Molecular cloning was carried out on the Danish strain of bovine viral diarrhea virus(BVDV) to construct strategy for the diagnostic tools and effective vaccine of BVD afterwards. A recombinant DNA clone(No. 29) was established successfully from cDNA for viral RNA tailed with adenine homopolymer at 3'-end. $^{32}P$-labeled DNA probes of 300~1,800bp fragments, originating from the clone 29, directed specific DNA-RNA hybridization results with BVDV RNA. Recombinant DNA of the clone 29 was about 5,200bp representing 41.6% of the full length of Danish strain's RNA, and restriction sites were recognized for EcoR I, Sst I, Hin d III and Pst I restriction enzymes in the DNA fragment.
단일 분사 노즐을 갖는 연소기 내부의 혼합과정에 대한 이해를 바탕으로 연료 분사 노즐을 이중으로 갖는 연소기의 혼합과정에 대한 수치연구를 수행하였다. 수치연구를 위하여 3차원 Navier-Stokes 방정식과 k-$\omega$ SST난류 모델을 이용하여 연료 이중 분사 유동을 모사하였다. 이중 분사구 사이의 거리 변화에 따른 혼합특성의 변화를 살펴보기 위하여 파라메터 연구를 수행하였다. 연료 이중 수직분사에서 두 분사기의 유동 및 혼합특성은 서로 상당히 다른 경향을 보이며, 후방 분사기의 유동 및 혼합특성은 전방 분사류의 영향을 받아 더 크게 팽창하고 침투거리가 증가하는 것으로 나타났다. 어떤 특정 거리가 되기 전까지 분사기 사이의 거리가 증가할수록 전체적인 혼합률과 침투거리가 증가하는 등 혼합특성이 개선되지만, 특정 거리보다 크게되면 오히려 혼합특성이 악화되는 것으로 나타났다. 이는 이중분사기에서 최적인 혼합특성을 위한 두 분사기 사이의 거리가 존재함을 의미하는 것으로 판단된다.
해수면 온도는 전 세계 해양, 기상 현상에 영향을 주고 해양 환경 변화와 생물에게 영향을 주는 중요한 요소이다. 특히, 우리나라 남해안을 비롯한 연안 지역의 경우 어업 및 양식업 등의 수산업이 많이 발달하여, 매년 고수온 현상으로 인한 사회·경제적 피해가 발생하고 있다. 따라서 위성 자료와 같은 광범위한 지역을 감시할 수 있는 자료를 활용한 해수면 온도 및 공간적 분포의 예측기술 개발을 통하여 피해를 예방할 수 있는 시스템을 구축할 필요가 있다. 해수면 온도 예측은 기존의 수치 모델을 통해서 예측을 진행하였지만, 다수의 역학적 요인들을 사용하여 예측 결과 산출 시 복잡함이 존재한다. 최근 기계학습 및 딥러닝 기법이 발달함에 따라 해양 분야의 예측에 적용하는 연구가 진행되고 있다. 본 연구는 그 중 시·공간적인 일관성 및 정확도가 높은 장단기 기억(Long Short Term Memory, LSTM)과 합성곱 장단기 기억(Convolutional Long Short Term Memory, ConvLSTM) 딥러닝 기법을 사용하여 남해지역의 해수면온도 예측 및 2017년부터 2019년까지의 고수온 발생 건에 대해서 예측 결과의 공간 분포와 공간 분포와 예측 가능성에 대해 분석을 하였다. 1일 예측 모델의 정확도는 RMSE 기준으로 ConvLSTM(전체: 0.33℃, 봄: 0.34℃, 여름: 0.27℃, 가을: 0.32℃, 겨울: 0.36℃)이 LSTM 기반의 예측 모델(전체: 0.40℃, 봄: 0.40℃, 여름: 0.48℃, 가을: 0.39℃, 겨울: 0.34℃)보다 우수한 성능을 보였다. 2017년 고수온 발생 사례에 대해 해수면 온도 예측과 고수온 탐지 성능에서 ConvLSTM은 5일까지 경보를 탐지하였지만, LSTM의 경우 2일 예측 이후 해수면 온도를 과소 추정하는 경향이 커짐에 따라 탐지하지 못하였다. 시공간적인 해수면 온도 예측 시 ConvLSTM이 LSTM에 비해 적절한 모델로 판단된다.
수영만을 포함한 부산 지방 안개발생의 특징 및 안개 발생에 대한 부산 연안의 역할을 최근 5개년(1984~1988)의 기상 관측자료 및 지상 일기도, 그리고 해양관측자료를 사용하여 조사하였다. 또한 1989년 7월 한 달 동안 수영만에서 관측한 일기상 자료와 표면 수온을 부산지방의 일기상 자료, 표면수온과 비교하였다. 부산 지방의 안개는 5, 6, 7월에 집중적으로 발생하며, 안개 발생시의 주풍향은 수증기 공급이 용이한 남서풍 및 북동풍이었다. 안개 발생과 관련된 기압계 유형은 봄철(3, 4, 5월)이 pattern 7 (이동성 고기압형), 여름철(6, 7, 8월)이 pattern3(정체전선인 33$^{\circ}$N미만에 위치한 형)과 pattern 10(정체전선이 33~36$^{\circ}$N사이에 위치한 형)이었다. 또한 이류무의 발생은 전층 (850~500mb)에 걸친 대기 불안정도의 증가 보다는 하층 대기 (850~700mb)의 한.온 이류와 해표면의 난.한 수온과의 관계에 더 많은 관련성이 있었다. 내륙 지방인 대구 지방의 안개는 강한 야간 복사 냉각에 의해서 가을철 새벽에 발생하나 해안 지방인 부산은 남풍 계열의 바람이 부는 늦봄에서 여름철(5, 6, 7월)에 다량의 수증기 유입으로 높은 상대습도를 가지게 되어 대기가 약간의 복사 냉각에 의해서도 쉽게 과포화에 이르기 때문에 5, 6, 7월에 집중적으로 발생했다. 수영만과 부산지방의 일기상 자료, 표면 수온 및 안개발생일 등은 거의 비슷하며 안개 발생 mechanism 역시 거의 일치한다고 생각된다.
The purpose of this paper is to design a hydraulic actuator to operate under high pressure conditions. The flow characteristics under design conditions of hydraulic actuator were numerically conducted by commercial fluid dynamic code(ANSYS CFX V11). The numerical analysis was performed by transient technique according to the variation of stroke times, which was changed from 0 to 1 second by interval of 0.01. Turbulence model, $k-\omega$ SST was selected to secure more accurate prediction of hydraulic oil flow. The ICEM-CFD 11 and CFXMesher, reliable grid generation software was also adapted to secure high quality grid necessary for the reliable analysis. According to the simulation results, the flow rate which was supplied to the hydraulic actuator was 30.4l/min. These results are in good agreement with design results within 3.5% error.
In this study, the characteristics in the simulation of high-resolution coastal weather, i.e. sea surface wind (SSW) and significant wave height (SWH), were studied in a southeastern coastal region of Korea using the WRF and SWAN models. This analyses was performed based on the effects of various input factors in the WRF and SWAN model during M-Case (moderate days with average 1.8 m SWH and $8.4ms^{-1}$ SSW) and R-Case (rough days with average 3.4 m SWH and $13.0ms^{-1}$ SSW) according to the strength of SSW and SWH. The effects of topography (TP), land cover (LC), and sea surface temperature (SST) for the simulation of SSW with the WRF model were somewhat high on v-component winds along the coastline and the adjacent sea of a more detailed grid simulation (333 m) during R-Case. The LC effect was apparent in all grid simulations during both cases regardless of the strength of SSW, whereas the TP effect had shown a difference (decrease or increase) of wind speed according to the strength of SSW (M-Case or R-Case). In addition, the effects of monthly mean currents (CR) and deepwater design waves (DW) for the simulation of SWH with the SWAN model predicted good agreement with observed SWH during R-Case compared to the M-Case. For example, the effects of CR and DW contributed to the increase of SWH during R-Case regardless of grid resolution, whereas the differences (decrease or increase) of SWH occurred according to each effect (CR or DW) during M-Case.
This study investigates the accuracy of short-term ocean predictions during the development of ocean stratification for the Korea Meteorological Administration (KMA) Global Seasonal Forecast System version 5 (GloSea5) as well as the effect of atmosphere-ocean coupling on the predictions through a series of sensitive numerical experiments. Model performance is evaluated using the marine meteorological buoys at seas around the Korean peninsular (KP), Tropical Atmosphere Ocean project (TAO) buoys over the tropical Pacific ocean, and ARGO floats data over the western North Pacific for boreal winter (February) and spring (May). Sensitive experiments are conducted using an ocean-atmosphere coupled model (i.e., GloSea5) and an uncoupled ocean model (Nucleus for European Modelling of the Ocean, NEMO) and their results are compared. The verification results revealed an overall good performance for the SST predictions over the tropical Pacific ocean and near the Korean marginal seas, in which the Root Mean Square Errors (RMSE) were $0.31{\sim}0.45^{\circ}C$ and $0.74{\sim}1.11^{\circ}C$ respectively, except oceanic front regions with large spatial and temporal SST variations (the maximum error reached up to $3^{\circ}C$). The sensitive numerical experiments showed that GloSea5 outperformed NEMO over the tropical Pacific in terms of bias and RMSE analysis, while NEMO outperformed GloSea5 near the KP regions. These results suggest that the atmosphere-ocean coupling substantially influences the short-term ocean forecast over the tropical Pacific, while other factors such as atmospheric forcing and the accuracy of simulated local current are more important than the coupling effect for the KP regions being far from tropics during the development of ocean stratification.
To understand the characteristics of low-level clouds (CLs), environmental variables are composited on each CL using individual surface observations and six-hourly upper-air meteorologies around the globe. Individual CLs has its own distinct environmental conditions. Over the eastern subtropical and western North Pacific Ocean in JJA, stratocumulus (CL5) has a colder sea surface temperature (SST), stronger and lower inversion, and more low-level cloud amount (LCA) than the climatology whereas cumulus (CL12) has the opposite characteristics. Over the eastern subtropical Pacific, CL5 and CL12 are influenced by cold and warm advection within the PBL, respectively but have similar cold advection over the western North Pacific. This indicates that the fundamental physical process distinguishing CL5 and CL12 is not the horizontal temperature advection but the interaction with the underlying sea surface, i.e., the deepening-decoupling of PBL and the positive feedback between shortwave radiation and SST. Over the western North Pacific during JJA, sky-obscuring fog (CL11), no low-level cloud (CL0), and fair weather stratus (CL6) are associated with anomalous warm advection, surface-based inversion, mean upward flow, and moist mid-troposphere with the strongest anomalies for CL11 followed by CL0. Over the western North Pacific during DJF, bad weather stratus (CL7) occurs in the warm front of the extratropical cyclone with anomalous upward flow while cumulonimbus (CL39) occurs on the rear side of the cold front with anomalous downward flow. Over the tropical oceans, CL7 has strong positive (negative) anomalies of temperature in the upper troposphere (PBL), relative humidity, and surface wind speed in association with the mesoscale convective system while CL12 has the opposite anomalies and CL39 is in between.
The long-term linear trend of global sea-to-air dimethyl sulfide (DMS) flux was analyzed over a 16-year time span (2000~2015), based on satellite observation data. The emission rates of DMS (i.e. DMS flux) in the global ocean were estimated from sea surface DMS concentrations, which were constructed with chlorophyll a (Chl-a) concentrations and mixed layer depths (MLD), and transfer velocity from sea to air, which was parameterized with sea surface wind (SSW) and sea surface temperature (SST). In general, the DMS flux in the global ocean exhibited a gradual decreasing pattern from 2000 (a total of 12.1 Tg/yr) to 2015 (10.7 Tg/yr). For the latitude band ($10^{\circ}$ interval between $0^{\circ}$ and $60^{\circ}$), the DMS flux at the low latitude of the Northern (NH) and Southern hemisphere (SH) was significantly higher than that at the middle latitude. The seasonal mean DMS flux was highest in winter followed by in summer in both hemispheres. From the long-term analysis with the Mann-Kendall (MK) statistical test, a clear downward trend of DMS flux was predicted to be broad over the global ocean during the study period (NH: $-0.001{\sim}-0.036{\mu}mol/m^2/day\;per\;year$, SH: $-0.011{\sim}-0.051{\mu}mol/m^2/day\;per\;year$). These trend values were statistically significant (p < 0.05) for most of the latitude bands. The magnitude of the downward trend of DMS flux at the low latitude in the NH was somewhat higher than that at the middle latitude during most seasons, and vice versa for the SH. The spatio-temporal characteristics of DMS flux and its long-term trend were likely to be primarily affected not only by the SSW (high positive correlation of r = 0.687) but also in part by the SST (r = 0.685).
동한 난류의 북상 끝부분 해역에서 재발생하는 와동류는 봄철과 가을철에 위성이 관측한 해수면 온도 영상에 나타났다. 이러한 재발생 와동류는 1997년 9월 하순 NOAA 위성에 탑재된 AVHRR의 열적외선 영상과 일본 ADEOS 위성에 탑재된 OCTS의 클로로필 영상에도 나타났다. 와동류의 중심은 주변보다 온도가 낮으며, OCTS 위성자료에서 3mg/m$^3$ 이상의 클로로필 농도가 나타났다. 반면, 와동류를 이루는 주변의 더운물에서는 클로로필 농도가 1mg/m$^3$ 이하로 나타났다. 와동류는 가을철 표면수온영상에도 나타났으며 봄에 나타난 것보다 와동류 중심핵의 찬물 온도가 높게 나타났다. 또한 와동류 중심에서 서쪽축의 더운물 온도가 봄에 나타난 것보다 가을에 더 높게 나타났다. 1998년 3월 NOAA 위성과 SeaWIFS 위성자료에서도 재발생되는 와동류와 클로로필량의 농도가 높은 물이 와동류 주변으로 포획되는 장면이 포착되었다. 동한난류의 북쪽 확장 선두와 약 1500m 수심의 대륙붕 위로 남하하는 리만한류가 만나 충돌하는 해역에서 와동류가 형성되는 것으로 사료된다. 와동류가 형성되는 이 해역은 동해 중앙부 해수면에서 우세하게 나타나는 극전선역에서 중규모 구조의 와동류가 극전선 서쪽 해역의 역학적인 해양현상과 강한 연관성이 있음을 나타내었다. ARGOS 위성 추적 표류부이와 와동류의 상호연관성 및 와동류의 지속성에 관한 증거가 토론되었다.
