Effects of Antler Development Stage on Fatty acid, Vitamin and GAGs Contents of Velvet Antler in Spotted Deer (Cervus nippon)

  • This study aimed to provide basic information as the foundation for further studies on the assessment of velvet antler quality by investigating the changes in fatty acid, vitamin A and E, minerals and GAGs contents by development stage of antler in spotted deer (Cervus nippon). Twelve stags (aged 4 to 5 years) were divided into two groups and velvet antler harvested 40 days (FDG) and 60 days (SDG) after casting of the buttons from the previous set. Total saturated fatty acid was lower in FDG than SDG (p<0.05). Total monounsaturated fatty acid, conjugated linoleic acid (p<0.05), polyunsaturated fatty acid and ${\omega}$3 fatty acid were higher in FDG than SDG. The vitamin A content of FDG was higher than that of SDG, but the vitamin E content of FDG was lower than that of SDG (p<0.05). SDG had significantly higher calcium and phosphate content than FDG (p<0.05). The magnesium content showed a similar trend to the contents of calcium and phosphate, but there was no significant difference between SDG and FDG. Uronic acid content was higher in FDG than SDG but there was no significant difference. The contents of GAGs and sialic acid were significantly higher (p<0.05) in FDG than SDG. These results indicated that the longer stage of antler development had lower content of activating components and this lead to a decrease of antler quality.

  • A behavioral investigation in deer equipped with EMG telemetry system was carried out to examine the chewing behavior pattern for efficient feeding management of Korean spotted deer(Cervus nippon). There was more frequent and irregular eating and rumination behavior pattern in deer than those in other ruminants. Time spent on eating and ruminating was 261 and 291 min., respectively. Total number of boli regurgitated was 647 times per a day and deer regurgitated an average boil of 21.6 times per a rumination period. Total rumination time was 291 min. and deer have an average rumination time of 26.9 sec. per a boli. Deer had 245.3 min. for total chewing time, 20,405 times fur number of chews, 31.5 times for number of chews per bolus, and 53.4 times for number of chews per minute during rumination period. An average regurgitation bolus an hour was lower with 15.3 numbers in the 09∼15 hours subperiod and higher with 26 numbers in the 15∼21 hours subperiod and it had been gradually trended to become to low after 15∼21 hours subperiod. An average numbers of chews a bolus at every 6 hours subperiod increased gradually according to time. A deer spent 23.3 min. on eating and 26.2 min. on ruminating per 100g ingested dry matter. Consequently, since deer have a different chewing behavioral pattern to other ruminants such as more frequent and irregular eating and ruminating patterns, small bolus and low number of chew, it is likely to needed to establishment and development efficient feeding system for deer.

Studies on Isolation and Characterization of Anaerobic Bacteria from Gut of Holstein Cows and Korean Male Spotted Deer (꽃사슴과 Holstein 젖소의 장내 혐기성 박테리아의 분리 및 특성)

  • The purpose of this study was to isolate cellulolytic and hemicellulolytic anaerobic bacteria inhabiting from gut of ruminants and investigate their hydrolytic enzyme activities. Extracellular CMCase activities of H-strains isolated from the rumen of a Holstein dairy cow were higher than those of D- and DC- strains from the rumen and large intestine of Korean spotted deer. Most isolated bacteria utilized more efficiently Dehority's artificial medium containing starch, glucose and cellobiose (DAS) than those in Dehority's artificial medium containing cellulose only (DAC). The results of biochemical reactions and sugar fermentation indicated that the isolated bacteria belong to one of bacterial strains of Peptostreptococcus spp., Bifidobacterium spp., Prevotela ruminicola/buccae, Clostridium beijer/butyricum and Streptococcus intermedis which are not highly cellulolytic. Activities of Avicelase, xylanase, β-D-glucosidase, α-L-arabinofuranosidase and β-xylosidase of the isolated anaerobic bacteria in DAS were higher than those in DAC. In conclusion, the results indicated the higher enzyme activities of the isolated strains cultured in DAS medium were mainly caused by their specific carbohydrate utilization for enzyme production and growth rate. The highly cellulolytic bacteria were not isolated in the present experiment. Thus further research is required to investigate characteristics of gut bacteria from Korean spotted deer.

Effects of Dietary Protein Level on Production and Chemical Composition of Velvet Antler in Spotted Deer (Cervus nippon) (꽃사슴에 있어서 사료의 단백질 수준이 녹용 생산성과 품질에 미치는 영향)

  • The aim of this study was to provide basic information to allow improved feeding management for velvet antler production by investigating the effects of dietary protein level on production and chemical composition of velvet antler in spotted deer(Cervus nippon). Eighteen male spotted deer of 5~6 years old were assigned to 3 unreplicated groups by production record of the previous year. Deer were fed on experimental diets at different protein level of 10%(T1), 15%(T2) and 20%(T3). Dry matter intake, crude protein intake and daily gain were increased with increasing the protein level in diets and there were significant differences(P<0.05) between groups. Although not significant, mean length of main beam tended to be longer in either left or right beam with increasing the protein level, and in girth at part of base of main beam, it was a similar pattern to the result of length. The velvet antler production was affected by dietary protein level and thus that was the smallest in T1 and the largest in T3(P<0.05). The content of crude protein in velvet antler was higher in T1 than in T3, whereas the content of crude ash in velvet antler was contrast to that of crude protein, however, there were no significant differences.

Effect of Selenium Feeding on Selenium Concentration of Blood and Velvet Antler in Sika deer(Cervus nippon) (셀레늄 급여가 꽃사슴의 혈액과 녹용내 셀레늄 함량에 미치는 영향)

  • This study was conducted to investigate the effects of selenium feeding and supplementation in diet on the concentration of selenium in blood and velvet antler of spotted deer(Sika deer). Three spotted deer were fed high selenium concentration(6mg/kg DM). Absorption and retention rates of selenium were examined by evaluating selenium concentrations in feces and urine. Stress-related hormones and serum biochemical parameters in blood were also evaluated for the purpose of detecting any negative effect by the high level of selenium feeding. Eight spotted deers were randomly assigned to two groups and were fed with one of two diets for 20 days, which were with or without the addition of 6mg selenium /kg diet. Concentration of selenium in velvet antler was evaluated. Selenium concentration in blood of spotted deer fed high level selenium for 30 days was significantly increased(p<0.05), retention rate of selenium reached 59.15%. No differences in level of stress-related hormone and biochemical parameters(NEFA, ALT, AST) in blood were observed by feeding high level selenium. The diet with selenium significantly increased concentrations of selenium in top(0.11 vs 0.45ppm; p<0.001), middle(0.08 vs 0.21ppm; p<0.01) and basepart(0.08 vs 0.15ppm; p<0.05) of velvet antler.

A Study on the Seasonal Comparison of Dry Matter Intake, Digestibility, Nitrogen Balance and Feeding Behavior in Spotted Deer (Cervus nippon) Fed Forest By-products Silage and Corn Silage

  • The purpose of this experiment was to assess seasonal variation of feed utilization by feed sources and to obtain information on the use of feed resources by comparing seasonal changes of dry matter intake, digestibility, nitrogen balance and feeding behavior in spotted deer (Cervus nippon) fed forest by-product silage (FBS) and corn silage (CS). Dry matter intake (DMI) of FBS was higher than that of CS in both winter and summer. While DMI of both diets was higher in summer, this was not significant at the 5% level. In contrast to DMI, digestible dry matter intake (DDMI) was higher for CS than for FBS in both seasons, but the difference was not significant. Digestibility of dry matter and crude protein was significantly higher (p<0.01) for CS than for FBS, whereas digestibility of crude fiber was significantly higher (p<0.01) for FBS than for CS in both seasons. Seasonal digestibility of dry matter and crude fiber for FBS was significantly greater (p<0.01) in summer than in winter: In summer, seasonal digestibility was 57.2% for dry matter and 55.5% for crude fiber, and in winter, 50.8% for dry matter and 30.7% for crude fiber. On the other hand, seasonal digestibility of crude protein was higher (p<0.01) in winter (42.1%) than in summer (32.3%). No significant difference (p>0.05) was found between the two seasons and diets for nitrogen intake (NI), 18.7 g/d in summer and 19.4 g/d in winter for FBS, 17.7 g/d in summer and 17.7 g/d in winter for CS. Fecal nitrogen was higher (p<0.01) for FBS than for CS and varied little seasonally. There was significant difference (p<0.01) between two seasons in urinary nitrogen, which was little difference between two diets. Retained nitrogen (RN) was different significantly (p<0.01) between two diets in both seasons, but there was little difference between seasons. Deer usually spent longer time on eating FBS than eating CS. Eating FBS took 221 min in summer and 187 min in winter, whereas eating CS took 113 min in summer and 109 min in winter. Deer spent less time on eating food in winter than in summer. Time spent on rumination was longer for FBS than for CS: for FBS, 504 min in summer and 456 min in winter, for CS, 423 min in summer and 279 min in winter. Time varied seasonally with both diets.

  • This experiment was conducted to examine possibility and suitability of forest by-product as a roughage source of deer. As a experimental diet, forest by-product silage(FPS) and commercial mixed ration(CMR) were prepared to compare dry matter intake, digestibility, nitrogen balance and feeding behavior in feeding trials with spotted deer. The digestibility of dry matter was higher(P<0.05) in CMR than in FPS. The digestibility of crude protein was higher in CMR than in FPS and there was significant difference(P<0.05). CMR had more digestibility of crude fiber than FBS but the difference was not significant. Dry matter intake were similar in both diets. Digestible dry matter intake was higher in CMR than FPS, however, there was no significant difference between experimental diets. Nitrogen intake, urinary and digestible nitrogen were significantly higher(P<0.05) in CMR than FPS. The efficiency of nitorgen utilization was higher for FPS than for CMR. Feeding behavior were similar in FPS and CMR. Consequently. It would be expected that FPS that composed with various feed sources can be used as a good roughage source of deer. Because it has high digestibility, dry matter intake and internal availability of nitrogen.

  • A comparison of intake, digestibility and nitrogen balance in spotted deer(Cervus nippon) fed forest by-product silage(FBS), arrowroot silage(ARS) and oak leaf hay(OLH) was made to examine the feeding value of forest by-product silage. Dry matter digestibility of ARS was significantly higher(p$<$0.05) than that of OLH while crude protein digestibility was significantly higher(p$<$0.05) in ARS and FBS than in OLH. The digestibility of crude fiber was highest in FBS. Dry matter intake of OLH was significantly higher(p$<$0.05) than that of ARS, and there was no significant difference with FBS. Nitrogen intake was higher in OLH and ARS than in FBS, but there was no significant difference. Fecal nitrogen was highest(p$<$0.05) in OLH and urinary nitrogen was highest(p$<$0.05) in ARS. Retained nitrogen was highest in FBS, however, there was no significant difference among treatments. In Conclusion. FBS was estimated as a useful roughage source for deer, showing high digestibility, dry matter intake and nitrogen utilization.

Effect of Cellulose Degrading Bacteria Isolated from Wild and Domestic Ruminants on In vitro Dry Matter Digestibility of Feed and Enzyme Production

  • Cellulolytic bacterial strains have been isolated from the faeces of wild (blackbuck, Antilope cervicapra; nilgai, Baselophus tragocamelus chinkara, Gazella gazella spotted deer, Axis axis and hog deer, Cervus porcinus) and rumen liquor of domestic (sheep, Ovis aries) ruminants. Five best cellulose degrading bacterial isolates (Ruminococcus sp.) were used as microbial feed additive along with buffalo rumen liquor as inoculum to study their effect on digestibility of feed and enzyme production in in vitro conditions. The bacterial isolate from chinkara (CHI-2) showed the highest per cent apparent dry matter (DM) digestibility ($35.40{\pm}0.60$), true dry matter digestibility ($40.80{\pm}0.69$) and NDF ($26.38{\pm}0.83$) digestibility (p<0.05) compared to control ($32.73{\pm}0.56$, $36.64{\pm}0.71$ and $21.16{\pm}0.89$, respectively) and other isolates at 24 h of incubation with lignocellulosic feeds (wheat straw and wheat bran, 80:20). The same isolate also exhibited the highest activities of fibre degrading enzymes like carboxymethylcellulase, xylanase, ${\beta}$-glucosidase and acetyl esterase. The bacterial isolate from chinkara (Gazella gazella) appears to have a potential to be used as feed additive in the diet of ruminants for improving utilization of nutrients from lignocellulosic feeds.
