Isolation and Identification of Pathogenic Bacteria from Spinach (시금치로부터 병원성세균의 분리 및 동정)

  • Raw and washed spinaches were tested to evaluate the incidences of Aeromonas hydrophila, Escherichia coli O157:H7, Plesiomonas shigelloides, Pseudomonas aeruginosa, Salmonella spp., Shigella spp., Yersinia enterocolitica, Bacillus cereus, Campylobacter jejuni, Clostridium perfringens, Listeria monocytogenes, and Staphylococcus aureus. Four pathogenic bacteria were isolated from spinach samples, and identified by morphological and biochemical methods, including API and ATB identification systems. Isolates from MacConkey, Cereus Selective, Clostridium Perfringens, and Baird-Parker agar media were in 99.9, 99.8, 99.9, and 97.8% agreements with A. hydrophila, B. cereus, C. perfringens, and S. aureus at the species level, respectively. SET-RPLA revealed, among the five strains of S. aureus isolates, two produced type A enterotoxin. All five strains of B. cereus isolates produced enterotoxin as revealed with CRET-RPLA.

Antimicrobial Effects of Herbal Medicine Extracts on Staphylococcus aureus and Escherichia coli O157:H7 (생약재 추출물의 Staphylococcus aureus 와 Escherichia coli O157:H7에 대한 항균효과)

  • To screening of antimicrobial activity, 95% ethanol and hot water extracts of roots, fruits, leaves, radix and stems of 50 species of traditional herbal medicines were examined. For their growth inhibitory effects on two food-born microorganisms, S. aureus and E. coli O157:H7, by the paper disc diffusion method and the minimum inhibitory concentration(MIC) test. Moutan radicis Cortex and Achyranthis Radix showed the highest inhibitory activities on both S. aureus and E. coli O157:H7. The Inhibition zones of Moutan radicis Cortex on S. aureus and E. coii O157:H7 were 22 mm and 24 mm respectively, and the corresponding inhibition zone of Achyranthis Radix were 23 mm and 22 mm. The MIC or Achyranthis Radix on S. aureus was 156.25 $\mu$g/mL, and the MIC or Achyranthis Radix and Moutan radicis Cortexas on E. coli O157:H7 were 625 $\mu$g/mL and 312.5 $\mu$g/mL, respectively. Their antimicrobial activities in ethanolic extracts were significantly higher than in hot water extracts. In the various solvent fractions prepared from ethanol extract, the ethyl acetate fraction of Achyranthis Radix and the CHCl$_3$ fraction of Moutan radicis Cortexas showed strongest activity.

Investigation of Microbial Contamination the Level in Fish Farms of Jeju East Coast. (제주 동부 연안 넙치 양식장의 미생물학적 오염도 조사)

  • Level of microbial contamination was examined in four fish farms of Jeju east coast for sanitary indication of bacterial contamination such as heterotrophic bacteria, Escherichia coli O157:H7, Salmonella spp., Vibrio parahaemolyticus, Staphylococcus aureus, Bacillus cereus and total coliforms. The samples were collected and investigated from June to October in 2007. Bacterial species of S. aureus, B. cereus and Salmonella spp. were frequently detected from the seawater of above fish farms. Salmonella spp., S. aureus, B. cereus were detected in the range of $0{\sim}4.3{\times}l0^3,\;0{\sim}2.7{\times}l0^4,\;0{\sim}7.6{\times}l0^3$ CFU/ml, respectively, from the fish feed used in all four selected farms. Additionally, Salmonella spp., S. aureus, B. cereus were detected in oliver flounder (Paralichthys olivaceus) of these farms, however total coliforms and E. coli O157:H7 were not detected during our experimental period. For the production of microbiologically safe and healthy oliver flounder, proper quality control of feed, sanitation programs, and continuous monitoring of microorganism are essential practices, which required to include in the farm management system.

Antimicrobial Effects of Camellia Japonica L. Leaves Extract on Food-borne Pathogenic Microorganisms (동백나무(Camellia japonica L.) 잎 추출물이 식품유해 미생물에 미치는 항균 효과)

  • Antimicrobial effects of Camellia japonica L. were determined against Escherichia coli, Salmonella typhimurium, Staphylococcus aureus, and Listeria monocytogenes using paper disc method, and minimum inhibitory concentrations (MICs) were measured. Methanol extract (MEex), water fraction (WAfr), and butanol fraction(BUfr) showed antimicrobial effects against all tested microorganisms, with MEex showing strong antimicrobial effect against S. aureus and L. monocytogenes, and WAfr, Bufr, and ethylacetate fraction (EAfr)against S. aureus. No effects were observed in n-hexane fraction (HEfr) and chloroform fraction (CHfr) against all tested microorganisms. All species grown in the medium adding fractions of Camellia Japonica L. leaves extact were inhibited from WAfr and BUfr, repectively.(meaning not clear) MEex showed over 25% inhibitory effect against all tested microorganisms. BUfr showed over 50% inhibitory effect against all microorganisms except L. monocytogenes. EAfr and WAfr showed over 30% effect against S. aureus and L. monocytogenes. MICs of MEex against S. typhimurium and BUfr against S. aureus were 625 g/mL, indicating C. japonica L. extract can exert antimicrobial activity even at low concentration.

Selection of Lactobacillus Species Inhibiting Enteropathogenic Bacteria and Potential Use as Probiotics (장내세균의 발육억제 효과로 정한 Lactobacillus 균주 선정과 Probiotic으로서의 이용가능성)

  • 목적 : 본 연구에서는 한국인 소아의 침 및 성인의 질부로부터 분리한 1367개의 유산 생성 균주 중 4가지의 장내 병원성 세균;Methicillin-resistant Staphylococcus aureus(MRSA), Listeria monocytogenes, Salmonella typhimurium, Escherichia coli O157에 대해 발육억제 효과를 나타내는 유산 균주를 분리하고 분리된 균주의 Probiotic으로서의 가능성을 조사하고자 하였다. 방법 : 네 가지 병원성 세균을 대상으로 agar spot test와 Catalse test를 시행하여 1367개 균주 중 최종적인 실험대상 균주를 선정하였다. 최종 선정된 유산균과 네 가지 병원성 세균을 혼합 배양하여 병원성 세균증식 억제 효과를 보았다. 또한 최종균주의 probiotic으로서의 가능성을 확인하기 위해 균을 동정하고 내산성 등 몇 가지 특성을 관찰하였다. 결과 : Agar spot test 결과 전체의 15.7%에 해당하는 215 균주가 MRSA에 대해 억제 효과를 나타내었으며 215 균주 중 9.8%에 해당하는 21 균주가 L. monocytogenes에도 동시에 억제효과를 나타냈다. 이들 균주 중 가장 탁월한 효과를 보인 Lb 1250을 실험 대상 균주로 선정하였다. Lb(Lactobacillus) 1250과 상술한 병원성 세균을 각각 혼합 배양하면 Lb 1250 존재 하에서 MRSA의 증식은 $10^4$배 억제되었으며, L. monocytogenes는 $10^3$배의 억제 효과를 보였다. 그러나 S. typhimurium, E. coli O157에 대해서는 아주 미약한 효과를 나타냈다. Lb 1250의 몇 가지 특성을 조사한 결과 Lactobacillus delbrueckii subsp. delbrueckii로 동정되었으며, 이는 $H_{2}O_2$를 잘 생산해 내는 균주이었다. 또한 이 균주는 우유에서 curd를 약하게 생성하며 이 curd의 향은 달콤하고 최종 pH가 4.6까지 감소하였으며, pH 2와 같은 강산의 조건하에서 배양한 결과 2시간 후에도 상당한 생존도를 유지하였다. 결론 : 이상의 결과에서 Lb 1250은 병원성 세균의 성장을 억제함을 관찰하였고, probiotic으로서의 이용가능성을 확인하였다.

Chemical Composition and Biological Activities of Essential Oil from 'Setoka' Branches (세토카 가지 정유의 성분 분석 및 생리 활성)

  • This study was designed to analyze the chemical compositions of 'Setoka' branch essential oils (SEBO) and to test their biological activities. 'Setoka' is a Citrus species widely cultivated in Jeju Island. At the present, 'Setoka' branches produced by thinning process were mostly discarded as a waste. Therefore, utilization of this branch waste has received much attention. 'Setoka' branch essential oils (SBEO) were prepared by treatment of its ethanol extracts with jojoba oil. SBEO were chemically analyzed using gas chromatograph-mass spectrometry (GC-MS), and following components were identified; ethyl linoleate (64.14%), ethyl palmitate (16.50%), neophytadiene (11.06%) and beta-citronellol (5.09%). The anti-inflammatory activity in the SBEO was examined using RAW 264.7 murine macrophage cells stimulated with LPS. As a result, the SBEO inhibited nitric oxide (NO) productions with a dose-dependent manner. In addition, SBEO showed good anti-microbial activities against drug-susceptible and -resistant skin pathogens such as Staphylococcus aureus and Propionibacterium acnes, which are acne-causing bacteria. Based on these results, we suggest that SBEO has the possibility for use as an anti-inflammatory and anti-microbial agent in cosmetic applications.

Effect of metals(Cd & Zn) and its nitrogen compounds on growth of bacteria isolated from the three tributaries and lower reach(Mulgum) of the Nakdong River (낙동강 하류(물금)와 세 지류에서 분리된 우점 세균의 증식에 미치는 금속(Cd & Zn) 및 그 질소 화합물의 영향)

  • Investigation was carried out to observe the dominant bacteria and the effect of metals(Cd & Zn) and its nitrate compound on growth of bacteria isolated from the three tributaries and lower reach of the Nakdong River. Mean CFU(log$_{10}$) level was highest in Kumho River(8.30 CFU), Nam River, Hwang River, and Mulgum followed. Staphylococcus xylosus, Staph. lentus, Pasteurella pneumotropica, Aeromonas hydrophilla were dominant species in each study site. Cadmium powder and Zinc powder showed strong effect to inhibit the growth of Micrococcus spp., Pasteurella pneumotropica, Aeromonas hydrophilla. But, nitrate compounds of Cd and Zn(Cd(NO$_3$)$_2$4$H_2O$, Zn(NO$_3$)$_2$6$H_2O$) did not clearly show the strong effect to inhibit the growth of dominants.s.

Antimicrobial Activity against Respiratory Bacteria by Asparagus Cochinchinensis Extracts and its Antioxidant Capacity (천문동 추출물의 호흡기 세균에 대한 항균활성 및 항산화)

  • This study was aimed at determining the antioxidant and antimicrobial effects of solvent extracts from Asparagus cochinchinensis. The Asparagus cochinchinensis was extracted with water, methanol, ethanol, n-hexane, dichloromethane, ethyl acetate, and ether. The antimicrobial activity of these extracts was determined by modified well diffusion methods against 4 species of respiratory disease bacteria (Staphylococcus aureus, Escherichia coli, Staphylococcus epidermidis, and Pseudomonas aeruginosa). In addition, the amount of total polyphenol and flavonoid content, and antioxidant activity was evaluated. Ethyl acetate extract of A. cochinchinensis exhibited higher antimicrobial activity against tested microorganisms than water, methanol, ethanol, n-hexane, dichloromethane, and ether extracts. For antioxidant activity, the ethyl acetate extract of A. cochinchinensis exhibited a notable effect on the scavenging of superoxide against DPPH ($IC_{50}=3.81mg/ml$). Finally, the total polyphenol and flavonoid contents were $14{\pm}0.7mg/g$, and $0.50{\pm}0.13mg/g$, respectively. These results can be regarded as basic research into A. cochinchinensis for the prevention of respiratory diseases. The results indicate that A. cochinchinensis may be utilized as a nutraceutical for respiratory diseases when the physiologically active substances of A. cochinchinensis are increased by further study.

Screening of Domestic Plants with Antibacterial Activity (국내 자생식물의 항균활성)

  • To select new useful plants with antibacterial activity, ninety five sample of eighty different species of wild plants were collected, and extracted with methanol. Antibacterial activity of the methanol extracts was tested against Bacillus subtilis, Staphylococcus aureus, Escherichia coli and Vibrio parahaemolyticus. The methanol extracts from Artemisia capillaris, Hemistepta lyrata, Youngia japonica, Prunella vulgaris, Lamium amplexicaule and Juniperus chinensis was effective against all bacterial strains tested, and eight methanol extracts including Ixeris dentata, Gnaphalium affine, Chelidonium majus and Spiraea prunifolia exhibited the antibacterial activity against at least 3 bacterial strains. Methanol extracts from leaf of Syringa vulgaris, Drava nemorosa and clove of Erythronium japonicum showed a selective antibacterial activity against two gram negative bacteria, V. parahaemolyticus, and B. subtilis, respectively. With investigations on antibacterial activity against a certain bacterial strains tested, metahnol extracts from clove of Erythronium japonicum, Spiraea prunifolia, leaf and twig of Camelia japonica, and Drava nemorosa showed strongest activities against B. subtilis, S. aureus, E. coli, and V. parahaemolyticus, respectively. Nine methanol extracts based on the results were successively fractionated with n-hexane, chloroform, ethyl acetate and water portions, which were examined antibacterial activity against B. subtilis and V. parahaemolyticus. Among the all fractions tested, chloroform fractions of Hemistepta lyrata showed strongest antibacterial activity against both B. subtilis (17mm) and V. parahaemolyticus (29 mm). Chloroform fractions of Youngia japonica, n-hexane fractions of Artemisia capillaris, Iexeris dentata and Prunella vulgaris, and ethyl acetate fraction of leaf and twig of Camelia japonica showed relatively a strong antibacterial activity. On the other hand, Juniperus chinensis and Equisetum arvense was distributed to all fractions except for water fraction.

A Study on Microbial Pollution of Indoor Air at Elderly Care Facilities (노인요양시설의 실내공기 중 미생물 오염에 관한 연구)

  • Culture was performed by using Sheep Blood Agar Plate (BAP, Asan Pharmaceutical) and Sabouraud Dextrose Ager (SDA, Asan Pharmaceutical) along with air $IDEAL^{TM}$ (Biomerieux), which is a microbe interceptor based on inertial impaction interception, in order to investigate bioaerosol in indoor and outdoor air at five elderly care facilities in a metropolis and an urban-rural consolidated city for two months from April 1 to May 31, 2007. From the culture followed by isolation and identification, the following conclusions were drawn. 1. As for the general isolation of microbes in each facility, care center S had the largest amount of microbes (263 cfu/$m^3$) isolated in a 300L room, followed by care center U having 123 cfu/$m^3$ isolated. 2. As for the number of bacteria isolated from a medium intercepting 300 L indoor, the largest amount of other unidentified or non-pathogenic Gram positive cocci (321 cfu/$m^3$) was isolated and most of the other Gram positive cocci were CNS (Coagulase Negative Staphylococcus). 3. As for the number of fungi isolated from a medium intercepting 300 L in a room, the largest number of Aspergillus spp. (66) was isolated, followed by Mucor spp. (62 cfu/$m^3$), Penicillium spp. (53 cfu/$m^3$), Alternaria spp. (50), and other unidentified or non-pathogenic fungi (42 cfu/$m^3$). 4. As for the rate of indoor and outdoor pollution, the average number of interceptions was all larger indoor than outdoor; the research differentiating the amount of air into 300 L and 500 L demonstrated that the larger amount of air led to more bacteria, making no great variation in the species.
