This study was performed to investigate the effect of different levels of protein and iron in the diet upon Fe, Cu and Zn metabolism in rat during four weeks of growing period. Forty-five male weanling rats of Sprague-Dawley strain weighing $68.5{\pm}1.1g$ were divided into 9 groups and each group was given with one of the 9 different kinds of diets for four weeks. The three dietary protein levels used were 5, 20 and 40% and Fe levels 0, 35, and 350 ppms. The results obtained were summarized as following ; 1) Food intake and body weight gain in 20%(SP) and 40%(HP) dietary protein groups tended to be significantly higher than 5%(LP) protein groups. Protein efficiency ratio (PER) was higher in LP groups than in HP and SP groups. With dietary Fe levels, there were no significant differences among groups in food intake, body weight gain, and PER. 2) In LP groups, the Fe concentrations in liver, kidney, and hind limb muscle were higher than in SP and HP groups. Regarding with dietary protein levels, the liver Cu concentrations in LP groups were slightly higher, but the liver Zn concentrations were lower in LP groups. The Fe concentrations in liver and kidney tended to decrease with decrease in dietary Fe levels, but Cu and Zn concentrations showed no consistent tendency with dietary Fe levels. 3) The Fe, Cu and Zn concentrations in serum were not different from dietary treatments except that the serum Fe concentrations increased slightly in LP groups. 4) The Fe and Cu concentrations in urine tended to be higher in HP groups. Fecal Cu and Zn concentrations showed no significant differences in dietary protein or Fe levels, but the Fe concentrations tended to increase with increase in dietary Fe levels.
The pharmacological therapies and whole body vibration as non-pharmacological therapies were known to have adverse side effects. Therefore, partial stimulation was suggested and its effects were evaluated. This study aimed to evaluate the site-specific effects of partial stimulator for treatment of osteoporosis induced by estrogen deficiency. Sixteen virginal Sprague-Dawley rats (12 weeks old) were divided into 2 groups(no stimulation, stimulation groups). All rats were ovariectomised to induce osteoporosis. After 3 weeks of operation, the right tibiae in rats of stimulation group (frequency: 10Hz, cycle: 1500, strain on bone surface: $2000{\mu}{\varepsilon}$) were stimulated perpendicularly at right tibia by using partial stimulator for 6 weeks (3days/week). The right tibiae in rats were scanned, before stimulation (0 week) and at 6 weeks after stimulation by using in-vivo micro computed tomography. For investigation of changes in morphological characteristics, structural parameters were measured and calculated. At 6 weeks the morphological characteristics (relative value) in stimulation group were significantly enhanced than those in no stimulation group (p<0.05). In this study, we find that after 6 weeks of partial stimulation, the morphological characteristics of tibia trabecular bone were enhanced. Thus, we concluded that partial stimulation could be used to treat osteoporosis.
도라지의 급이가 고콜레스테롤혈증 흰쥐의 간기능과 지질대사에 미치는 영향 구명하기 위하여 SD계 숫흰쥐에게 동결건조한 도라지 분말 5%와 도라지 분말 5%에 해당하는 식물섬유, saponin, 에탄올 추출물을 각각 급이하여 3주간 실험 사육한 후 혈청효소 및 간장의 지질성분 분석, 간세포의 형태를 검토하였다. 혈청 중 AST, ALT효소활성은 대조군에 비해 전 실험군이 유의적으로 감소하였고 그 중 saponin 급이군이 가장 낮았고 그 다음이 식물 섬유군이었다. LDH효소 활성도 전 실험군에서 유의적으로 낮게 나타났으며 22년근 도라지의 식물 섬유군과 saponin군이 낮은 값을 보였다. 간장 중의 총 콜레스테롤 함량은 4년근과 22년근 도라지 분말 및 식물섬유 급이군이 비교적 낮았으며, 중성지질 함량은 대조군이 비해 전 실험군에서 낮았으나, 각 실험군간의 차이는 크지 않았고, 인지직 함량은 전 실험군에 있어 비슷한 수준이었다. 간장 조직세포의 전자현미경 관철 결과, 대조군은 지방구가 증가되어 있는 반면 도라지 급이군에서는 감소되었으며, 22년근 도라지 급이군은 간장 글리코겐 축적량이 증가되어 있었다.
Iron deficiency is a severe nutritional problem in the world. Coffee intake of the people is increasing every year and it can increase the loss of several essential body minerals including iron. Either iron deficiency or coffee intake may increase the oxidative stress of the body. However, the effect of iron deficiency and/or coffee intake on peroxidation have not been studied much. Therefore, the aim of this study was to investigate the effect of coffee intake on oxidative stress and antioxidative enzyme activities of iron-deficient rats. Forty-eight male rats of Sprague-Dawley strain were divided into two groups by dietary iron levels. Iron deficient group were fed 5 ppm iron diet and iron-sufficient group were fed 50 ppm iron diet. Each iron group were divided into three sub-groups by coffee levels (0%, 1%, 4%) included in the experimental diet. The experimental diets were fed for 4 weeks. The hemoglobin level was significantly low in iron deficient group and the level was exacerbated by high coffee intake. The malondialdehyde concentration of the plasma and liver were not affected by iron or coffee level in this study. However, plasma aspartate aminotransferase and alanine aminotransferase, the indicator of the liver damage, were increased by high coffee intake. The erythrocyte and liver superoxide dismutase (SOD) activities were elevated in iron deficient groups. Coffee intake increased erythrocyte SOD activity in iron sufficient groups. Glutathione peroxidase and catalase activities were not influenced much by either iron or coffee intake. In conclusion, high coffee intake in iron deficiency may not only increase the anemia symptoms, but also may increase the oxidative stress of the body.(Korean J Nutrition 35(9) : 919~925, 2002)
This study was performed to investigate nutritional effect of various dietary fibers on lead absorption, and protein and lipid metabolisms in growing rats. Sixty male rats of Sprague-Dawley strain weighing 140$\pm$1.1g were blocked into 10 groups according to body weight and fed 10 kinds of diet different with fiber sources [non-fiber, cellulose, pectin, guar gum or carboxymethylcellulose(CMC)] and lead levels (0 or 1%) for 4 weeks. Results were summerized as follows : 1) Food intake, weight gain, FER and PER were remarkably decreased in lead(Pb)-added groups. Weight gain, FER and PER in Pb-added pectin group were significantly lower than those in Pb-added non-fiber group. 2) Liver and kidney weights, femur weight and length, hematocrit and hemoglobin content were decreased in Pb-added groups. Especially femur and liver weights in pectin groups were the lowest among groups. 3) Total protein content in serum was significantly decreased in Pb-added groups but was not different with dietary fiber sources. Total lipid content in serum was not different with dietary Pb levels and fiber sources, but cholesterol content in serum of guar gum group was significantly decreased by Pb addition. 4) Nitrogen, lipid and cholesteol contents in liver were significantly decreased in Pb-added groups, and lipid content in liver of pectin and CMC groups was lower than other groups. 5) Daily urinary and fecal excretions of nitrogen, kipid and cholesterol were decreased in Pb-added groups, and fecal nitrogen was significantly increased in Pb-added groups, and fecal nitrogen of cellulose and guar gum groups was significantly higher than other groups. Fecal excretions of lipid and cholesterol were increased by dietary fibers, and especially fecal lipid was remarkably increased in pectin and guar hum group. 6) Pb contents in liver and femur were decreased by dietary fibers. Especially Pb contents in liver, kidney and femur were significantly decreased in guar gum group. 7) Daily urinary and fecal excretions of Pb were significantly increased in cellulose and guar gum groups, and fecla excretion of Pb in guar gum group was twice of non-fiber group. Pb absorption ratio was significantly decreased in guar gum group. In conclusion, dietary fibers have effect on protein and lipid metabolisms, and decreased intestinal absorption of Pb by increasing fecal excretion. But the degree of effect was different with dietary fiber sources.
This balance study was performed to investigate nutritional effects of guar gum and calcium supplement for 8 weeks. 36 male rate of Sprague-Dawley strain weighing 50g were blocked into six groups : 5CN, 5GN, 5CS, 5GS, FFN, 10GN. Food intake, body weight gains, FER, PER, bioavailabity of energy, protein, fat, Ca and P, and Ca & P content of femur and kidney was measured. The results obtained in this study were summarized as follows: (1) Feed intake, weight gains, FER, PER and bioavailabilities of protein and P and the contents of Ca & P in femur of 5GB were significantly higher than 5CN and 5CS (p<0.05) but was not significant different with those 5GN. Fat bioavailabilities of 5GS was significantly lower than 5CN and 5GN (p<0.05) but was not significant different with those of 5CS. Ca contents of kidney of 5GS was significantly higher than 5CN and 5GN(p<0.05) but was not significant different with those of 5SC. (2) According to guar gum levels feed intake. FER and PER were not sifgnificantly different among groups but body weight gains, bioavailabilities of energy, protein, fat and Ca, Ca & P content of femur was significantly decreased (p<0.05). Bioavailabilities of P was significantly increased according to the guar gum levels (p<0.05). Ca contents in Kidney of 10GN was significantly higher those of 5GN(p<0.05) but P contents in kidney was not significant difference by guar gum levels. Therefore 5% supplemental guar gum in diet of obese man and diebetics could be recommended. But it should be careful in using 10% guar gum in diet.
The aim of this study was to evaluate the effects of soyoligosaccharides on intestinal microflora transit time lipid profiles and immune responses. Forty-eight male rats of Sprague-Dawley strain were divided into six groups(SYS: soybean oil-sucrose SYO: soybean oil-oligo SES; esame oil-sucrose SEO:sesame oil-oligo BFS : beef tallow-sucrose BFO : beef tallow-oligo) soyoligo-groups(SYO, SEO, BEO) were fed the diet containing 5% soyoligosaccharides but control groups not fed them and all groups were fed 20% fats for 4 weeks. The number of fecal bifidobacteria tended to increase after soyoligosaccharides feeding. The significant increase was found in SEO group compared to SES in that of fecal bifidobacteria,. Stool wet weight dry fecal weight and stool water content were increased significantly in soyoligo-group(SYO, SEO, BFO) compared to control groups(SYS, SES, BFS) and gastrointestinal transit time(TT) tended to increase. Soyoligosaccharides lowered plasma cholesterol in SEO group significantly more than did other groups. soyoligo-groups(SYO, SEO, BFO) tended to decrease liver triglycerides compared to control groups. Plasma IgG concentration increased in beef tallow groups(BFS, BFO) than that in other dietary fat groups. Soyoligosaccharides and kinds of fats had effects on plasma C3 concentration and mitogenesis of the spleen cells. Soyoligosaccharides had no effects on plasma leptin and insulin concentration but kinds of fats did so that sesame groups were decreased significantly in plasma leptin concentration but increased significantly in plasma insulin concentration compared to other dietary fat groups. In conclusion dietary soyoligosaccharides changed the composition of the intestinal microflora beneficially with sesame oil and partially had effect on plasma and liver lipid profiles .(Korean J Nutrition 33(6) : 597∼612, 2000)
Effects of alchohol and fat content in a balanced diet on chemical composition and morphology of liver were investigated in growing rats. Fourth eight male rats of Sprague-Dawley strain weighing about 160g were divided into 4 groups ; high fat diet group, alcohol-administered high fat diet group, low fat diet group and alcohol-administered high fat diet group, low fat diet group and alcohol-administered low fat diet group. High and low fat diets supplied 30% and 12%, respectively, of total calorie intake from fat, and alcohol was given by adding ethanol in drinking waster at 10%. Diets contained adequate amounts of all nutrients required for rats, including lipotrpoic agents(choline and methionine) to minimize effects of factors other than alcohol on liver damage. Ratios of liver weight to body weight were statistically different among groups. Liver/dody weight ratios alcohol-administered rats were significantly higher than those of non-alcohol groups after 6 weeks treatment. Although total lipid and triglyceride per gram liver were increased in alcohol-administered rats, especially low fat diet fed rats, the values were not significantly different. Opticmicroscopical observation revealed increase in cell size and no change in morphology of liver. Examination of hepatocytes by electron microscopy showed that fat droplets were observed in all groups but enlarged in the alcohol-administered low fat diet fed rat. Contents of protein, cholesterol and phospholipid were not affected by alcohol consumption. The level of lipid peroxide was significantly lower in the livers of alcohol-administered rats than in the livers of non-alcohol groups. The results of this study indicate that even moderate alcohol drinking and dietary fat content did not affect any significant change in composition and morphology of liver until 6 week treatment but that even moderate alcohol drinking caused some signs of steatosis of liver.
The antioxidant effects of freeze-drying acorn were examined to find how much the freeze-drying acorn can reduce peroxidized corn oil poisoning, which influenced cholesterol, triglyceride, phospholipids, lipidperoxides, GOT(glutamate oxaloacetate transaminase), GPT(glutamate pyruvate transaminase) in serum, and cholesterol, triglyceride, lipidperoxides, fatty acid of phospholipids, SOD(superoxide dismutase), catalase in liver. In this experiment, male rats of Sprague-Dawley strain were used. The rats were divided into 6 groups, which were fed differently for 5 weeks : basal diet, 10% peroxidized corn oil added to basal diet, 1% acorn flour and 10% peroxidized corn oil added to basal diet, 5% acorn flour and 10% peroxidized corn oil added to basal diet, 10% acorn flour and 10% peroxidized corn oil added to basal diet, and 0.25% tannic acid and 10% peroxidized corn oil added to basal diet. The results were as follows : It was found that the peroxidized corn oil-fed 5 weeks induced the elevation of cholesterol, triglycerides, lipid peroxides, GOT, GPT in serum, and cholesterol, triglycerides, lipid peroxides in liver as compared to the basal diet-fed rats, but the acorn flour-fed rats reduced the elevation of these components. In addition, saturated fatty acid in rat liver phospholipids induced the elevation by feeding of peroxidized corn oil and, on the other hand, the acorn flour-fed rats reduced the elevation of saturated fatty acids. The acorn flour-fed rats reduced the activity of SOD in liver while they enhanced the activity of catalase in liver as compared with the peroxidized corn oil-fed rats.
This study was performed to investigate the influence of dietary fibers on the serum lipid level and bowel function in aged rats. Fiber sources of experimental diets were made from mugwort(M), butterbur(B), apple(A), sea mustard(S) by drying and milling. Each of fiber sources was mixed into the diet at the level of 5, 15% of diet. Sprague-Dawley strain, 13 month old male rats were divided into 9 groups by randomized complete block design : C, M5, B5, A5, S5, M15, B15, A15, S15. The animals were fed ad libitum each of experimental diets for 4 weeks. Control group showed lower food intake compare to the other groups. There were no significant difference between 5% groups and 15% groups in food intake. Fecal weight, dry fecal weight and fecal water content of control group were significantly lower than those of experimental groups, and fecal water content was increased by increasing level of dietary fibber. Apple group showed the lowest values, sea mustard group showed the highest. The shorter transit time was observed in the group of higher intake of dietary fiber. At the same level of dietary fiber, transit time of sea mustard group was shorter than the other groups. With increasing age, serum triglycerides, total cholesterol were increased and HDL-cholesterol was decreased. Fiber fed groups showed lower serum TG, TC and higher HDL-c level compare to the control group. Absorption rates of calcium, magnesium and phosphorus was tend to be lower in the group fed dietary fiber sources than control group. Mucosa weight and maltase activity in the small intestine were decreased by increasing age. As intake of dietary fiber increaed, mucosa weight in the small intestine was not different but maltase activity was decreased.
