Transesterification Using the Cross-Linked Enzyme Aggregate of Photobacterium lipolyticum Lipase M37

  • Biodiesel is methyl and ethyl esters of long-chain fatty acids produced from vegetable oils or animal fats. Lipase enzymes have occasionally been used for the production of this biofuel. Recently, biodiesel production using immobilized lipase has received increased attention. Through enhanced stability and reusability, immobilized lipase can contribute to the reduction of the costs inherent to biodiesel production. In this study, methanol-tolerant lipase M37 from Photobacterium lipolyticum was immobilized using the cross-linked enzyme aggregate (CLEA) method. Lipase M37 has a high lysine content (9.7%) in its protein sequence. Most lysine residues are located evenly over the surface of the protein, except for the lid structure region, which makes the CLEA preparation yield quite high (~93%). CLEA M37 evidences an optimal temperature of $30^{\circ}C$, and an optimal pH of 9-10. It was stable up to $50^{\circ}C$ and in a pH range of 4.0-11.0. Both soluble M37 and CLEA M37 were stable in the presence of high concentrations of methanol, ethanol, 1-propanol, and n-butanol. That is, their activities were maintained at solvent concentrations above 10% (v/v). CLEA M37 could produce biodiesel from olive oil and alcohols such as methanol and ethanol. Additionally, CLEA M37 generated biodiesel via both 2-step methanol feeding procedures. Considering its physical stability and reusability, CLEA M37 may potentially be used as a catalyst in organic synthesis, including the biodiesel production reaction.

An Experimental Investigation of Direct Condensation of Steam Jet in Subcooled Water

  • The direct contact condensation phenomenon, which occurs when steam is injected into the subcooled water, has been experimentally investigated. Two plume shapes in the stable condensation regime are found to be conical and ellipsoidal shapes depending on the steam mass flux and the liquid subcooling. Divergent plumes, however, are found when the subcooling is relatively small. The measured expansion ratio of the maximum plume diameter to the injector inner diameter ranges from 1.0 to 2.3. By means of fitting a large amount of measured data, an empirical correlation is obtained to predict the steam plume length as a function of a dimensionless steam mass flux and a driving potential for the condensation process. The average heat transfer coefficient of direct contact condensation has been found to be in the range 1.0~3.5 ㎿/$m^2$.$^{\circ}C$. Present results show that the magnitude of the average condensation heat transfer coefficient depends mainly on the steam mass fin By using dynamic pressure measurements and visual observations, six regimes of direct contact condensation have been identified on a condensation regime map, which are chugging, transition region from chugging to condensation oscillation, condensation oscillation, bubbling condensation oscillation, stable condensation, and interfacial oscillation condensation. The regime boundaries are quite clearly distinguishable except the boundaries of bubbling condensation oscillation and interfacial oscillation condensation.

Abell 2261: a fossil galaxy cluster in a transition phase

  • Fossil galaxy cluster has a dominant central elliptical galaxy (${\Delta}M12$ >2 in 0.5Rvir) embedded in highly relaxed X-ray halo, which indicates dynamically stable and passively evolved system. These features are expected as a final stage of the cluster evolution in the hierarchical structure formation paradigm. It is known that Abell 2261(A2261 hereafter) is classified as a fossil cluster, but has unusual features such as a high central X-ray entropy (i.e., non-cool core system), which is not expected in normal fossil clusters. We perform a kinematic study with a spectroscopic data of 589 galaxies in the A2261 field. We define cluster member galaxies using the caustic method and discover a new second bright galaxy at ~1.5 Rvir (nearly the splash-back region). It implies the current fossil state of the cluster can break in the near future. In addition, with three independent substructure finding methods, we find that A2261 has many substructures within 3 Mpc from the center of the cluster. These findings support that A2261 is not in a dynamically stable state. We argue that A2261 is in a transitional phase of dynamical evolution of the galaxy cluster and maybe previously defined fossil cluster does not mean the final stage of the evolution of galaxy clusters.

Real-time identification of the separated lanthanides by ion-exchange chromatography for no-carrier-added Ho-166 production

  • No-carrier-added holmium-166 (n.c.a 166Ho) separation is performed based on the results of separation conditions using stable isotopes dysprosium (Dy) and holmium (Ho) to minimize radioactive waste from separation optimization procedures. Successful separation of two adjacent lanthanides was achieved by cation-exchange chromatography using a sulfonated resin in the H+ form (BP-800) and α-hydroxyisobutyric acid (α-HIBA) as eluent. For the identification process after separation of stable isotopes, the use of chromogenic reagents alternatively enables on-line detection because the lanthanides are hardly absorb light in the UV-vis region or exhibit radioactivity. Four different chromogenic reagents were pre-tested to evaluate suitable coloring reagents, of which 4-(2-Pyridylazo)resorcinol is the most recommendable considering the sensitivity and specificity for lanthanides. Lanthanide radioisotopes (RI) were monitored for separation with an RI detector using a lab-made separation LC system. Under the proper separation conditions, the n.c.a 166Ho was effectively obtained from a large amount of 100 mg dysprosium target within 2 hrs.

Increased Stability of Nucleolar PinX1 in the Presence of TERT

  • PinX1, a nucleolar protein of 328 amino acids, inhibits telomerase activity, which leads to the shortening of telomeres. The C-terminal region of PinX1 is responsible for its nucleolar localization and binding with TERT, a catalytic component of telomerase. A fraction of TERT localizes to the nucleolus, but the role of TERT in the nucleolus is largely unknown. Here, we report a functional connection between PinX1 and TERT regarding PinX1 stability. The C-terminal of $PinX1^{205-328}$, a nucleolar fragment, was much more stable than the N-terminal of $PinX1^{1-204}$, a nuclear fragment. Interestingly, PinX1 was less stable in TERT-depleted cells and more stable in TERT-myc expressing cells. Stability assays for PinX1 truncation forms showed that both $PinX1^{1-328}$ and $PinX1^{205-328}$, nucleolar forms, were more rapidly degraded in TERT-depleted cells, while they were more stably maintained in TERT-overexpressing cells, compared to each of the controls. However, $PinX1^{1-204}$ was degraded regardless of the TERT status. These results reveal that the stability of PinX1 is maintained in nucleolus in the presence of TERT and suggest a role of TERT in the regulation of PinX1 steady-state levels.

Phase Diagrams and Stable Structures of Stranski-Krastanov Structure Mode for III-V Ternary Quantum Dots

  • The strain, surface and inerfacial energies of III-V ternary systems were calculated for three kinds of structure modes: the Frank-van der Merwe (FM) mode, the Stranski-Krastanov (SK) mode an the Volmer-Wever (VW) mode. The free energy for each mode was estimated as functions of the thickness and composition or lattice misfit. Through comparison of the free energy of each mode, it was found that the thickness-composition phase diagrams of III-V ternary systems can be determined only by considering the balance of the free energy and three kinds of structure modes appear in the phase diagrams. The SK mode appears only when the lattice misfit is large and/or the lattice layer is thick. The most stable structure of the SK mode is a cluster with four lattice layers or minimum thickness on a wetting layer of increasing lattice layers. The VW mode appears when the lattice misfit is large and the lattice layer is thin and only in the InPSb/InP and GaPSb/GaP systems which have the largest lattice misfit of III-V ternary systems. The stable region of the SK mode in the GaPSb/GaP and InPSb/InP phase diagrams is largest of all because the composition dependence of the strain energy of these systems is stronger than that of the other systems. The critical number of lattice layers below which tow-dimensional (2D) layers precede the three-dimensional (3D) nucleation in the SK mode at x=1.0 depnds on the lattice misfit.

Characteristics of near-surface ozone distribution

  • This study presents an analysis of the characteristics of vertical ozone distribution near the surface using ozonesonde data(l995 to 1998), plus surface ozone and meteorological data from the Pohang region. These features were examined in detail using three case studies. The first related to episodes of high surface ozone concentrations during the Spring season when the frontogenesis between the high and low pressure associated with the upper-level jet stream was found to be located near the surface. The second was a 5-day winter period(l3 -17 December, 1997) in the Pohang province when the hourly concentrations exceeded 90 ppb on several occasions owing to low-level jets(LLJs) induced by a nocturnal stable layer. Accordingly, this explains why the high surface ozone concentrations occurred at night as the ozone was transported across the zone by a strong wind speed( over 12.5 ms .1). The third case study was ozone enhancement due to photochemical reactions. In this case, the maximum concentration of ozone exceeded 60 ppb in the summer(23 -28 August, 1997). When an ozone peak appeared within the boundary layer, the occurrence frequency of a low-level jet due to the nocturnal stable layer was about 77%, similarly the occurrence frequency of a near-surface ozone peak relative to the appearance of an LLJ was about 76%. Accordingly, there is clearly a close correlation between the occurrence of LLJs and near-surface ozone peaks.

A Closed Counter-Current Two-Phase Thermosyphon Loop of a Cold Neutron Source in HANARO Research Reactor (하나로 원자로에 설치될 대향 이상 열사이펀 루프에 관한 실험)

  • An experimental study was carried out to delineate the flow characteristics in a closed countescurrent two-phase thermo syphon with concentric tubes. This is to be installed in the HANARO research reactor as a part of a Cold Neutron Source(CNS). In the present investigation, experiments ata room temperature with Freon-II3 as a moderator were performed. Results show that, based on the magnitude of pressure fluctuation, the flow regimes could be divided into 4 distinct ones in the ($V_f,\;Q_i$) plane, where $V_f$ represents the volume of the charged liquid and $Q_i$ the heat load: a stable flow regime, an oscillatory flow regime, a restablized flow regime and a dryout flow regime. For $V_f$>2.5l, the flow is stable at low $Q_i$. However, as $Q_i$ increases, the flow becomes oscillatory and finally restablizes As $V_f$ increases, the oscillation amplitude decreases, reaching to the restablized flow region at low $Q_i$, and the liquid level in the moderator cell remains high. In the oscillatory flow regimes, for a fixed VI; the oscillating period of time varies with $Q_i$, having a minimum value at a certain value of $Q_i$. The heat load, where the oscillating period of time is minimum, decreases as $V_f$ increases.

Construction of the Stable and High Copy Number Yeast Vectors (고효율 효모 유전자 운반체의 개발)

  • Yeat-Escherichia coli shuttle vectors were constructed by combination of various functional segments such as autonomous replicating sequence (ARS1), centromere region (CEN3), origin of replication of 2 $\mu$m plasmid (2 $\mu$m OR). Transformation efficiency, stability and copy number of constructed vectors were analyzed in yeast strains, SHY4(cir$^+$) containing 2 $\mu$m plasmid and NNY1(cir$^{\circ}$) without it. The results showed that centromere containing plasmids were very stable and existed at one copy per cell; fused replication system (2$\mu$m OR and ARS1) containing Plasmids were more stable and higher copy number than one replicon containing plasmids ; presence of endogenous 2$\mu$m plasmid influenced on stability and copy number of 2 $\mu$m based plasmids.
