Objective : This study was conducted to investigate the effects of Panax Ginseng (인삼) on the sperm motility and spermatogenesis in the male rat. Methods : We used 8-week-old Sprague-Dawley rats, and administered the extract powder of Panax Ginseng to 5 rats (treated group) and normal saline (control group) once a day for 28 days. We isolated their testes surgically, then observed the change of the body weights before and after administration of Panax Ginseng extracts and normal saline. We observed the weight of the testes, epididymis, vascular gland, and prostate. Also, we examined the total, normal motile sperm concentration, and the concentration of testicular catalase and peroxidase. Results : We found that the concentration of normal, motile sperm in the testes of the Panax Ginseng group showed a significant difference compared with the control group. The angiogenesis of the seminiferous tubule was increased and the increasement of the number of spermatogonia, primary and secondary spermatocyte was observed in the Panax Ginseng group through a microscope. The body weight, the weight of the testes, epididymis, prostate and the concentration of testicular catalase and peroxidase were higher in the Panax Ginseng group but showed no significant difference. Conclusion : This study shows that Panax Ginseng may have an effect on the morphology and motility of sperm, the important factor in male fertility, and can promote the concentration of antioxidants, catalase and peroxidase, which is the important factor in spermatogenesis.

Methods : We used 8-week-old Spraque-Dawley rats and administered the extract powder of Cynomorii Herba to 5 rats and normal saline to control group once in a day for 28 days. Then we observed the changes of body weight before and after administration of Cynomorii Herba extracts and normal saline. We isolated their testis surgically and observed the weight of testis, epididymis, vascular gland and prostate. Also we examined the total, normal and motile sperm concentration, the concentration of testicular catalase, peroxidase and configuration of testicular tissue before and after administration of Cynomorii Herba extracts and normal saline. Results : We found that the concentration of total, normal, motile sperm in testis of Cynomorii Herba treated group shows significant difference compared with the control group. The body weight, the weight of testis, epididymis and the concentration of testicular catalase and peroxidase were higher in the Cynomorii Herba treated group, on the other hand the weight of vascular gland was lower in the Cynomorii Herba. But the contents of above statement showed no significant difference. The weight of prostate showed lower in the control group statistically than that of the Cynomorii Herba treated group. We observed the seminiferous tubules taken shape minutely and the number of normal sperm increased in Cynomorii Herba treated group's testicular tissue. Conclusion This study shows that Cynomorii Herba may have an effect on the concentration, morphology and motility of sperm, the important factor in male fertility.
2,3,7,8-Tetrachlorodibenzo-p-Dioxin (TCDD), a ubiquitous environmental contaminant, causes a variety of adverse effects on the male reproductive system in rats. The effect of green tea extract (GTE) was investigated on the testicular function in Spragure-Dawley rats after a single exposure of 10$\mu\textrm{g}$ TCDD/kg body weight. The exposure of rat to TCDD significantly increased the weights of the epididymis and ventral prostate, yet significantly decresed the weight of the seminal vesicle when compared to the controls (p<0.05). In a combined treatment of TCDD with GTE, the organ weight changes caused by TCDD treatment disappeared. Significant decreases in sperm motility and sperm numbers were observed in the TCDD-treated rats, when compared to the control (p<0.05). GTE treatment reversed the decrease of sperm motility and sperm numbers caused by TCDD. There were no differences in sperm morphology, histological changes of the reproductive organs, and spermatogenesis between all the treated groups. In the ventral prostate and seminal vesicle, TCDD increased the CYP1A1 mRNA level, however, it did not affect the estrogen receptor $\beta$ (ER-$\beta$) mRNA level. GTE treatment did not influence the effect of TCDD on the levels of CYP1A1 and Er-$\beta$ mRNA. These results seem to indicate that green tea protects the testicular function against TCDD-induced reproductive toxicity, not because of a receptor-mediated mechanism but rather due to a secondary change of testes or accessory sex organs.
With an increase in the number of people suffering from ageing-related diseases in our rapidly aging society, interests in natural products such as maca(Lepidium meyenii), which has properties of enhancing cognition and sexual performance, have increased. This study was conducted to assess the effects of 7 weeks of maca extract supplementation(0.5~2.0 g/kg BW) on scopolamine-induced amnesia in mice and on sperm count in male mice. All doses of maca supplementation significantly protected against scopolamine-induced amnesia as determined by a Morris water maze, but not according to passive avoidance tests. Maca supplementation did not affect acetylcholinesterase activity in the whole brain, nor the testicular sperm count of male mice. This study suggests that maca may have some neuroprotective properties in mice, which will be further examined by future studies.
Several herbs including Artemisia are known to possess conceptive property. In the present study, mouse spermatozoa were incubated with ethanol extract of Artemisia vulgaris leaves. The effect of extract on acrosome exocytosis was studied by labeling spermatozoa with fluorescein isothiocyanate (FITC) peanut agglutinin and by staining with Coomassie blue. Viability and membrane integrity were studied by Trypan-blue staining and hypo-osmotic swelling test. Artemisia extract at very low concentration caused precocious acrosome reaction and loss of sperm viability. Acrosome reaction increased remarkably from 22.63% to 88.42% with increasing extract concentration from 0 to 2,000 ㎍/mL. However, the viability loss of spermatozoa was increased from 11.71% in control to 63.73% in samples treated, evaluated by Trypan-blue staining method. Membrane damage caused by the extract, evaluated by hypo-osmotic swelling test was even low, ranging from 2.27% to only 24.23%. These results indicate that Artemisia extract might block fertilization by causing precocious acrosome exocytosis in spermatozoa. A direct contraceptive effect was tested by injecting the plant extract into the vagina of female mice and then allowing them to mate with normal males. The treated female mice delivered significantly fewer litters in comparison to the control.

Objectives: This study was conducted to investigate the effects of Morindae officinalis Radix (巴戟) on the spermatogenesis and antioxidant activities in the Sprague Dawley (SD) rat. Materials and Methods: We choose the 2-month-old SD rats, and administered the extract powder of Morindae officinalis Radix once in a day for 28 days. The control rats were administered normal water in the same way and duration. We observed changes of body weight, surgically isolated testis, epididymis, vascular gland and prostate gland before and after administration of Morindae officinalis Radix extracts in SD Rats. Also we compared the testicular tissue, especially seminiferous tubules between the control and treated groups by histochemical methods. In addition, we examined the total, normal, morphologic and motile sperm in the cauda epididymis, and the activity of catalase and peroxidase in the isolated testis tissue. Results: There was no significant difference between control and treatment groups in the body weight, testis, vascular and prostate gland, but the weight of epididymis showed significant difference in the control group. The concentration of total sperm, the motility and normality of spermatozoa was significantly different when compared with the control group, respectively. In the histological examination of testicular tissues, the tendency of increasement of angiogenesis between seminiferous tubules was observed. And the concentration of spermatogonia, primary and secondary spermatocyte and sperm were higher than that of control testicular tissues. Finally, the activity of catalase and peroxidase related inhibitory molecules of oxidation were slightly increased in the treatment group than those of control group. Conclusions: This study shows that Morindae officinalis Radix has the beneficial effect on the concentration, morphology and motility of sperm, the important factor in male fertility. We can suggest that Morindae officinalis Radix has an effect on the spermatogenesis in the SD rat.
The antifertility activity of methanol extract of Cuscuta reflexa Roxb. stem (MECR) and Corchorus olitorius Linn. seed (MECO) were studied on male Swiss albino mice. The extracts were found to decrease sperm count, percentage of motile sperm and testosterone level in treated mice when compared with vehicle control after 17 days of treatment. The weight of gonads, epididymis were decreased whereas no significant changes of the body weight of mice were observed after methanol extract treatments. The fertility test showed 100% negative result in MECR and MECO treated mice at medium and high dose level of treatment. MECR and MECO in low (25 mg/kg and 15 mg/kg, respectively), medium (50 mg/kg and 20 mg/kg, respectively) and high (75 mg/kg and 25 mg/kg, respectively) dose level caused a simultaneous fall in testicular ${\Delta}5$-$3{\beta}$-hydroxy steroid dehydrogenase and glucose-6-phosphate dehydrogenase activities which are involved in testicular steroidogenesis. Total cholesterol and ascorbic acid content in testis were increased significantly in gonads. The activities of lactate dehydrogenase, malic dehydrogenase and ascorbic acid oxidase were reduced whereas that of carbonic anhydrase was increased significantly in the testis of MECR and MECO treated mice. All these observations indicate that the methanol extract of C. reflexa stem and C. olitorius seed produced antifertility activity in sexually matured male mice, which may be due to inhibition of gonadal steroidogenesis. This activity may be attributed due to the presence of flavonoids and steroids, respectively.
Ketoconazole (KET) is an antifungal drug with a broad spectrum of activity that also induces reproductive toxicity in humans and animals. KET inhibits C17-20 lyase which blocks the conversion of 17 ${\alpha}$-hydroxyprogesterone to androstenedione. The effect of Cuscutae semen(CS) extract against KET-induced testicular damage was evaluated in male rats. CS extract was administered orally (100 and 200 mg/kg) for 26 days. Three weeks after CS extract administration, KET was CS-administered intraperitoneally at a dose of 100 mg/kg once a day for 5 days. KET-induced reproductive toxicity was associated with clear reductions of the weights of testes and epididymides, sperm indices and serum testosterone levels. In addition, marked oxidative damage to testicular lipids and alterations of natural antioxidant enzymes were reported in association with KET toxicity. Most of the KET-induced effects were greatly decreased with the concomitant application of CS extract. This study suggests a protective role of Cuscutae semen extract that could be attributed to its antioxidant properties.
Brassica oleracea var. italica (broccoli) is a type of cabbage that contains vitamins, minerals, and phytochemicals. Consequently, it is used as a potential nutraceutical source for improving human health by reducing oxidative stress and inflammatory responses. Here, the effects of broccoli sprout extract (BSE) on the inflammatory response were investigated through lipopolysaccharide (LPS)-induced inflammatory mouse models. First, we found that the BSE obviously reduce NO production in RAW 264.7 cells in response to LPS stimulation in in vitro study. Pretreatment with BSE administration improved sperm motility and testicular cell survivability in LPS-induced endotoxemic mice. Additionally, BSE treatment decreased the levels of the pro-inflammatory cytokines TNF-a, IL-1β, and IL-6, and COX-2 in testis of LPS-induced endotoxemic mice models. In conclusion, BSE could be a potential nutraceutical for preventing the excessive immune related infertility.

Objective: The aim of this study was to ascertain the effects of adding green tea extract (GTE) to skim milk-egg yolk (SM-EY) extender on both the quality of post-thawed bull semen and the pregnancy rates of the recipient cows. Methods: Twelve ejaculates from four Simmental bulls, aged 3 to 5 years and weighing 900 to 950 kg, were diluted SM-EY extender, added with 0, 0.05, 0.1, and 0.15 mg GTE/100 mL extender and then frozen. After four weeks storage in liquid nitrogen, the sperm were thawed and evaluated for viability, motility, intact plasma membrane (IPM), and DNA fragmentation. Meanwhile, the estrus cycles of 48 recipient cows were synchronized by intramuscular administration of a single injection of 5 mg prostaglandin F2α. Estrus cows were divided into four equal groups and inseminated artificially 18 to 20 h after the onset of estrus by using semen from each extender group. Pregnancy was diagnosed by measuring serum progesterone levels at 21 days, followed by transrectal palpation 90 days after insemination. Results: The findings revealed that adding 0.1 mg of GTE/100 mL extender produced the highest percentages of sperm viability (70.67%±1.75%), motility (69.17%±1.47%), and IPM (69.23%±1.21%) and the lowest percentage of DNA fragmentation (3.00%±0.50%). The pregnancy diagnosis revealed that all cows (36/36) inseminated using frozen semen in GTE addition extender were pregnant (pregnancy rate 100%), whereas the pregnancy rate of the control group was 83.33% (10/12). Conclusion: It may be concluded that 0.1 mg GTE/100 mL extender yields the best quality of spermatozoa and that all variants doses of GTE in extender produce a higher pregnancy rate among recipient cows.
