유화법과 분무법에 의해 제조된 경구백신용 알긴산 마이크로스피어의 평가 (Evaluation of Alginate Microspheres Prepared by Emulsion and Spray Method for Oral Vaccine Delivery System)

  • Alginate microspheres, containing fluorescein isothiocyanate-bovine serum albumin (FITC-BSA) or green fluorescent protein (GFP) were prepared and used as a model drug to develop the oral vaccine delivery system. The alginate microspheres were coated with poly-L-lysine or chitosan. Two methods, w/o-emulsion and spray, were used to prepare alginate microspheres. To optimize preparation conditions, effects of several factors on the particle size and particle morphology of microsphere, and loading efficiency of model antigen were investigated. In both preparation methods, the particle size and the loading efficiency were enhanced when the concentration of sodium alginate increased. In the w/o-emulsion preparation method, as the concentration of Span 80 was increased from 0.5% to 2%, the particle size was decreased, but the loading efficiency was increased. The higher the emulsification speed was, the smaller the particle size and loading efficiency were. The concentration of calcium chloride did not show any effect on the particle size and loading efficiency. In the spray preparation method, the particle size was increased as the nozzle pressure $(from\;1\;kgf/m^2\;to\;3\;kgf/m^2)$ and spray rate was raised. Increasing calcium chloride concentration (<7%) decreased the particle size, in contrast to no effect of calcium chloride concentration on the w/o-emulsion preparation method. Alginate microspheres prepared by two methods were different in the particle size and loading efficiency, the particle size of microspheres prepared by the spray method was about $2-6\;{\mu}m$, larger than that prepared by the w/o emulsion method $(about\;2{\mu}m)$, and the loading efficiency was also higher with spray method. Furthermore, drying process for the microspheres prepared by the spray was simpler and easier, compared with the w/o emulsion preparation. Therefore, the spray method was chosen to prepare alginate microspheres for further experiments. Release pattern of FITC-BSA in alginate microspheres was evaluated in simulated intestinal fluid and PBS (phosphate buffered saline). Dissolution rate of FITC-BSA from alginate/chitosan microsphere was lower than that from alginate microsphere and alginate/poly-L-lysine microsphere. By confocal laser scanning microscope, it was revealed that alginate/FITC-poly-L-lysine microspheres were present in close apposition epithelium of the Peyer's patches of rabbits following inoculation into lumen of intestine, which proved that microspheres could be taken up by Peyer's patch. In conclusion, it is suggested that alginate microsphere prepared by spray method, showing a particle size of & $10\;{\mu}m$ and a high loading efficiency, can be used as a model drug for the development of oral vaccine delivery system.

  • 본 논문에서는 비증발 디이젤 분무의 고속도음영사진을 화상해석 함으로서, 분무선단지달거리와 분무용 이외에 어떤 연문의 분무속 전체 액적군의 평균입자경(sauter mean diameter)과 분무내 연료농도분포를 동시에 측정할 수 있는 새로운 측정법을 제시한다.

  • In this study, to analyze characteristics of acoustic pressure for spot spray type megasonic, FEM analysis was performed for variable parameters based on the structure of commercial one. and 2 models of transmitter were designed and fabricated, and then acoustic pressure distribution(APD) of the transmitter was measured and compared to the commercial. The results of this experiment show that maximum acoustic pressure of model 1 was higher to 1.6 times compared to the commercial, and model 2 was higher to 1.23 times. Through the course of this study, design technology of transmitter has been developed by means of FEM analysis and experiment for characteristics of acoustic pressure. Also, it is expected to be useful in the development of high power spray type megasonic that is necessary with advance in semiconductor technology.

정상류 조건에서의 디젤 연료 분사 노즐내의 유동가시화 (Visualization of the Flow in a Diesel Injection Nozzle In case of the Steady Flow Condition)

  • The effects of the internal flow in a D.I. Diesel injection nozzle on the atomization of a spray were analyzed experimentally. Flow visualization studies were made using a transparent acrylic model nozzle as a diesel nozzle . Water instead of disel fuel was used as the injection liquid. The geometry of the model nozzle was scaled up 10 times of the actual nozzle and the injection pressure for the model nozzle was adjusted so as to achieve a Reynolds number at the discharge hole that was the same as the actual nozzle. Experimental results show that when the needle lift was small, the high turbulence in the sac chamber generated by the high velocity seat flow made the spread angle of the spray plume large. Cavitation, which arose from the sac chamber, makes the spread angle of the spray plume large but the discharge coefficient small.

MODELING OF DIRECT INJECTION DIESEL ENGINE EMISSIONS FOR A QUASI-DIMENSIONAL MULTI-ZONE SPRAY MODEL

  • Phenomenological models for direct injection diesel engine emissions including NO, soot, and HC were implemented into a full engine cycle simulation and validated with experimental data obtained from representative heavy-duty DI diesel engines. The cycle simulation developed earlier by Jung and Assanis (2001) features a quasi-dimensional, multi-zone, spray combustion model to account for transient spray evolution, fuel-air mixing, ignition and combustion. In this study, additional models for HC emissions were newly implemented and the models for NO, soot, and HC emissions were validated against experimental data. It is shown that the models can predict the emissions with reasonable accuracy. However, additional effort may be required to enhance the fidelity of models across a wide range of operating conditions and engine types.

Multidimensional Engine Modeling: NO and Soot Emissions in a Diesel Engine with Exhaust Gas Recirculation

  • The effects of EGR(Exhaust Gas Recirculation) on heavy-duty diesel engine performance, NO and soot emissions were numerically investigated using the modified KIVA-3V code. For the fuel spray, the atomization model based on the linear stability analysis and spray wall impingement model were developed for the KIVA-3V code. The Zeldovich mechanism for the formation of nitric oxide and the soot model suggested by Hiroyasu et al. were used to predict the diesel emissions. In this paper, the computational results of fuel spray, cylinder pressure, and emissions were compared with experimental data, and the optimum EGR rates were sought from the NO and soot emissions trade-off. The results showed that the EGR is effective in suppressing NO but the soot emission was increased considerably by EGR. Using cooled EGR, soot emission could be enhanced without worsening of NO.

차량용 연소식 난방기의 열 및 유동특성에 대한 수치연구 (Numerical Investigation on the Thermal and Flow Characteristics of Combustion Heater for Commercial Vehicle)

  • The diesel pre-heater has being used in cabin heating and coolant heating of engine to reduce the engine warm up time for commercial vehicle. The pre-heaters are classified as diesel spray combustor and it forms diffusion flame. By using swirler, a recirculation flow of hot product gases is established near the fuel nozzle and it helps the maintaining of diffusion flame. The design difference of swirler can affect on reaction characteristics and temperature distribution inside pre-heater. The purpose of this study is the investigation of the effect of swirler configuration on combustion characteristics. To solve spray combustion problem, the Euler-Lagrange approach discrete model is used to track droplet trajectory and evaporation history. The PDF equilibrium model is used for chemical reaction model. These models are implemented into the FLUENT code.

노즐 특성 변화에 따른 미분무수와 화염과의 상호작용에 관한 수치해석 (Numerical Analysis on Interaction between Fire Flame and Water Mist according to the Variation of Nozzle Performance)

  • In the present study, the numerical investigation on the effects of water-mist characteristics has been carried out for the fire suppression mechanism. The FDS are used to simulate the interaction of fire plume and water mists, and program describes the fire-driven flows using LES turbulence model, the mixture fraction combustion model, the finite volume method of radiation transport for a non-scattering gray gas, and conjugate heat transfer between wall and gas flow. The numerical model is consisted of a rectangular enclosure of $L{\times}W{\times}H=1.5{\times}1.5{\times}2.0m$ and a water mist nozzle that be installed 1.8m from fire pool. In the study, the parameters of nozzle for simulation are the droplet size and the spray velocity. Finally, the droplet size influences to fire flume on fire suppression than spray velocity because of the effect of terminal velocity, and the optimal condition for fire suppression is that the droplet size and the spray velocity are $100{\mu}m$ and 20m/s, respectively.

  • 가스터빈 연소기의 스월러와 노즐을 제작하여 스월러 형상 변화에 따른 비연소시 케로신 분무 특성과 연소시에 열유동장의 특성을 조사하였다. 액적의 크기(SMD)와 속도를 파악하기 위해서 위상도플러입자분석기(PDPA)를 이용하였고, 연소 온도를 조사하기 위해서 R-type 열전대(Platinum vs. Platinum-13%rhodium)를 이용하였다. 분무와 화염의 가시화는 스틸 카메라를 이용하였다. 스월러각이 커서 스월수가 큰 경우에 분무 및 화염이 반경방향으로 빠르게 발전하였다. 화염은 스월수가 작을 때는 원추형의 화염이 형성되지만 스월수가 커지면서 스월 모멘텀이 중심축으로 이송되고 혼합이 촉진되어 원통형으로 바뀌었다.

  • 본 연구에서는 가장 다순한 내부혼합형 이류체분사노즐의 기류속도, 액체유량, 노즐직경 및 혼합길이, 기액접촉각을 변화시켜 평균입경(SMD), 분무각, 입도분포, 분 무분사량분포 등을 조사하여 노즐형상에 따른 분무특성의 변화를 자세히 밝혀, 분무특 성을 조절할 수 있는 이류체분사 노즐의 설계에 대한 기초적 자료를 제시하고자 한다.
