Comparative Genomic Analysis of Food-Originated Coagulase-Negative Staphylococcus: Analysis of Conserved Core Genes and Diversity of the Pan-Genome

  • To shed light on the genetic differences among food-originated coagulase-negative Staphylococcus (CNS), we performed pan-genome analysis of five species: Staphylococcus carnosus (two strains), Staphylococcus equorum (two strains), Staphylococcus succinus (three strains), Staphylococcus xylosus (two strains), and Staphylococcus saprophyticus (one strain). The pan-genome size increases with each new strain and currently holds about 4,500 genes from 10 genomes. Specific genes were shown to be strain dependent but not species dependent. Most specific genes were of unknown function or encoded restriction-modification enzymes, transposases, or prophages. Our results indicate that unique genes have been acquired or lost by convergent evolution within individual strains.

Comparison of fatty acid composition of Staphylococcus sp isolated from bovine mastitis milk (유방염 감염 우유에서 분리된 Staphylococcus sp의 지방산 조성 비교)

  • The result of API staph-ident system was compared with cellular fatty acid composition for the identification of Staphylococcus species isolated from cattle. Isolated strains from cattle were correctly identified to S aureus, S intermedius, S hyicus, S simulans, S saprophyticus, S epidemis, S sciuri and S xylosus by API staph-ident system. The correlation between bacterial cellular fatty acid profile and Staphylococcus species isolated to API STAPH-IDENT system were. In conclusion, the result presented indicate that Staphylococci can be indentified to the species level by the cellular fatty acid profiles. Moreover, computerized fatty acid profile correlative anaylsis can be applied for determining identify of Staphylococcus species.

  • Between June and August 1990, the tonsils of 86 healthy pigs were examined for the presence of staphylococci. All of the pigs examined harboured Staphylococci in the tonsils, the most predominant Staphylococcus species was Staphylococcus aureus(45.3%) followed by Staph hyicus subsp chromogenes (20.9%), Staph hyicus subsp hyicus (16.3%), Staph hominis(4.7%), Staph simulans(2.3%) and Staph xylosus(1.2%), Unidentifiable species were isolated from 3(3.5%) of the 86 tonsils examined. Thirty-nine strains of Staph aureus were subjected to the biotyping scheme of Hajek & Marsalek all the strains were classified as biotype B.

Diagnosis of Subclinical Mastitis-Causing Pathogens Using MALDI-TOF Mass Spectrometry in a Certified Organic Dairy Farm in Korea

  • We identified mastitis-causing pathogens using matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) in an organic dairy farm and evaluated the effects of antimicrobial restriction on antimicrobial susceptibility. A total of 43 Holstein cows without any clinical sign of mastitis were used in this study, and 172 quarter milk samples were cultured on blood agar plates for 24 hours at 37℃. Subsequently, bacterial species were identified and antimicrobial susceptibility tests were performed. The subclinical mastitis infection rates in the cows and quarters were 58.1% (25/43) and 25.6% (44/172), respectively. In the species identification, Staphylococcus aureus (40.9%) was the most prominent isolate, followed by S. chromogenes (22.7%), S. epidermis (18.2%), S. simulans (11.4%), S. haemolyticus (2.3%), S. muscae (2.3%), and S. xylosus (2.3%). In the antimicrobial susceptibility test, all isolates were 100% susceptible to 24 of 28 antibiotics, except for benzylpenicillin, cefalotin, cefpodoxime, and trimethoprim/sulfamethoxazole. The resistance rates of S. aureus, S. chromogenes, and S. muscae isolates to trimethoprim/sulfamethoxazole were 27.8%, 10%, and 100%, respectively, and the resistance rates of S. epidermis and S. xylosus to benzylpenicillin were 50% and 100%, respectively. S. chromogenes, S. epidermis, S. simulans, S. haemolyticus, and S. xylosus were resistant to cefalotin and cefpodoxime. In conclusion, restrictions on antimicrobial use for organic dairy farm certification have resulted in a high Staphylococcus spp. infection rate. Therefore, our study indicates the importance of mastitis management strategies implemented by farmers together with veterinary practitioners, even if mastitis does not appear clinically in organic dairy farms.

Enterobacteriaceae and Related Microorganisms Isolated from Rump of Raw Beefs

  • In this study, 50 rump samples of raw beef obtained from local Korean supermarkets were analyzed to survey microbial distributions. As results, mesophilic microorganisms ranged from $(1.4{\pm}0.01){\times}10^2$ to $(1.6{\pm}0.05){\times}10^5\;CFU/g$, and total coliforms ranged from 0 to $(1.3{\pm}0.04){\times}10^4\;CFU/g$. Major foodborne pathogens, including Listeria monocytogenes, Escherichia coli O157:H7, and Salmonella spp., were not found among the samples. However, Staphylococcus aureus was isolated with 4% frequency. Other isolated microorganisms included Enterobacter amnigenus (4%), Enterobacter cloacae (24%), E. coli (24%), Listeria innocua (8%), Staphylococcus saprophyticus (56%), Staphylococcus xylosus (10%), and Staphylococcus warneri (8%).

  • In order to develop effective manufacturing method and to improve quality of low-salt fermented squid(10% of table salt), we investigated the effects of temperature, salinity and pH on the growth of Staphylococcus xylosus, Micrococcus varians, Pseudomonas diminuta and Pseudomonas D2 isolated from of low-salt fermented squid and the growth characteristics of these bacteria during fermentation were elucidated. All bacteria showed good growth during the process of low-salt fermented squid(pH 6~7 ; concentration of NaCl, 7~10% ; temperature, 7~1$0^{\circ}C$) and their cell numbers increased as fermentation proceeded under the same fermentation condition.

Characteristics of digestive enzyme activity, antibiotic resistance, and pathogenicity of bacteria inhabited in animal feed resources (사료자원에 서식하는 세균의 소화효소활성, 항생제내성 및 병원성에 관한 특성)

  • Among different types of spoilage, microbial contamination can cause feed decomposition, which results in decreases in feed intake and productivity, infection, and breeding disorder. During the storage time, various microbes have a chance to inoculate with depreciation of feed and to infect the animals. We investigated bacteria that inhabit diverse feed ingredients and complete feed which have been stored for a few months. We isolated and identified 30 genera and 62 species of bacteria. Among these 62 species, 21 species were of non-pathogenic bacteria, 18 species were of pathogenic bacteria, 9 species were of opportunistic pathogens, and 14 species were of unknown bacteria. Pantoea allii and 24 species showed proteolytic enzyme activity. We also confirmed that 6 species including Pseudomonas psychrotolerans showed ${\alpha}$-amylase activity, and 29 species including Burkholderia vietnamiensis showed cellulase activity. Microbacterium testaceum and 3 species showed resistance to Ampicillin, Kanamycin, Streptomycin, Gentamicin, Carbenicillin, and Erythromycin ($50{\mu}g/mL$). Using mealworm larvae (Tenebrio molitor L.) as a model for pathogenicity, we confirmed that 8 species including Staphylococcus xylosus had pathogenicity for mealworm larvae. Especially, Enterobacter hormaechei, Staphylococcus xylosus, and Staphylococcus hominis were reported as being pathogenic for humans. This research suggests that hygienic management of animal feed is essential because beneficial and harmful bacteria can inhabit animal feed differently during storage and distribution.

  • Of 111 bacterial isolates on 32 dried salted marine products in Garak wholesale market.72 strains were coliforms, and 12 were staphylococcus spp. etc. In average, humidity 17.28%, ashes 4.92%, standard plate count $1.1{\times}10^7$ CFU/ml, coliforms count $9.0{\times}10^3$ CFU/ml, staphylococcus spp. $5.4{\times}10^6$ CFU/ml respectively. Of 72 coliforms, it was isolated by 21 Pantoea spp, 14 Enterobacter cloacae and, 13 Ranella aquatilis, etc. and of 12 Staphylococcus spp. 4 staphylococcus xylosus was the most.

Production of staphylokinase in Staphylococcus hyicus subsp. hyicus strains of swine, poultry and bovine origin (돼지, 닭 및 소유래 Staphylococcus hyicus subsp. hyicus의 staphylokinase 산생능)

  • Staphylococcus hyicus subsp. hyicus strains isolated from pigs, chickens and cattle were examined for the production of staphylokinase after inhibition of staphylococcal proteases by two procedures with EDTA(disodium). In one, EDTA was added to the bovine fibrin-dog plasminogen agar medium in concentration of 0.07% and paper strips soaked in 2mg/ml soy bean trypsin inhibitor were then applied on the agar plates. In the other, paper strips soaked in 5% EDTA solution were applied on the bovine fibrin-dog plasminogen agar plates and the strains to be tested were then streaked at right angles with the strip. By these procedures, staphylokinase activity was detected in 8(88.9%) of 9 strains from diseased pigs and in 57(80.3%) of 71 strains from skin of healthy pigs, but not in any strains from skin of healthy chickens and milk samples of mastitic cattle. Additionally kinase activity in 9 Staphylococcus species and subspecies isolated from bovine intramammary infections was also tested by these procedures. Staphylokinase activity was detected in 74.2% of Staph aureus strains and in 25% of Staph xylosus strains.

  • The in vitro activity of penicillins(penicillin G, ampicillin, carbenicillin, methicillin and cloxacillin) and gentamicin to Staphylococcus aureus and coagulase-negative staphylococci isolated from bovine mastitic milk samples was determined. The growth of all isolates of staphylococci tested was inhibited by cloxacillin and methicillin at a concentration of $0.78{\mu}g/ml$ and by gentamicin at a minimum inhibitory concentration (MIC) of $1.56{\mu}g/ml$. Of the 140 strains of Staphylococcus aureus isolated 99(70.7%) gave positive reactions for penicillinase on the starch-iodine test. Of 121 isolates of coagulase-negative staphylococci, 58(47.9%) showed penicillinase production, but all the Staph. xylosus lacked the ability to produce penicillinase. MIC of penicillin G of the penicillinase-positive strains was $0.2{\mu}g/ml$ or more. The combination of cloxacillin with gentamicin showed a synergistic effect by inhibiting regrowth of the tested organism.
