Application of Lactic Acid Bacteria to Inhibit Fungal Contamination of Cured Cheeses (항곰팡이능 보유 유산균의 숙성치즈 적용 연구)

  • Lactic acid bacteria with antibacterial activity can be effectively used as probiotics to inhibit the growth of harmful bacteria that cause food spoilage or food poisoning. In this study, Pediococcus pentosaceus M132-2, isolated from soybean paste, was analyzed for its effects on three major contaminating fungi. M132-2 was confirmed to exert antifungal activity by inhibiting the growth of all three fungi tested. In addition, M132-2 displayed excellent salt resistance and low temperature tolerance. Thus M132-2 can survive at the salinity level in cheese and at the low temperatures used in the aging process. Finally, when supernatant from an M132-2 culture was applied to Gouda cheese, the growth of contaminating fungi was significantly inhibited. Consequently, M132-2 may be useful for the prevention of spoilage of various foods, including cheese.

Effect of coating with combined chitosan and gallic acid on shelf-life stability of Jeju black cattle beef

  • Objective: Beef of Jeju black cattle (JBC) is considered as a healthy meat type due to its significantly higher unsaturated fatty acids (UFA). Lipid (e.g., UFA) is highly susceptible to oxidizing agents, which results in the quality deterioration and economic value loss of meat products. Therefore, development and application of novel preservative techniques is necessary to improve the shelf-life stability of high-UFA beef. The objective of this study was to assess the applicability of chitosan-based coatings in preservation of JBC beef. Methods: Different coating solutions: 2% chitosan alone, and 2% chitosan containing 0.1% or 0.3% gallic acid were prepared to investigate their applicability in preservation of fresh beef during storage. Jeju black cattle beef (2-cm thick steaks) were non-coated (control) or coated with the above coating solutions, placed on trays, over-wrapped with plastic film and stored at 4℃. The microbiological indices, color, total volatile basic nitrogen (TVBN) and lipid oxidation of the beef were investigated after 1, 10, and 21 days of storage. Results: Coating with 2% chitosan alone reduced the spoilage bacteria count, TVBN and thiobarbituric acid reactive substances levels in the beef compared with control during storage (p<0.05). Noticeably, coating with 2% chitosan containing 0.1% or 0.3% gallic acid was more effective on retardation of spoilage bacteria growth, lipid oxidation and discoloration in the beef compared to the chitosan coating alone over the storage period (21 days) (p<0.05). Conclusion: Taken together, the combined chitosan and gallic acid coating could be used as a bio-preservative technique in the meat industry.

Changes in the Quality of Peaches (Prunus persica L. Batsch) Treated by UV-C Irradiation during Storage (Ultraviolet-C 조사에 의한 복숭아의 저장 중 품질변화)

  • To prolong the shelf-life of domestic peaches, samples were treated with UV-C (0~10.0 $kJ/m^2$) and the spoiling rate and changes in physico-chemical and sensory properties were investigated. No spoiled peaches were observed within the first four days of storage in the control and 2.5 $kJ/m^2$ UV-C treatment groups. However, spoilage was observed in these groups on day six, and 29.17% and 25.0% of the samples showed spoilage on 10 day, respectively. Moreover, samples treated with greater than 5.0 $kJ/m^2$ of UV-C showed a higher percentage (41.67% or higher) of spoilage than those of the control or 2.5 $kJ/m^2$ UV-C treatment groups on 10 day. Additionally, weight changes in the peaches were the lower in the control group and 2.5 $kJ/m^2$ UV-C treatment group than in those treated with 5.0 $kJ/m^2$ of UV-C treatment or higher during 10 days of storage. There was no difference in pH among treatments, regardless of storage time. The hardness of the samples was not changed immediately after UV-C treatment, but that of samples treated with 5.0~10.0 $kJ/m^2$ of UV-C decreased rapidly after four days, when compared to the control and 2.5 $kJ/m^2$ UV-C treatment groups. No significant changes in the lightness and redness of the samples were observed in response to UV-C treatment, however, UV-C treatment led to a slight decrease in the yellowness of the samples. The initial taste, flavor, color, texture, and overall acceptance did not differ among control and UV-C treatments. The sensory score of the samples was the highest after 2 and 4 days of storage, while it decreased thereafter. In general, samples in the control and the 2.5 $kJ/m^2$ UV-C treatment groups showed higher sensory quality than those treated with UV-C at 5.0 $kJ/m^2$ or higher.

Cultural and chemical approaches for controlling postharvest diseases of garlics (마늘 저장병 방제를 위한 경종적, 화학적 접근)

  • The purpose of this study was to investigate damages of garlics occurred under cold storage and farmhouse storage condition, influence of cultural and environmental factors on storage spoilage of garlics, and to establish control strategies to reduce damages occurred under storage of garlics. Decays of garlics were highly related with cultural condition (paddy field or upland soil), ventilation, storage temperature and relative humidity, continuous cropping years, and harvesting stage. Early-harvested garlics were more decayed than late-harvested garlics. Garlics cultivated on paddy field were less decayed than ones cultivated on upland soil under farmhouse storage condition. The densities of Penicillium spp. and Fusarium spp. were higher on plot with long term continuous cropping cultivation history than on plot with short term continuous cropping cultivation history. However there is no relation between continuous cropping years and percent of decay of garlics. As a result of investigating influence of environmental factors on decay of garlics, P. hirsutum caused severe spoilage under high relative humidity condition, while F. oxysporum and Stemphyllium botryosum were not related with relative humidity. The three postharvest pathogens grew well above woe. In addition when P. hirsutum and S. botryosum were cultured for two months, they grew even at $-1^{\circ}C$. Except for environmental factors, wounds occurred through farming works. had an effect on storage spoilage of garlics. Garlics only hurt with a toothpick without inoculation of pathogens were decayed more severe than those inoculated with pathogens without wounds. Seven agro-chemicals showed highly suppressive effect were selected by measuring mycelial growth of three major pathogens of garlics on potato dextrose agar amended with 0.1% (v/v) of each fungicide. When they were foliar-sprayed on garlics 30 days before harvesting, it was confirmed that they suppressed storage spoilage of garlics. Also when garlics were sprayed with and drenched into the suspension of Benoram WP very after harvesting garlics, garlic damages by postharvest pathogens were reduced remarkably.

Effect of Pichia anomala SKM-T and Galactomyces geotrichum SJM-59 Dipping on Storage Property and Sensory Quality of Strawberry

  • Simple competition plate bioassays of Pichia anomala SKM-T and Galactomyces geotrichum SJM-59 were conducted to evaluate their potential as biological control agents that inhibit growth of Botrytis cinerea during post-harvest storage of strawberries (Fragaria ${\times}$ ananassa Duche, Red-Pearl). Occurrence rates of fungi on the surface of yeast-treated strawberries were evaluated during storage at $4^{\circ}C$. P. anomala SKM-T and G. geotrichum SJM-59 showed antifungal activities on agar plate, and P. anomala SKM-T maintained its desirable antifungal activity on surface of strawberries and its physicochemical properties during storage. Sensory evaluation was based on kinesthetics and consumer acceptability. Due to its potential antifungal activity, P. anomala SKM-T could function as biological control agent against spoilage fungi during post-harvest storage of strawberries.

A Study of Antioxidant and Antibacterial Activates of the Extraction of Perscaria hydropiper L.

  • This purpose of this study is to examine the antioxidant and antimicrobial activities of Persicaria hydropiper L. extract in 70% ethanol and in water, a medicinal herb, as an effort to examine the potential of medicinal herbs for development of antioxidants and natural preservative substitutes. The total poly-phenol content in the 70% ethanol extract and in the water extract were 19.88 mg/g and 7.46 mg/g, respectively. The DPPH radical scavenging activity was 90.99% and 64.98% in the 70% ethanol extract and water extract, in which 70% ethanol extract showed a higher activity. The antioxidant effect of Persicaria hydropiper L. extract appears to be very good and due to its excellent growth inhibitory effect on food-poisoning-causing microorganisms in the food, it is thought to be utilized as a potential natural preservative substitute in many areas.

Microbiological Quality Control of Cooked Rice by Aseptic Process Using Heat Shock Treatment (열쇼크방식 살균에 의한 무균 취반의 미생물학적 품질관리)

  • Newly introduced aseptic process for cooked rice was optimized by evaluating the process parameters. The optimal conditions were as follows: ratio of washing water to rice, 3:1 by weight; steeping at 20oC for 20min; heat shock at 140oC for 7sec with 8 repetitions; addition of 0.2% glucono lactone solution; cooking at 103oC for 30min. The final products were incubated at 37oC and 50oC for 8 weeks and no spoilage was occurred at both temperatures. Furthermore, any significant losses of sensory and physicochemical qualities were not observed.

Scientific Consideration in Determining Shelf Life of Market Milk (시유의 유통기간 결정에 관한 학문적 고찰)

  • The shelf lift of market milk should be determined based on the flavor which is influenced by environmental and sanitary conditions of dairy farm, milk processing plant, and storage and transportation facility as well as compositional quality, such as protein and fat, of the milk itself. The legal shelf life of market milk is often limited by microbiological quality, e.g. total bacterial count, coliform count, and food poisoning bacteria. The bacteria involved with milk spoilage and poisoning are originated from bacteria contaminating milk after pasteurization or spores surviving the heat treatment of pasteurization. The important factors which influence the shelf life of market milk are microbiological quality of raw milk, pasteurization condition, post-pasteurization contamination, and temperature during storage and transportation. The organoleptic quality and shelf life of market milk should be further improved by satisfying the consumer's taste, which depends on somatic cell count and bacterial count of milk, feed quality, foreign substance in milk, and physical treatment during processing and transportation.

  • Bacteriocins from lactic acid bacteria have attracted much attention in recent years because of their useful worth in increasing safety and extending shelf life of foods. These substances show an inhibitory effect against some food spoilage bacteria and food-borne pathogens. The inhibitory effect fo the bacteriocin produces by lactic acid bacteria against Listeria monocytogenes(L. monocytogenes) was examined in this study. The culture supernatants of 5 kinds of bacteria among the 10 kinds of testes lactic acid bacteria had the inhibitory activity against Listeria sp., various Gram positive and Gram negative bacteria. Bacteriocin produced by Lactobacillus plantarum(Lact. plantarum) LMG 7945 was the most active toward L. monocytogenes. Bacteriocin production of the Lact. plantarum LMG 7945 cultured on MRS broth was increased late logarithmic phase over early stationary phase. This bacteriocin was stable at heat treatment and acidic pH relatively; The activity was retained after heating at 121$^{\circ}C$ for 15min and was active in the pH range of 2~4 but was lost above pH 5.

Microbiological Evaluations of Retail and Refrigerated Chickens in Winter (겨울철 소매점 및 냉장 닭고기의 미생물학적 평가)

  • Aerobic plate counts(APC) gram-negative bacterial counts (GNC) and sensory evaluations on chic-ken carcasses during retail and refrigerated storages (3$\pm$1$^{\circ}C$ and 1$0^{\circ}C$) were evaluated. APC and GNC on whole chicken in retail store after storage of 7 days at 3$\pm$1$^{\circ}C$ increased to 3.11 and 3.89 log units com-pared to the initial controls. APC and GNC on whole chicken after storage of 7 days at 1$0^{\circ}C$ increased to 5.43 and 5.03 log units. Sensory scores of chicken carcasses obtained from retail store were in the "liked less" category after storage of 7 days compared to fresh controls. These results indicated that chicken carcasses during refrigerated (1$0^{\circ}C$) storages rapidly allowed the growth of aerobic spoilage bacteria dur-ing storage period which cluld not be microbiologically acceptable after of 7 days of 7 days.
