The protective effects of sonicated Bordetella bronchiseptica bacterin on the immunosuppression of spleen cells induced by anti-cancer drugs (5-fluorouracil, doxorubicin, and vincristine) (항암제 (5-fluorouracil, doxorubicin, vincristine)로 인한 비장세포의 면역억제에 대한 Bordetella bronchiseptica의 보호 효과)

  • 5-Fluorouracil, doxorubicin, and vincristine are chemotherapy agents used to treat various cancers, such as breast cancer and lymphoma for decades, and their effects on cancer have been proven. On the other hand, these anti-cancer drugs cause fatal side effects, including immunosuppression. This study investigated whether sonicated Bordetella bronchiseptica bacterin (B. bronchiseptica) can attenuate the immunosuppression of spleen cells induced by these chemotherapy agents and which subsets of spleen cells were affected. B. bronchiseptica increased the metabolic activity of spleen cells treated with 3 anti-cancer drugs. Cell death analysis using Annexin V/propidium iodide showed that B. bronchiseptica markedly decreased the death of spleen cells. The subsets of spleen cells were analyzed by flow cytometry using a surface marker-specific antibody. B. bronchiseptica increased nitric oxide production in the spleen cells treated with anti-cancer drugs (p < 0.0001). Despite the pharmacological effects of anti-cancer drugs, many patients suffer from the fatal side effects of immunosuppression. This study provides valuable information on how to overcome chemotherapy-induced immunosuppression.

Differential cytotoxic effects of fenbendazole on mouse lymphoma EL-4 cells and spleen cells

  • Fenbendazole (FBZ) is a benzimidazole anthelmintic widely used to treat parasitic infections. The anticancer effect of FBZ has been recently highlighted leading to its consideration as a potential anticancer agent. Although previous studies have demonstrated the effect of FBZ on cancer cells, there is a paucity of studies on the effect of FBZ on lymphoma cells and normal immune cells. Herein, we investigated the effects of FBZ on a mouse lymphoma cell line, EL-4 cells, and spleen cells, using vincristine as a positive control. The cellular metabolic activity of EL-4 cells was decreased by FBZ, but that of the spleen cells was not decreased. Moreover, FBZ reduced the mitochondrial membrane potential and induced reactive oxygen species production in EL-4 cells, but not in spleen cells. FBZ induced G2/M phase arrest and increased the sub G0/G1 phase ratio, indicating apoptosis. Furthermore, compared to the control cells, the reactivity of spleen cells pretreated with FBZ to lipopolysaccharide was maintained. In summary, FBZ is cytotoxic to EL-4 cells, but not to spleen cells. This study provides experimental evidence that FBZ exerts an anticancer effect, and less cytotoxic effects and functional damage to normal spleen cells.

  • Objectives : It was designed for making theoretical base about the relation of the Spleen, Stomach and the liver which could be used in clinic. Methods : Including Huang Di Nei Jing(黃帝內經), the 22 kinds of books written about the relation of the Spleen, Stomach and the Liver, and 2 kinds of papers were referenced. They were divided and studied according to physiology, pathology, meridian system and treatment. This study is focused on internal organ theory. Conclusion 1. In physiology, the Liver has the function of smoothing and regulating the flow of vital energy(Ki), while the Spleen is in charge of digestion and transportation. 2. In pathology, there can be informed disharmony of the Spleen, Stomach and the Liver, blood deficiency in the Liver and Spleen, jaundice, and bleeding etc. 3. In meridian system, the junction points of Spleen Meridian and Liver Meridian are Liv14, SP6, SP12, SP13. 4. In treatment, 1) If there is disharmony of the Liver and Spleen, the Liver must be soothed and the function of Spleen invigorated. 2) If there is disharmony of the Liver and Stomach, the Liver must be soothed and the function of Stomach invigorated.

Correlation of ketone bodies in blood and spleen

  • Starvation, diabetes, alcoholism and hypothermia cause ketoacidosis in the human body; therefore, the cause of death can be determined by analyzing ketone bodies in the blood of the deceased. In the case of decomposition of the cadaver, however, since collecting intact blood is impossible, ketone body analysis is performed using the spleen. However, the index for diagnosing ketoacidosis is based on blood concentration, and its correlation with ketone bodies present in the spleen remains unknown. In particular, since decomposition proceeds rapidly during summer, when temperature and humidity are high, understanding the correlation between ketone bodies in the blood and spleen is important to estimate the state at the time of death from a decaying body. Therefore, in the present study, the correlation between ketone bodies in the blood and spleen of the deceased was explored. Ketone bodies (beta-hydroxybutyric acid [BHB] and acetone) in the blood and spleen were analyzed and compared from autopsies (>100 mg·L-1 BHB, blood basis) conducted at the Daejeon Forensic Research Institute from June to December 2021. Moreover, the concentration of ketone bodies in the spleen juice and tissues was compared assuming the scenario of extreme decomposition. Ketone retention concentration in the blood and spleen was positively correlated, and the ratio of BHB concentration in the spleen to BHB concentration in the blood ranged from 0.52 to 1.08 (mean = 0.85 ± 0.12), although the ratio may vary depending on the degree of decomposition of the corpse.

  • Sa-Hwang-San, which is invented by Jhun Eul for the treatment of tongue thrust of Spleen heat, is first mentioned on 'So Ah Yak Jeung Jik Gyul'. It is consisted of Gardeniae Fructus, Agastache rugosa, Glycyrrhizae Radix, Gypsum Fibrosum, Ledebouriellae Radix. According to 'Dong Ui Bo Gam', this medicine cures spleen heat, sore mouth and halitosis. In the viewpoint of pharmacology, this medicine as a whole can treat spleen stagnancy, spleen heat and stomach heat. However, there are some differences between the opinions of 'So Ah Yak Jeung Jik Gyul' and 'Dong Ui Bo Gam' the principal medicine of the former is Ledebouriellae Radix, while that of the latter is Gardeniae Fructus. Spleen stagnancy is the malfunction of spleen under the presence of the body fluid. Symptoms are abdominal dropsy and powerlessness. Spleen heat is the insufficiency of body fluid due to the heat of the spleen, while the function of the spleen is normal. Symptoms are sore mouth, halitosis and maxillary stagnancy. In clinical cases, Sa-Hwang-San has been found effective to the individuals with large, lowered, soft, and slanted mouths and lips. It was also beneficial for the YangMyong type. In clinical cases, Sa-Hwang-San is proven to be effective to the following symptoms soremouth, halitosis and maxillary stagnancy due to spleen heat abdominal dropsy and powerlessness due to spleen stagnancy.

Study on the Bitonghyul (비통혈(脾統血)에 관한 고찰)

  • Although Bi(脾 Spleen) is a major organ which controls digestion and absorption in Korean medicine, there also introduced another function of bi, which is called 'bitonghyul(脾統血)' that means the spleen controls(holdings) the blood. While the spleen is involved in the function of blood generation through the digestion and absorption of food, 'bitonghyul' is another expression regarding function of the spleen. The meaning of 'bitonghyul' is related with the function of keeping the blood flowing within the blood vessels, hemostasis. Comparing the function of the spleen with the mechanisms of hemostais, it is concluded that 'bitonghyul' is a physiological effects which affect hemostatic mechanisms via absorption of vitamin K.

Effect of Bobitang water extracts on immunosuppression induced by Methotrexate in SD rats (보비탕(補脾湯)이 methotrexate로 유발(誘發)된 흰쥐의 면역기능저하(免疫機能低下)에 미치는 영향(影響))

  • Objectives : In order to investigate the effect of Bobitang on SD rats with deteriorated immunity caused by methotrexate. Methods : The test sample were dosed once a day for 14 days by gastric gavage at a dosage 1,000, 500 and $250mg/kg/10m{\ell}$ from 2 days after last MTX-dosing, and the changes on body weight and gains, spleen weight and total blood leukocyte numbers, total lymphocyte numbers, the percentage of B-cell, T-cell, CD3+CD4+ T-cell, CD3+CD8+ T-cell and CD4+/CD8+ T-cell ratios in the blood and spleen were observed. Results : The changes on body weight gains, the spleen weight, the total blood leukocyte numbers, the total lymphocyte numbers in the blood and spleen, the ratio of T-cell in the blood and spleen, the ratio of CD3+CD4+ T-cell in the blood and spleen were increased significantly in BBT Extracts groups as compared with control group. The ratio of B-cell in the blood and spleen was not increased significantly in BBT Extracts groups as compared with control group. The percentage of CD3+CD8+ T-cell in the blood and spleen was decreased significantly in BBT Extracts groups as compared with control group. The ratio of CD4+/CD8+ T-cell in blood and spleen was increased significantly in BBT Extracts group as compared with control group. Conclusions : According to the above results, Bobi-Tang has an effect of increasing immune responses on SD rats with deteriorated immunity caused by methotrexate.

  • The aim of the present study was to investigate the development of theory of the deficiency of yin-yang in the spleen-stomach(脾胃). The spleen-stomach theory is a very valuable composition of the oriental medicine. Its first theoretical basis was established by Li Dong Yuan(李東垣) who wrote Piweilun("脾胃論"). He insisted the importance of spleen yang(脾陽) which is the "postnatal base of life" by transforming and transporting of food essence and fluids and raises the clear. After him, his theory had influenced many descendant medical men. one of them, Ye Tian Shi(葉天士) found out the differences of between stomach and spleen. Especially he focused on the stomach yin(胃陰) which is easy to be exhausted by dry-fire(燥火). And he also made another remedy, "the stomach yang should be moved well(宣通胃陽)", which emphasize on that cold and stagnant cause the deficiency of the stomach yang. After that, spleen yin(脾陰) theory was set up by Tang Zong Hai(唐宗海). His spleen yin(脾陰) was deeply related with digestive enzymes and pancreas, because his theory was established by comparing oriental and western medicine. These four theories became the theoretical basis of the deficiency of yin-yang in the spleen-stomach(脾胃), and similar symptom of the spleen-stomach(脾胃) could be categorized according to the pattern identification(辨證) which was developed from these four theory.

Immunomodulatory Effects of Aqueous-extracted Zingiberis rhizoma on Methorexate Induced Immune Suppression in Mouse Spleen Cell (건강 열수추출액이 Methotrexate에 의해 유도된 마우스 면역억제 조절에 미치는 영향)

  • Zingiberis rhizoma(ZB) has been used to treat a various condition and disease in traditional oriental medicine. The present study was conducted to evaluate the immunomodulatory effect of aqueous-extracted ZB(ZBE) on methotrexate (MTX)-induced immune suppression in mouse spleen cells. In spleen cell proliferation assay, ZBE enhanced mitogenic activity in mouse spleen cells. In RT-PCR, ZBE induced IL-2, IFNr and IL-6 cytokine gene expression in mouse spleen cells. In spite of MTX treatment, IL-2, IFNr and IL-6 gene expressions sustained in MTX treated spleen cells. CD45R/B220, pan B marker was slightly increased in ZBE treated mouse spleen cells. IL-6, B cell tropical cytokine, production was induced by ZBE-treated mouse spleen cells and IL-6 production was sustained on MTX-ZBE co-cultured cells. ZBE administration enhanced suNival of S-180 bearing mouse. These data indicate that ZBE has a protective effect of immune suppression caused by MTX, and ZBE may be enhance cellular and humoral function by regulate cytokine gene expression as well as the mitogenic effect on spleen cells.

  • An acute fatal infectious disease in rabbits has been outbroken in Korea since 1985. This disease has been characterized as an acute hepatitis caused by viruses. However, viral pathogenesis in rabbit viral hepatitis leading to sudden death remain unclear. This report dealt with the electron microscopic findings on the spleen of experimentally infected rabbits, because spleen is one of the affected organs which have high titer of virus by a haemagglutination test. A typical crystalline array of virus was not found in the splenic cells of infected rabbits with acute hepatitis. Virus-like particles were seen within the phagosome of macrophages of the spleen. Ultrastructural changes in the spleen were severe with the lapse of time after inoculation. From these results, virus-like particles in the spleen were supposed to be phagocytosed by macrophage during viremia, while active replication of virus occurred in the liver. It was concluded that sudden death in this viral disease was caused by hepatic coma and/or circulatory disturbance.
