UPTAKE OF α-AMINOISOBUTYRIC ACID (AlB) BY ROOSTER SPERMATOZOA

  • This experiment was designed to determine whether ${\alpha}$-aminoisobutyric acid (AIB) can be used to predict membrane function of spermatozoa by measuring the uptake of AIB by fresh, stored and frozen-thawed rooster spermatozoa. When spermatozoa were stored at low temperature ($0{\sim}3^{\circ}C$) for 24 h. no difference was found in AIB uptake compared with fresh spermatozoa, whereas storage for 48 h resulted in a slight increase in AIB uptake by spermatozoa. On the one hand, the uptake of AIB by frozen-thawed spermatozoa was less than that by fresh spermatozoa. This suggests possibility of a different membrane transport system between spermatozoa preserved at low temperature ($0{\sim}3^{\circ}C$) and those frozen-thawed. Glycerol used as cryoprotectant may modify rooster sperm membrane in a different manner from cold preservation. Ouabaine ($10^{-4}M$) caused a slight decrease in AIB uptake, but caffeine ($10^{-2}M$) did not influence spermatozoal AIB uptake. These results indicate a successful application of AIB to rooster spermatozoa as a mean for measuring sperm membrane function and suggest a possible alteration of membrane transport system in rooster spermatozoa between cold ($0{\sim}3^{\circ}C$) and cryopreservation ($-196^{\circ}C$).

Dose dependent effect of benzene extract of Ocimum sanctum leaves on cauda epididymal spermatozoa of albino rats

  • An attempt has been made to assess whether the dose dependent effect of benzene extract of Ocimum sanctum leaves on the morphological changes in the cauda epididymal spermatozoa and sperm parameters in male albino rats. Scanning Electron Microscope observations illustrate the disturbance in plasma membrane as well as acrosomal membrane. Most of the sperms appear morphologically abnormal in the mid region of the tail; there is formation of balloon like cytoplasmic droplet. Sperm parametric study exhibits decrease in the total sperm count, sperm motility, forward velocity and increase in the percentage of abnormal sperms in dose dependent manner on treatment benzene extract of Ocimum sanctum leaves. The results suggest that the effects may have resulted from a general disturbance in the proteins and alteration in cauda epididymal milieu probably due to androgen deficiency consequent upon antiandrogenic property of Ocimum sanctum leaves.

  • Fatty acids such as n-3 and n-6 polyunsaturated fatty acids (PUFA) are critical nutrients, used to improve male reproductive performance through modification of fatty acid profile and maintenance of sperm membrane integrity, especially under cold shock or cryopreservation condition. Also, PUFA provide the precursors for prostaglandin synthesis and can modulate the expression patterns of many key enzymes involved in both prostaglandin and steroid metabolism. Many studies carried out on diets supplemented with PUFA have demonstrated their capability to sustain sperm motility, viability and fertility during chilling and freezing as well as improving testis development and spermatogenesis in a variety of livestock species. In addition to the type and quantity of dietary fatty acids, ways of addition of PUFA to diet or semen extender is very crucial as it has different effects on semen quality in male ruminants. Limitation of PUFA added to ruminant ration is due to biohydrogenation by rumen microorganisms, which causes conversion of unsaturated fatty acids to saturated fatty acids, leading to loss of PUFA quantity. Thus, many strategies for protecting PUFA from biohydrogenation in rumen have been developed over the years. This paper reviews four aspects of PUFA in light of previous research including rumen metabolism, biological roles, influence on reproduction, and strategies to use in male ruminants.

Detailed morphological analysis of axolotl sperm

  • The axolotl has extraordinary regeneration capacity compared to other vertebrates. This remarkable potential has been attributed to its life-long neoteny, characterized by the exhibition of embryonic characteristics at the adult stage. A recent study provided a detailed morphological analysis of the sperm morphology of the Ambystoma mexicanum using routine and detailed histological techniques. The primary purpose of the present study is to describe a simple and inexpensive method for evaluating the morphology of axolotl sperm. In this study, spermatophore structures were collected and spread on slides and air-dried. The slides were stained with periodic acid Schiff, toluidine blue, Masson's trichrome, Giemsa, Spermac, and Diff-Quik dye for a morphological examination. The slides were coated with gold/palladium for a scanning electron microscopy examination. The sperm of the axolotl consisted of an elongated head, a neck, and a flagellum covered with an undulating membrane. The lengths of the midpiece, tail, and head were 8.575 ㎛, 356.544 ㎛, and 103.661 ㎛, respectively. In the flagellum part, the wavy membrane structure, whose function has not been explained, surrounds the tail. The data obtained from this study will constitute an important step in designing future research on the reproductive and regeneration capacity of the axolotl.

Catalase 첨가에 따른 돼지 정액 동결 및 융해 후 생존 정자에서 Hydrogen Peroxide의 감소 (The Reduction of Hydrogen Peroxide in Viable Boar Sperm Cryopreserved in the Presence of Catalase)

  • 요 약: 정액 동결 과정은 활성산소종의 생성을 유발하며, 생성된 활성산소종은 정자의 손상을 일으키는 것으로 알려져 있다. 따라서 본 연구의 목적은 동결 과정 중 항산화 효소 중 하나인 catalase (CAT)를 첨가함으로써 융해 후 정자의 기능과 활성산소종의 수준에 미치는 효과를 알아보고자 하였다. 5마리 돼지에서 채취한 정액은 0 (대조군), 200, 400 U/mL CAT가 첨가되어 있는 동결 희석액으로 각각 동결하였다. 융해 후, 정자 운동성, 생존성, 정상 형태율, 형질막 온전성, 미토콘드리아 기능, 세포내 ROS를 평가하였다. CAT는 400 U/mL의 농도에서 전체 정자 운동성을 향상시켰지만 (P < 0.05), 전진 운동성, 생존성, 기형율, 형질막 온전성, 미토콘드리아 기능의 향상을 나타내지 않았다. 활성산소종의 평가에서, CAT는 융해된 생존 정자의 ${\cdot}O_2$의 감소에는 효과를 나타내지 않은 반면 $H_2O_2$를 감소시켰다(P < 0.05). 결론으로 CAT는 동결 및 융해된 정자의 질을 향상시키는 데 큰 효과를 나타내진 않았지만 생존 정자에서 $H_2O_2$을 제거함로써 생존정자의 산화적 손상을 감소시킬 수 있으리라 판단된다.

Molecular cloning and characterization of Izumo1 gene from bovine testis

  • A well-characterized sperm specific protein of the Member of immunoglobulin superfamily, IZUMO1, has crucial role in fertilization by mediating sperm binding to the egg plasma membrane in the mouse. However little is known about IZUMO1 in bovine. Here, we describe the molecular cloning and expression analysis of bovine IZUMO1 (bIZUMO1). RT-PCR and Western blot analysis of the bovine tissues indicated that bIZUMO1 was specifically expressed in the testis and sperm, Furthermore, the result of our biotinylation assay from ejaculated bovine sperm strongly suggest the assumption that bIZUMO1 is localized on the cell surface. These data imply the potential role of bovine IZUMO1 in mammalian fertilization.

돼지 정자의 체외조본에 있어 Pyruvate, Taurine 및 Melatonin의 항산화효과와 평가방법의 상관관계 분석 (Antioxidative Effect of Pyruvate, Taurine and Melatonin, and Relationships of the Evaluation Methods on Boar Sperm Viability for In Vitro Storage)

  • 본 연구는 돼지 정액의 체외보존에 있어 pyruvate, taurine 및 melatonin의 항산화 효과와 생존력의 평가방법간의 상관관계를 검토하고자 실시하였다. 돼지 정액의 체외보존시 항산화 효과를 평가하기 위하여 체외보존 신선정액에 pyruvate(1mM), taurine(50 mM) 및 melatonin(100nM)에 H2O2(100uM)을 혼합첨가 및 항산화제 단독처리하여 체외에서 3~12시간까지 배양하면서 3시간 간격으로 정자의 운동성, 정자의 원형질막의 완전성 및 지질 산화도를 측정하였다. 정자의 운동성은 항산화제 첨가구가 다른 처리구에 비해 높은 결과를 나타내었고, Hypoosmotic swelling test(HOST) 방법을 이용한 정자의 원형질막 완전성에서도 운동성과 유사한 결과를 나타내었다. 정자의 원형질막의 지질산화도 검사에서는 항산화제 첨가구와 무첨가구가 H2O2 첨가구와 H2O2와 항산화제의 혼합 첨가구에 비해 낮은 결과를 나타내었다. 또한 평가방법들 간의 상관관계에서는 운동성과 정자의 원형질막의 완전성에는 정의 상관관계를 나타내었고, 정액의 성상검사 방법과 지질산화검사 방법에서는 부의 상관관계를 나타내었다. 본 실험의 결과는 정액희석액에 항산화제의 첨가는 정자의 운동성 및 정자의 원형질막 완전성을 증가시키고, 정자의 원형질막의 지질산화도를 감소시키는 것으로 나타났다. 또한 정자의 원형질막의 완전성 검사 (HOST)는 정자의 원형질막의 지질산화도 검사 및 운동성 검사를 대신하여 정자의 질적 평가에 유용하게 이용될 수 있을 것으로 사료된다.

Effects of Taurine on Sperm Characteristics during In vitro Storage of Boar Semen

  • The objective of this study was to investigate the anti-oxidative effects of taurine on sperm characteristics for in vitro storage of boar semen. Semen was randomly divided into 10 groups in conical tubes and treated with different concentrations of taurine (25-100 mM) with or without $250{\mu}M$ $H_2O_2$. The percentage of motile spermatozoa in taurine groups after 6 and 9 h were significantly higher at >94% and 87%, respectively, compared to the control group ($85.1{\pm}0.5$ and $72.4{\pm}0.3$, p<0.05). The sperm motility in taurine with $H_2O_2$ after 6 h incubation was slightly decreased compared to the taurine alone treatment, but after 9 and 12 h incubation % sperm motility dropped sharply in taurine with $H_2O_2$ ($75.3{\pm}0.3$ and $69.6{\pm}2.9$, p<0.05). For 3, 9 and 12 h incubation, sperm viability in the control was lower than in taurine groups, irrespective of taurine concentration. In eosin Y and nigrosin staining (ENS), the sperm survival rates (%) for 6 h incubation were significantly higher in 25 mM ($76.0{\pm}0.6$) and 50 mM taurine groups ($78.0{\pm}0.7$), respectively. Sperm survival rates for 9 and 12 h incubation were higher in taurine groups (${\geq}48%$ in 9 h and ${\geq}42%$ in 12 h) compared to controls ($43.0{\pm}2.1$ and $31.0{\pm}0.6$, respectively). In the hyoosmotic swelling test (HOST), sperm membrane integrity was similar to the results of sperm survival. These experiments indicate that supplementation of taurine to the semen extender can increase the sperm characteristics(motility, viability, survival and membrane integrity).

융해 속도가 동결.융해된 개 정자의 기능에 미치는 영향 (Effect of Thawing Rate on the Function of Cryopreserved Canine Sperm)

  • 정액 동결 방법들은 지속적으로 향상되어 왔지만 융해 속도에 대한 연구는 여전히 정립되어 있지 않다. 따라서, 본 연구의 목적은 동결 후 융해 속도가 정액의 기능에 미치는 영향을 알아보고자 하였다. 비글견으로부터 채취된 정액은 동결 보존 후 다른 융해 속도 ($37^{\circ}C$/1분 or $70^{\circ}C$/15초)에서 융해 되었다. 융해 후, 운동성, 생존성, 정상 형태율, 형질막 온전성, phosphatidylserine (PS) translocation, 세포내 $H_2O_2$ 수준을 평가하였다. $70^{\circ}C$에서 융해된 정자는 $37^{\circ}C$에서 융해된 정자에 비해 향상된 정자 운동성, 생존성, 정상 형태율, 세포막 온전성, non-PS translocation을 보였으나(P < 0.05), $70^{\circ}C$$37^{\circ}C$에서 융해된 실험군간 세포내 $H_2O_2$ 수준에서 유의적인 차이는 나타나지 않았다(P > 0.05). 결론으로, $70^{\circ}C$에서의 융해는 개 정자 동결 후 정자 기능을 증진시켰으며, 적절한 융해 온도는 동결 정자의 기능을 향상시킬 수 있을 것으로 판단된다.

자기활성수가 소 정액 동결에 미치는 영향 (Effect of Magnetized Water on Cryopreservation in Bovine Spermatozoa)

  • The purpose of this study was to improve of frozen-thawed sperm using magnetized water in Korean native cattle. Before cryopreservation, without egg-yolk Triladyl$^{(R)}$ solution was flowed though magnet [0, 2000, 4000 and 6000 Gauss(G)] for S min. The freezing of dilluted semen added with Triladyl containing 20% egg-yolk. Analysis of frozen-thawed sperm was estimated viability with SYBR14/PI double stain, membrane intact with hypoosmotic swelling test (HOST), acrosome reaction with FITC-PNA, mitochondria membrane function with Rhodamin 123 by flow- cytometry. Sperm viability was significantly higher in 4000G group than other groups (p<0.05). However, the Hypoosmotic Swelling Test(HOST) was significantly higher in fresh, 4000 and 6000G than 0 and 2000G (p<0.05). In addition, mitochondria membrane damage and acrosome damage were significantly lower in 6000G group than other groups (p<0.05). in conclusion we suggest that magnetized water could be improve ability of sperm on cryopreservation in Korean native cattle.
