Quantification of Starch Content in Apples Using Image Analysis and Its Relationships to Physicochemical Properties

  • The starch content of apples, which is related to the maturity and ripeness, was quantified by using a computer vision system. The stained area was measured by image analysis when an apple slice was stained with KI/I$_2$ solution. The ratio of stained area of the apple slice to its whole area, so called the starch index, was defined as an indicator of the starch content. When Tsugaru apples were manually sorted into immature, turing and mature groups, their starch indices were 0.374, 0.312 and 0.129, respectively. The starch index had relationships to various physicochemical properties of Tsugaru apples. At the statistically significant level of 0.1%, it was correlated with the pH value, bio-yield force, rupture force and color of intact and skin-removed apples. At the 1% significant level, it had the correlation with the density and moisture content.

  • In order to successfully meet the environmental and recycling problems, natural polymer and their derivatives are recognized as a promising biodegradable material. In this study, the biodegradable composites of cellulose diacetate and starch were prepared, and their physical and thermal properties were investigated. For the melting processing, triacetine was added as a plasticizer into the composites. The processability of cellulose diacetate was further enhanced by increasing the amount of starch in the composites. The tensile stress and Young's modulus were decreased and elongation was increased with increasing the amount of starch in them. A $T_g$ value was decreased with increasing the amount of starch in the composites. Also, the morphology of the composites were observed with the SEM.

  • Physicochemical properties of potato starches oxidized with sodium hypochlorite containing 1.5, 3.0, 4.5, and 6.0% active Cl/g starch at pH 7.0 were examined. Carboxyl group contents of oxidized potato starches were proportional to active chlorine concentration in use and water binding capacity, alkali number, and solubility increased with increasing the degree of oxidation. Blue value and iodine binding property indicated the change of amylose structure by the oxidation. Gelatinization trends obtained from transmittance and DSC thermograms showed that gelatinization temperature was lower as starch was oxidized higher. X-ray diffraction patterns and scanning electron micrographs implied oxidation might occur on the surface amorphous regions of starch granule.

Quality Characteristics of Omija Jelly Prepared with Various Starches (전분의 종류에 따른 오미자 젤리의 품질 특성연구)

  • This study was carried out to determine the effects of various starches (mungbean starch, cowpea starch and corn starch) on the quality characteristics of Omija jelly made of Omija extract. The viscosity of starch suspended in Omija extract and distilled water was measured by using a RVA(Rapid Visco Analyzer), and, color value, syneresis, texture(rupture test and TPA test) and sensory properties of Omija jelly and pure starch jelly were measured. Gelatinization temperature of each starch suspended in Omija extract was higher than that suspended in distilled water, whereas final viscosity of Omija jelly was decreased. Omija extract appeared to retard the gelatinization of starch and recrystallization of gelatinized starch. The viscosity of com starch was lowest among the three types of starch, suggesting thai higher concentration is needed in the use of com starch. The lightness(L) of corn starch gel was the highest among the gels. The syneresis of Omija jelly was lower than that of starch jelly, therefore, Omija extract seemed to be helpful on the stability of starch gel. Rupture properties of Omija jelly was lower than that of starch jelly, whereas the adhesiveness of omija jelly was greater. Omija jelly made of corn starch was less cohesive and more sticky than other gels, and its acceptability was very low. Sensory characteristics of the gel were relatively well correlated with the mechanical characteristics. Overall acceptability of Omija jelly was high in the concentration of 7, 8% of mungbean starch and 8, 9% of cowpea starch. Thus, the optimum concentration of starch for making Omija jelly using mungbean starch was 7, 8% and that using corn starch was 8, 9%.

  • The objectives of this research were to analyze physicochemical properties of physically modified rice starch which was prepared by hydro-shear homogenizer and ultrasonic homogenizer. The 5:1 of water to starch ratio, 20,500 rpm of hydro-shear homogenizer, and 5 sec of pulse and 40 kHz of frequency of ultrasonic homogenizer, which were operated for 10 min. The 1/3 of size reduction of raw rice starch and gelatinized rice starch, and reduced of particle size were achieved by above processing conditions. The homogenization of raw rice starch and gelatinized rice starch were higher values of specific area, transmittance, solubility and swelling power than control. While, these had lower value of apparent viscosity than control. In raw rice starch, the gelatinization characteristics of ultrasonic homogenizer treated rice starch had some higher values of gelatinization temperature, peak temperature and enthalpy than those of control. While, those of hydro-shear homogenizer treated rice starch had similar values to those of control.

  • It is well known that the native conformation of many proteins can be stabilized by carbohydrates or polyalcohols. However, the mechanism of the stabilization still remains unclear. In the present studies, to characterize the cryoprotective mechanism of corn starch enzyme hydrolysates on fish protin, solubility of hydrolysates, thermal behavior of hydrolysates and actomyosin solution, and enzyme kinetics in frozen system were investigated. The solubility of the hydrolysates increased with the increase in D.E. value. The $T_g^{'}$ of the hydrolysates were linearly correlated with D.E. value and the T-g value of the hydrolysates (D.E. 5,10,15,20) were reported to be $-7.2^{\circ}C\;-8.8^{\circ}C\;-11.9^{\circ}C$, and $-14.3^{\circ}C$, respectively. The results of enzyme experiments showed that the higher the D.E. value, the higher was the rate of reaction in frozen storage ($-12^{\circ}C$). It is found to support the cryostabilization mechanism that the hydrolysats act to enmesh the protein in a glass state where all deteriorative processes are greatly slowed down.

Quality Evaluation of Tangmyon Prepared from Sweet Potato and/or Corn Starches (원료 전분이 다른 당면의 품질 평가)

  • Cooking, texture and sensory properties of sweet potato Tangmyon (starch noodle), common Tangmyon (50% sweet potato starch+50% corn starch) and corn Tangmyon were evaluated. The weight gain of Tangmyon showed a linear relationship with the square root of cooking time, in which the common Tangmyon showed the highest value. The solid loss during cooking was the highest in common Tangmyon followed by corn Tangmyon. The sweet potato Tangmyon showed the highest value for compression strength and stretching ratio, but lowest value for elongation elastic modulus. At the same compression strength, corn Tangmyon had the highest tensile strength and sweet potato Tangmyon the lowest value. Sweet potato Tangmyon showed the highest sensory scores of gloss, clarity, adhesiveness, gumminess, extensibility and overall desirability, and corn Tangmyon the lowest scores. Except adhesiveness (by appearance) and the gumminess of common and corn Tangmyon, significant differences (p<0.05) were observed for other sensory properties among samples.

Granule-Bound Starch Synthase I (GBSSI): An Evolutionary Perspective and Haplotype Diversification in Rice Cultivars

  • Granule-bound starch synthase I (GBSSI), encoded by the waxy gene, is responsible for the accumulation of amylose during the development of starch granules in rice endosperm. Despite many findings on waxy alleles, the genetic diversity and evolutionary studies are still not fully explored regarding their functional effects. Comprehensive evolutionary analyses were performed to investigate the genetic variations and relatedness of the GBSSI gene in 374 rice accessions composed of 54 wild accessions and 320 bred cultivars (temperate japonica, tropical japonica, indica, aus, aromatic, and admixture). GBSS1 coding regions were analyzed from a VCF file retrieved from whole-genome resequencing data, and eight haplotypes were identified in the GBSSI coding region of 320 bred cultivars. The genetic diversity indices revealed the most negative Tajima's D value in the tropical-japonica, followed by the aus and temperate-japonica, while Tajima's D values in indica were positive, indicating balancing selection. Diversity reduction was noticed in temperate japonica (0.0003) compared to the highest one (wild, 0.0044), illustrating their higher genetic differentiation by FST-value (0.604). The most positive Tajima's D value was observed in indica (0.5224), indicating the GBSSI gene domestication signature under balancing selection. In contrast, the lowest and negative Tajima's D value was found in tropical japonica (-0.5291), which might have experienced a positive selection and purified due to the excess of rare alleles. Overall, our study offers insights into haplotype diversity and evolutionary fingerprints of GBSSI. It ako provides genomic information to increase the starch content of cooked rice.

Effect of Glucose, Starch, Sucrose on the Protein Utilization In Weanling Rats (흰쥐에 있어 탄수화물의 종류에 따른 단백질의 체내 이용에 관한 연구)

  • This study was conducted to compare effects of various types of dietary carboh ydrates fed with different levels of protein on the protein utilization in weanling rats. Sixty male Sprague-Dawley rats weighing $60{\pm}1.3grams$ were adapted for 1 week with 77% starch-15% casein diet. Then the animals divided into 12 groups according to body weight and fed each experimental diet for two weeks. Carbodydrates used were glucose, starch, and sucrose and the amount of protein given were 0g, 1g, 3g, 5g casein/day. Protein portion of the diet was fed in two seperate feedings per day while nonprotein portion was fed ad libitum. It seemed that there was no significant difference in the protein utilization by using the different kinds of carbohydrate, but in P.E.R., N.P.U., weights of organs and protein and lipid in total carcass, glucose groups were tended to be slightly lower than starch and sucrose groups. The larger the amount of casein given, the higher were the value of body weight gain, F.E.R., weights of organs, total lipid in carcass and the amount of nitrogen retention. On the while, the larger the amount of casein given, the lower were the value of the intake of non-protein portion, P.E.R., N.P.U, and the percentage of nitrogen retention.
