Samsoeum inhibits systemic anaphylaxis and release of histamine, cytokine in vivo and in vitro

  • Samsoeum (SSE) is used in traditional oriental medicine for various medicinal purposes. However, the exact mechanism that accounts for the anti-allergy and anti-inflammatory effects of the SSE is still not fully understood. The aim of the present study is to elucidate whether and how SSE modulates the allergic reactions in vivo, and inflammatory reaction in vitro. In this study, we showed that SSE significantly decreased compound 48/80-induced systemic anaphylaxis, ear-swelling response, histamine release from preparation of rat peritoneal mast cells and anti-dinitropheny IgE-induced passive cutaneous reaction. Also, SSE inhibited the expression of inflammatory cytokine and cyclooxygenase-2 in PMA plus A23187-stimulated human mast cells (HMC-1). In addition, we showed that anti-inflammatory mechanism of SSE is through suppression of nuclear factor-${\kappa}B$ activation and $I{\kappa}B-{\alpha}$ phosphorylation/degradation in HMC-1. These results provided new insight into the pharmacological actions of SSE as a potential molecule for therapy of inflammatory allergic diseases.

Effects of Activated Carbon Particle Sizes on Caffeine Adsorptions (활성탄 입자 크기가 카페인 흡착에 미치는 영향)

  • The effect of activated carbon particle diameter (i.e. US sieve No. $8{\times}10$ ($d_p{\approx}2.19mm$), $18{\times}20$ ($d_p{\approx}0.92mm$), $50{\times}60$ ($d_p{\approx}0.27mm$) and $170{\times}200$ ($d_p{\approx}0.081mm$) on caffeine adsorption is investigated. BET surface area was increased with decreasing particle diameter ($d_p$), and caffeine adsorption rates increased with decreasing $d_p$. Moreover, pseudo-second order model is predicted the experimental data more accurately than pseudo-first order model, and the fastest rate constant ($k_2$) was $1.7g\;mg^{-1}min^{-1}$ when $d_p$ was 0.081 mm. Surface diffusion coefficient (Ds) was decreased with decreasing $d_p$ based on the minimum sum of square error (SSE). Practically, certain ranges of Ds are acceptable with high reliability ($R^2$) and it is determined that the effect of $d_p$ on Ds is unclear. The effect of pH on caffeine adsorption indicated the dependency of m/L ratio (mass liquid ratio) and $pH_{pzc}$. The $pH_{pzc}$ (i.e. $7.9{\pm}0.2$) was not affected by $d_p$. The higher caffeine adsorption at pH 4 and pH 7 than at pH 10 is due to $pH_{pzc}$, not $pk_a$ of caffeine.

Profile Analysis of Bacteria in Human Hands Using the Terminal Restriction Fragment Length Polymorphism (T-RFLP) Analysis (제한효소 절편길이 다형성(T-RFLP) 분석기법을 이용한 손에 서식하는 세균의 군집조성 분석)

  • As evidence that can be obtained at the crime scene, the importance of micro-evidences has been recognized in recent years with the development modern molecular-level analytical techniques. These micro-evidences include substances useful for personal identification such as DNA, but it is difficult to collect only the evidences showing individual characteristics every time at the crime scene. Therefore, development of new research approaches for the discovery and application of micro-evidence candidates is in increasing demand. For this purpose, skin microbial communities of bacteria inhabiting the palms of 16 people were collected and terminal-restriction enzyme fragment length polymorphism (T-RFLP) analysis was carried out to examine the potential for the application in personal identification. As a result, 16 different electropherograms were obtained, and various taxa including Staphylococcus and Bacillus were shown to produce different T-RF profiles among individuals. These results were analyzed with the factors affecting the microbiota such as sex and working environment of individuals.

Metamorphic P-T Paths from Devonian Pelitic Schists from the Pelham Dome, Massachusetts, USA (뉴잉글랜드 펠암돔 주변부 데본기 변성 이질암의 변성 온도-압력 경로)

  • Major element zoning has been analyzed in garnet porphroblasts obtained from the Grt-St and Ky-Grt-St grade assemblages in Zones I on the northern flank of the Pelham Dome, north central Massachusetts. These porphyroblasts grew during multiple phases of deformation and meta-morphism revealed by the inclusion trail geometry plus the chemical zoning patterns within garnet porphyroblasts. Unusual zoning patterns, including zoning reversals and gradient changes in XMn, zlgzag patterns in Fe/(Fe +Mg) and staircase-shaped patterns in XCa, are coincident with textural truncations and other changes in microstructure within the garnet porphrublasts. Chemical variations in plagioclase, biotite, muscovite and staurolite combined with inclusion trail geometry and petrography reveal that the garnet zoning patterns are modified by combinations of the following. (1) Uni-and divariant reactions involving garnet consumption(Grt+ Chl+Ms=St+Bt+Qtz + $H_2$O) and production(St+Ms + Qtz= Bt+ Grt +A1$_2$$SiO_{5}$ + $H_2$O). (2) Deformation induced episudic ionit dissolution, preferential diffusion and re-distribution during foliation development. (3) P-T changes during growth of the porphyroblasts. The P-T paths combined with petrographic and inclusion trail morphology observations consist of two pattens; (1) heating/compression during NW-SE shortening; and (2) decompression with cooling during NNW-SSE shortening. Based on temperature-time(T-t) geochronological data and late-Paleozoic tectonic model, Alleghanian metamorphism, which is the result of heterogeneous shearing concentrated along the boundary between the Abalone Terrane(Pelham dome) and cover rocks(Bronson Hill Terrane), has produced Ky-St-Ms mineral assemblage during Pennsylvanian(290-300 Ma) in Shutesbury area. However, temperature of alleghanian metamorphism was not high enough to form garnet and staurolite in the Northfiled syncline area. Alleghanian metamorphism has affected only the matrix due to heterogeneous shearing in the study area.
