Sufflavibacter maritimus gen. nov., sp. nov., Novel Flavobacteriaceae Bacteria Isolated from Marine Environments

  • Four Gram-negative, chemoheterotrophic, non-motile, yellow-colored strains were isolated from the East Sea or from deep-sea sediments of Nankai Trough by standard dilution plating. Characterization by polyphasic approaches indicated that the four strains are members of the same species. Phylogenetic analyses based on 16S rRNA gene sequences revealed that the strains formed a coherent and novel genus-level lineage within the family Flavobacteriaceae. The dominant cellular fatty acids were i-C15:0, 3-OH i-C17:0, and 2-OH i-C15:0 and/or C16:1 ${\omega}7c$. Predominance of 2-OH i-C15:0 and/or C16:1 ${\omega}7c$ clearly differentiated the strains from closely related members. The DNA G+C contents ranged 35.1-36.2 mol%. It is proposed, from the polyphasic evidence, that the strains should be placed into a novel genus and species named Sufflavibacter maritimus gen. nov., sp. nov., with strain $IMCC1001^T(=KCCM\;42359^T=NBRC\;102039^T)$ as the type strain.

  • The primary goal of six sigma is to improve patient satisfaction, and thereby profitability, by reducing and eliminating defects. Defects may be related to any aspect of customer satisfaction: high product quality, schedule adherence, cost minimization, process capability indices, defects per unit, and yield. Many six sigma metrics can be mathematically related to the others. Literally, six means six standard deviations from the mean or median value. As applied to quality metrics, the term indicates that failures are at least six standard deviations from the mean or norm. This would mean about 3.4 failures per million opportunities for failure. The objective of six sigma quality is to reduce process output variation so that on a long term basis, which is the customer's aggregate experience with our process over time, this will result in no more than 3.4 defect Parts Per Million(PPM) opportunities (or 3.4 Defects Per Million Opportunities. For a process with only one specification limit (upper or lower), this results in six process standard deviations between the mean of the process and the customer's specification limit (hence, 6 Sigma). The results of applicative six sigma experiment studied on 18 items TP, ALB, T.B, ALP, AST, ALT, CL, CK, LD, K, Na, CRE, BUN, T.C, GLU, AML, CA tests in clinical chemistry were follows. Assessment of process performance fits within six sigma tolerance limits were TP, ALB, T.B, ALP, AST, ALT, CL, CK, LD, K, Na, CRE, BUN, T.C, GLU, AML, CA with 72.2%, items that fit within five sigma limits were total bilirubin, chloride and sodium were 3 sigma. We were sure that the goal of six sigma would reduce test variation in the process.

The Studies on the Transaminase Activities(1) (Transaminase 활성에 관한 연구(제 1보))

  • Transaminase has been a popular methods in the clinical laboratory for the diagnosis of particular diseases. The methods we are familiar with should be considered further on the serum states provided on the test to be done. (serum freshness, temperature and duration preserved) I have studied transaminase activities of the serum preserved under frozen (-10$\circ$C), refrigerated (4$\circ$C) and room temperature conditions. At a given interval those activities of serum are measured and then the results are compared with the optical density and calculated for the standard deviation and percent variation (Table 1. & Fig. 1.) G.O.T. activity is maintained decreased at a relatively constant rate from 2nd day to 14th day preservation under frozen and refrigerated but its activity variation at 25$\circ$C are far greater than the above conditions throughout the test periods. Elevated G.P.T. level of serum compared with G.O.T. are remained at any temperature and duration. Unless the determination of amino acid level study can not ruled out clearly.

  • This paper describes the functions and design technology of the T-DMB (Terrestrial Digital Multimedia Broadcasting) receiver. T-DMB is a novel broadcasting media that can provide high-quality video and audio services. In this paper, we will describe the VLSI implementation of RF, Baseband and Multimedia Chip for T-DMB Receiver. The designed DMB SoC has low power consumption and has been implemented using a standard-cell library in 0.18um CMOS technology.

Transition State Characterization of the Low- to Physiological-Temperature Nondenaturational Conformational Change in Bovine Adenosine Deaminase by Slow Scan Rate Differential Scanning Calorimetry

  • Bovine adenosine deaminase undergoes a nondenaturational conformational change at $29^{\circ}C$ upon heating which is characterized by a large increase in heat capacity. We have determined the transition state thermodynamics of the conformational change using a novel application of differential scanning calorimetry (DSC) which employs very slow scan rates. DSC scans at the conventional, and arbitrary, scan rate of $1^{\circ}C/min$ show no evidence of the transition. Scan rates from 0.030 to $0.20^{\circ}C/min$ reveal the transition indicating it is under kinetic control. The transition temperature $T_t$ and the transition temperature interval ${\Delta}T$ increase with scan rate. A first order rate constant $k_1$ is calculated at each $T_t$ from $k_1\;=\;r_{scan}/{\Delta}T$, where $r_{scan}$ is the scan rate, and an Arrhenius plot is constructed. Standard transition state analysis reveals an activation free energy ${\Delta}G^{\neq}$ of 88.1 kJ/mole and suggests that the conformational change has an unfolding quality that appears to be on the direct path to the physiological-temperature conformer.

  • The field utilization factor (equation omitted) (the mean electric field / the maximum electric field) of standard sphere gaps was calculated by the charge simulation method, taking into account the ground plane and shanks. n changes mainly with g/r and slightly with 1$_1$, 1$_2$ and 1, where D=2r is the sphere diameter, g is the gap length, 1$_1$ and 1$_2$, respectively, are the lengths of the upper and lower shank, and t is the shank diameter. Generally, (equation omitted) increases as 1$_1$,1$_2$ and t each becomes larger. IEC standard 60052(2002) limits t$\leq$0.2D 1$_1$$\geq$1D and prescribes A=1$_2$+D+g where A is the height of the spark point on the upper sphere. Therefore, (equation omitted) is the largest when A=9D and the smallest when A=3D. The simple equation of a straight line, (equation omitted)=1- (g/3r), can generally be used as a representative value of (equation omitted) for a wide variety of sphere diameters that are permitted by the IEC standard. The maximum electric field E$_{m}$ at sparkover of standard air gaps has also been calculated by the relation E$_{m}$=V/(equation omitted)g). E$_{m}$ describes a U-curve for g/r, up to the sphere diameter of 1 m. Moreover, for 1.5-m and 2-m diameters and especially .for negative polarity, sparkover voltages have been calculated by integration of the ionization index.index.

Quality parameters of chicken breast meat affected by carcass scalding conditions

  • Objective: The influence of broiler carcass scalding conditions on chicken breast meat quality parameters was investigated. Methods: Two hundred and seventy Cobb broiler chickens from 42 to 48 days old were slaughtered according to the standard industry practice and scalded in five temperature/time combinations-$T_1$, $54^{\circ}C/210s$; $T_2$, $55^{\circ}C/180s$; $T_3$, $56^{\circ}C/150s$; $T_4$, $57^{\circ}C/120s$; $T_5$, $58^{\circ}C/90s$. Results: Scalding temperature increase resulted in higher values of external and ventral lightness and in protein functionality reduction-determined by emulsification capacity and protein denaturation-in chicken breast fillets 24 h post-mortem. Protein secondary structures had conformational changes, with a decrease of the ${\alpha}$-helix and an increase of the ${\beta}$-sheet and ${\beta}$-turn proportions, mainly in $T_1$ and $T_5$ samples, determined by Fourier-transform infrared spectroscopy in an attenuated reflectance mode analysis. The chemical composition, pH, water holding capacity and Warner-Bratzler shear force did not differ among the treatments. In the fatty acid profile, the 18:1n-9 was lower in $T_5$, which suggested that the high scalding-temperature could have caused the lipid oxidation. The values of the polyunsaturated fatty acids (PUFA), such as 22:2, 20:4n-6, and 22:6n-3, were highest in the $T_5$, thus being related to the phospholipid cellular membrane collapse in this experimental condition and subsequent release of these PUFA. Conclusion: Intermediate scalding-parameters avoided the negative changes in the chicken meat quality.

  • Over 60~70% of the domestic soil contamination have occurred by petroleum products. B T E X including benzene, toluene, ethylbenzene, xylene and total petroleum hydrocarbon (TPH) have to be inspected for the contaminated soil by petroleum products. An accurate contamination analysis is necessary to estimate the are of contaminated soil and also establish an appropriate purification scheme. In this study, we analyzed a sectional TPH pattern for current domestic distributed petroleum products. Also, the TPH content was analyzed by compensating the defect of current Korea standard analytic methods for soil where the analytic range is just for $C_8{\sim}C_{40}$. The light distillate petroleum products such as gasoline and solvent 1 showed the maximum of 85% difference in the TPH content between the standard analytic and improved methods.

Determination of Hydrochlorothiazide in Human Plasma by HPLC and Applicability to Oral Bioavailability in Korean Healthy Male Volunteers (HPLC를 이용한 혈중 히드로클로로티아지드의 분석 및 이를 이용한 한국인 성인남성에 대한 생체이용률 평가)

  • A simple and sensitive high-performance liquid chromatographic method for quantitation of hydrochlorothiazide in human plasma was developed and bioavailability parameters of hydrochlorothiazide were assessed in Korean healthy male volunteers. Caffeine was used as an internal standard. Hydrochlorothiazide and internal standard in plasma sample were extracted using tert-butylmethylether (TBME). A centrifuged upper layer was then evaporated and reconstituted with mobile phase of acetonitrile-25 mM phosphate buffer (20/80, pH 2.5). The reconstituted samples were injected into a Luna C18 column $(250{\times}4.6\;mm,\;5{\mu}m)$ at a flow-rate of 1.0 ml/min. The wavelength was set at 230 nm and no endogenous substances were found to interfere, A linear relationship for hydrochlorothiazide was found in the range of $10{\sim}300\;ng/ml$. The lower limit of quantitation (LLOQ) was 10 ng/ml with acceptable precision and accuracy. Assayed in plasma, the intra- and inter-day validation for all coefficients of variation (R.S.D.%) were found less than 15%. Main pharmacokinetic parameters of 50mg of hydrochlorothiazide were revealed as follows: $AUC_t\;1761{\pm}509.0\;ng{\cdot} hr\;ml,\;C_{max}\;296.5{\pm}95.5\;ng/ml,\;T_{max}\;1.94{\pm}0.85hr,\;K_{el}\;0.12{\pm}0.04\;hr^{-1}\;and T_{12}\;6.81{\pm}2.92\;hr.\;C_{max}\;and\;T_{max}$ were in accordance with the values $(270{\sim}350\;ng/ml\;and\;1.9{\sim}2.7\;hr)$ of Caucasian.
