Evaluation of Antibacterial Efficacy of Certain Botanicals Against Bacterial Pathogen Bacillus sp. of Silkworm, Bombyx mori L.

  • An in vitro and in vivo studies were conducted to evaluate the antibacterial efficacy of certain botanicals viz., rhizomes of turmeric (Curcuma longa) and leaves of amla (Phyllanthus emblica), asparagus (Asparagus racemosus), bael (Aegle marmelos), boerhavia (Boerhavia diffusa), garlic (Allium sativum) and basil (Oscimum basicilum) against bacterial pathogens viz., Staphylococcus sp., Bacillus sp. and Klebsiella cloacae, of silkworm, Bombyx mori. Asparagus and basil, amla and boerhavia, basil and bael at concentration of 20, 000 ppm showed higher antibacterial activity against Staphylococcus sp., Bacillus sp., K. cloacae respectively, both in vitro and in vivo studies.

Antimicrobial Activities of Korean Marine Algae against Fish Pathogenic Bacteria (한국산 해조류의 어류병원성세균에 대한 항균활성)

  • To obtain antimicrobial algae against fish pathogenic bacteria, we screened 80% methanolic extracts of 30 algae using fish pathogenic bacteria, Staphylococcus sp., Streptococcus sp., Edwardsiella tarda and Vibrio anguillarum. Among them, Corallina officinalis, Dumontia simplex, Gloipeltis furcata, Grateloupia lanceolata and Grateloupia turuturu were effective for growth inhibition of a Gram-positive bacterium, Staphylococcus sp.. Sargassum thunbergii and Polysiphonia morrowii exhibited significant inhibitory effects against the growth of Gram-negative bacteria, both E. tarda and V. anguillarum. Moreover, antimicrobial activity-guided fractionation for P. morrowii extract yielded significantly active 90% methanolic fraction. This fraction significantly inhibited the growth of E. tarda exhibiting a MIC of 1 mg/ml. In addition, its antimicrobial activity was stable under various pH conditions.

Patterns on the Outbreaks of Bovine Mastitis and Susceptibility to Antimicrobias of Isolated Causative Agents in a Municipal Area

  • 광주시 지역에서 사육하고 있는 젖소 1,614두중 유방염으로 의심되는 730분방 중에서 170분방을 검사분석하여 유방률, 균의 분리와 간이검사법과의 관계, 계절별 분리균의 분포, 항균 요법제에 대한 감수성 등을 검사하였다. 본 원인균은 730분방 중 134분방(18.4%)에서 분리되었으며 체세포 숫자는 평균 $1.620 imes 10^6pm 1.167 imes 10^6/ml$(C.V. 72.0%)이었다. CMT 반응치는 평균 $2.9pm 1.2$(C.V. 41.4%)이었으며 WT 반응치는 평균 $2.8pm 1.2$(C.V. 42.9%)이었다. RBVT와 CMT의 상관계수는 0.82(P<0.001)이었고 RBVT와 WT의 상관계수는 0.75 (P<0.001)이었으며 CMT와 WT의 상관계수는 0.93 (P<0.001)이었다. 체세포 숫자를 기준으로 하여 CMT 및 WT의 양성율을 비교하여 보면 원인균이 분리된 경우에는 체세포 숫자가 $0.49 imes 10^6$ 이하/ml의 경우에 반응치가 1+일때의 CMT는 72.4%, WT는 42.1%이었고 체세포 숫자가 $0.50 imes 10^6~1.00 imes 10^6/ml$의 경우에 반응치가 2+일 대의 CMT는 45.5%, WT는 48.8%이었으며, 체세포 숫자가 $3.01 imes 10^6$ 이상/ml의 경우에 반응치가 3+일 때의 CMT는 73.7%, WT는 92.3%이었다. 원인균의 월별 분리 빈도를 보면 8월 (17.9%)이 가장 높았고 다음은 9월(16.4%), 7월 (12.7%), 6월 (11.2%), 1월 (9.0%)의 순이었다. 원인균의 분리 빈도를 보면 Staphylococcus sp. (51.4%)가 가장 높았고 다음은 Escherichia coli(23.9%), Pseudomonas sp. (11.2%). Streptococcus sp. (6.7%)의 순이었다. 항균 요법제에 대한 감수성은 trimethoprim/sulfamethoxazole은 Streptococcus sp., Staphylococcus epidermidis, Proteus sp., Salmonella sp. 등에 높았고 gentamycin은 Streptococcus sp., Staphylococcus aureus, Enterobacteriaceae, Klebsiella sp., Proteus sp., Salmonella sp. 등에 높았으며 enrofloxacin은 일반적으로 거의 모든 균에서 감수성이 높았다.

  • Lipase of Staphylococcus sp. was purified from the culture supernatant, and its molecular mass estimated to be 44 kDa by SDS-PAGE. Its optimum temperature and pH for the hydrolysis of p-nitrophenyl substrates was $28^{\circ}C$ and pH 8.5, respectively The gene encoding the lipase was cloned in Escherichia coli in the $NH_2$-teminally truncated form by using the shotgun method, and sequenced. The mature enzyme had a 49-93% amino acid sequence homology with other staphylococcal lipases. This lipase was used for the hydrolysis of the 3-O-acetate of cephalosporin-C to give an intermediate, deacetylated cephalosporin-C that is useful for further chemical elaborations.

  • Methicillin-resistant Staphylococcus pseudintermedius Z0118SP0130 was isolated from eye specimen of a companion dog in South Korea. The complete genome of Z0118SP0130 consisted of a 2,663,277 bp chromosome and there was no plasmid. The strain was identified as the sequence type 45 and contained a mecA gene which comprised of staphylococcal cassette chromosome mec type Vb (5C2&5). Antimicrobial resistance to erythromycin, clindamycin, quinupristin-dalfopristin, trimethoprim-sulfamethoxazole, mupirocin, oxacillin, streptomycin, and gentamicin was observed in the strain.

  • Various problems in fermented fish products have been a major obstacle to manufacture the product in large scale, which is mainly concerned with the food safety. In this review, salt-fermented anchovy was selected to elucidate the characteristics of microorganisms involved in fermentation; thereby, it is suggested for research areas to achieve the quality improvement of tile product. Different microorganisms were involved in fermentation of anchovy. Dominant species were reported to be Bacillus sp., Pseudomonas sp., and Micrococcus sp., other microorganisms were Vibro sp., Clostridim sp., Serratia sp., Achromobacter sp., Streptococcus sp., Breuibacterium sp., Halobacterium sp., Flavobacterium sp., Corynebacterium sp., Acinetobacter sp., Sarcina sp., Staphylococcus sp., Torulopsis sp., and Saccharomyces sp. To standardize the quality of fermented fish products, screening and isolation of promising microorganisms should be carried out to develop different types of products; at the same time, proper sanitation control should be employed to keep the commercial value of the product by prolonging the shelf life.

Isolation and Identification of Streptomyces sp. Producing Anti-vancomycin Resistant Staphylococcus aureus Substance (반코마이신 내성 Staphylococcus aureus 억제 물질 생산 Streptomyces sp.의 분리 및 동정)

  • An Actinomycetes producing an anti-VRSA (vancomycin-resistant Staphylococcus aureus) substance was isolated from soil. The cultural, morphological, physiological and phylogenetic analyses of an isolated strain were investigated for identification. Cultural characteristics based on ISP (International Streptomyces Project) were as follows: white aerial mycelium, yellow reverse side, and good growth on various medium. Also, the isolate did not produce the soluble pigment. Morphological characteristics were showed cylindrical spore chain and smooth spore surface by SEM (Scanning Electron Microscope). Physiological characteristics were showed LL-type by DAP isomer analysis and detected glycine, glutamic acid and alanine. A phylogenetic analysis of the 16S rDNA provided a clue that the isolated strain was actually a member of the genus Streptomyces, because the determined sequence exhibited a higher homology with Streptomyces echinatus. The isolate was identified to be a genus of Streptomyces sp.. The optimal culture conditions for the maximum production of anti-VRSA substance by Streptomyces sp. were attained in a culture medium composed of $2.0\%$ (w/v) glucose, and $0.4\%$ (w/v) yeast extract. The anti-VRSA substance was highly produced after 5 days of culture. Optimal pH and temperature conditions for the production of anti-VRSA substance were pH 7.0 and $28^{\circ}C$, respectively.

Monitoring of Microbial Contamination in Air Purifier for Preventing Cross-contamination (교차오염방지를 위한 공기정화제품에서 미생물 오염도 분석)

  • Many atmospheric pollutants including chemical agents, house dust, and microorganisms cause building-related illnesses through respiration in humans. This study was conducted to analyze the profiles of microbial pollutants in air purifiers used in home, office and playschool. Dominant eleven species of microorganisms were isolated and identified in environmental air and air purifiers. Among them, Staphylococcus sp., Micrococcus sp. and Bacillus sp. are the most dominant species. By phylogenetic analysis of the 16S rRNA gene, the dominant bacteria were identified as Staphylococcus epidermidis, Micrococcus luteus and Bacillus epidermidis, respectively. It has been known that these bacterial species are closely related with food spoilage and human infectious disease. Thus, these results indicate that microbial pathogens related with human illnesses through respiration will be contaminated in air purifiers and also need to develop a method to control those of pathogens for human health.

A Peptide Antibiotic AMRSA1 Active against Multidrug-resistant Staphylococcus aureus Produced by Streptomyces sp. HW-003

  • The antibiotic-producing strain HW-003 was screened from soil and found to be effective against the multidrug-resistant Staphylococcus aureus. The spore chain of HW-003 was retinaculiaperti, and the spore surface was spiny. Strain HW-003 has a LL-diaminopimelic acid isoform in the cell wall. The aerial mass color of the strain was gray, and the reverse side was yellow-brown. The strain produced melanin, but did not produce soluble pigments. According to the Taxon program, HW-003 showed best match with Streptomyces cyaneus. Antibiotic production reached a maximum after 72-h cultivation. The antibiotic was purified with silica gel column chromatography, octadecylsilyl column chromatography, and HPLC. The purified antibiotic, AMRSA1, showed strong inhibitory activity against multidrug-resistant Staphylococcus aureus and gram-positive bacteria. The molecular weight of AMRSA1 was about 1, 100. AMRSA1 was a peptide antibiotic containing alanine and serine.

Psammaplin A, a Natural Bromotyrosine Derivative from a Sponge, Possesses the Antibacterial Activity against Methicillin-resistant Staphylococcus aureus and the DNA Gyrase-inhibitory Activity

  • Psammaplin A, a natural bromotyrosine derivative from an associated form of two sponges (Poecillastra sp. and jaspis sp.) was found to possess the antimicrobial effect on the Gram-positive bacteria, especially on methicillin-resistant Staphylococcus aureus (MRSA). The minimal inhibitory concentration of psammaplin A against twenty one MRSAs ranged from 0.781 to 6.25 ${\mu}g/ml$, which that of ciprofloxacin was 0.391~3.125${\mu}g/ml$. Psammaplin A could not bind to penicillin binding protein, but inhibited the DNA synthesis and the DNA gyrase activity with the respective 50% (DNA synthesis) and 100% (DNA gyrase) inhibitory concentration 2.83 and 100 ${\mu}g/ml$. These results indicate that psammaplin A has a considerable antibacterial activity, although restricted to a somewhat narrow range of bacteria, probably by inhibiting DNA gyrase.
