The Study of Improvement of Measurement Precision on Bulk Density, Soil Hardness and Air Permeability in Upland Soils

  • The measurement based on reliable standard operating procedures (SOPs) is important for consistent information. The objective of this study is to investigate reliable SOPs of soil physical methods, including core method for bulk density, Yamanaka hardness, and air permeameter method for air permeability. The coefficients of variation in bulk density (core method), Yamanaka hardness, and air permeability were ranged of 1~6%, 8~13%, and 10~84%, respectively. The variation in situ measurement such as bulk density, hardness, and air permeability due to spatial variability at measuring site was larger due to the number of replicates, organic matter content, and soil texture. Nevertheless, air permeability had different values as different number of replicates, and thus, it is thought that more replicates can result in higher reliability. It suggested that investigation of soil physical properties for the target sites should required to consider about soil texture, organic matter content, and number of replications before measurement. In conclusion, core sampling for bulk density measurement in upland soil recommended to perform in 3 repetitions with 2 inch core, and 3 inch core sampling for higher organic matter content.

관광농원의 레크리에이션 기능에 대한 경제가치 평가 (Economic valuation for Recreation Roles of Tourist Farms)

  • The expansion of demand associated with leisure, which has resulted from the elevated standard of living, has made the domestic tourist demand diversified. Recently, people, especially urban population, show the higher interest and need for environmentally friendly rural tourism. This study aims to grasp the utilization of recreation roles of tourist farms and then to evaluate the economic value for recreation roles of tourist farm using Traveling Cost Method. Data were collected from a survey in 2003. The results show that its value is 62,037 won per head annually and so the tourist farms play their role of connecting the urban population with rural amenity.

추출방법별 식용 및 약용버섯류의 총 당함량 비교분석 (Total sugar contents of edible and medicinal mushrooms comparative analysis by the extraction method)

  • 이번 연구에서는 식용 및 약용버섯 자실체를 가지고 버섯의 주요 기능성 성분인 총 당의 함량을 추출방법을 달리하여 비교분석하였다. 열수추출, 산추출, 알칼리 추출을 통해 총 당 측정법으로 미량의 시료에 적합한 페놀-황산법으로 측정하였다. 표준 곡선은 glucose solution을 사용하여 작성하고 측정한 흡광도를 표준곡선에 적용하여 시료의 농도를 얻었다. 열수추출에서는 동충하초가 658.4(${\mu}g/g$)로 가장 높았고, 산추출과 알칼리 추출에서는 운지버섯이 각각 649.7(${\mu}g/g$)와 679.0(${\mu}g/g$)로 가장 높음을 알 수 있었다. 추출방법별로 열수추출과 마이크로웨이브 추출로 총 당 함량을 정량한 결과 열수 추출에서는 동충하초, ASI 7025가 높았고, 1 hr 마이크로웨이브 추출 및 2 hr 마이크로웨이브 추출에서는 운지버섯, 꽃송이가 높게 나타났다. 이러한 결과를 통해 각기 다른 추출용매와 추출방법에 따라서 추출되는 총 당 함량이 다름을 알 수 있었다.

  • 본 연구에서 분자육종을 위한 신속 DNA추출법을 개발하고자, 2% CTAB 추출법을 시스템적으로 개량한 Modified CTAB법에 대한 결과를 요약하면 아래와 같다. 1. DNA 추출용 샘플마쇄는 Voltex mixer, 3개 텅스텐 구슬 및 96 deep well block을 이용하였으며, 5분 이내에 96개 샘플을 분쇄할 수 있었다. 2. Modified CTAB DNA 추출법은 1.5 ml E-tube 대신에 1.2 ml 8연식 tube를 사용하였으며 DNA 추출, PCR작업 및 전기영동까지 일련의 작업이 8채널 멀티피펫을 이용할 수 있는 시스템적으로 개선하였다. 3. Modified CTAB법은 어린잎 뿐 만 아니라 이앙 후 30일째 채취한 잎에서도 DNA 추출이 가능하기 때문에 기존의 육종포장에 재식된 재료를 활용할 수 있는 장점이 있다.

영유아 보육시설의 영양관리 시범사업(I): 유아의 식생활 현황 (Baseline Dietary Behaviors of Children for Nutritional Management Programs at Child Care Centers in Korea)

  • The aim of this investigation was to collect the baseline data about growth, dietary behaviors, nutrient intakes and dietary quality of children at child care centers for planning nutritional management program. The dietary intakes were measured by weighing method for intakes at child care centers and by dietary records by children\\`s mothers for intakes at home. The weight and height were measured and calculated z-scores by using standard of the same age groups. The children were 163 boys and 168 girls and the mean age were 55.6 months(12-78 months) and 56.0 months(16-78 months). respectively. The Z-score for height(0.33) and weight(0. 11) showed that the growth of children were above average. Nutrients intakes such s energy, protein, fiber, iron, sodium, potassium, vitamin A, vitamin B$_1$, vitamin C, niacin of boys from lunch and snack at day care centers were significantly higher than those of girls, and the meals at child care centers provided 20-37% of RDAs. The daily energy intakes were 1332.2kcal(83.1% RDA), protein 44. 26g(109.3% RDA), calcium 437.73mg(72.4% RDA), iron 7.24mg(72.4% RDA), vitamin A 459.6RE(113.1% RDA),vitamin B$_1$ 0.74mg(93.4% RDA), vitamin B$_2$0.79mg(79.7% RDA), niacin 7.66mg(69.9% RDA) and vitamin C 56.84mg(142.1% RDA). The nutrients which more than 50% of subjects ate less than 75% RDA were calcium, iron, niacin, and vitamin B$_2$.The average mean adequacy ratio was 0.80 and mean dietary variety score was 22.42, and those are positively correlated to % RDA of nutrient intakes. In summary, the children using child care centers consumed most nutrients below level of RDA and the nutritional management program at child care center is required to enhance the nutritional status of them. 수박 밀도의 간편 계측시스템 개발 (Development of Simple Density Measurement System for Watermelons)

  • Density is a physical property which contains information relating to the internal quality of fruits and vegetables, and can be used as an index for nondestructive quality evaluation. Density sorting has been employed by farmers for some agricultural products since ancient times. In this study, an automatic density measuring system based on the platform scale or water displacement method was developed for density sorting of watermelon. It consisted of water tan, load cell, net tray, electric motor, limit switch, control system and its program. The resolution of density was 0.001 g/㎤. In order to calibrate and evaluate the accuracy, the density was measured using a balloon kept in cold water. It showed 1.002 g/㎤ which almost correspond to real density of water. Test results with 6 watermelons and 3 replications showed that the standard deviations of the dens were 0.001∼0.004 g/㎤. The relationship between density and internal quality of watermelon was investigated using the system. The densities of hollow watermelons were less than 0.950 g/㎤, it was apparent that the density of the watermelon was related to the degree of hollowness. But the soluble solid contents and internal defects could not be estimated from the density.

자탈형 콤바인용 벼 무게 측정시스템 개발 (Development of a Rice Weighing System for Head-Feed Combine)

  • Yield mapping is necessary for precision farming. An on-site rice weighing system for a head-feed combine was developed to measure the total weight of rice grain harvested while the combine was operated. A load cell system was used to monitor rice weights accumulated into the combine grain tank using a load cell. This method gave cumulative grain weight readings as a function of time. The system consisted of a load cell, two supporting brackets, and a computer-based data acquisition system. The weights measured with the system from two fields were compared with those obtained with a commercially available electronic balance. The response of the load cell to varying grain weights was linearly modeled, showing a coefficient of determination of 0.998 and a standard error of ${\pm}4.09kg$.

Mechanical Test Methods for Compost Maturity

  • Manure-based composts can have detrimental effects on the agricultural lands and crops if they are applied without proper stabilization process. Composting is well-known method for the stabilization of manure-based composts and the extent of composting could be examined by maturity test. Among various methods to examine compost maturity, the performances of two mechanical methods (Solvita and CoMMe-100) were examined and the test results were compared with seed germination test. The mechanical methods are considered to be simple and relatively objective. Also they are cost- and time-efficient. Ten commercially available composts collected in Korea were used for this study. Basically, the maturity determined with these mechanical methods was in good agreement with the seed germination test. However, it appeared that the index levels of compost maturity indicated more maturity by CoMMe-100 than Solvita for the same compost. The differences between maturity index levels determined by both methods were able to be minimized through extending the reaction time for CoMMe-100 and adjusting index level for maturity determination in the standard color chart.

Cryopreservation of in Vitro Grown Shoot Tips of Sweet Potato (Ipomoea batatas L.) by the Encapsulation-Vitrification Method

  • Sweet potato (Ipomoea batatas L.) shoot tips grown in vitro were successfully cryopreserved by encapsulation-vitrification. Encapsulated explants are very easily manipulated, due to the relatively large size of the alginate beads, and a large number of samples can be treated simultaneously. In this study, the effects of sucrose preculture, cryoprotectant preculture, and post-warm recovery media on regrowth, following liquid nitrogen (LN) exposure, were investigated to establish an efficient encapsulation-vitrification protocol for sweet potato. Shoot tips of plants grown in vitro were precultured in 0.3 M sucrose for 2 d before encapsulation. Encapsulated shoot tips were pre-incubated in liquid MS (Murashige and Skoog) medium containing 0.5 M sucrose for 16 h, before preculturing in sucrose-enriched medium (0.7 M sucrose) for 8 h. Shoot tips were osmoprotected with 35% plant vitrification solution 3 (PVS3) for 3 h, before being dehydrated with PVS3 for 2 h at $25^{\circ}C$. The encapsulated and dehydrated shoot tips were transferred to 2 mL cryotubes, suspended in 0.5 mL PVS3, and plunged directly into liquid N. High levels of shoot formation were obtained for the cv. Yeulmi (65.7%) and Yeonwhangmi (80.3%). The regrowth rates of cryopreserved samples in Yeulmi (78.9%) and Yeonwhangmi (91.3%), following culture on ammonium-free MS medium for 5 d, were much higher than those cultured on standard MS medium (65.7% and 80.3%, respectively). This encapsulation-vitrification is a promising method for the long-term preservation of sweet potato.

건강기능식품 원료로서 구릿대의 지표성분 분석법 검증 (Method validation of marker compounds from Angelicae Dahuricae Radix as functional food ingredients)

  • 백지는 산형과(Umbelliferae)에 속하는 구릿대(Angelica dahurica)의 뿌리를 건조한 것으로, 다양한 약리작용을 하는 것으로 알려져 예로부터 한약재로 사용되어 왔다. 백지의 대표적인 활성 성분인 coumarin을 포함하여 다양한 성분이 알려져 있으며, 최근 백지의 다양한 생리활성이 보고되고 있다. 본 연구는 백지를 건강기능식품의 기능성 원료로 활용하고자, 백지의 지표성분 설정 및 이에 대한 UPLC 분석법을 확립하여 분석법 유효성 검증을 실시하였다. 백지의 지표성분으로는 oxypeucedanin hydrate, byakangelicol, oxypeucedanin, imperatoin, phellopterin, isoimperatorin의6종을 설정하였다. 분석법 유효성 검증은 특이성, 직선성, 검출한계, 정량한계, 정확성, 정밀성 등의 항목을 이용하여 확인하였다. 본 시험법에서 지표성분 6종은 서로 간섭없이 분리되어 특이성을 확인하였고, 결정계수의 값도 0.999 이상으로 나타나 높은 직선성을 나타내었다. 분석법 유효성 검증 결과, 지표 성분 6종의 RSD는 5% 이하로 나타났으며 90-110%의 범위의 회수율을 나타내 높은 정확성과 정밀성이 있는 것을 확인할 수있었다. 이러한 결과는 UPLC를 이용한 백지의 지표성분 분석법이, 새롭게 제시한 6종에 대해 적합한 동시분석 분석법임을 나타낸다. 따라서, 본 연구를 통하여 확립된 백지의 지표성분 분석법이 향후 백지(구릿대)의 건강기능식품 기능성 원료 개발을 위한 기초자료로 활용될 것으로 기대된다.
