the existence of circadian cortisol with glucose in flat fish, and indicated that the starvation does not almost affect the health of fish.
- I aimed to isolate trans-acting factors involved in the basal expression level of eukaryotic genes. One of the yeast histidine biosynthetic gene, HIS5 was taken as a model for this study. HIS5 gene has a substantial basal level in amino acid rich medium and is derepressed if starved for any single amino acid. The derepression is mediated by cis-acting DNA sequences 5'-TGACTC-3' found in 5' non-transcribed region of the gene and trans-acting factors including GCN4 as positive factor and its negative factor GCDI 7, and GCNZ as a negative factor of GCD17. I first investigated the role of these trans-acting factors in HIS5 basal expression level by using HIS5-pH05 fusion in which expression of pH05 gene encoding inorganic phosphate-repressible acid phosphatase (APase) is regulated by HIS5 promoter. Strain with gcn2 or gcn4 mutation showed 3 to 4 fold lower APase activity than wild type. The level of APase activity was similar in gcn2 and gcn4 mutants. Trans-acting factors involved in basal level were identified by isolating 14 mutants showing increased expression of HISSPH05 fusion from gcn4 background. All the mutants carry a single nuclear recessive mutation and fall into four complementation groups, designated as bell (basal expression level), be12, be23 and be14.
