The study was conducted to investigate per oral (PO) effects of Black raspberry wine on testosterone levels in Sprague-Dawley rats oral administrated with Methoxychlor in order to establish the experimental clinical model for evaluating the influences on the sexual hormones of SD-Rat administrated with Methoxychlor(MET), it was dissolved in acetone and olive oil (1:19), which was administrated orally at doses of 200mg/kg body weight/ day for 7days. Black raspberry wine of 13% alcohol concentration, was prepared from ripen fruits of Rubus coreanus fermented with Saccharomyces cervisiae. PO administration of Black raspberry wine for 15 week produced dramatic increases of serum testosterone levels. Increase in the testosterone level was observed, using gamma counter with 125I testosterone, starting from 1 week post administration. Maximum increase in testosterone level was observed at 4 week post administration, 5.18${\pm}$0.76ng/mL, which was 10.1 times higher than before and at 15 weeks post administration it was recorded as 1.67${\pm}$0.19ng/mL indicating Black raspberry wine as an effective phyto-testosteronic beverage of the future.
Background: Increases in the average global temperature cause heat stress-induced disorders by disrupting homeostasis. Excessive heat stress triggers an imbalance in the immune system; thus protection against heat stress is important to maintain immune homeostasis. Korean ginseng (Panax ginseng Meyer) has been used as a herbal medicine and displays beneficial biological properties. Methods: We investigated the protective effects of Korean ginseng extracts (KGEs) against heat stress in a rat model. Following acclimatization for 1 week, rats were housed at room temperature for 2 weeks and then exposed to heat stress ($40^{\circ}C$/2 h/day) for 4 weeks. Rats were treated with three KGEs from the beginning of the second week to the end of the experiment. Results: Heat stress dramatically increased secretion of inflammatory factors, and this was significantly reduced in the KGE-treated groups. Levels of inflammatory factors such as heat shock protein 70, interleukin 6, inducible nitric oxide synthase, and tumor necrosis factor-alpha were increased in the spleen and muscle upon heat stress. KGEs inhibited these increases by down-regulating heat shock protein 70 and the associated nuclear $factor-{\kappa}B$ and mitogen-activated protein kinase signaling pathways. Consequently, KGEs suppressed activation of T-cells and B-cells. Conclusion: KGEs suppress the immune response upon heat stress and decrease the production of inflammatory cytokines in muscle and spleen. We suggest that KGEs protect against heat stress by inhibiting inflammation and maintaining immune homeostasis.
Objective: Approximately 30% of preeclamptic pregnancies exhibit abnormal liver function tests. We assessed liver injury-associated enzyme levels and circulating transforming growth factor beta (TGF-β) levels in an arginine vasopressin (AVP)-induced pregnant Sprague-Dawley rat model. Methods: Pregnant and non-pregnant Sprague-Dawley rats (n=24) received AVP (150 ng/hr) subcutaneously via mini-osmotic pumps for 18 days. Blood pressure was measured, urine samples were collected, and all animals were euthanized via isoflurane. Blood was collected to measure circulating levels of TGF-β1-3 isomers and liver injury enzymes in pregnant AVP (PAVP), pregnant saline (PS), non-pregnant AVP (NAVP), and non-pregnant saline (NS) rats. Results: The PAVP group showed significantly higher systolic and diastolic blood pressure than both saline-treated groups. The weight per pup was significantly lower in the AVP-treated group than in the saline group (p<0.05). Circulating TGF-β1-3 isomer levels were significantly higher in the PAVP rats than in the NS rats. However, similar TGF-β1 and TGF-β3 levels were noted in the PS and PAVP rats, while TGF-β2 levels were significantly higher in the PAVP rats. Circulating liver-type arginase-1 and 5'-nucleotidase levels were higher in the PAVP rats than in the saline group. Conclusion: This is the first study to demonstrate higher levels of TGF-β2, arginase, and 5'-nucleotidase activity in PAVP than in PS rats. AVP may cause vasoconstriction and increase peripheral resistance and blood pressure, thereby elevating TGF-β and inducing the preeclampsia-associated inflammatory response. Future studies should explore the mechanisms through which AVP dysregulates liver injury enzymes and TGF-β in pregnant rats.
Objective: High frequency wave has been used in cancer treatment and cosmetic area but not in musculoskeletal pain yet. The purpose of this study is to evaluate temperature distribution according to depth and confirm safety of high frequency wave through animal study. Method: High frequency wave was applied to the posterior limb of 9 Sprague-Dawley rats for 20 minutes (experimental group) and no wave was used in the same number of rats for control group. Tissue temperature was measured from skin surface to 1 cm depth (surface, 1 mm, 5 mm, and 1 cm) for 5 seconds. Results: In the experimental group, temperature was elevated 3.2℃ at skin surface, 2.87℃ at 1 mm, 2.25℃ in 5 mm, and 1.74℃ in 1 cm depth. These were significantly different from those in the control group (p<0.001). There was no bulla or redness in the skin after high frequency wave stimulation and neither change of myocytes nor collagen degeneration was found in the tissue histology. There was no apoptosis in the skin surface and muscle layer in TUNEL assay. Conclusion: High frequency wave elevated tissue temperature from the skin to muscle layer without both histologic change and apoptosis.
우리는 Loperamide로 유도한 변비 Sprague-Dawley 랫드 모델에서 프락토올리고당 및 과채류복합 추출물을 2 주간 경구 투여 한 후 랫 분변의 그룹 기간별로 수집 한 후 장내 마크로바이옴 변화 경향을 분석하였다. 프락토올리고당 및 과채복합추출물(FVCE)에 대한 미생물 군집 분석을 16S rDNA 클로닝 및 pyrosequencing을 통해 수행하여 표준화 및 체계화를 위한 기초 데이터를 얻었다. 과채복합추출물(FVCE) 제조 공정은 원핵생물 군집에 대한 미생물 분석을 통해 문 수준에서 미생물 verrucomicrobiota의 약간의 차이가 우세한 것으로 나타났다. 속 수준에서는 prevotella와 muribaculaceae가 종 수준에서 더 많은 차이를 보였다. 이러한 결과는 사용된 미생물 군집이 생산되는 과일 및 채소 복합 추출물(FVCE)의 품질에 영향을 미친다는 것을 시사한다고 할 수 있다. 따라서, 일관된 품질의 과일 및 채소 복합 추출물(FVCE)을 생산하기 위해서는 안정적인 미생물 군집이 유지되어야 한다.
Experiments were performed to determine the effects of the Oyacksunkisan(烏藥順氣散) and Kamioyacksunkisan(加味烏藥順氣散) liquid extract on the hypertension and the pulse rate in Sprague-Dawley rat(SDR) and Spontaneous Hypertensive rats(SHR). The results were obtained as follows ; 1. Blood pressure was significantly decreased firstday and 11th day after administration of Oyacksunkisan extract and pretreatment of phentolamine. 2. Blood pressure and pulse rate were regulated first day after administration of Kamioyacksunkisan and blood pressure was significantly decreased after pretreatment of clonidine and phentolamine. 3. Oyacksunkisan and Kamioyacksunkisan didn't show any significant changes of blood pressure and pulse rate after pretreatment of prdpranolol, hydralazine and verapamil From the above result, it was concluded that Oyacksunkisan(烏藥順氣散) and Kamioyacksunkisan(加味烏藥順氣散) could be applied effectively to the hypertension.
The investigation was aimed to study the effect of ginseng ethanol extract on the hepatic ethanol-metabolizing enzyme activity in vivo. The extract (100mg/kg/day) was administered orally to Sprague-Dawley rats for $7{\sim}10$ days and their microsomal ethanol oxidizing system(MEOS) and catalase activities were measured. The MEOS activity in the rat treated with the extract was not significantly different from that of the normal group. Microsomal fraction containing MEOS was separated and the MEOS activity was measured after preincubation for 5, 60 and 180 min, respectively. There were no significant differences in MEOS activities between the normal and treated groups preincubated for 5, 60 and 180 min. The activity in the rat treated with single i.p. injection of 95% $CCl_4$ (0.5ml/kg) was decreased by 48%, compared to the normal group and in the rat treated with the extract (100mg/kg) for $7{\sim}10$ days, the decrease of the MEOS activity was potentiated. Catalase activity in the rat treated with the extract (100mg/kg) was similar to that obtained from the normal group.
This study was conducted to evaluate the effect of cytochalasin B (CB) treatment in the activation medium on the development of somatic cell nuclear transfer (SCNT) rat embryos. Fetal fibroblast cells were isolated from a Day 14.5 fetus, and the oocytes for recipient cytoplasm were recovered from 4-week old Sprague Dawley rats. After enucleation and nuclear injection, the reconstructed oocytes were immediately exposed to activation medium consisting of 10 mM $SrCl_2$ with or without CB for 4 hr, and formation of pseudo-pronucleus (PPN) was checked at 18 hr after activation. Then, they were transferred into day 1 pseudopregnant recipients (Hooded Wistar) or cultured for 5 days to check their developmental competence in vivo or in vitro. The number of PPN was not affected by CB treatment during the activation. However, CB treatment supported pre-implantation development of rat SCNT embryos. Embryos generated by the procedures of SCNT were also capable of implanting, with 1 implantation scar found from a recipient following the transfer of 87 SCNT embryos to four foster mothers. The result of the present study shows that rat SCNT embryo can develop to post-implantation stage following treatment with CB.
This study was performed to assess that clinco-therapeutic effect of natural Italian honeybee (Apis mellifera) venom in adjuvant-induced arthritic rat. Ninety Sprague- Dawley rats of male were injected with complete Freund's adjuvant (CFA). Adjuvant arthritis was produced by a single subcutaneous injection of 1 mg Mycobacterium butyricum suspended in 0.1 ml paraffin oil into the right hindpaw. Righting reflex was uniformly lost and considered to be the point of arthritis development on day 14 after CFA injection. Experimental groups were divided into three groups. When arthritis was developed in the rat hind-paw, tested groups were administrated with prednisolone (10 mg/kg, p.o) and honeybee venom (one bee, s.c) at an interval of two days. Control group was subcutaneously injected with 0.1 ml of physiological saline solution in the rat at an interval of two days. Clinical findings, hematological values and histopathological findings were observed during or after the drugs administration. In tested groups, the development of inflammatory edema and polyarthritis on day 14 after treatment was suppressed. No significant differences of hindpaw edema volume and lameness score between prednisolone and honeybee venom groups were observed during or after therapeutic drugs treatment. WBC counts of prednisolone and honeybee venom treatment groups as compared with the control group were getting remarkably decreased during or after the therapeutic drugs administration(p<0.01). Erosions of articular cartilage and inflammatory cell infiltrations during or after the therapeutic drugs treatment was effectively suppressed in natural honey venom.
Mertansine, a thiol-containing maytansinoid, is a tubulin inhibitor used as the cytotoxic component of antibody-drug conjugates for the treatment of cancer. Liquid chromatography-tandem mass spectrometry was described for the determination of mertansine in rat plasma. 50-μL rat plasma sample was pretreated with 25 μL of 20 mM tris-(2-carboxyethyl)-phosphine, a reducing reagent, and further vortex-mixing with 50 μL of 50 mM N-ethylmaleimide for 3 min resulted in the alkylation of thiol group in mertansine. Alkylation reaction was stopped by addition of 100 μL of sildenafil in acetonitrile (200 ng/mL), and following centrifugation, aliquot of the supernatant was analyzed by the selected reaction monitoring mode. The standard curve was linear over the range of 1-1000 ng/mL in rat plasma with the lower limit of quantification level at 1 ng/mL. The intra- and inter-day accuracies and coefficient variations for mertansine at four quality control concentrations were 96.7-113.1% and 2.6-15.0%, respectively. Using this method, the pharmacokinetics of mertansine were evaluated after intravenous administration of mertansine at doses of 0.2, 0.5, and 1 mg/kg to female Sprague Dawley rats.
