Spongy Alphonso mangoes were found to be infected with Staphylococcus bacteria. A Gram positive Staphylococcus strain was isolated from spongy pulp and identified from CABI Bioscience, UK, by partial 16S rDNA sequence analysis and by morphological and biochemical characterization through IMTECH, Chandigarh, India. Although identification by both of these methods indicated the organism belonged to same genus, different species names were given. Changes in total phenolics, reducing, and non-reducing sugars, respiration rate, total carotenoids, peroxidase(POX), and catalase activities were monitored during ripening of these fruits. The climacteric rise in spongy fruits was marked by an increase in respiration rate and a decrease in sugar content. Total phenolics content increased in spongy fruits as compared to ripe non-spongy fruits. Development of corky white tissue in spongy fruits was associated with about a 2.5-fold reduction in total carotenoids and a concomitant increase in lipoxygenase-mediated, $\beta$-carotene co-oxidation. A marked decrease in soluble protein content and about a 1.5-fold increase in POX activity was observed. Maximum POX activity was confined to 50-70%$(NH_4)_2SO_4$ fraction. The intense dark bands visible after POX specific substrate staining of the Native gel indicated a high expression of isoenzymes of POX in spongy fruits. Similarly, changes in levels of catalase activity were also observed in spongy fruits. The results suggest that infection of Alphonso mangoes with Staphylococcus bacteria affects the normal ripening processes of the fruit interfering with the carbohydrate and carotenoid metabolism. Also, the studies indicate the expression of POX and catalase enzymes as a plant defense response to microbial invasion.
애완견의 털에 부착되어 있는 세균을 분리 동정한 후, 이 분리 세균들에 대한 항세균효과가 있는 생약재 추출물을 탐색하였다. 애완견의 털에서 76개의 세균을 분리한 결과, Staphylococcus속이 41균주로 가장 많았으며 그 다음으로 Micrococcus속이 21균 주, Enterococcus속이 8균주, Bacillus속이 3균주, Exiguobacterium속 of 2균주, Shigella속과 Zoogloea속이 각각 1균주씩으로 동정되었다. 이러한 결과는 개의 털에 존재하는 세균들이 사람에게 감염을 일으킬 수 있는 원인이 될 수 있음을 시사한다. 분리균주들에 대한 항생제 저항성과 생약재 추출물의 항세균 활성은 디스크 확산법으로 확인하였다. 황금의 추출물이 애완견의 털로부터 분리한 세균들에 대해 높은 항세균 활성을 나타내어 항생제 저항성이 있는 세균들에 대한 새로운 항미생물제로 사용학 수 있을 것으로 기대된다.
솔잎착즙액(PNE)은 엽록소 함유량(40.8 mg/100 g)이 높고 비타민 C도 다량함유(149 mg/100 g)하고 있어 기능성 식품으로 개발가능성이 높으며, 솔잎 발효액은 향미가 좋아지고 기능성이 높아져서 고부가가치가 크므로 발효의 조건과 활성도를 조사 분석하여 고기능성 솔잎발효액(SFPE)을 개발하고자 하였다. 솔잎자체발효(SFPE)에 관여하는 균을 발효단계에 따라 분리, 선발하고 동정하였다. GPYA배지에서 발효균을 스크린하여 Pichia galeiformis, Candida boidinii, Pichia species, Candida species1, Candida species2, Candida ooidensis, Saccharomyces cerevisiae, Candida karawaiewii를 선발하였으며, 발효가 진행됨에 따라 2년차에는 Candida boidinii, Candida species2, Candida ooidensis, Saccharomyces cerevisiae가 선발되었고, 3년차에는 Candida species1&2가 선발되어 Candida species2가 우점종으로 나타났다. 그러나 발효 4년차에는 어떤 균도 선발되지 않았다. Candida species2는 Candida ernobii와 99.65%의 homology를 보였다. E. coli, Bacillus subtilis와 Staphylococcus aureus에 대해서 SFPE를 농도별로 처리하였을 때 높은 항균활성을 확인하였다. SFPE 1, 4, 7의 항산화 활성은 $0.025{\mu}{\ell}/ml$의 농도로 처리하였을 경우 SFPE 1, 4, 7이 약 34.8%의 NBT scavenging의 활성을 보여주었고, $0.2{\mu}{\ell}/ml{\sim}0.3{\mu}{\ell}/ml$의 농도에서는 SFPE 1, 4, 7이 약 90%에 달하는 NBT scavenging 의 활성능력을 확인할 수 있었다. 또한 전기생리학적 실험을 통하여 솔잎발효액이 쥐의 혈관수축 이완에 미치는 효과를 분석한 결과 Phenylephrine(PE)($10^6M$)으로 유발된 수축반응에 대해 SFPE를 0.15 mg/ml과 0.3 mg/ml을 첨가하였을 때 혈관이 다시 이완되었으며, ATP 민감성 $K^+$ 채널 억제제인 glibenclamide($10^5M$)을 처리하였을 때 SFPE 7 효과가 나타나지 않은 것으로 보아 SFPE의 혈관 이완작용이 세포막 ATP-민감성 $K^+$ 통로를 활성화시켜 이루어짐을 확인하였다.
본 연구는 단기 숙성형 생햄의 식중독균 오염 상태와 냉장 및 실온 저장 조건 중 미생물수의 변화를 살펴보기 위하여 실시되었다. 생햄의 등심 원료육에서의 미생물 오염도를 조사한 결과 총균수는 $3.11\;log\;CFU/cm^2$이었고 유산균, Pseudomonas spp., Clostridium perfringens 및 효모와 곰팡이는 모두 2 log 미만이었으나 Enterobacteriaceae가 3.11 log로서 주종균이었다. 생햄 원료 및 10과 $25^{\circ}C$에서 저장 된 제품에서 Salmonella spp., Staphylococcus aureus, Vibrio parahaemolyticus, Clostridium perfringens, Listeria monocytogenes와 Escherichia coli O157:H7 등 6가지 식중독균은 발견되지 않았다. 생햄의 초기 총균수는 3.06 log CFU/g이었으며 90일 후에도 균수 증가는 미미하여 $10^{\circ}C$와 $25^{\circ}C$에서 각각 4.6과 4.69 log CFU/g이었다. 생햄의 저장 중 주 종균은 유산균과 Staphylococcus균이었다.
A total of 38 bacterial strains within the classes Bacilli and Deinococci were isolated from various sources in Korea. Samples were collected from animal intestine, urine, soil, tidal flat mud, and kimchi. In the sequence comparison and phylogenetic analysis of 16S rRNA sequences, the 38 isolates were assigned to the classes Bacilli and Deinococci with sequence similarities more than 98.7%. Twenty-four strains and 13 strains were classified the order Bacillales and Lactobacillales in the class Bacilli, respectively. In the order Bacillales, there were nine species in the genus Bacillus, seven species in the genus Paenibacillus, and the remaining eight species in the genera Domibacillus, Halobacillus, Virgibacillus, Lysinibacillus, Paenisporosarcina, Planococcus, Savagea, and Staphylococcus. In the order Lactobacillales, there were four species in the genus Lactobacillus, three species in the genus Leuconostoc, three species in the genus Lactococcus, and the remaining three species in the genera Aerococcus, Enterococcus, and Streptococcus. One species was related to the genus Deinococcus of the order Deinococcales. Most of the isolated strains were Gram-stain-positive, but some were Gram-stain-variable or Gram-stain-negative. Cells were rod or cocci-shaped. Based on the results of 16S rRNA analysis, we report 38 strains as previously unrecorded species to Korea, and the basic characteristics of strains are described herein.
Objectives: Exposure to airborne bacteria is associated with adverse health effects such as respiratory and infectious diseases. This study evaluated airborne bacterial concentrations in the living rooms, kitchens, and toilets of 30 homes. Methods: Bacteria were sampled with an MAS100 impactor in three spaces in the subject homes between April 2014 and February 2015. Bacteria were grown on TSA plates for 48 hours at $35^{\circ}C$. The bacterial strains were isolated and amplified by polymerase chain reaction. Results: The most culturable bacteria were found in toilets ($624.0CFU/m^3$, GM: $417.3CFU/m^3$), followed by in the kitchen ($503.8CFU/m^3$, GM: $324.9CFU/m^3$). The dominant genera identified were: Staphylococcus sp.(19%), Micrococcus sp.(16%), and Bacillus sp.(11%) in the indoor air and Bacillus sp. (30%) in the outdoor air. Gram-positive bacteria comprised more than half of all colonies. Conclusion: In this study, culturable bacteria concentrations were higher than those reported in other spaces. Therefore, it is important to control relative humidity and remove moisture to prevent bacteria from multiplying. Additionally, the dominant species in indoor air were Staphylococcus sp. and Micrococcus sp. These are found on the human skin, mucous membranes, and hair, so human activity can affect bacterial distribution. Therefore, cleaning and controlling moisture are important for reducing indoor bacterial concentrations.
미생물학적 실내공기질을 평가하기 위하여, 울산에 위치한 3곳의 유치원을 대상으로, 2002년 4월에서 2003년 1월에 걸쳐 계절별로 공기 중의 미생물 농도를 결정하였다. 미생물의 포집에는 충돌식 공기 채취기를 사용하였으며, 세균수는 Staphylococcus medium과 plate count agar, 진균수는 dichloran rose bengal chloramphenicol agar를 사용하여 측정하였다. Staphylococcus medium에서 생장한 세균의 평균 농도는 827.0 MPN/㎥ 로 83.5~4,149.1 MPN/㎥의 범위였으며, plate count agar의 경우는 평균이 580.3 MPN/㎥로 50.0~2,636.0 MPN/㎥의 범위였다. 계절별 평균값은 여름이 가장 높았고, 가을, 봄, 겨울의 순이었으며, 세균의 집락수와 실내온도는 양의 상관관계가 있었다. 계절에 따라 그람양성의 구균은 전체 세균 집락의 45.6~61.0%, 그람음성의 막대균이 8.5~20.6%를 차지하였으며, Micrococcus spp.가 가장 우점하였다. 진균의 농도는 평균이 660.8 MPN/㎥로 0~1,887.5 MPN/㎥의 범위였으며, 계절별 평균값은 여름이 가장 높았고, 겨울이 가장 낮았다. 실내외 공기의 진균 집락으로부터 Penicillium spp.와 Aspergillus spp.를 확인할 수 있었다. 연구의 결과는 계절에 따른 미생물 농도의 변이와 학교의 실내환경에서 bioaerosol의 허용수준을 결정함에 있어 이를 고려해야 할 필요가 있음을 보여주었다.
The synthesis of osme quinoxaline derivatives containing indoline-2, 3-dione or thiazolidlinone residue is described. The synthesized derivatives were sureened in vitro for their growth inhibitory activity against six species of bacteria, viz. Staphylococcus aureus, Streptococcus faecalis, Escherichia coli, Pseudomonas aeruginosa, Serratia marcescens and Mycobacterium semegmatils. Most of the compounds exhibited antimicrobial activity especially those having indoline-2, 3-dione moiety.

As an outcome of the study on the bacterial species diversity in Korea, we report 24 unrecorded bacterial species of Korea belonging to the phylum Firmicutes. The unrecorded species excavated through this study were assigned to 12 different genera of 7 families, namely Bacillus, Halobacillus, Lysinibacillus and Thalassobacillus of Bacillaceae, Brevibacillus and Paenibacillus of Paenibacillaceae, Viridibacillus of Planococcaceae, Salinicoccus and Staphylococcus of Staphylococcaceae, Enterococcus of Enterococcaceae, Lactobacillus of Lactobacillaceae, and Lactococcus of Streptococcaceae, respectively. The bacterial isolates were obtained from various ecosystems in Korea. The isolates were identified based on 16S rRNA gene sequences, and those exhibiting at least 99% sequence similarity with known bacterial species but never reported in Korea were selected as unrecorded species. The selected isolates were subjected to further taxonomic characterization including the analysis of cell shape and fine structure using electron microscope, colony color and shapes, enzyme activities and carbon source utilization. The descriptive information on the 24 unrecorded species are provided.
On studying of microbes and appearance frequency which are floating around dwelling space and life environments in Seoul area, the results are summarized as follows ; 1. 1931 strains of 29 bacteria are isolated in dwelling spaces, Among them, there are Staphylococcus aureus, S. anthracis which are pathogenic to human beings. 2. In fungi, 76 strains of 13 species are isolated. That of highest frequency is confirmed to be Aspergillus which is pathogens of Aspergillosis (one of a group of diseases of animals and human being caused by various species of Aspergillus). The places, where the number of kinds and the frequency of appearances have been observed highest, are coffee shops, lunch counters and office rooms, while factory districts are of far lower level than expected. 3. 0.1% $HgCl_2$, Leasking oil, and Telephone disinfectant were better than any other in fungicidal effect.
