다양한 약침제제(藥鍼製劑)가 Rat의 Adjuvant 관절염(關節炎)에 미치는 영향(影響) (Effects of various Herbal Acupuncture on Adjuvant Arthritis in Rats)

  • To investigate effects of vanous herbal-acupuncture, which were Daebangpungtang, Oyaksungisan, Dokhwalgisaengtang, and Binsosan on Adjuvant Arthritis in rats, the edema rate, the number of WBC, the quantity of TNF-${\alpa}$, COX -2, and IL-6, and histological test of the muscular tissue were measured in the arthritis part. After elicilating arthritis of Sprague Dawely(SD) rats by injection of Freund's complets adjuvant for 2 weeks, saline was injected for the control group, Daebangpungtang acua-acupuncture was injected for the Daebangpungtang group, Oyaksungisan acua-acupuncture was injected for the Oyaksungisan group, Dokhwalgisaengtang acua-acupuncture was injected for the Dokhwalgisaengtang group, and Binsosan acua-acupuncture was injected for the Binsosan group during 30days. Selected point was ST35 in all groups. 1. The number of WBC was $8.24{\pm}0.51$($10^3$/ml) in the normal group and $27.35{\pm}3.51$($10^3$/ml) in the control group. the Daebangpungtang group and the Oyaksungisan group decreased each $20.42{\pm}2.75$($10^3$/ml) and $19.78{\pm}4.99$($10^3$/ml) in the Exp.ll group. This fact showed that the group Exp.ll was more effective than the control group effectively. (P<0.05) 2. The volume of the paw were checked. The volume of paw was $0.62{\pm}0.11mm in the control group and $0.45{\pm}0.08$mm in the Daebangpungtang group and $0.47{\pm}0.07$mm in the Oyaksungisan group, the swelling of the paw was restricted significantly in the Daebangpungtang and Oyaksungisan group(P<0.05) 3. The bands of the TNF-${\alpa}$, COX -2 in the muscular tissue of the control group wcre certain and thick than other groups(except normal group), and the band of the IL-6 in the muscular tissue of all group(except normal group) were similar forms. 4. In histological finding, because of severe inflammatory reaction, remarkably irregular tissue and large amount of inflammatory cells were found in thc control and Dokhwalgisaengtang group. But the Oyaksungisan and Daebangpul1gtang group showed small amount of inflammatory cells, the refrained inflammatory state and even recovering state. From these results, it is showed Oyaksugisan and Daebangpungtang acua-acupuncturc refrain inflammatory reaction and muscular tissue necrosis in SD rats paw were induced by Freund's complete adjuvant

천마의 일반 성분과 기능성 조사 (Composition and Functionality of Chonma)

  • 최근에 난과 식물에 속하는 천마의 농가재배가 성공함에 따라 천마를 이용한 음용차의 개발 등이 추진되고 있다. 본 연구에서는 천마의 성분을 분석하였고 Sprague-Dawely (SD) rat을 이용하여 천마의 기능성을 조사하였다. 사용된 건조 천마는 수분 11.8, 조단백질 7.6, 회분 3.2, 조지방 0.5, 조섬유 3.9, 조탄수화물 72.9%(w/w) 로 분석되었다. 무기질 함량은 칼슘 121, 나트륨 83, 철 6.2, 인 170, 마그네슘 69, 칼륨 1,278 mg% 수준 이었다. SD rat에 분말 천마가 0, 0.15, 1.5, 5.0% 첨가된 chow diet를 4주간 섭식시키며 실험한 결과 실험군간에 몸무게의 유의적인 차이는 없었으며 혈중 콜레스테롤은 5% 섭취군이 49.3 mg/dl로서 대조군 (63.7 mg/dl)에 비하여 현저히 저하되었다. 0.15% 첨가군은 가장 높은 진정 효과를 나타내었다. 천마 식이는 본 연구에서 조사된 대장의 상재 균총조성에는 큰 영향이 없었고 대장 내용물의 ${\beta}-glucuronidase$ 활성 수준도 유의적 변화가 나타나지 않았다. ${\beta}-Glucosidase$의 경우에는 5%첨가군이 대조군에 비하여 유의적으로 높았다.

Isoprene 아급성 흡입독성 연구 (A Study on Subchronic Inhalation Toxicity of Isoprene Using Sprague-Dawely Rats)

  • The purpose of this study was to obtain scientific information regarding classification and health hazards that may result from a 13 weeks inhalation exposure of isoprene in Sprague-Dawley (SD) rats. The testing method was conducted in accordance with OECD guidelines for the testing of chemicals No. 413. The Rats were divided into 4 groups (10 male and 10 female rats in each group) and exposed to 0, 360, 1,620, 7,300 ppm isoprene in each exposure chamber for 6 h/day, 5 days/week, for 13 weeks. As a result, there were no mortality or abnormality during the period of study and did not show any significant changes of body weight. There were no dose response changes in urinalysis, hematological and serum biochemical value examination. Relative organ weight was increased significantly the right kidney in 7,300 ppm group of male rats. In female rats, relative organ weight of the left kidney and the both lungs in 1,620 ppm group and the left lung and the both kidneys in 7,300 ppm group were increased significantly. But the histopathological findings did not reveal any exposure-related changes. According to the above results, the no observable adverse effect level (NOAEL) of isoprene was 7,300 ppm (20.3 mg/L) in both male and female rats. In conclusion, Isoprene was not classified specific target organ toxicity of the 'Standard for Classification and Labeling of Chemical Substance and Material Safety Data Sheet' (Ministry of Employment and Labor, 2009).

Rat에서 ChondroT의 DRF 및 단회독성 시험 (DRF and Single Dose Oral Toxicity Study of ChondroT in Rat)

  • Objectives The purpose of this experiment is to evaluate 4 weeks DRF (Dose Rate Finding) and single oral dose toxicity of ChondroT in rats. Methods In 4-week DRF, male and female Sprague-Dawely rats were treated with ChondroT at oral dose of 0, 500, 1000, and 2000 mg/kg. clinical signs, body weight, food consumption, necropsy findings, organ weight, hematological and blood-chemical parameters, and histological findings were monitored for 4 weeks. Also, after single oral administration of ChondroT, mortality, clinical signs, body weight, and necropsy findings were minitored for 2 weeks. Results In 4-week DRF and single dose oral toxicity study of ChondroT in sprague-Dawley rats, ChondroT did not exhibit any toxicity under the study conditions employed. Conclusions The results suggested a no-observed adverse effects level (NOAEL) was over 2,000 mg/kg/day in SD rats after oral administration, this study could be used as basic study of the repeated dose 13-week oral toxicity study of ChondroT.

SD 랫드에서 혹명나방 저항성 유전자 변형 낙동벼의 13주 경구 반복투여 독성에 관한 연구 (Thirteen-Week Repeated Oral Toxicity Study of Leaf Folder (Cnaphalocrocis medinalis) Resistant Rice (Nakdongbyeb) in Sparague-Dawely Rats)

  • 본 연구는 혹명나방 저항성 유전자 변형 낙동벼(GMO)의 식품으로서의 안전성을 평가하고자 13주 경구 반복투여 독성시험을 수행하였다. SD 랫드를 0, 5, 20%의 유전자 변형 낙동벼와 20% 낙동벼(Non-GMO)를 투여군으로 분리하여 13주간 투여한 후 체중, 사료섭취량, 요, 혈액 및 혈액생화학, 부검 및 조직병리학적 검사를 수행하였다. 시험종료시까지 모든 동물은 생존하였으며, GMO 벼의 투여와 관련된 독성은 관찰되지 않았다. 따라서 혹명나방 저항성 유전자 변형 낙동벼의 무독성량은 10,000 mg/kg/day 이상일 것으로 판단되며, 13주 경구 반복투여 독성은 없는 것으로 평가되었다.

다이옥신 저해능에 대한 삼백초의 지질대사에 미치는 효과 (Effects of Saururus chinensis Baill on Lipid Metabolism Against TCDD Damage)

  • 삼백초가 TCDD 투여로 유발된 rat의 지질대사에 미치는 효과를 알아보기 위해 NTT, TTA, STT군으로 각각 10마리씩 나누어 TCDD를 투여하고 4주 동안 물질을 투여하였다. 4주 후 희생시켜서 혈청과 간의 지질대사를 관찰한 결과는 다음과 같다. 1. Total Cholesterol에서 삼백초를 투여한 STT군은 NTT군 비교해서 14.46% 증가하였으며, 혈청 중의 HDL-Cholesterol은 SST군이 TTA군과 비교하여 21.29% 증가하였고 94.98%로 엑제되었다. LDL-Cholesterol에서는 STT군이 TTA군 보다 12.86% 감소하였으며, 48.34%로 억제되었다. 2. 혈청중의 TG의 함량은 TTA군이 NTT군에 비해 34.84% 증가하였다. TTA 군에 비해 STT군은 7.06% 감소하였으며, 18.9% 억제시킨다 3. 간조직 중의 MDA량은 STT군이 TTA군에 비해 17.14% 감소하였고, 32.72%의 억제율을 보였다.

  • To investigate effects of Taraxaci herbal-acupuncture on Adjuvant Arthritis in rats, the edema rate, the number of WBC, the quantity of total protein, albumin and globulin in the blood serum and histological test of the muscular tissue were measured in the arthritis part. 1. After elicitating arthritis of Sprague Dawely(SD) rats by injection of Freund's complets adjuvant for 2 weeks, saline was injected for the Exp. I group and Taraxaci herbal-acupuncture was injected for the Exp. II group during 30days. Selected point was $ST_{35}$ in both the groups. And then the volume of the paw were checked. The volume of paw was $0.84{\pm}0.14mm$ in the Exp. I group and $0.38{\pm}0.17mm$ in the Exp. II group, the swelling of the paw was restricted significantly in the Exp. II group(p<0.05) 2. The number of WBC was $10.34{\pm}0.14(10^3/ml)$ in the normal group and $37.47{\pm}5.46(10^3/ml)$ in the Exp. I group. It was $20.39{\pm}4.23(10^3/ml)$ in the Exp. II group. This fact showed that the group Exp. II was more effective than the Exp. I group in the treatment of arthritis(p<0.05) 3. The content of the total protein in the blood serum was $6.14{\pm}0.43g/dl$ in the normal group, $7.95{\pm}0.94g/dl$ in the Exp. I group, and $6.38{\pm}1.75g/dl$ in the Exp. II group. This fact showed that the group Exp. II was more effective than the Exp. I group in the treatment of arthritis(p<0.05) 4. The contents of albumin in the blood serum was $2.94{\pm}0.13g/dl$ in the normal group, $2.01{\pm}0.48g/dl$ in the Exp. I group. and $2.71{\pm}0.34g/dl$ in the Exp. II group. This fact showed that the group Exp. II was more effective than the Exp. I group in the treatment of arthritis(p<0.05) 5. The contents of globulin in the blood serum was $3.19{\pm}0.48g/dl$ in the normal group, $4.70{\pm}1.26g/dl$ in the Exp. I group. and $3.67{\pm}0.56g/dl$ in the Exp. II group. There was no significance in the serum globulin between Exp. II group and Exp. I group from the stastical analysis 6. In histological finding, because of severe inflammatory reaction, remarkably irregular tissue and large amount of inflammatory cells were found in the Exp. I group. But the Exp. II group showed small amount of inflammatory cells, the refrained inflammatory state and even recovering state. From these results, it is showed Taraxaci herbal-acupuncture refrain inflammatory reaction and muscular tissue necrosis in SD rats paw were induced by Freund's complete adjuvant

미역 (Undaria pinnatifida) 국수가 SD계 흰쥐의 혈청 지질대사에 미치는 영향 (Effect of Brown Algae (Undaria pinnatifida)-Noodle on Lipid Metabolism in Serum of SD-Rats)

  • 미역분말-첨가 미역국수의 지질대사에 미치는 생리적 효능을 구명하기 위하여 SD계 흰쥐에 4주 동안 미역국수 (BA-noodles)를 투여하여 중성지질 (TG), 총콜레스테롤 (TCh), LDL 및 HDL-콜레스테롤, 그리고 동맥경화지수 (AI)에 미치는 BA-noodles의 영향을 평가하였다. SD계 흰쥐에 조제사료로써 4주 동안 사육하면서 $10\%,\;20\%\;40\%$ BA-noodles의 TG의 함량은 $84.78\pm3.52\~91.30\pm3.77$ mg/dl serum로서 대조군 ($99.77\pm4.25$ mg/dl serum: $100\%$ ) 대비 $10\~15\%$정도나 유의적으로 감소하였다. 같은 방법으로 $10\%,\;20\%\;40\%$ BA-noodles의 TCh의 함량은 $87.28\pm4.97\~91.00\pm3.72$ mg/dl serum로서 대조군 ($99.30\pm4.61$ mg/dl serum: $100\%$)대비 $8\~12\%$정도나 유의적으로 감소하였다 또한 $10\%,\;20\%\;40\%$ BA-noodles의 LDL-콜레스테롤의 함량은 $44.20\pm4.21\~46.00\pm4.41$ mg/dl serum으로서 대조군 ($53.75\pm2.18$ mg/dl serum: $100\%$)대비 $15\~18\%$나 유의적으로 감소하였다. 한편 $10\%,\;20\%\;30\%$ BA-noodles 투여군의 HDL-콜레스테롤의 함량은 각각 $25.36\pm1.12\~27.58\pm1.52$ mg/dl serum로서 대조군 ($23.80\pm0.77$ mg/dl serum: $100\%$) 대비 각각 $7\~16\%$정도나 유의적으로 증가하였다. 끝으로 $10\%,\;20\%\;40\%$ BA-noodles 투여군의 AI는 $2.16\pm0.14\~2.59\pm0.17$로서 대조군 ($3.00\pm0.13$ : $100\%$) 대비 $14\~28\%$나 현저히 감소하였다. 이상의 결과에서 볼 때 이들 기능성 미역국수의 투여는 성인병을 효과적으로 억제할 수 있을 것으로 기대된다.
