Evaluation of liquid and powdered forms of polyclonal antibody preparation against Streptococcus bovis and Fusobacterium necrophorum in cattle adapted or not adapted to highly fermentable carbohydrate diets

  • Objective: Feed additives that modify rumen fermentation can be used to prevent metabolic disturbances such as acidosis and optimize beef cattle production. The study evaluated the effects of liquid and powdered forms of polyclonal antibody preparation (PAP) against Streptococcus bovis and Fusobacterium necrophorum on rumen fermentation parameters in ruminally cannulated non-lactating dairy cows that were adapted or unadapted to a high concentrate diet. Methods: A double 3×3 Latin square design was used with three PAP treatments (control, powdered, and liquid PAP) and two adaptation protocols (adapted, unadapted; applied to the square). Adapted animals were transitioned for 2 weeks from an all-forage to an 80% concentrate diet, while unadapted animals were switched abruptly. Results: Interactions between sampling time and adaptation were observed; 12 h after feeding, the adapted group had lower ruminal pH and greater total short chain fatty acid concentrations than the unadapted group, while the opposite was observed after 24 h. Acetate:propionate ratio, molar proportion of butyrate and ammonia nitrogen concentration were generally greater in adapted than unadapted cattle up to 36 h after feeding. Adaptation promoted 3.5 times the number of Entodinium protozoa but copy numbers of Streptococcus bovis and Fibrobacter succinogens genes in rumen fluid were not affected. However, neither liquid nor powdered forms of PAP altered rumen acidosis variables in adapted or unadapted animals. Conclusion: Adaptation of cattle to highly fermentable carbohydrate diets promoted a more stable ruminal environment, but PAP was not effective in this study in which no animal experienced acute or sub-acute rumen acidosis.

  • For the development of combustion technology of RDF(refuse derived fuel), combustion characteristics are examined in bubbling fluidized bed, circulating fluidized bed, continuos combustor and batch type combustor. The characteristics of combustion and exhaust gas has been compared and analyzed in many type of combustion facilities, which has been utilized as basic data for the advanced research of specified RDF combustion facility. Stable combustion has been observed in bubbling and circulating fluidized bed from controled operating condition like the proper feeding rate and superficial gas velocity. In circulating fluidized bed, concentration of NOx has been increased with the operating condition by the fuel-NO and oxygen reaction and $SO_2$ can be considered not to be produced in RDF fluidized bed from very low concentration in flue gas. HCl concentration is 36.4 ppm as average value and lower than standard emission value, but the counter plan is needed. Shaped RDF and fluff RDF have been compared in continuos combustor and batch type combustor and shaped RDF shows benefit for the stable heat recovery and gas emission shows similar value and characteristics.

Study on a broadband quasi-Yagi antenna for mobile base station (이동통신 기지국용 광대역 quasi-Yagi 안테나에 관한 연구)

  • In this paper, a method for the improvement in the gain and bandwidth of a microstrip-fed broadband planar quasi-Yagi antenna (QYA) is studied. The broadband characteristics of the QYA are achieved from the coplanar strip-fed planar dipole driver and a parasitic director close to the driver. In order to obtain stable gain variation over the required frequency band, a director and a ground reflector are appended to the driver having a nearby parasitic director. The QYA is fed through an integrated balun composed of a microstrip line and a slot line which are terminated in a short circuit. By adjusting the feeding point, a broadband impedance matching is obtained. A QYA with an operating frequency band of 1.75-2.7 GHz and a gain > 4.5 dBi is designed and fabricated on an FR4 substrate with dielectric constant of 4.4 and thickness of 1.6mm. The experimental results show that the fabricated antenna has good performance such as a broad bandwidth of 59.7%(1.55-2.87 GHz), a stable gain between 4.7-6.5 dBi, and a front-to-back ratio > 10 dB. The measured data agree well with the simulation, which validates this study.

A Preliminary Study of the Effect of Pelagic Organisms on the Macrobenthic Community in the Adjacent East China Sea and Korea Strait (표영생물이 동중국해 주변 해역과 대한해협의 대형저서동물 군집에 미치는 영향 파악을 위한 선행 연구)

  • Despite the impacts of the climate changes on the pelagic ecosystem, few studies have examined the pelagic-benthic coupling in the adjacent East China Sea and Korea Strait. Therefore, the species composition and abundance of the macrobenthic community, as well as the potential food sources of benthic fauna were investigated in the present study using stable isotope analysis (${\delta}^{13}C\;and\;{\delta}^{15}N$) for suspended particulate organic matter (SPOM), sedimentary organic matter (SOM), phytoplankton, and zooplankton. A total of 157 macrobenthic fauna were collected, and the density of the macrobenthic fauna ranged from 4 to 434 ind./0.25 $m^2$, with an average density of 149 ind./0.25 $m^2$. The density of the benthic fauna increased moving from offshore shelf sites to coastal sites adjacent to the Korea Strait. Cluster analysis showed that the macrobenthic communities consisted of three distinct groups: group A in the Korea Strait, group B in the East China Sea, and group C near Ieodo. The dominant species in group A were the amphipods Photis japonica and Ampelisca miharaensis, followed by the polychaete Scolotoma longifolia. Environmental variables, such as the temperature of the seawater and sediment, and oxygen, and chlorophyll a levels, appeared to affect the structure of the community, suggesting the importance of coupling with the pelagic system. The ${\delta}^{13}C$ values of SPOM and zooplankton ranged from -22.97 to -23.5% and -19.92 to -21.86%, respectively, showing a relatively narrow range(<1%) between the two components. The difference between the ${\delta}^{13}C$ values of SOM and pelagic organic matter was also within 1%, suggesting that the SOM originated from the pelagic system, which is an important factor controlling the macrobenthic community.

  • Rice (Oryza sativa L.) is the most important crop and its yield must be improved to feed the increasing global population. Recently developed high-yielding varieties with extra-large sink capacity often have a problem in unstable grain-filling. Therefore, understanding limiting factors for improving grain-filling and controlling them are essential for further improvement of rice grain yield. However, since grain-filling rate was determined by complex sink-source balance, the ability of grain-filling was very difficult to evaluate. Source ability for 'grain' was not only determined by the ability of carbon assimilation in leaves, but also that of carbon translocation from leaves to panicles. Sink strength was determined by the complex carbon metabolism from sucrose degradation to starch synthesis. Hence, to evaluate the grain-filling ability and determine its regulatory steps, the whole picture of carbon flow from photosynthesis at leaves to starch synthesis at grains must be revealed in a metabolite level. In this study, the yield and grain growth rate of three high-yielding varieties, which show high sink capacity commonly, were compared. Momiroman showed lower grain filling rate and slower grain growth rate than the other varieties, Hokuriku 193 and Tequing. To clarify the limiting point in the carbon flow of Momiroman, $CO_2$ labeled by stable isotope ($^{13}C$) was fed to three varieties during ripening period. The ratio of $^{13}C$ left in the stem was higher in Momiroman 24 hours after feeding, suggesting inefficient carbon translocation of Momiroman. More interestingly, $^{13}C$ translocation from soluble fraction to insoluble one in the grain seemed to be slower in Momiroman. To get the further insight in a metabolite level, we are now trying the $^{13}C$ labeling metabolome analysis in the developing grains.

Change of fatty acid compositions of rotifer according to enrichment diets and methods in the high density culture (고밀도 배양에 있어서 영양강화 방법 및 종류에 따른 rotifer의 지방산 조성의 변화)

  • This study was carried out to compare the growth and fatty acids composition of the rotifer (Brachionus rotundiformis) cultured in high density by the various enrichments and culture methods. The rotifer fed on condensed freshwater Chlorella was enriched with $\omega$-yeast, Algamac, Super Selco and marine Chlorella. In another culture method, the rotifer was cultured with enrichment supplements for 6 hours after feeding with condensed freshwater Chlorella supplement for 18 hour. The rotifer fed with condensed marine Chlorella for 24 hours without fieshwater Chlorella was used as a control group. Culture tanks (5 $\ell$ working volume) was immersed in a water bath ($28^{\circ}C$). The density of rotifer and dissolved oxygen level in water was stable in control group of rotifer cultured with condensed marine Chlorella for 24 hours and the n-3 HUFA content of rotifer was the highest among the rotifer culture methods. However, the density of rotifer and dissolved oxygen level in the groups of rotifers enriched with $\omega$-yeast, Algamac and Super Selco by methods were drastically decreased, The n-3 HUFA contents of rotifers enriched by Super Selco were higher than those of rotifer enriched by either $\omega$-yeast or Algamac in both methods. The results from this experiment indicated that supplementation of condensed marine Chlorella for 24 hour by the semi-continuous culture was effective for the improvement of the nutritional value of rotifer and it could provide the stable growth condition for rotifer culture in high density.

Habitat Characteristics and Management of Abandoned Rice Paddy Field Wetlands in Mountain - In Case of the Uldae Wetland in Bukhansan National Park - (도시 내 묵논습지 생물서식 특성 및 관리방안 -북한산국립공원 울대습지를 대상으로-)

  • The purpose of this study is to identify the ecological characteristics and biological interactions between species of the abandoned rice paddy field in mountainous areas and to suggest a management strategy for stable food chain formation and biodiversity enhancement. The study site is located in Uldae wetland of Songchu district Bukhansan National Park, site characteristics and biological habitat characteristics were identified through site survey and literature survey. With regard to physical environment, among geographical features, the Uldae Wetland and the neighborhood inside the basin was a gently sloping area($5{\sim}15^{\circ}$). And 64.0% of basin faced the north. With regard to water environment, the Uldae Wetland was wetland of rainfed paddy field depending on precipitation and the system of stream flowing into the wetland from valley. According to the results of examining flora in plant ecology, in general, they were herbaceous wetland species. 88.6% of existing plants inside the Uldae Wetland basin was a forest in the mountain. And Quercus spp. community and Pinus densiflora community accounted for 64.6% of that, and was dominant. Except for that, Salix koreensis community was distributed. The existing vegetation of Uldae Wetland inhabited wetland species and terrestrialization indicator species, and it was thought that partial terrestrialization inside the Uldae Wetland was in progress after the discontinuation of paddy cultivation, such as the expansion of Salix koreensis distribution area. In the status of appearing faunae in the Uldae Wetland with regard to wildbirds of appearing principal species, The Uldae wetland was based on a abandoned rice paddy field various wildlife, and was a wildlife feeding, spawning, and resting place. The water environment was an important factor in maintaining the wetland living creatures function, habitat of waterbirds and benthic macroinvertebrates, amphibians and odonate are spawning ground and habitat, it was affecting the vegetation ecosystem based on wetlands. In order to maintain the diversity of wildlife, it was important to maintain smooth water supply and water level. A stable food chain will be formed and the Uldae wetland biodiversity will be abundant by establishing the relationship between the species of Uldae wetland, which is abandoned rice paddy field, and the habitat environment favored by species belonging to the ecosystem stepwise linkage. The ecological characteristics of the Uldae wetlands and the relation between the species were analyzed and the environmental conditions were reflected in the planning and management plan of Uldae wetland ecology.

The Effects of Additives in Napier Grass Silages on Chemical Composition, Feed Intake, Nutrient Digestibility and Rumen Fermentation

  • The effect of silage additives on ensiling characteristics and nutritive value of Napier grass (Pennisetum purpureum) silages was studied. Napier grass silages were made with no additive, fermented juice of epiphytic lactic acid bacteria (FJLB), molasses or cassava meal. The ensiling characteristics were determined by ensiling Napier grass silages in airtight plastic pouches for 2, 4, 7, 14, 21 and 45 d. The effect of Napier grass silages treated with these additives on voluntary feed intake, digestibility, rumen fermentation and microbial rumen fermentation was determined in 4 fistulated cows using $4{\times}4$ Latin square design. The pH value of the treated silages rapidly decreased, and reached to the lowest value within 7 d of the start of fermentation, as compared to the control. Lactic acid content of silages treated with FJLB was stable at 14 d of fermentation and constant until 45 d of ensiling. At 45 d of ensiling, neutral detergent fiber (NDF) and acid detergent fiber (ADF) of silage treated with cassava meal were significantly lower (p<0.05) than the others. In the feeding trial, the intake of silage increased (p<0.05) in the cow fed with the treated silage. Among the treatments, dry matter intake was the lowest in the silage treated with cassava meal. The organic matter, crude protein and NDF digestibility of the silage treated with molasses was higher than the silage without additive and the silage treated with FJLB. The rumen parameters: ruminal pH, ammonia-nitrogen ($NH_3$-N), volatile fatty acid (VFA), blood urea nitrogen (BUN) and bacterial populations were not significantly different among the treatments. In conclusion, these studies confirmed that the applying of molasses improved fermentative quality, feed intake and digestibility of Napier grass.

Characterization and Enhanced Production of Enterocin HJ35 by Enterococcus faecium HJ35 Isolated from Human Skin

  • A strain named as HJ35 was isolated from the skin of sixty-five men and fourteen women for acne therapy, in order to find an effective antimicrobial agent against Propionibacterium acnes. Isolate HJ35 was identified as Enterococcus faecium based on 16 rDNA sequence and produced enterocin HJ35 having antimicrobial activities against most lactic acid bacteria, En­terococcus spp., Staphylococcus aureus, S. epidermidis, Clostridium perfringens, some bacilli, Mi­crococcus flavus, Listeria monocytogenes, L. ivanovii, Escherichia coli, Pseudomonas fluorescens and Propionibacterium acnes, in the modified well diffusion method. Especially, enterocin HJ35 showed a bactericidal activity against Propionibacterium acnes P1. The antimicrobial activity of enterocin HJ35 was disappeared completely with the use of protease XIV. But enterocin HJ35 activity is very stable at high temperature (up to $100^{\circ}C$ for 30 min), in wide range of pH (3.0${\~}$9.0), and by treatment with organic solvents. The apparent molecular mass of enterocin HJ35 was estimated to be approximately 4${\~}$4.5 kDa on detection of its bactericidal activity after SDS-PAGE. In batch fermentation of E. faecium HJ35, enterocin HJ35 was produced at the mid­log growth phase, and its maximum production was obtained up to 2,300 AU/mL at the late stationary phase. By employing fed-batch fermentation, the enhanced production of enterocin HJ35 was achieved up to 12,800 AU/mL by feeding with 10 g/L glucose or 6 g/L lactate.

Tissue Culture Studies of Anthranilate Synthase the Tryptophan Biosynthetic Control Enzyme

  • Experiments initiated 30 years ago to obtain selectable markers have led to a series of studies of Trp biosynthesis and anthranilate synthase (AS) the control enzyme using largely plant tissue cultures since they have experimental properties that can be readily exploited. Enzymological and compound feeding studies provided evidence that AS is the control point in the Trp biosynthesis branch and that altering the AS feedback control by the selection of mutants resistant to the Trp analog 5-methyl-tryptophan (5MT) can lead to the overproduction of this important amino acid. Plants regenerated from these Trp overproducing lines of most species also had high free Trp levels but Nicotiana tabaum (tobacco) plants expressed the feedback altered AS only in cultured cells and not in the regenerated plants. further tests by transient and stable expression of the cloned promoter for the naturally occurring tobacco feedback-insensitive AS, denoted ASA2, confirmed the tissue culture specific nature of the expression control. The 5MT caused by the expression of a feedback-insensitive AS from tobacco has been used to select protoplast fusion hybrids with several species since the resistance is expressed dominantly. Recently the ASA2 gene has been used successfully as a selectable marker to select transformed Astragalus sinicus and Glycine max hairy roots induced by Agrobactetium rhizogenes. These results show that the ASA2y-subunit can interact with the y-subunit of another species to form active feedback-insensitive enzyme that may be useful for selecting transformed cells. Plastid DNA transformation of tobacco has also effectively expressed ASA2 in the compartment in which Trp biosynthesis is localized in the cell.
