Growth Characteristics and Grain Quality of Naked Barley Infected by Barley Yellow Mosaic Virus (보리호위위병(BaYMV)이병에 따른 쌀보리 품종의 생육특성 및 품질)

  • The growth characteristics and damage of naked barley infected with barley yellow mosaic virus (BaYMV) were investigated between resistant and susceptible varieties in habitual field plot of BaYMV BaYMV of the barley plants with typical disease symptom were identified by enzyme linked immunosorbent assay (ELISA) test. The visual degree (0-9) based on disease symptoms of BaYMV was different as 9 and 1 between susceptible variety Baegdong and resistant variety Naehanssalbori, respectively. Susceptible variety, Baegdong showed significant damage in culm length, number of kernel per spike and tiller per square meter but not in 1,000 kernel weight, so these results caused yield reduction to only 80% comparing to the control. Seed germination did not affected by BaYMV infection both in susceptible and resistant variety. In grain quality test, abortive grain yale and crude protein content were significantly increased compared to the control. The relationships between BaYMV infection and growth characteristics showed the negative correlations in culm length, number of tiller, 1000 kernel weight and yield, but it showed the positive correlation in crude protein contents. These results implied that BaYMV can affect not only barley growth and yield but grain quality.

Germinability during Overwintering, Field Emergence, and Growth of Shattered Rice Seeds on Paddy Field (논 표면 탈립 벼 종자의 월동중 발아력변화와 월동후 포장 출현 및 생육)

  • Germinability, electrophoretic variation of protein of shattered seeds during overwintering and characteristic of main agronomic traits of off-type rice plant occurring in paddy field harvested by combine were investigated. Germinability of shattered seed decreased as time goes in paddy field in both Japonica and Tongil type varieties. Electrophoretic protein bands become more and more light as time goes. Occurrence of off-type rice plant was higher in Japonica varieties than in Tongil type varieties. Off-type rice plant was shorter in plant height, leaf length, tiller number per hill, internode length, panicle length and spike let number per panicle. Germinability of seeds of off-type rice plant as not significantly different compared to the control variety. Yield of off-type plants of Japonica varieties was on average 5.1kg / 10a and that of Tongil type varieties was on average 0.9kg /10a. Mixing ratio to the control variety was about 0.7% in Japonica varieties and that of Tongil type varieties was about 0.1%.

  • Coronavirus disease, COVID-19 (coronavirus disease 2019), caused by SARS-CoV-2 (severe acute respiratory syndrome coronavirus 2), has a higher case fatality rate in European countries than in others, especially East Asian ones. One potential explanation for this regional difference is the diversity of the viral infection efficiency. Here, we analyzed the allele frequencies of a nonsynonymous variant rs12329760 (V197M) in the TMPRSS2 gene, a key enzyme essential for viral infection and found a significant association between the COVID-19 case fatality rate and the V197M allele frequencies, using over 200,000 present-day and ancient genomic samples. East Asian countries have higher V197M allele frequencies than other regions, including European countries which correlates to their lower case fatality rates. Structural and energy calculation analysis of the V197M amino acid change showed that it destabilizes the TMPRSS2 protein, possibly negatively affecting its ACE2 and viral spike protein processing.

  • Global warming has significant effects on the growth and development of wheat and can cause a reduction in grain yield and quality. Grain quality is a major factor determining the end-use quality of flour and a reduction in quality can result economic losses. Therefore, it is necessary to study the physiological characteristic of wheat to understand its response to temperature elevation, which can aid in the development of strategies to mitigate the negative effects of high temperature and sustain wheat production. This study investigated the effects of elevated temperature on grain characteristics and quality during the grain filling period of two Korean bread wheat cultivars Baekkang and Jokyoung. These two bread wheat cultivars were subjected to an increasing temperature conditions regime; T0 (control), T1 (T0+1℃), T2 (T0+2℃) and T3 (T0+3℃). The results showed that high temperature, particularly in T3 condition, caused a significant decrease in the number of grains per spike and grain yield compared to the T0 condition. The physical properties, such as grain weight and hardness, as well as chemical properties, such as starch, protein, gluten content and SDSS, which affect the quality of wheat, were changed by high temperature during the grain filling period. The grain weight and hardness increased, while the grain size not affected by high temperature. On the other hand, amylose content decreased, whereas protein, gluten content and SDSS increased in T3 condition. In this study, high temperature within 3℃ of the optimal growth temperature of wheat, quantity properties decreased while quality-related prosperities increased. To better understand the how this affects the grain's morphology and quality, further molecular and physiological studies are necessary.

Evaluation of Feed Value and Fermentation Quality of New Wheat Cultivar, 'Taejoong' (밀 신품종 '태중'의 사료가치 및 발효품질 평가)

  • This experiment was conducted to obtain information of feed value and fermentative quality of wheat cultivar, 'Taejoong' to confirm availability as a whole crop silage. As a result, the heading date of 'Teajoong' is April 27, and plant height is longer than whole crop barley, 'Youngyang' or whole crop wheat, 'Cheongwoo', spike length are also large, fresh and dry matter yield are also high. In case of feed value, 'Taejoong' had higher crude protein content than whole crop barley, 'Youngyang' or whole crop wheat, 'Cheongwoo', lower NDF and ADF contents, and significantly higher digestible nutrient contents(p<0.05). In case of fermentation qulity, pH of 'Taejoong' was 4.2, and lactic acid content was lower than 'Cheongwoo' silage and significantly higher than 'Youngyang' silage(p<0.05). Acetic acid content was significantly lower than 'Youngyang' and 'Cheongwoo' silage(p<0.01), butyric acid content was significantly lower than that of 'Youngyang' silage (p<0.05). The final Flieg's score showed that 'Taejoong' silage was the best. Also income of 'Taejoong' improved than 'Youngyang' or 'Cheongwoo'. Therefore, 'Taejoong' is considered sufficiently available as whole crop for forage.

  • A barley(Hordeum vulgare L.) cultivar 'Youjin' with hooded spike type having good silage quality was developed at National Institute of Crop Science, RDA in 2016. 'Youjin' showed both high yielding and cold resistance through the preliminary and advanced yield trials(PYT, AYT) from 2012 to 2013. We conducted regional yield trials(RYT) of 'Youjin' in six locations around Korea for three years from 2014 to 2016. It had erect plant type, growth habit of II, the green leaf and hooded awn type. In the paddy field its heading date was April 24 and the maturing date was May 25. Plant height was 99 cm and the number of spikes per ㎡ was 696. It has high rate of leaf blades, resistance to BaYMV(Barley Yellow Mosiac Virus) and good winter hardiness. The average dry matter of Youjin was about 17.2 MT ha-1 in the field. And feed quality of 'Youjin' was 10.6% of crude protein content, 24.8% of ADF(Acid Detergent Fiber), 43.5 % of NDF(Neutral Detergent Fiber), 69.1% of TDN(Total Digestible Nutrients). And also 'Youjin' had grade I of silage quality.

A New Six-rowed and Covered Barley Cultivar, "Hyedang" with Lodging Tolerance and High-Yield (내도복 다수성 겉보리 신품종 "혜당")

  • "Hyedang", a new covered cultivar derived from the crosses between "Suwon300" and "Haganemugi//CI08397/Haganemugi" developed at the Honam Agricultural Research Institute (HARI), NICS, RDA in 2007. The origin of "Hyedang" is "Iksan 407" (SB951033-B-B-B-72). The initial cross was made in 1995 and the selected line showed a high yield and good quality characteristics under yield trial test in 2004. "Iksan407" consistently performed well for three years (2005-2007) from the four locations of regional yield trial (RYT) in Korea and released as "Hyedang". The characteristics of "Hyedang" were the following: rate III growth habit, green leaf and stem, compact spike and with long rough awns. The heading date was April 25 in upland and April 17 in paddy field, which was 2 and 3 days earlier than that of check cultivar, "Olbori". The culm length was 80 cm which was 8 cm shorter than those of check cultivar. It showed spike length of 4.3 cm and 696 spikes per $m^2$, 51 grains per spike, 35.0 g of 1,000 grain weight and 704 g of test weight. It showed stronger winter hardiness and higher resistance to barley yellow mosaic virus (BaYMV) than those with check cultivar. It showed similar protein content and higher whiteness than those of the check cultivar and diastatic power was higher than that of Olbori. The average yield of the pearled grain in the RYT was $4.17ton\;ha^{-1}$ in upland and $4.27ton\;ha^{-1}$ in paddy field, which was 23% and 9% higher than that of the check cultivar, respectively. This cultivar would be suitable for the area above the daily minimum mean temperature of $-8^{\circ}C$ in January in Korean peninsula.

'Youho', A New Forage Barley Cultivar with Ruminant-Palatable Hood Spike Type and Non-Scatteredness (가축 기호성이 높은 내탈립 삼차망 청보리 신품종 '유호')

  • 'Youho' (Hordeum vulgare L.), a new ruminant-palatable forage barley cultivar, was developed by the breeding team at the Department of Rice and Winter Cereal Crop, National Institute of Crop Science, RDA in 2008. It was derived from the cross between 'Suwon339' and 'Suwon355'. Among the cross made in 1999, a promising line, SB992047-B-B-B-6-2, showed good characteristics in potential forage yield in the yield trial tested at Iksan from 2005 to 2006. In 2007, it was designated as 'Iksan431' and placed in regional yield trials at eight locations in Korea for two years from 2007 to 2008, and was released as the name of 'Youho'. It has the growth habit of group II, erect plant type, green leaf and hood spike. Its average heading and maturing dates were on Apr. 24 and May 26, respectively, which are similar to check cultivar 'Yuyeon'. 'Youho' also showed weaker winter hardiness, but better resistance to lodging, shattering and BaYMV than those of check cultivar. It showed higher crude protein content, grade of silage quality than those of check cultivar. The average forage dry matter yield in the regional yield trial was about 14.1, $10.9MT\;ha^{-1}$ in upland and paddy field, respectively, which were 1% to 4% lower than that of the check cultivar. This cultivar would be suitable for the area whose daily minimum temperature was above $-8^{\circ}C$ in January in Korean peninsula.

Development of Enzyme-Linked Immunosorbent Assay for Rapid and Sensitive Analysis of Biotin (Biotin의 분석을 위한 효소면역측정법(ELISA)의 개발)

  • In order to develop more rapid and reproducible analysis of biotin known as vitamin H, attempts were made to establish the condition for enzyme linked immunosorbent assay(ELISA) compared with traditional microbiological assay(MBA). Antibiotin and antiserum were obtained from the immunized rabbits injected with emulsion of biotin KLH conjugate and Freund's adjuvant. The antiserum showed cross reactivity on biocytin, a derivative of biotin, which is converted to biotin in intestine, at the rate of 177%(median inhibitory concentration(IC50)=12.58ppb), but not on other derivatives such as desthiobiotin, diaminobiotin and 2 imino biotin. Specific antibody for biotin was purified from the antiserum through protein A column and desalting column. The conditions of competitive direct ELISA (cdELISA) were established. Detection range of biotin concentration by cdELISA was 0.01∼300ng/ ml(ppb). In the spike test with milk, fruit flake and pine carrot juice, the correlation coefficient between two methods of MBA and ELISA was reliably consistent at the value of r=0.992. But detection of biotin by microbiological assay(MBA) was rather restricted in range and nonspecific. Detection range of biotin by MBA was 0.1∼0.5ng/ml(ppb). It showed cross reactivities on biocytin and desthiobiotin at the rate of 80.1% and 66.7%, respectively. In conclusion, ELISA revealed a significant improvement compared with MBA for the biotin detection in terms of sensitivity, detection range and cross reactivity. In addition, a variety of samples could be analyzed rapidly and conveniently at one time by using ELISA. These results strongly suggest that the ELISA is very promising for the practical application to detect biotin contents in a wide range of food stuffs.

Clinical and Electrophysiological Features of the Patients with POEMS Syndrome (POEMS 증후군의 임상적, 전기생리학적 특성)

  • Backgrounds and objectives: POEMS (polyneuropathy, organomegaly, endocrinopathy, M protein, and skin changes) syndrome is the rare cause of polyneuropathy. Although the polyneuropathy is essential for the diagnosis of the disease, the pattern of electrodiagnostic abnormalities has not been characterized in detail. The purpose of this study was to elucidate the features of nerve conduction abnormalities in POEMS syndrome. Methods: We reviewed the medical records and nerve conduction studies (NCS) of 12 consecutive patients with POEMS. Results: A total of 68 motor and 46 sensory nerves were examined. Compound muscle action potentials (CMAPs) and sensory nerve action potentials were abnormally attenuated or not elicited in majority of motor and sensory nerves (80.88% in motor, and 82.6% in sensory nerves). Frequency of the nerves with no potential was significantly higher in lower limbs than in upper limbs (p<0.01 in both motor and sensory nerves), and CMAP amplitude was more reduced in lower limbs than in upper limbs (p<0.01). Conduction slowing was very frequently observed with 95% and 76% of motor and sensory nerves, respectively, having the abnormally reduced values of conduction velocity. Distal motor latencies were abnormally prolonged in 75% of motor nerves, and terminal latency indices were significantly higher in patients than in normal controls (p < 0.05). Conduction block was observed only in 5% of motor nerves. Conclusions: NCS in POEMS syndrome showed characteristic patterns, in which conduction abnormalities were more frequently and severely affected in the lower limbs, and more predominantly in the intermediate nerve segments than in the distal portions. The recognition of these characteristic patterns may be helpful in early diagnosis of polyneuropathy in POEMS syndrome.
