Physiological Characteristics of Biosurfactant-Producting Bacillus subtilis TBM 3101 (Biosurfactant를 생산하는 Bacillus subtilis TBM 3101의 생리학적 특성)

  • A biosurfactant-producing strain, Bacillus subtilis TBM 3101 was isolated from the soil sample at Tae-Baek Mountain through an antifungal test and emulsification assessment. The strain was assessed, regarding to the microbial growth, by physical and chemical test, surface tension, emulsification activity and stability. The surface tension of the isolate sharply decreased to the minimum 29mN/m at 48 h growth. Of note, its emulsification was stabilized to the highest degree when tributyrin was utilized as a substrate, indicating that in comparison to a variety of synthetic surfactants, the biosurfactant produced by the isolate was significantly similar to synthetic surfactant, tween 20. In addition, the biosurfactant showed high emulsification activity when soybean oil, crude oil and tetradecane were used as a substrate. Thus, these studies could contribute to the detection and development of biosurfactant beneficial to the environment and humans.

Geomicrobiological Behavior of Heavy Metals in Paddy Soil Near Abandoned Au-Ag Mine Supplied with Carbon Sources (탄소원을 공급한 폐금은광산 주변 논토양 내 중금속의 지구미생물학적 거동 연구)

  • The study was conducted to investigate the effects of indigenous bacteria on geochemical behavior of toxic heavy metals in contaminated paddy soil near an abandoned mine. The effects of sulfate amendment to stimulate microbial sulfate reduction on heavy metal behaviors were also investigated. Batch-type experiments were performed with lactate or glucose as a carbon source to activate indigenous bacteria in the soil under anaerobic condition for 100 days. Sulfate (250 mg/L) was artificially injected at 60 days after the onset of the experiments. In the case of glucose supply, solution pH increased from 4.8 to 7.6 while pH was maintained at 7~8 in the lactate solution. The initial low pH in the case of glucose supply likely resulted in the enhanced extraction of Fe and most heavy metals at the initial experimental period. Lactate supply exerted no significant difference on the amounts of dissolved Zn, Pb, Ni and Cu between microbial and abiotic control slurries; however, lower Zn, Pb and Ni and higher Cu concentrations were observed in the microbial slurries than in the controls when glucose supplied. Sulfate amendment led to dramatic decrease in dissolved Cr and maintenance of dissolved As, both of which had gradually increased over time till the sulfate injection. Black precipitates formed in solution after sulfate amendment, and violarite($Fe^{+2}{Ni^{+3}}_2S_4$) was found with XRD analysis in the microbial precipitates. Conceivably the mineral might be formed after Fe(III) reduction and microbial sulfate reduction with coprecipitation of heavy metal. The results suggested that heavy metals which can be readily extracted from contaminated paddy soils may be stabilized in soil formation by microbial sulfate reduction.

Effect of Pyroligneous Acid Liquor on the Maturity of Pig Manure Compost (목초액 처리가 돈분퇴비의 부숙도에 미치는 영향)

  • To investigate the effect of pyroligneous acid liquor (PAL) on the maturity of pig manure compost, PAL was treated to the compost piles. The treatments included applications of 100 and 300 times diluted PAL in addition to the control. The compost piles were stirred in three times at the 1st, 10th, and 25th day of composting. Temperature in the compost pile of control treatment increased from 28 to $60^{\circ}C$ within 10 days and remained nearly at the level until 30th day, then it began to decrease. On the other hand, temperature of the PAL-100 and PAL-300 treatments reached 65 to $70^{\circ}C$ within 8 days and became stabilized until 30th day, then the temperature decreased to about $28^{\circ}C$. However, the temperature of control was stabilized approximately at the 40th day. Initially, the pH of control treatment dropped from 8.2 to slightly above pH 8.0 during 4th day, but that of the PAL-100 treatment declined to 7.8. Among treatments, pH value of PAL-100 treatment was the lowest, which was about 7.3 after becoming stabilization. Also the germination index (GI) was increased at all treatments. The C/N ratio range of PAL-100 treatment was better balanced than others and was at 24.3. Moreover the round paper chromatogram of extracted solution of compost of PAL-100 treatment was the sharpest and clearest among treatments. The GI values of control, PAL-100, and PAL-300 in 60 days of composting were about 108, 120, and 118 in germination test using chinese cabbage, respectively. It can be concluded that the addition of diluted PAL solution is effective in composting of pig manure.

The Effects of Kinetics on the Leaching Behavior of Heavy Metals in Tailings-Water Interaction (광미-물 상호반응에서 반응시간이 중금속 용출에 미치는 영향)

  • Experimental leaching of tailings was performed as a function of times (1, 2, 4, 7, 14, 21 and 30 days) in the laboratory using reaction solutions equilibrated to three different pH set-points (pHs 1,3 and 5). The initial pHs of 5 and 3 stabilized at either 4.6-6.1 or 2.8-3.5 in 2 days and decrease gradually with time afterwards. The results of the leaching tests indicate that the significant increase in the sulfate concentrations and in acidity after 7 days of leaching results from the oxidation of sulfide minerals. There were no significant variations in the extractable Pb found in the leach solutions of pH 5 and 3 within the reaction time (1-30 days), while Zn, Cd and Cu concentrations tend to significantly increase with time. In tailings leaching at an initial pH=1, two trends were observed: i) The 'Zn-type' (Zn, Cd and Cu), with increasing concentrations between days 1 and 30, corresponding to the expected trend when continuous dissolution is the dominant process, ii) the 'Pb-type' (Pb), with decreasing concentrations over time, suggesting rapid dissolution of a Pb source followed by the precipitation of 'anglesite' in relation to the large increase in dissolved sulfates. The high sulfate concentrations were coupled with high concentrations of released Fe, Zn and Cd. Release of Zn and Cd and acidity from these leaching experiments can potentially pose adverse impact to surface and groundwater qualities in the surrounding environment. The kinetic problems could be the important factor which leads to increasing concentrations of trace metals in the runoff water.

Stabilization of Heavy Metals-contaminated Soils Around the Abandoned Mine area Using Phosphate (인산염을 이용한 휴.폐광산 주변 중금속 오염토양의 안정화처리에 관한 연구)

  • The objective of this study was to evaluate the efficiency of $(NH_4)_2HPO_4$, $Na_2HPO_4{\cdot}12H_2O$, $CaHPO_4{\cdot}2H_2O$, $Ca(H_2PO_4)_2{\cdot}H_2O$ and $H_3PO_4$ for the stabilization of soils contaminated with multi-metals containing Pb, Cd and As. The application rate of stabilizers to soils was determined based on $PO_4/Pb_{total}$ molar ratio of 0.5, 1, 2, 4. The results of Korea Standard Test and TCLP (EPA Method 1311) showed the reduction of metal leachabilities below the regulatory limits for Pb and Cd when $H_3PO_4$ and $Ca(H_2PO_4)_2{\cdot}H_2O$ were applied. However, stabilization efficiency for Cd was low and in case of As leaching concentration increased rather. It is considered that $PO_4$ reacted effectively $Pb^{2+}$ due to leaching Pb under low pH condition created by adding $H_3PO_4$. Accordingly Pb was stabilized by dissolution and precipitation of hydroxypyromorphite. From the change of metals fraction using sequential extraction procedure when $H_3PO_4$ applied as a stabilizer, we confirmed that residual fraction increased more than 60% and this result was accorded with XRD analysis that detected only hydroxypyromorphite peak in $H_3PO_4$.

A study for conservation of plant-based cultural properties : on the subject of straw sandals excavated at Goongnamji in Buyeo (초본류 문화재의 보존처리를 위한 연구 -부여 궁남지 출토 짚신을 대상으로-)

  • Plant-based cultural assets using straw and grass as household goods of our people's have been used as indispensable tools for practical living for a very long time. However, only a limited number of artifacts were unearthed so far due to tile fragility of the material. For this reason, research on plant-based cultural properties had close to no progress, and the appropriateness of the PEG method, high-grade alcohol method, alcohol--ether-resin method, and Paraloid B-72 used in preserving plant-based cultural properties has not been sufficiently investigated. Therefore, this study examined the weight change rate by applying the methods of Primal MC-76 and vacuum freeze-drying used mostly as a earth-layer hardening material among PEG and acrylic resin, which are applied widely for preservation of waterlogged archaeological wood, as a means to preserve plant-based cultural properties along with the examination of the subject material, and an experiment was also performed on moisture absorption. The findings as a result were, first, the plant-based material being studied was found to be Typha (Typha orientalis Presl). Secondly, the weight change experiment applying $PEG\#400$ and $PEG\#4000$ confirmed a steady increase of weight if PEG -2Step is used for treatment. Third, in preserving all subject materials with soil, treatment with $PEG\#4000$, Primal MC-76, and vacuum freeze-drying showed that tile vacuum freeze-drying method resulted in the largest or $20\%$ reduction in weight, while Primal MC-76 resulted in $18\%$ and $PEG\#4000$ in $8\%$ of weight reduction. It was concluded that, considering the stability of soil measurement, this came to be because resin permeation was carried out along with tile drying process. Fourth, the weight changes were found to be around $10\%$ in various humidity conditions after the preservation treatment. The greatest weight change rate was seen in the case of $PEG\#4000$, particularly having chemicals gush out in a high humidity (RH $84\%$ or higher) environment. In the case of Primal MC-76 and vacuum freeze-drying methods, $6\~8\%$ weight changes were detected, and the lowest weight change was found in the case of the vacuum freeze-drying method. Fifth, as for color changes after treatment, blackening occurred most strongly with $PEG\#4000$, while Primal MC-76 and vacuum freeze-drying manifested colors closest to dry straw or grass. However, the texture of straw was not very evident in the case of Primal MC-76, due to a glossy surface, but vacuum freeze-drying was found to offer tile best result in terms of texture. Putting together the results of the above experiments, vacuum freeze-drying presented after being treated with PEG2-Step the most stabilized changes in weight, while it offered the smallest change in color as well.

Changes of Physico-chemical Properties and Microflora of Pig Manure due to composting with some Bulking Agents (보조재료별 돈분 퇴비화 과정중 이화학적 특성과 미생물상 변화)

  • This study was conducted to investigate physicochemical and microbiological properties during composting process when pig manure was composted with some bulking agents(Saw dust, Rice hull, Crushed wood). The pig manure consisted of 2.95% total nitrogen, 4.55% $P_2O_5$, 2.07% $K_2O$, 81.2% organic matter and 14.0 C/N, dry base. The inorganic content of bulking agents were similar one another, and C/N ratio was Saw dust 392. Rice hull 91.5, Crushed wood 266. The temperature of Saw dust composting slowly increased at initial stage, whereas one of Rice hull and Crushed wood rapidly increased and stabilized similarly to outdoor temperature after about 2 month. The pH of compost increased during initial 1 month after decreasing, but thereafter decreased rapidly. The C/N ratio rapidly decreased at initial stage, and it slowly decreased after 1 month. Total nitrogen somewhat increased according to composting process. Ammonium-nitrogen increased until 60th day, and thereafter it was rapidly inclined to decrease. The population of aerobic bacteria, Bacillus spp., actinomycetes and fungi increased up to > $10^{10}cfu\;g^{-1}$ fresh weight, > $10^7$, > $10^8$ and > $10^6$, respectively in Rice hull and Crushed wood composting, and more than ones in Saw dust composting, and specially actinomycetes. The persuasive Bacillus spp. isolated from composting process were B. lentimorbus, B. licheniformis, B. pumilus, B. megaterium, etc.

Study on the Lowering of NaCl Content by Co-composting Food Wastes (저농도 식염을 함유하는 음식물쓰레기 퇴비제조)

  • High salt content in food waste limit use as the potential source of compost. Since sodium chloride content should be lower than one percent to meet requirement for agricultural use of compost, we attempted use of three kinds of co-composting material for food wastes compost, which were fermented, dried, and fresh types of amendments which included pig manure, sawdust and puffed rice hull. Food wastes were composted well and stabilized after around the 40th days of composting. Little difference were found between composts amended sawdust and puffed rice hull. The result indicated that puffed rice hull could be used as a good alternative of sawdust. Because sawdust or puffed rice hull were mixed as amendments to control water contents and to dilute high NaCl content of compost material, the C/N ratios of the final products were significantly high over 40. However, NaCl contents of final products were dropped to less than percent on the fresh weight basis by mixing food wastes and the above amendments with the optimum rates.

Swelling Patterns and relevant Hydraulic Conductivities of Na-Bentonite under Various Acidic and Alkali conditions (산과 알칼리 조건하에서 Na-벤토나이트의 팽창경향과 상대적 수리전도도)

  • We investigated the effects of solution pH on swelling characteristics and relevant hydraulic conductivity of different particle sizes of Na-bentonite which have significantly high swelling capacity. The results showed that the time taken to reach the maximum swelling indexes for all pH levels ranged from 84 hours and 156 hours for pH 6.5 or above by NaOH and pH 3.5 by HCl, respectively. The maximum swelling index slightly increased with increasing particle size, while the maximum swelling indexes were less or approximately half of that of the indigenous Na-bentonite. The changes in swelling indexes before and after solution treatment were distinctive in acidic condition, especially in pH 3.5 by HCl while there were not much differences in alkali condition. For hydraulic conductivities of fully swelled Na-bentonite in a given solution pH, elution did not occur under pressure below 1 bar. But elution started as the pressure was raised to 1.5 bars or above after 500 seconds. The stabilized hydraulic conductivities observed from 1.5, 3.0, and 5.0 bars ranged from $7{\times}10^{-3}cm\;day^{-1}$ to $6{\times}10^{-3}cm\;day^{-1}$, indicating that the hydraulic conductivities were slightly higher in acidic condition than that of normal condition.

Analysis of Streamflow Characteristics of Boryeong-dam Watershed using Global Optimization Technique by Infiltraion Methods of CAT (CAT 모형의 침투해석방법별 전역최적화기법을 이용한 보령댐 유역의 유출 특성 변화 분석)

  • In this study, the changes of the streamflow characteristics of the watershed were analysed depending on the infiltration methods of CAT. The study area, Boryeong-dam watershed located in Chungcheongnam-do area, has been suffered from severe drought in recent years and stabilized regarding on the storage rate through efforts such as constructing a channel connecting the upstream of Boryeong-dam from the downstream of the Geum river. In this study, the effects of soil infiltration parameters on the watershed streamflow characteristics were analyzed by the infiltration methods of CAT such as Rainfall Excess, Green&Ampt and Horton. And the parameter calibrations were conducted by SCEUA-P, a global optimization technique module of the PEST, the package for parameter optimization and uncertainty analysis, to compare the yearly variations of soil parameters for infiltration methods of CAT. In addition, the streamflow characteristics were analyzed for three infiltration methods by applying three different scenarios, such as applying calibrated parameters for every years to simulate the model for each years, applying calibrated parameters for the entire period to simulate the model for entire period, and applying the average value of yearly calibrated parameters to simulate the model for entire period.
