Enhancement of Spermidine Content and Antioxidant Capacity by Modulating Ginseng Spermidine synthase in Response to Abiotic and Biotic Stresses

  • Polyamines (putrescine, spermidine and spermine) play pivotal roles in plant defense to different abiotic and biotic stresses. In order to understand the function of ginseng spermidine synthase gene, a key gene involved in biosynthesis of polyamines, transgenic plant was generated in Arabidopsis. The transgenic plants exhibited high levels of polyamines compared to the untransformed control plants. We investigated the tolerance capacity of transgenic plants to abiotic stresses such as salinity and copper stress. In addition, transgenic plants also showed increased resistance against one of the important fungal pathogens of ginseng, the wilt causing Fusarium oxysporum and one of important bacteria, bacterial blight causing Pseudomonas syringae. However, an activity of the polyamine catabolic enzyme, diamine oxidase (DAO) was increased significantly in F. oxysporum and P. syringae infected transgenic plant. Polyamine catabolic enzymes which may trigger the hypersensitive response (HR) by producing hydrogen peroxide ($H_2O_2$) seem act as an inducer of PR proteins, peroxidase and phenyl ammonium lyase activity. The transgenic plants also contained higher antioxidant enzyme activities, less MDA and $H_2O_2$ under salt and copper stress than the wild type, implying it suffered from less injury. These results strongly suggest an important role of spermidine as a signaling regulator in stress signaling pathways, leading to build-up of stress tolerance mechanisms.

애기장대 잎 절편 배양시 NAA 농도에 따른 부정근, 모용 및 캘러스 형성에 미치는 Polyamine의 영향 (Effect of Polyamines on Formation of Adventitious Roots, Trichomes and Calli by NAA in Leaf Segment Cultures of Arabidopsis thaliana)

  • 애기장대 (Arabidopsis thaliana) 생태종 'Columbia'의 잎 절편 배양시 부정근, 모용 및 캘러스 형성에 미치는 polyamine의 역할을 알아보고자 putrescine, spermidine, spermine, cyclohexylamine (CHA) 및 methylglyoxal-bis(guanylhydrazone) (MGBG)의 영향을 조사하였다 애기장대 잎 절편을 부정근 (0.1 mg/L NAA), 모용 (2.0 mg/L NAA) 및 캘러스 (10.0mg/L NAA)를 형성하는 MS배지 각각에 putrescine, spermidine 및 spermine을 1∼100 mg/L 처리하였다. Polyamine 처리시 부정근 형성구에서는 부정근과 함께 모용이 형성되었으며, 모용 형성구에서도 모용과 함께 캘러스가 형성되었다 또한 CHA와 MGBG를 1∼100mg/L 범위에서 처리한 결과 CHA는 부정근 형성구에서 다소 부정근이 증가시켰으나 모용이나 캘러스 형성구에는 영향을 미치지 않았다. MGBG는 부정근 모용 및 캘러스 모두를 억제하였고 모용 형성구에서는 고농도에서 부정근을 형성시켰다.

  • The effect of some phytohormones and light on the growth, ADC activity and polyamine content in ginseng was studies. In seedlings, the growth, ADC activity and putrescine content were in creased by Ghs treatment. ADC activity ann putrescine content were slightly slightly derreased by ABA, but not changed by kinetin. Light treatment increased ADC activity and putrescine content greatly. In two year ginseng leaves treated by GA3, the ADC actin$.$its reached maximum and the spermidine content reached maximum 2 days faster than in the control. Thtse results suggest the possibility that these regulators are closely related to growth and polyamine cotent. UeVo'ordsEPanaxgineng C.A. Chlamydomonas reinhardtii로부터 분리, 정제된 DNA Methyltransferase 활성에 대한 Polyamine의 영향 (Effect of Polyamines on purified DNA Methyltransferase from Chlamydomonas reinhardtii)

  • DNA methyltransferase was purified 282.6-fold from Chlamydomonas reinhardtii 21gr (mt+) gametic cell to examine the effect of polyamine on the enzyme acctivity. Polyacrylamide gel electrophoresis(PAGE) revealed at least three bands(1 major band, 2 minor bands). Among these, the major band represents DNA methyltransferase. Polyacrylamide gel electrophoresis in the presence of 0.1% sodium dodecylsulfate(SDS-PAGE) revealed a major band with M.W. 60,000. DNA methyltransferase activity was inhibited more effectively by spermine than by spermidine, and the inhibition by putrescine was smaller than spermine and spermidine. DNA methyltransferase activity was inhibited by 40% and 53% at 5mM and 20mM spermine, respectively. In the case of spermidine, the inhibition was 35% at 10mM and 44% at 20mM. However, the inhibition by putrescine appeared only above 5mM and reached about 25% at 20mM.

  • Spermidine, spermine and IAA promoted an increase in $\beta$-1, 4-endoglucanase activity in hypocotyls of Glycine max. The optimal concentration for the increase of the enzyme activity was 10-6 M for spermidine, 10-8 M for spermine and 10-6 M for IAA. However, IAA had innocuous effect on arginine decarboxylase and ornithine decarboxylase activites, and the content of polyamine. Such cumulative results suggest that the increase in $\beta$-1, 4-endoglucanase activity by IAA is not attributed by the effect on the biosynthesis of polyamine by IAA but spermidine, spermine and IAA induce cell wall loosening and therefore extension growth of cells.

  • Polyamine titers and the activities of arginine decarboxylase(ADC) and ornithine decarboxylase (ODC), enzymes which catalyze rate-limiting steps in polyamine biosynthesis, were investigated during polar regeneration of Populus leaf segments. The polar regeneration occurred at the basal cut end of Populus leaf segments through cell division around the vascular bundle. In the process of polar regeneration, the titers of putrescine and spermidine increased rapidly but the content of spermine remained constant. The leaf segments were then divided into three separte part ; the proximal, middle and distal. Spermidine titers showed an increase mainly in the proximal parts where polar regeneration occurred. On the other hand, putrescine titers showed an increase in the other two parts. In the course of polar regeneration, the activities of ADC and ODC increased, the ADC activities being higher than those of ODC. However, ODC activity was higher in the proximal part. Therefore, the spermidine contents and ODC activities are suggested to be related to polar regeneration in Populus leaf segments.

  • The effects of polyamines on the activities of elongation factors EF-1 and EF-2, phenylalanyl-tRNA synthetase, and tRNA were investigated. The activities of EF-1 and EF-2 were mostly stimulated by spermidine among three kinds of polyamines. The activities of EF-1 and EF-2 were investigated in the presence of spermidine by 230 and 181%, respectively. The activity of phenylalanyl-tRNA synthetase was slightly increased in the presence of polyamines. The effect of spermine on the synthetase was higher than that of the other polyamines. The tRNA activity in the presence fo polyamines was increased by 206% with spermidine, by 144% with spermine, and by 114% with putrescine. According to these results, it is concluded that polyamines in higher plants stimulate the protein biosynthesis by promoting the activities of elongation factors EF-1 and EF-2, aminoacyl-tRNA synthetases, and tRNAs, but the effects of polyamines on the various components for protein biosynthesis are different in according to the kind of polyamines.

당근(Daucus carota L.)배양세포의 DNase활성에 미치는 Polyamines의 영향 (The Effect of Polyamines on the DNase Activity in Cultured Carrot Cells)

  • The present study was attempted to investigate the effects of polyamines such as putrescine, spermidine and spermine on protein content and DNase activity in vivo and in vitro in carrot embryos. It was also investigated whether polyamines could replace role of cations required for DNase activity in vitro. The results obtained are as follows. Putrescine, spermidine and spermine increased protein content, although response to spermine reached plateau at the concentration of 0.1 mM. DNase activity was inhibited by polyamines, the inhibition being concentration-dependent and the highest att he concentration of 10 mM. The inhibition of DNase activity was the most prominent with spermine. Similar inhibitory effect to polyamines which was concentration-dependent was found in DNase activity but no change was shown on time-course in vitro. Putrescine and spermidine enhanced the DNase activity at low Mg2+ and Mn2+ concentrations, suggesting that the role of Mg2+ and Mn2+ for DNase activity could be, in part, replaced by these polyamines. These results, therefore, suggest that plyamines can modulate DNase activity through binding to DNA rather than direct effect on DNase activity.

포플라 잎절편의 부정아 분화와 Polyamine 함량의 연관성 (Correlations Between Polyamine and Adventitious Shoot Regeneration from Populus Leaf Segments)

  • Polyamine contents and the activities of their main biosynthetic enzymes, arginine decarboxylase (ADC) and ornithine decarboxylase(ODC), were investigated in Populus leaf segments during adventitious shoot regeneration with the addition of 1 millimolar polyamine synthesis inhibitors. From the study of polyamine synthesis inhibitors, ODC was found to be the principal route of polyamine biosynthesis in adventitious shoot regeneration of Populus leaf segments. The ADC inhibitor difluoromethyl arginine(DFMA) and ODC inhibitor difluoromethyl ornithine(DFMO) strongly reduced the putrescine content. On the contarary, DCHA, an inhibitor of spermidine synthase, increased it. Spermidine content was decreased with the treatment of each polyamine synthesis inhibitor, but the inhibitory effect of DCHA was stronger than any other polyamine inhibitor. The decreased polyamine level by polyamine synthesis inhibitors was restored with the exogenously applied polyamine. Comparing the polyamine contents with the adventitious shoot regeneration rate, were observed a close correlation between spermidine content and adventitious shoot regeneration of Populus leaf segments.

  • A rapid and sensitive high performance liquid chromatographic method with UV detection for the analysis of polyamines(putrescine, spermidine, spermine) has been developed. The benzoyl chloride derivatives of putrescine, spermidine and spermine are separated on a mc-Bondapak C18 reverse-phase column with 50% methanol as the mobile phase. This method was applied to the analysis of polyamines in the serum of normal human and uterine cancer patients. The results show that the mean level of polyamines in cancer patients serum is much higher than that in normal human serum.
