Immunomodulating Activities of Copolang, a Proteopolysaccharide from Coriolus versicolor in Lewis Lung Carcinoma (LLC) Bearing mice (Lewis Lung Carcinoma(LLC) 이식 생쥐에 있어서 천연운지 단백 다당체(Copolang)의 면역조절활성)

  • Immune functions of mice bearing Lewis Lung Carcinoma (LLC) were significantly suppressed when evaluated with mitogen responsiveness, IL-2 production and non-specific suppressor activity. Based on these immunosuppressive characteristics of LLC bearing mice, immunomodulating activates of Copolang were investigated in this model. After 15 days of LLC inoculation, Copolang was intraperitoneally administered for 7 consecutive days with doses of 20 or 200 mg/kg. Immune functions were evaluated 3 days after the final administration of Copolang. The results showed that the growth of LLC solid tumor was not inhibited by Copolang. But, mitogens-induced proliferation, IL-2 production and responsiveness to recombinant IL-2 of splenocytes were significantly augmented by the treatment of Copolang. However suppressor cell activity was not affected by Copolang. These results indicate that Copolang expresses potent immunomodulating activates through the augmentations of IL-2 production and responsiveness to recombinant IL-2, which have been generally known to be suppressed in tumor bearing mice, without affecting the growth of tumor.

  • Effects of chrysene on immune functions were studied in female BALB/c mice. When mice were treated po with chrysene for 7 consecutive days, the antibody response was suppressed dose-dependently. Chrysene induced the enzyme activities of CYP LA and 2B time- and dose-dependently. In ex vivo lymphocyte proliferation, chrysene inhibited splenocyte proliferation by LPS and Con A. Moreover, the numbers of $CD4^+IL-2^+$ cells were reduced by chrysene. In conclusion, chrysene-induced immunotoxicity might be mediated, at least in part, via IL-2 production, and caused by mechanisms associated with metabolic activation.

  • The purpose of this research was to investigate the immunoregulatory effect, apoptosis of L 1210 leukemia cells of Comus officinalis Sieb.et Zucc. The proliferation of cultured splenocytes and thymocytes were enhanced by the addition of SSY. Splenic, thymic and mesenteric lymph node-T lymphocytes, especially TH cells was significantly increased in SSY-administered (p.o. for 7 days) mice. SSY treatment induced the apoptosis of L1210 mouse leukemia cells. In addition, SSY accelerated the phagocytic activity and nitric oxide production in peritoneal macrophages. These results suggest that SSY have an immuno-regulatory property and anti-cancer effect.

  • Polychlorinated biphenyls (PCBs) has been widely used as plasticizer, insulator, lubricant, paint and ink. The persistence of PCBs in the environment and their bioaccumulation in living organism make a raise concerns regarding their toxic effects in immune system and subsequent effects on human health. However little has been known about effect of PCB, an endocrine disrupter, on splenocytes. In this study, for identifying the effect on the organs and immune cell of mice by the concentration and time of commercial PCB mixture (Aroclor 1254), each 3 mice were tested at the concentration of 3, 30, 300, 1,000 mg/kg respectively, and their organ's weight were measured in 4, 7, 14 days, respectively. Also according to concentration and time, PCB was evaluated for the effects on splenocyte viability and lipopolysaccaride (LPS) and concanavaline A (Con A)-induced splenocyte proliferation on mice spleen. In liver and lung, there were significantly defferent by concentration and time of PCB (p < 0.0001). In respect of concentration of PCB, no significant effects on mice's liver by Aroclor 1254 concentration below than 300 mg/kg were observed except at the concentration of 1,000 mg/kg doses (p < 0.0001). But there was not significant different change in mice spleen by concentration and time of PCB (p=0.2206) and the mode of weight change of spleen was different to of liver and of lung. Viabilities of splenocytes were decreased following treatment with high concentration of PCB. Also, LPS and Con A-induced cell proliferations were decreased by Aroclor 1254 at 1,000 mg/kg. These data suggest that Aroclor 1254 is the immunotoxic compound that may have an effect on mouse immune system.

  • High soy consumption leading to high exposures of soy isoflavones has been associated with a reduced risk of cancers at many sites. As part of a study focusing on the chemopreventive mechanisms, we have investigated the modulating effects of daidzein and genistein, a prominent and more bioavailable isoflavone in soy foods, on murine immune function. Daidzein (50 mg/kg) or genistein (50 mg/kg) was administered p.o. once a day for 7 days in BALB/c mice. Daidzein decreased the mitogen-stimulated proliferation of murine splenocyte, but genistein increased it. Daidzein stimulated the secretion of interleukin-4, but inhibited the secretion of ${\gamma}$-interferon, interleukin-2 and tumor necrosis factor-$\alpha$. Genistein stimulated the secretion of ${\gamma}$-interferon, interleukin-2 and tumor necrosis factor-$\alpha$, but inhibited the secretion of interleukin-4. Daidzein and geiustein inhibited the production of nitric oxide and enhanced the phagocytic activity in peritoneal macrophage. These results suggest that cancer preventive effects of daidzein is partly concerned with the secretion of $T_{H}$2 cells cytokine and the activation of macrophage phagocytosis, and genistein is partly concerned with the secretion of $T_{H}$l cells cytokine, tumor necrosis factor-$\alpha$ and the activation of macrophage phagocytosis.sis.

Stimulation of the Immune Response by Herbal Formulas for Taeeumin (대표적인 태음인 처방의 면역 활성화 비교 연구)

  • 1. Objectives: Three herbal formulas (Yuldahanso-tang, Chungsimyonja-tang, and Taeeumjowi-tang) for Taeeumin were applied to investigate the immunological activities on antigen (Ag)-specific or Ag-non-specific immune responses in murine macrophage cell line (RAW 264.7) and ovalbumin (OVA)-immunized mice. 2. Methods: This study was carried out in nitric oxide (NO) synthesis in RAW 264.7 cells and cellular proliferation in mouse splenocytes according to three herbal formulas. C57BL/6 mice were immunized intraperitonially with OVA/aluminium ($100\;{\mu}g/200\;{\mu}g$/mouse) on day 1, 8, and 15. Three herbal formulas were administrated to mice orally for 3 weeks from day 1. On day 22, OVA-, lipopolysaccharide (LPS)-, and concanavalin A (Con A)-stimulated splenocyte proliferation and antibodies (OVA-specific antibodies of the IgG, IgG1, and total IgM classes) in plasma were measured. 3. Results: Yuldahanso-tang and Chungsimyonja-tang increased NO synthesis in RAW 264.7 cells. Three herbal formulas significantly enhanced cellular proliferation by LPS and Con A in splenocytes from OVA-immunized mice (p<.001). Three herbal formulas for Taeeumin also significantly enhanced plasma OVA-specific IgG, IgG1, and total IgM levels compared with the OVA/Alum group. 4. Conclusion: These results suggested that three herbal formulas for Taeeumin could be used as stimulator of immune response.

Enhancing Effect of Pleurotus ostreatus Extracts on Mouse Spleen and Cytokine Cells Activation (느타리버섯 물 추출물 투여에 의한 마우스 비장세포 및 사이토카인 활성효과)

  • Pleurotus ostreatus have been used as a traditional remedy and food source. Its anti-inflammatory, anti-oxidation effects have been previously reported. The production of cytokines (IL-$2{\beta}$, IFN-${\gamma}$, and TNF-${\alpha}$) secreted by LPS- and non LPS-stimulated macrophages, were detected by ELISA assay using a cytokine kit. Mouse splenocytes proliferation increased with Pleurotus ostreatus water extracts supplement at 5, 10, 50, 100, 250, 500, $1,000{\mu}g/mL$ after a 48hr pre-treatment with the mitogen (ConA or LPS). The cytokine production (IL-$2{\beta}$, IFN-${\gamma}$, and TNF-${\alpha}$), measured by a cytokine ELISA kit, increased on water extracts supplementation. This in vitro study suggested that supplementation with Pleurotus ostreatus water extracts may enhance the immune function by regulating the splenocyte proliferation and enhancing cytokine production.

  • Plantago asiatica L., observed frequently in East Asia, is a known herb used in traditional medical remedies several studies report that P. asiatica L has anti-inflammatory and antioxidant effects. To determine the production of cytokines (IL-2, $IFN-{\gamma}$, and $TNF-{\alpha}$) induced by lipopolysaccharide (LPS) and non-LPS-stimulated macrophages, an ELISA assay was conducted using cytokine kits. Mice splenocytes were cultured for 48 h with various concentrations of P. asiatica L. (5, 10, 50, 100, 250, 500, and $1,000{\mu}g/mL$) or with mitogens (ConA or LPS). P. asiatica L. increased the proliferation of mice splenocytes, especially under the condition of its concentration ranging from 250 to $1,000{\mu}g/mL$. In addition, Plantago asiatica L. notably induced cytokine production of (IL-2, $IFN-{\gamma}$, and $TNF-{\alpha}$) at its concentration of $250{\sim}500{\mu}g/mL$. These results suggest that supplementation with P. asiatica L. water extracts may play a potential role in enhancing immune function by mediating splenocyte proliferation and cytokine production through its anti-inflammatory activit.
