Analysis of Flat board-shaped Lacquer ware excavated from Daho-ri was conducted. The result of species identification is identifed as Pinus spp. As a result of microscopic observation, lacquer fragment is consist of 5 layers and its thickness is about 100 ㎛. Infrared spectroscopy of the lacquer ware revealed that they had a very similar absorption band as refined lacquer, confirming that they were painted with lacquer. Result of Analysis for Circular metal ornament is alloy smelted by tin and lead. It is called Pewter. While The species of wooden objects investigated until now is broad-leaved tree; the species of flat board-shaped lacquer ware is conifer.
The impact of Korea's joining the Convention on the Conservation of Migratory Species of Wild Animals(CMS) was analyzed to examine its merits and faults as well as to discuss Korea's opportunities. Results of the analysis based on the agenda and decisions of the Conferences of the Parties, the parties performance, and other committees meetings over the last decades indicated that the affiliation of Korea into the CMS could provide various advantages and opportunities for Korea. First, Korea could upgrade its conservation activities regarding migratory species to the global aspects. Second, Korea could take initiatives for conservation of the migratory species in the Far East Asia. Third, Korea would have a better system in forecasting and problem-solving against the epidermic Avian Influenza through systematic cooperation with the CMS parties and other related international regimes. Finally, Korea will be in a better position to generate statistical data and to develop techniques to reduce the by-catches of the sharks and whales. Korea has already provided a fair and protective institutions for most of the migratory endangered species listed under Appendix I and II of the CMS. This implies that Korea may not require additional major changes to the basic acts and/or legislation. Joining the CMS may negatively impact on the fisheries and related businesses related to whales and sharks around the Ulsan and Pohang provinces. However, the obligation to protect whales and sharks demanded by the CMS is regarded as an acceptable article in Korea according to the analysis of the existing policies and scientific aspects. Nevertheless, if the joining the CMS should generate irreversible hardship for local people's livelihood and cultural aspects, Korea may ask for reservations on particular activities. Overall, we suggest that by joining the CMS, Korea could see various advantages and promotion in national policy.
Ecological approach to conservation of Korea's National Parks (KNP) was reviewed. Four hundred twenty five reports on 15 national parks for 80 years (1915~1994) were analyzed in consideration of phytocoenotic information such as flora, plant community and landscape. Total species was recorded as 157 familles, 752 genera, and 2,369 species (64.3% of Korean flora), in which is included only 75 species of the protected plant species designated by the Ministry of EnvironMent. Occurrence patterns of plant species in national parks were quite similar to one another, but those of plant community were rather unique. Seventy-four of 97 plant communities were reported in only one of 15 national parks, which might be regarded as an endemic or local vegetetation type. These facts are far different from actual status of the KNP's ecosystem, which indicate that not only original data in previous reports are less informative, but also tools of investigation and description are too subjective, and thus these can never afford to monitor the ecosystem. From the correlation analysis between 10 ecological characteristics, the following results were obtained: (1) the species richness was positively associated with the area of national park, (2) the number of visitors was related to landscape diversity, (3) occurrence of the designated species was closely related to the vegetation diversity. Numerical analysis (cluster analysis and ordination) using dissimilarity ratio by ecological characteristics divided 15 national parks into 4 groups. The first group, composed of national parks of Chirisan, $S\v{o}laksan$, Sokrisan, Hallasan, $T\v{o}kyusan$, Odaesan, and Sobaeksan, is recognized as the best national parks in terms of ecological conservation values. Nevertheless, they have been faced with the threat of visitor stress. This study will contribute to the establishment of strategy for appropriate conservation and sustainable use of KNP.

The aim of this study was to provide basic guidelines for conservation and management of endangered plants in the national parks of Korea. Iris dichotoma Pall. (Iridaceae), which is a popular garden plant, is considered a second-class endangered species by Korean government and it is listed as a EN (Endangered) species in Red Data Book of Korea. We analyzed ecological conditions of I. dichotoma habitats based on vegetation properties and soil characteristics. This species which is known to inhabit in grassland adjacent to the ocean of lowlands slope and its population was located at an elevation of 8 m to 11 m. In the study sites, the mean of soil organic matter, total nitrogen and soil pH were 6.16%, 0.234% and 5.39 respectively. Additionally, the genetic variation and structure of three populations were assessed using ISSR (Inter Simple Sequence Repeat) markers. The genetic diversity of I. dichotoma (P = 59.46%, H = 0.206, S = 0.310) at the species level was relatively high. Analysis of molecular variance (AMOVA) showed 82.1% of the total genetic diversity was occurred in within populations and 17.9% variation among populations. Lastly, we developed predicted distribution model based on climate and topographic factors by applying SDMs (Species Distribution Models). Consequently, current status of I. dichotoma habitats is limited with natural factors such as the increase of the coverage rate of the herbs due to ecological succession. Therefore, it is essential to establish in situ and ex situ conservation strategies for protecting natural habitats and to require exploring potential and alternative habitats for reintroduction.
Twenty-eight of 44 Indian Ceropegia species are endemic and their survival is threatened. As a step towards conservation, we implied in vitro methods for the sustainable propagule production in C. hirsuta, C. lawii, C. maccannii, C. oculata and C. sahyadrica. Effects of explant, growth regulators, sucrose and photoperiod were studied. High frequency microtuber production was achieved with the seedling-apical buds, grown on MS medium containing 4-6 mg $1^{-1}$ BAP, 3-8% (w/v) sucrose, under continuous illumination. Each microtuber, when subcultured proliferated to form a cluster of secondary microtubers. Every primary and secondary microtuber bore at least one shoot-bud and a root primordium. Each tuber (formed with any of the significantly effective treatments) weighed more than 500 mg, enough to plant directly in non-sterilized soils. Microtubers could be produced and proliferated round the year. Proliferation could be solely attributed to in vitro procedures as these plants bear solitary tubers in vivo. Microtubers could be sprouted in vitro to prepare ready to pot plantlets. As, this novel method succeeded for all five species, though they belong to different eco-physiological backgrounds, we recommend its implementation in the conservation programs for a broader range of Ceropegia species, supported by other integrated strategies.

The study assessed the utility of mitochondrial DNA for identifying a deceased rorqual discovered off the western coast of India. Both the COI and Cytb genes exhibited remarkable 99-100% similarity with the GenBank sequence of Balaenoptera musculus through a global BLAST search, confirming their affiliation with this species. Inter-species genetic distances for COI and Cytb genes ranged from 6.75% to 9.80% and 7.37% to 10.96% respectively, compared with other Balaenopteridae species. The Bayesian phylogenies constructed based on both COI and Cytb genes demonstrated clear and separate clustering for all Balaenopteridae species, further reaffirming their distinctiveness, while concurrently revealing a cohesive clustering pattern of the generated sequences within the B. musculus clade. Beyond species confirmation, this study provides valuable insights into the presence of live and deceased B. musculus individuals within Indian marine ecosystems. This information holds significant potential for guiding conservation efforts aimed at safeguarding Important Marine Mammal Areas (IMMAs) in India over the long term.
This study was conducted to develop the new Environmental Impact Assessment techniques for the grading of rareness in plant. 17 EISs which were submitted in 1996 were analyzed to know the problems in existent EIA techniques for the conservation of rare plants. Category of rare plant was reevaluated and evaluation technique for the grading of rareness in plant was developed. The results obtained from this study were as follows: 1. The evaluation technique of rareness in plant was not reflected in the current preparation provision for EIS. And t his fact also appeared in most of the EIS which were reexamined in this study. 2. The category of rare species which have been considered as the subject of protection in EIA have to be enlarged to more than legally protected species designated by MOE. 3. The taxonomic characteristics and status of species, characteristics of population, geographical characters, extent of threat and its possibility by man resulted from the habitat loss were investigated. Each item was endowed scores from 1 to 5 and all the scores were summed, and then this value was used to evaluate the order of conservation. 4. Conservation measures for the rare plants have to be applied according to the levels of rareness grade, and they are absolute conservation, in situ conservation, transplanting and ex situ conservation, post observation and none by the levels from A to E. 5. Considering the phenology of rare plants, investigation for the rare plants have to be made a1 least three times. 6. Size of the MVP(Minimum Viable Population), pollination ecology also have to be considered for the conservation of rare plants. And in the case of adopting ex situ conservation measure, the site and methods for the conservation of rare plants have to be proposed in detail.

The Gold-spotted pond frog, Rana plancyi chosenica, designated as a vulnerable species by IUCN Red list. This species is a typical example facing local population threats and extinction due to human activities in South Korea. A strategic conservation plan for this endangered species is urgently needed. In order to provide information for future conservation planning, accurate information on the genetic diversity and taxonomic status is needed for the establishment of conservation units for this species. In this study, we used a molecular genetic approach using the mitochondrial cytochrome b gene and control region sequences to find the genetic diversity of gold-spotted pond frogs within South Korea. We sequenced the mitochondrial DNA cytochrome b gene and control region of 77 individuals from 11 populations in South Korea, and one from Chongqing, China. A total of 15 cytochrome b gene haplotypes and 34 control region haplotypes were identified from Korean gold-spotted pond frogs. Mean sequence diversity among Korean gold-spotted pond frogs was 0.31% (0.0-0.8%) and 0.51% (0.0-1.0%), respectively. Most Korean populations had at least one unique haplotype for each locus. The Taean, Ansan and Cheongwon populations had no haplotypes shared with other populations. There was a sequence divergence between Korean and Chinese gold-spotted pond frogs (1.3% for cyt b; 2.9% for control region). Analysis of genetic distances and phylogenetic trees based on both cytochrome b and control region sequences indicate that the Korean gold-spotted pond frog are genetically differentiated from those in China.
The genus Termitomyces (Lyophyllaceae, Basidiomycota) is often associated with fungus-feeding termites (Macrotermitinae) due to their strong symbiotic relationships. The genus is widely found exclusively in certain regions of Africa and Asia. They are recognized as edible mushroom within Southeast Asia as well. But it is often misidentified based on morphology by the local communities especially in Malaysia for Chlorophyllum molybdites which is a highly poisonous mushroom. Thus, it is necessary to study the genus for Malaysia with the synergy of using both morphological and molecular identification. In this study, we aim to describe another new species as an addition to the genus Termitomyces found within Sabah, Malaysia. We generated two new sequences (nrLSU and mtSSU) for the new species and a total of 28 nrLSU and mtSSU sequences were retrieved from GenBank for the phylogenetic analysis using maximum likelihood and Bayesian inferences. We identified that the new collection from Sabah province is a new species and named as Termitomyces gilvus based on the termites found in the mound. A phylogeny tree made from the concatenated genes of LSU and mtSSU suggests that T. gilvus is closely related to T. bulborhizus from China. According to our results, the combination of molecular and morphology proved to be a robust approach to re-evaluate the taxonomic status of Termitomyces species in Malaysia. Additional surveys are needed to verify the species diversity and clarify their geographic distribution.

Korean long-tailed goral (Nemorhaedus caudatus) is one of the most endangered species in South Korea. However, detailed species distribution and sex ratio data on the elusive goral are still lacking due to difficulty of identification of the species and sex in the field. The primary aim of this study was to develop an economical PCR-RFLP method to identify species using invasive or non-invasive samples from five Korean ungulates: goral (N. caudatus), roe deer (Capreolus pygargus), feral goat (Capra hircus), water deer (Hydropotes inermis) and musk deer (Moschus moschiferus). The secondary aim was to find more efficient molecular sexing techniques that may be applied to invasive or non-invasive samples of ungulate species. We successfully utilized PCR-RFLP of partial mitochondrial cytochrome b gene (376 bp) for species identification, and sex-specific amplification of ZFX/Y and AMELX/Y genes for sexing. Three species (goral, goat and water deer) showed distinctive band patterns by using three restriction enzymes (Xbal, Stul or Sspl). Three different sexing primer sets (LGL331/335 for ZFX/Y gene; SE47/48 or SE47/53 for AMELX/Y gene) produced sex-specific band patterns in goral, goat and roe deer. Our results suggest that the molecular analyses of non-invasive samples might provide us with potential tools for the further genetic and ecological study of Korean goral and related species.
