Transesterification Using the Cross-Linked Enzyme Aggregate of Photobacterium lipolyticum Lipase M37

  • Biodiesel is methyl and ethyl esters of long-chain fatty acids produced from vegetable oils or animal fats. Lipase enzymes have occasionally been used for the production of this biofuel. Recently, biodiesel production using immobilized lipase has received increased attention. Through enhanced stability and reusability, immobilized lipase can contribute to the reduction of the costs inherent to biodiesel production. In this study, methanol-tolerant lipase M37 from Photobacterium lipolyticum was immobilized using the cross-linked enzyme aggregate (CLEA) method. Lipase M37 has a high lysine content (9.7%) in its protein sequence. Most lysine residues are located evenly over the surface of the protein, except for the lid structure region, which makes the CLEA preparation yield quite high (~93%). CLEA M37 evidences an optimal temperature of $30^{\circ}C$, and an optimal pH of 9-10. It was stable up to $50^{\circ}C$ and in a pH range of 4.0-11.0. Both soluble M37 and CLEA M37 were stable in the presence of high concentrations of methanol, ethanol, 1-propanol, and n-butanol. That is, their activities were maintained at solvent concentrations above 10% (v/v). CLEA M37 could produce biodiesel from olive oil and alcohols such as methanol and ethanol. Additionally, CLEA M37 generated biodiesel via both 2-step methanol feeding procedures. Considering its physical stability and reusability, CLEA M37 may potentially be used as a catalyst in organic synthesis, including the biodiesel production reaction.

Application of Stable Isotope Ratio Analysis for Origin Authentication of Pork

  • Origin authenticity of the animals used as food has always been a major concern to consumers around the world. In the past twenty years, a stable isotope ratio has been used for origin authentication. In this study, pork samples, both local and imported, were collected from the major markets from all around South Korea and analyzed for stable isotope ratios of nitrogen (${\delta}^{15}N$‰) and carbon (${\delta}^{13}C$‰), using Isotope Ratio Mass Spectrometry (IR-MS). A total of 599 samples with 335 Korean and 264 imported from 13 countries within America and Europe were investigated in accordance to the standard established methods for isotope ratio analysis. The results showed a significant variation related to the origin of the samples, explaining the difference in the feeding styles of the pork in each country. The stable isotope ratio values of carbon (${\delta}^{13}C$‰) were found in the decreasing order of: America ($-15.55{\pm}1.01$‰)>Korea ($-19.62{\pm}0.89$‰)>Europe ($-24.79{\pm}1.35$‰). Canada was having ${\delta}^{13}C$ ratio of $-22.87{\pm}0.92$‰, which is very low in the region of America and very close to Europe (-23.78 to -27.17‰). For nitrogen ${\delta}^{15}N$‰ the order was: America ($4.92{\pm}0.71$‰)>Europe ($4.54{\pm}0.66$‰)>Korea ($3.69{\pm}0.54$‰), with a slight variation among countries in each region studied. From the results it was concluded that the stable isotope ratio of the pork samples from different countries provide enough information about the origin and is therefore a potential tool which can be employed for origin authentication.

Understanding the Nutritional Sources of Gastropods and Anomura from the Mangrove Forest of Weno Island, Micronesia (마이크로네시아 웨노섬의 맹그로브 숲에 서식하는 고둥류 및 집게의 영양원에 대한 이해)

  • Carbon cycling and productivity within Weno Island of Micronesia enclosed by the coral reef may be likely self-maintained and insignificantly affected by the open ocean. Therefore, it is important to understand the role of the mangrove known as providing the organic matter and habitats for many organisms in this enclosed area. In order to trace the nutritional source of fauna (mostly invertebrates) in the mangrove forest of Weno island, we analyzed the fatty acid (FA) and carbon and nitrogen stable isotopes of potential nutritional sources (mangrove leaf & pneumatophore, seagrass leaf & root, surface sediment, and particulate organic matter (POM) in water) and consumers (4 gastropods and anomura). The mangrove and seagrass contained the abundance of 18:2${\omega}$6, and 18:3${\omega}$3, whereas FAs associated with phytoplankton and bacteria were accounted for a high proportion in the surface sediment and POM. FA composition of consumers was found to be similar to those of the surface sediment, mangrove, and seagrass. These were also confirmed through the mixing model of stable isotope for contribution of nutritional sources to consumers. Overall results with the feeding types of investigated mangrove fauna indicate that investigated mangrove fauna obtained their nutrition from the various sources, i.e. the mangrove for Littorina cf. scabra, the microalgae for Strombus sp., and omnivorous Pagurus sp. and Terebralia cf. palustris. However, it is obvious that the nutrition of most species living in the mangrove ecosystem is highly dependent on the mangrove, either directly or indirectly. More detail food-web structure and function of the mangrove ecosystem would be established with the analysis of additional fauna and flora.

Food Source Analysis of Rattus norvegicus, a Coinhabitant of Marine Birds in South Korea

  • The Chinese crested tern (Thalasseus bersteini) is one of the most globally endangered species, listed as "Critically Endangered (CE)" on the IUCN Red List, with only approximately 30-49 individuals surviving in the wild. Chinese crested terns were discovered to breed in South Korea for the first time in 2016 while conducting a census on uninhabited islands. The Ministry of Environment has declared the breeding habitat of the Chinese crested terns as "Specified Island" to protect this CE species. However, brown rats (Rattus norvegicus) inhabiting the breeding grounds of the Chinese crested terns and Black-tailed gulls may potentially pose a threat to the breeding of these avian species. Therefore, we conducted a study on the feeding behavior of brown rats involving stable isotope analysis to determine their food sources. Fecal analysis showed that brown rats mainly fed on plants, whereas they scarcely fed on animals, such as insects. In addition, the stable isotope analysis showed that the δ13C values of brown rats, insects, and Indian goosegrasses were approximately -16 to -11‰, whereas the δ13C value of Chinese crested terns that obtained their food from the marine ecosystem was approximately -22 to -18‰. Hence, we conclude that the source of carbon for brown rats on this island is the terrestrial ecosystem. We ruled out the possibility of any direct prey-predator interaction between the brown rat and the Chinese crested tern or Black-tailed gull.

Effect of Supplementary Feeding Levels on Productivity and Grazing Intensity in Grazing Elk stags(Cervus canadensis) (보충사료 급여수준이 엘크 수사슴의 생산성 및 방목강도에 미치는 영향)

  • This study was conducted to investigate the effect of supplementary feeding levels on livestck and forage productivity and grazing intensity in Elk stags (Cervus canadensis). A fifteen 2-year-old Elk stags about 195 kg were randomly assigned to one of three dietary treatments (five animals per treatment). The dietary treatments consisted of a feeding concentrate of 1.0% of body weight (T1), 1.5% of body weight (T2) and 2.0% of body weight. Total dry matter intake (TDMI) was increased with increased with an increasing supplementary feeding levels. Average daily gain (ADG) were significantly increased with an increasing supplementary feeding levels (p<0.05) and reached a maximum on July and was lower in spring than autumn. The velvet antler production was no differences among treatment groups. Forage productivity of pasture and crude protein content were highest on May and decreased thereafter, however, crude fiber content was the reversed. The grazing intensity of Elk stags was increased in spring (38 to 59 head per ha) than summer and autumn (13 to 32 head per ha). The average grazing intensity of Elk stags ranged from 21 to 34 head per ha, which is affected by supplementary feeding levels. This result suggests that feeding supplementary diet at 1.5 % of body weight was needed to maintain the stable wight gain in antler growing periods and control the proper grazing intensity of Elk deer stags.

Studies on the Forage Production and Utilization on Paddy Field in Korea (한국에 있어서 답리작을 이용한 양질 조사료 생산기술)

  • The problems in the current domestic forage production were evaluated, and the prospective improvement was suggested in this paper. Grassland development in forest, production of high quality forages in upland and paddy land, efficient utilization of rice straw, development of new varieties of forages suitable for our environmental conditions and imported forages were described Among them, preferential production and utilization of forages using paddy field after rice harvest . should be enlarged for domestic supply of forages in Korea. Several studies were carried out to select the promising forage crops and barley cultivars for whole crop silage production, to determine productivity, nutritive value and production cost of forages produced in paddy field, and feeding effect of forages with Hanuwoo and milking cow for whole crop silage with forages produced in paddy field, 1999 to 2001, and also discussed restraint factors and activation plans for enlargement of forage production in paddy land. The promising forage crops in paddy field were rye and barley for Middle region, and rye, barley, early maturing Italian ryegrass and wheat for Southern region. The promising barley cultivars for whole crop silage in paddy field were Albori in Suwon, Keunalbori, Milyang 92, Saessalbori, and Naehanssalbori in Iksan, and Keunalbori, Albori, Naehanssalbori, and Saegangbori in Milyang, respectively. Silage production, quality and animal palatability of silage by trench and round bale were also compared. The production yields of whole crop barley silage(WBS) were 17,135kg as a fresh matter, and 6,011kg as a dry matter per ha, and the quality of WBS was 2∼3 grade, while that of rice straw silage was 4 grade as a farm basis. The production cost of WBS per kg was 83won as a fresh matter, and 238won as a dry matter. Feeding of WBS as forages on Hanwoo was very desirable for the improvement of live-weight gain, beef quality and farm income, particularly in growing stage of Hanwoo. Milk production and income were also increased, and feed cost was decreased by feeding of WBS. The daily voluntary intake of WBS in milking cow was 26.3kg as a fresh matter(DM 7.7kg) per head. Milk production when WBS was fed, was very similar to that of imported hay feeding such as Kentucky bluegrass or domestic corn silage. The issues to be solved in near future f3r stable forage production and supply in paddy land are sustainable livestock-forages policy, development & seed production of new varieties of barley, rye, Italian ryegrass and other promising forages, efficient demand & supply system of forages, solidification for mass production and utilization of forages, efficient application management of animal slurry on paddy field considering environmental agriculture/livestock industry, and break k development of bottleneck technique in production field. Domestic production & supply of high cost agricultural machine (round baler, wrapper, handler and so on), plastic wrapping film, and silage additives are also important.

The Effect of Food Deprivation Length of Pair House Pigs on the Running Speed and Feeding Activity in Solitary and Social Conditions

  • The purpose of this experiment was to study the feeding behavior and running speed under various feed deprivation lengths and social environments. Three trials were conducted. Trial 1: ten pigs were trained individually to run a course and eat their feed at the end of the course. The pigs were deprived feed for 1, 5, 10 or 20 h. Trial 2: 1. Two pigs ran and ate together. Both pigs had 5 h of feed deprivation before the run (D5). 2. Two pigs ran and ate alone, but both pigs had 5 h of feed deprivation before the run (S5). 3. Two pigs ran and ate together. Both had 1 h of feed deprivation before the run (D1). 4. Two pigs ran and ate alone and both pigs had 1 h of feed deprivation before the run (S1). 5. Two pigs ran together, one had 5 h of feed deprivation, and the other had 1 h of feed deprivation before the run (51). Trial 3: 1. On the 1st day 5 pairs of pigs had 5 h feed deprivation and could eat feed together at (B) point (D1). 2. On the 2nd day the pigs ran and ate alone at (B) point after 5 h of feed deprivation. Feed was obtainable (D2). On the 3rd to 6th days, the pigs ran in pairs after 5 h of feed deprivation and only the dominant pig ate feed at point (B). The inferior pig was chased back to room and fed there. This stage was continued for four consecutive days, d 3 to 6. In trial 1, the running speed of pigs increased with the length of feed deprivation until 10 h, then being stable afterwards. Total feeding time increased with the length of feed deprivation (p<0.001). Eating speed did not increase with the length of feed deprivation (p>0.05). In trial 2, nine of ten pigs in treatment D5 ran faster than those in S5. Seven of the ten pigs in treatment S1 ran faster than those in treatment D1. The pigs in treatment D5 had significantly higher feed intake (p<0.001) and eating speed (p<0.05) than the pigs in other treatments. In trial 3, there were significant differences on running speed between D1 and D6 (p<0.01) and between D2 and D1, D3, D4 and D5. The inferior pig ran faster in D2 but from 3 to 6 it was the dominant pig that showed the greatest speed in completing the whole course. The results demonstrated that the pigs with low feeding motivation may cause low running speed to feed and low feed intake of the neighbor when compared with pigs kept individually.

Design of Miniaturization Terminal Antenna for 2.4 GHz WiFi Band with MZR (MZR을 이용한 2.4 GHz WiFi 대역 소형 단말기 안테나 설계)

  • In this paper, we implemented an on-board miniaturization antenna operating 2.4 GHz using MZR(Mu Zero Resonator). It is must be operating under the constraint that the size of the small terminal PCB should be $78{\times}38{\times}0.8mm^3$ and the size of the system should be $63{\times}38{\times}0.8mm^3$ and the size of the radiating part should be $15{\times}38{\times}0.8mm^3$. The feeding structure uses a CPW structure for stable feeding and a feeding point at the upper left of the system board. A magnetic field coupling structure is used for coupling the feeding part and the antenna. The resonance frequency of the MZR is determined by the series inductance and capacitance of the cell, so the gap between the cells, the length of the cell, the length of the interdigital capacitor, and the spacing between the radiation part and the ground plane are analyzed. The antenna was designed and fabricated using the results. The total size of the antenna including the feed structure is $20.8{\times}9.0{\times}0.8mm^3$, and the electrical length is $0.1664{\lambda}_0{\times}0.072{\lambda}_0{\times}0.0064{\lambda}_0$. The measurement result for 10 dB bandwidth, gain and directivity are 440 MHz(18.3%), 0.4405 dB, and 2.722 dB respectively. It is confirmed that the radiation pattern has omnidirectional characteristics and it can be applied to ultra small terminal antenna.

  • The basic mechanism of the granular sludge formation which is the most important factor in the start-up and stable operators is not confirmed yet. In this study, the effect of granular sludge formation was investigated with the different substrate concentrations and the various ratios of substrate supply/deficiency. The granular sludge formation in the UASB reactor was closely related to the substrate concentrations and the ratio of substrate supply/deficlency The granular sludge formation was not accelerated at low substrate concentration. It was convinced that granular sludge formation was accelerated when the substrate supply with high concentration was stopped at UASB reactor. From this experiment, it was estimated that granular sludge was formed by the combination of hydrogen utilizing bacteria that form hydrogen condition and acid forming bacteria at substrate deficit condition by mutual symbiosis. Though the removal efficiency of organic matter was decreased as the influent substrate concentration was Increased, the higher the influent substrate the better the granular sludge formation.

Thermal Characteristic Analysis of a High-Precision Centerless Grinding Machine for Machining Ferrules (페룰 가공용 초정밀 무심 연삭기의 열 특성 해석)

  • To perform the finish grinding process of ferrules which are widely used as fiber optic connectors, a high-precision centerless grinding machine is necessary. The high-precision centerless grinding machine is consisted of the hydrostatic GW and RW spindle systems, hydrostatic RW feeding mechanism, RW swivel mechanism, on-machine GW and RW dressers, and concrete-filled steel bed. In this study, the thermal characteristics of the high-precision centerless grinding machine such as the temperature distribution, temperature rise and thermal deformation, are estimated based on the virtual prototype of the grinding machine and the heat generation rates of heat sources related to the machine operation conditions. The reliability of the predicted results is demonstrated by the temperature characteristics measured from the physical prototype. Especially, the predicted and measured results show the fact that the high-precision centerless grinding machine has very stable thermal characteristics.
