Agglomeration of human dermal fibroblasts with ECM mimicking nano-fragments and their effects on proliferation and cell/ECM interactions

  • Here, we engineered spheroids by using ECM mimicking nano-fragments (NFs) with fibroblasts and investigated their effect on proliferation and cell/ECM interactions. NF incorporation resulted in formation of a stable spheroid, which improved proliferation and viability of cells by assisting oxygen transport confirmed by LOX-1 staining. In addition, hypoxic and apoptotic genes were significantly downregulated in spheroids with PD-NFs. Furthermore, ECM and cell junction proteins were highly expressed. Overall, our findings suggest that incorporation of NFs within spheroids for assembly with various cell types can be an alternative approach for 3D cell culture in many applications.

  • A simple emission model applicable for low scattering (scattering << absorption) anisotropic layer is developed and applied to the interpretation of measurements of microwave emission from row crops. The vegetation layer of row crops is modeled as a random slab embedded with small spheroid with major axis aligend paralel to the crop-row direction. The total emission is given in a simple algebraic form based on the zero-order radiative transfer theory. The single scattering albedo for spheroid and its polarimetric phase function are presented. The effects of layer azimuthal dependence on emission are accounted for by using an anisotropic albedo in the zero-order transfer theory. The developed emission theory favorably compares with the brightness temperature measured over soybeans canopy.

Two-Cell Spheroid Angiogenesis Assay System Using Both Endothelial Colony Forming Cells and Mesenchymal Stem Cells

  • Most angiogenesis assays are performed using endothelial cells. However, blood vessels are composed of two cell types: endothelial cells and pericytes. Thus, co-culture of two vascular cells should be employed to evaluate angiogenic properties. Here, we developed an in vitro 3-dimensional angiogenesis assay system using spheroids formed by two human vascular precursors: endothelial colony forming cells (ECFCs) and mesenchymal stem cells (MSCs). ECFCs, MSCs, or ECFCs+MSCs were cultured to form spheroids. Sprout formation from each spheroid was observed for 24 h by real-time cell recorder. Sprout number and length were higher in ECFC+MSC spheroids than ECFC-only spheroids. No sprouts were observed in MSC-only spheroids. Sprout formation by ECFC spheroids was increased by treatment with vascular endothelial growth factor (VEGF) or combination of VEGF and fibroblast growth factor-2 (FGF-2). Interestingly, there was no further increase in sprout formation by ECFC+MSC spheroids in response to VEGF or VEGF+FGF-2, suggesting that MSCs stimulate sprout formation by ECFCs. Immuno-fluorescent labeling technique revealed that MSCs surrounded ECFC-mediated sprout structures. We tested vatalanib, VEGF inhibitor, using ECFC and ECFC+MSC spheroids. Vatalanib significantly inhibited sprout formation in both spheroids. Of note, the $IC_{50}$ of vatalanib in ECFC+MSC spheroids at 24 h was $4.0{\pm}0.40{\mu}M$, which are more correlated with the data of previous animal studies when compared with ECFC spheroids ($0.2{\pm}0.03{\mu}M$). These results suggest that ECFC+MSC spheroids generate physiologically relevant sprout structures composed of two types of vascular cells, and will be an effective pre-clinical in vitro assay model to evaluate pro- or anti-angiogenic property.

방사선 모사 미생물 유래 리보솜 스트레스에 의한 대장암 스페로이드 구조 결함 유발 (Structural Disorganization of Intestinal Tumor Spheroid by Microbial Ribotoxins)

  • 기존에는 방사선 요법은 효과에 비해 소화기 점막 궤양 등 부작용이 많다. 본 연구는 방사선의 작용을 모사하는 방사선 모사 미생물 유래 리보솜 스트레스 반응을 이용하여 방사선 치료의 한계를 대체할 기전적 방법을 모색하고자 한다. 암 치료의 평가를 위해서 대장암의 비균질적인 세포구성, 종양줄기세포 및 주위 미세환경 및 배양 기질 등의 상호작용을 고려할 수 있는 소화기암 스페로이드를 이용하였다. 대조군 대장암 스페로이드에 비해서 리보솜 스트레스하에서는 스페로이드 구조가 상대적으로 큰 군집을 형성하였다. 하지만 이는 구조 자체가 매우 섬긴 세포간 구성을 가진 스페로이드였으며 스페로이드 형성과정의 크기 수축은 대조군에 비해 매우 느리게 나타났다. 대조군은 강하게 뭉친 소규모 클러스트의 집합체가 최종적으로 스페로이드를 형성하여 물리적 손상을 받아도 단단한 소규모 클러스트는 유지되나 리보솜 스트레스 하에서는 물리적 손상에 의해 형태가 파손 후에는 스페로이드 형성이 거의 일어나지 않았다. 기전적으로 리보솜 스트레스 하에서 암세포의 군집하는 이동속도는 매우 느렸으며, 뭉치더라도 세포-세포간 접합부위가 상대적으로 적었다. 이 대장암 스페로이드를 이종 및 동종 이식을 통해 동물에서 증식 시 종양 조직 형성이 매우 억제되었으며, 형성이 되어도 세포-세포간 접합에 핵심단백질인 E-cadherin의 발현이 매우 감소 됨을 알 수 있었다. 결론적으로 방사선 모사 미생물 유래 리보솜 스트레스는 종양 스페로이드 세포 이동 및 접합을 저해하여 3차원 구조 형성 결함을 유발하였으며, 향후 방사선을 대체하여 약물적으로 방사선 항암효과를 구현하는 기반을 제공한다.

Effects od Segree of Cell-Cell Contact on Liver Specific Function of Rat Primary Hepatocytes

  • Cell-Cell interaction and the extracellular matrix (ECM) are belisved to play essential roles during in vitro culturing of primary hepatocytes in the control of differentiation and in the maintenance of tissue spcific functions. The objective of this study was to examine the effects of degree of cell-cell contact (DCC) on liver sperific function of rat promary hepatocytes. Hepatocyte aggregates with various with various degrees of cell-cell contantact, I. e., dispersed cell, longish aggregate, rugged aggregate, and smooth spheroid were obtained at 1, 5-6, 15-20, and 36-48 hrs, respectively in suspension cultures grown in spinner flasks embedded in Caalginate bead and collagen gel in order. The may result from mass transfer limitation and shear damage caused by agitation during aggregation. The rugged aggregate showed a higer viability and albumin secretion rate than the dispersed cells or the other aggregates. This result indicates the possible enhancement of a bioartificial liver's (BAL) performance using primary hepatocytes and the reduction in time to prepare a BAL through optimization of the immobilization time.

Self-renewal and circulating capacities of metastatic hepatocarcinoma cells required for collaboration between TM4SF5 and CD44

  • Tumor metastasis involves circulating and tumor-initiating capacities of metastatic cancer cells. Hepatic TM4SF5 promotes EMT for malignant growth and migration. Hepatocellular carcinoma (HCC) biomarkers remain unexplored for metastatic potential throughout metastasis. Here, novel TM4SF5/CD44 interaction-mediated self-renewal and circulating tumor cell (CTC) capacities were mechanistically explored. TM4SF5-dependent sphere growth was correlated with $CD133^+$, $CD24^-$, ALDH activity, and a physical association between CD44 and TM4SF5. The TM4SF5/CD44 interaction activated c-Src/STAT3/ Twist1/ B mi1 signaling for spheroid formation, while disturbing the interaction, expression, or activity of any component in this signaling pathway inhibited spheroid formation. In serial xenografts of less than 5,000 cells/injection, TM4SF5-positive tumors exhibited locally-increased CD44 expression, suggesting tumor cell differentiation. TM4SF5-positive cells were identified circulating in blood 4 to 6 weeks after orthotopic liver-injection. Anti-TM4SF reagents blocked their metastasis to distal intestinal organs. Altogether, our results provide evidence that TM4SF5 promotes self-renewal and CTC properties supported by $CD133^+/TM4SF5^+/CD44^+^{(TM4SF5-bound)}/ALDH^+/CD24^-$ markers during HCC metastasis.

LOCATIONS OF OUT-OF-PLANE EQUILIBRIUM POINTS IN THE ELLIPTIC RESTRICTED THREE-BODY PROBLEM UNDER RADIATION AND OBLATENESS EFFECTS

  • This study deals with the generalization of the Elliptic Restricted Three-Body Problem (ER3BP) by considering the effects of radiation and oblate spheroid primaries. This may illustrate a gas giant exoplanet orbiting its host star with eccentric orbit. In the three dimensional case, this generalization may possess two additional equilibrium points ($L_{6,7}$, out-of-plane). We determine the existence of $L_{6,7}$ in ER3BP under the effects of radiation (bigger primary) and oblateness (small primary). We analytically derive the locations of $L_{6,7}$ and assume initial approximations of (${\mu}-1$, ${\pm}\sqrt{3A_2}$), where ${\mu}$ and $A_2$ are the mass parameter and oblateness factor, respectively. The fixed locations are then determined. Our results show that the locations of $L_{6,7}$ are periodic and affected by $A_2$ and the radiation factor ($q_1$).

회전타원체 보빈 형상의 거동에 관한 연구 (A Study on the Behavior of Spheroid Configuration Bobbin)

  • The initial trajectory of a spheroid configuration bobbin for precision guidance has been investigated by analyzing its aerodynamic load and six-degree-of-freedom motion. The effects of changes in the spheroidal head configuration, flow angle and lateral center-of-gravity offset are numerically studied using the commercial software "FLUENT". A wind tunnel test is also conducted to validate the numerical scheme and to examine effect of the Reynolds number on the flow around the bobbin. It is shown that the size of the separation bubble formed on the surface decreases significantly when the Reynolds number is varied between 110,000 and 140,000. At a zero flow angle, an oblate spheroidal head shows relatively moderate rotation while a prolate spheroidal head shows rapid rotation. The bobbin with a spherical head shape has little effect on the flow direction; however, the oblate bobbin is sensitive to the flow angle. The roll motion of the bobbin is greatly influenced by the lateral center-of-gravity offset and maximum dispersion is observed at half of the radius.
