Effect of ABA and Kinetin on Alleviating NaCl Injury during Rice Germination (ABA와 Kinetin 처리가 벼 발아중 NaCl 독성 경감에 미치는 효과)

  • The study was carried out to determine an optimum concentration of plant growth regulators(ABA and kinetin) on reducing NaCl toxicity during germination in rice. Plant height of a japonica cultivar Ilpumbyeo in ABA 10$^{-5}$ M was increased, while all treatments of kinetin had no clear effects on increasing the plant height. However, other rice cultivars showed slightly different reaction by PGRs compared with Ilpumbyeo. Germination rate of four rice cultivars was ranged from 53.1 to 58.2% in NaCl 1.3%. All treatments of kinetin accelerated germination rate of tested rice cultivars except Dasanbyeo compared with NaCl 1.3% treatment. The higher concentration of ABA and kinetin treatment induced the higher starch content, and while the lower concentration of two plant growth regulators induced the higher sugar content in all four rice cultivars. The free proline content of rice seedlings was highest in Ilpumbyeo among the tested cultivars under NaCl 1.3% treatment.

Development of Squash Sikhye Added with Retrograded Rice Flour Containing Resistant Starch Using Response Surface Methodology and Quality Comparison with Commercial Beverages (반응표면검사를 이용한 RS함유 노화쌀가루를 첨가한 단호박식혜 개발 및 시판음료와 품질비교)

  • This research aimed to develope squash sikhye containing retrograded rice flour using response surface methodology. For this, nonlinear regression equation was calculated with the setting of independent variables as retrograded rice flour containing resistant starch and squash, and dependent variables as viscosity, sugar content, pH, color, and sensory attributes. Under the experimental condition, the quality characteristics of squash sikhye were found as 6.20~6.25 for pH, $14.00{\sim}17.33^{\circ}Brix$ for sweetness, 2.64~4.45 cP for viscosity, 57.51~60.18 for lightness, 4.94~7.52 for redness, and 50.98~60.29 for yellowness. It also was revealed overall acceptability in sensory evaluation rated as 9.67~10.83 out of 15 point. These results showed statistically significant differences in quality attributes with the increase of retrograded rice flour and squash(p<0.05). Thus, optimal mixing quantity of squash and retrograded rice flour for squash sikhye was identified as 114 g and 65 g, respectively. Comparison of sensory qualities of four samples, namely squash sikhye with and without retrograded rice, and two commercial products, showed that squash sikhye including retrograded rice flour had the best quality in terms of color, taste, mouth feeling, flavor and overall acceptability (p<0.05). Thus the squash sikhye with retrograded rice flour can be the potential products as a health benefit beverage for the old adult and the young generation.

  • To study the biochemical and physiological role of the plastidic glucose transporter (pGlcT) in carbohydrate metabolism, we characterized transgenic plants with mutations in the pGlcT gene (GT), gt-1 and gt-2, as well double mutants of GT and the maltose transporter (MEX1) and GT and the triose phosphate/phosphate translocator (TPT), GT and the cytosolic fructose-1,6-bisphosphatase gene (cFBP), and MEX1 and TPT, gt-1/mex2, gt-1/tpt-2, gt-1/cfbp-1, mex1-1/tpt-2, respectively. Compared to the wild type, all mutants except the gt-1/cfbp-1 mutant lines displayed higher starch accumulation and higher levels of maltose. Starch accumulation is due to a decrease in starch turnover, leading to an imbalance between the rates of synthesis and degradation. Sucrose levels of gt alleles were higher than those in wild-type plants during the light period, suggesting possible nightly supplementation via the maltose transport pathway to maintain proper carbohydrate partitioning in the plant leaves. The gt plants displayed less growth retardation than mex1-1 mutant and gt-1/mex2 double mutant displayed accumulativesevere growth retardation as compared to individual gt-1 and mex1-1 mutants, implying that the maltose transporter-mediated pathway is a major route for carbohydrate partitioning at night. The gt-1/tpt-2, mex1-1/tpt-2 and gt-1/cfbp-1 double mutants had retarded growth and low chlorophyll content to differing degrees, indicating that photosynthetic capacity had diminished. Interestingly, the gt-1/tpt-2 line displayed a glucose-insensitive phenotype and higher germination rates than wild type, suggesting its involvement not only in carbon partitioning, but also in the sugar signaling network of the pGlcT and TPT.

  • Changes of chemical componts and populations of total bacteria and lactic acid bacteria were examined during a month-long storage of sweet potatoes crushed and sealed up with polyethylene film at $7-8^{\circ}C$. 1. In a three days, pH down and increase of non-volatile acid content were notable, populations of total bacteria and lactic acid bacteria were maximum. 3. Vitamic C and soluble sugar contents tended to be reduced during the storage and the leftover were 75-85% and 41-45% respectively. 4. ${\beta}$-Amylase activity decreased gradually and vanished 15-30days. 5. Variations of chemical compontents of the samples inoculated lactic acid bacteria were larger than non- treated to some extent. 6. As above results, mainly homo lactic acid fermentation was done in this storage condition.

  • To produce liquor and vinegar using potatoes needs to liquefy and sacchrify potatoes . So selecting the efficient fermenter for proceeding these process successfully is very important . This study was investigated several fermenter and crush types of potatoes for alcohol fermentation. Final sugar contents was high in pottoes saccharificatiion by nuruk or crude enzyme. But pure enzyme and blucoamylase ended liquefaction and saccharificatiion within short ime. So complex type fermenter mixed several fermenters was superior to single type fermenter. Complexfermenter III using crude enzyme and glucoamyulase saccharificed excellently potatoes with 150% of water contents by treatment of 3 hours. Through alcohol fermentation using pressure steamed potatoes (PSP), it could be obtained 6.4% , 150%, of alcohol content and yield. However to perform a series process efficiently , crush steamed pottoes (CSP) was suitable. When it was fermented after saccharification using crush steamed potatoes and complex fermenter III, it could be obtained 6.6% of alcohol and 6.7% of acidity.

  • Mixed feeds and their components are a very diverse matrix compared to other agricultural products worked on with NIRS classically. On a database of mixed feeds and their components (n=2.500) universal PLS calibrations and “local” calibrations were compared. The results from validation (n=600) show the potential of the calibrations and their limitations. Crude protein, crude fiber, crude fat, sugar and starch are predicted with SEPs of 0.6%, 1%, 0.3%, 1% and 1.5%, respectively. Ash content of 15% and more in several mixed feeds or components as well as rare components limit the use of NIRS for routine analyses.

  • Kojis were prepared from the different starch raw materials such as glutinous rice, barley rice, wheat flour and sweet potato, and were tested in an effort to reduce production cost and to improve quality of Kochuzang(red pepper paste). During the starting period of Koji preparation, there were less significant changes in pH; however, pH decreased somehow in the case of sweet potato, whereas it increased for the other Kojis. In general, the highest acidity was obtained after 72 hours of Koji preparation. Total nitrogen, soluble nitrogen and amino-nitrogen content increased in order of wheat flour, barley rice, and glutinous rice; Kojis they were markedly produced between 72 and 96 hours of Koji preparation. The maximum amount of reducing sugar was observed between 48 and 72 hours of Koji preparation during this period the reducing sugar content varied widely depending on starch source. Sweet potato Koji produced the highest level of ethyl alcohol content after 72 hours of Koji preparation; for the other Koji the same trends were observed after 24 hours. The starch liquefying activities have reached the highest level after 96 hours and for glutinous rice and barley rice; however, they kept on increasing until 120 hours for wheat flour and sweet potato. Koji Starch liquefying and saccharogenic amylase activities tended to increase in order of sweet potato, glutinous rice, barley rice and wheat flour. Kojis Various protease activities were measu-red during the Koji preparation, and they increased in order of alkali, neutral and acidic protease.

  • This study was conducted to investigate the kernel characteristics of sweet corn during ripening with different sowing date. Godangok and Guseulok were sown at April 10, July 10, July 20, and July 30 in 2013. Irrespective of sowing date, fresh and dry weight of 100 kernel, starch content, crude fat content, and hardness of sweet corn kernels were significantly increased according to maturation (p<0.05). But moisture content, total sugar content, crude protein content and crude ash content of sweet corn kernels were significantly decreased according to maturation (p<0.05). The changing rates of 100 kernel weight and starch content on kernels were faster at early sowing date than at late sowing date. The moisture content of sweet corn kernels was 68~69% at optimum for harvesting. The degree of sweetness on Godangok was about 1.8 and 1.4 times higher in sowing at July 20 and July 30 than in sowing at April 10. On the other hand, the degree of sweetness was similar regardless of sowing time on Guseulok. These results will be helpful to farmers for fall-harvest sweet corn cultivation and management.

  • This experiment was carried out to find the growth response and changes of nitrate and soluble sugar content in tomato leaves with salt stress. Tomato (Solanum lycopericum) seedlings were grown under different electrical conductivity (EC) levels adjusted with $CaCl_2$ as 1, 2, and $6dS\;m^{-1}$. The growth response and contents of nitrate and soluble sugar in tomato plants were examined at 7 and 14 days after salt treatment. Leaf area and dry weight ratio of shoot to root of tomato plants were decreased as EC level increased. Photosynthetic rate of leaves was reduced under high EC level due to the stomatal closure and the reduction of transpiration rate. The soluble sugar and starch content were lower in the tomato leaves grown under high EC level. Total nitrogen and nitrate contents were decreased in high EC level, whereas the ammonium content was increased. High-salt stress induced the accumulation of salt crystal in mesophyll cells of tomato leaf.

Studies on the Change of Sugars in Sweet Potatoes on Heating (가열(加熱)에 의한 고구마의 당(糖)의 변화(變化))

  • A study was conducted on the conversion of starch in sweet potatoes to sugar by the amylolytic enzymes during baking. Sugars were extracted with ethanol from the raw and baked sweet potatoes at the temperature of $55{\sim}57$, $70{\sim}75$, and $90{\sim}95^{\circ}C$. The individual sugars in the extracts was identified by thin-layer chromatography- and the individual sugar content was determined by high -performance liquid chromatographic analysis. Sugars identified from the raw and baked sweet potatoes at the temperature of $55{\sim}57^{\circ}C$ are glucose, fructose, and sucrose. Sucrose, maltose, glucose, and fructose were identified is the baked sweet potatoes at tile temperature of $70{\sim}75$ and $90{\sim}95^{\circ}C$. There was no significant increase in glucose, fructose and sucrose content during baking. Maltose was formed only above the gelatinization temperature.
