A Study on Variation of Trans Fatty Acid with Heat Treatment of Corn Oils (가열조리에 의한 옥수수유의 트랜스지방산 변화에 관한 연구)

  • Trans fatty acid components separated and quantified using a SP-2560 capillary column in a gas chromatograph (GC) with flame ionization detector (FID). Trans fatty acid and total fatty acid contents were measured in 21 corn oils. Ranges of values for trans fatty acid (tFAs) contents of total fat (as g/100g fatty acids) were com oils $0.65{\pm}0.31$. Corn oils were heated at $175{\pm}5^{\circ}C$ for 5mins $(0{\sim}15\;times)$. The contents of tFAs (g/100) were increased from 0.292 (0 time) to 2.585 (15 times) in com oil. When frying oils (15 times) were incubated at $20{\pm}5^{\circ}C$ for 150 days, the contents of tFAs (g/100g) were increased from 2.585 to 3.683 in com oil. The amounts of tFAs (g) per serving size of frying oils (15 times) were increased from 0.01 to 0.18 in corn oil. The levels of the 18:1 trans isomers increased significantly the time of reusing of com oil.

Characterizing LipR from Pseudomonas sp. R0-14 and Applying in Enrichment of Polyunsaturated Fatty Acids from Algal Oil

  • In this study, Pseudomonas R0-14, which was isolated from Arctic soil samples, showed a clear halo when grown on M9 medium agarose plates containing olive oil-rhodamine B as substrate, suggesting that it expressed putative lipase(s). A putative lipase gene, lipR, was cloned from R0-14 by genome walking and Touchdown PCR. lipR encodes a 562-amino-acid polypeptide showing a typical α/β hydrolase structure with a catalytic triad consisting of Ser153-Asp202-His260 and one α-helical lid (residues 103-113). A phylogenetic analysis revealed that LipR belongs to the lipase subfamily I.3. LipR was successfully expressed in Escherichia coli, purified, and biochemically characterized. Recombinant LipR exhibited its maximum activity towards p-nitrophenyl butyrate at pH 8.5 and 60℃ with a Km of 0.37 mM and a kcat of 6.42 s-1. It retained over 90% of its original activity after incubation at 50℃ for 12 h. In addition, LipR was activated by Ca2+, Mg2+, Ba2+, and Sr2+, while strongly inhibited by Cu2+, Zn2+, Mn2+, and ethylenediaminetetraacetic acid. Moreover, it showed a certain tolerance to organic solvents, including acetonitrile, isopropanol, acetone, methanol, and tert-butanol. When algal oil was hydrolyzed by LipR for 24 h, there was an enrichment of n-3 long-chain polyunsaturated fatty acids, including eicosapentaenoic acid (1.22%, 1.65-fold), docosapentaenoic acid (21.24%, 2.04-fold), and docosahexaenoic acid (36.98%, 1.33-fold), and even a certain amount of diacylglycerols was also produced. As a result, LipR has great prospect in industrial applications, especially in food and/or cosmetics applications.

Chemical Properties of Hot Water Extracts from Bamboos(Phyllostuhys sp.) (대나무 열수추출물의 화학적 특성)

  • Chemical composition of the water extract of three different kinds of bamboos was investigated to access nutritive sources. Bamboo stalks and leaves were extracted massively and concentrated to 3°Brix, and crude protein and fat contents of the extracts were respectively 0.56∼0.69% and 0.28∼0.58%. Main mineral contents of the extracts were K, P and Mg in order, and those were the highest in Wangdae stalks. In free sugar analysis of the extracts, Maengjongjuk stalks contained 1.1% sucrose, Somdae stalks contained 0.51% fructose and Wangdae stalks contained 0.6% glucose as the highest level. Organic acid contents of the extracts showed high level in acetic and malic acid which were 54.0 and 44.2mg% in extracts of Wangdae stalks respectively. Among amino acids, aspartic acid was contained the highest level in stalk extracts, and their concentrations were 42.2, 39.9 and 34.6mg% in Wangdae, Somdae and Maengjongjuk respectively. In leaf extracts, glutamic acid concentrations were higher than the other amino acids, and those were 19.3, 18.5 and 15.7mg% in Maengjongjuk, Wangdae and Somdae, respectively.

Trend of Acupuncture Treatment Study for Functional Dyspepsia (기능성 소화불량증의 침치료 연구 동향 : Pubmed를 중심으로)

  • Objectives: This study was conducted to help clinical studies for treating Functional Dyspepsia with acupuncture. We inspected tendency of studies of acupuncture treatment for functional dyspepsia and analyzed the assessing instrument for it by scrutinizing Pubmed recent publications. Materials and Methods: We inspected 8 theses and scrutinized their objectives, periods, participants, materials and methods, the instrument of assessment for the studies, and criteria and symptom score for questionaire, results and JADAD score. Results: 1. Most studies were published by oriental doctors except one which was published by an American doctor. More and more studies are being published in America and written in English recently. So it is considered that western doctors think that the acupuncture treatment is useful as unorthodox medicine for functional dyspepsia. 2. Among the 4 clinical studies, 2 studies were RCT and 1 study was not RCT and 1 study with vagueness. All 4 clinical studies were not double-blinded because they were about comparison between acupuncture treatment and medicine. 3. ST36, PC6, CV12, BL21 were chiefly used, and auricular point, ST40, SP6 were also used. Manual acupuncture were used in 2 studies, and low frequency pulse and electroacupuncture were used in other studies respectively. 4. 4 clinical studies used 0~3 points symptom score as the instrument of assessment. Additional assessments were on gastric emptying time, EGG and plasma level of motilin. All thesis were not verified of validity and reliability. 5. Among the 4 reviewed thesis, 2 theses were about acupuncture as one of the various treatment of functional dyspepsia, and the other 2 theses were about efficacy of acupuncture treatment. And the result generally showed effectivity of acupuncture treatment. Conclusion: Acupuncture treatment for functional dyspepsia is considered useful as in unorthodox medicine by western doctors, and it is necessary to provide objective instrument for assessing acupuncture treatment for functional dyspepsia.

Sensory Characteristics of Citrus Vinegar fermented by Gluconacetobacter hanenii CV1 (Gluconacetobacter hansenii CV1에 의해 발효된 감귤식초의 관능적 특성)

  • Citrus juice, a concentrate manufactured by the Jeju Provincial Corporation, was converted into vinegar orderly by alcohol and acetate fermentation. The juice with 6 folds dilution by distilled water was used as the sole nutrient source through out experiments. Diluted juice contained $12.96^{\circ}Brix$ of total sugar, $0.632\%$ of total acid and $20.23{\mu}g/m{\ell}$ of hesperidin. Naringin was not detected from the juice. Citrus wine having $5.6\~6.3\%$ alcohol was produced from diluted juice by 3 days of fermentation at $28^{\circ}C$. A kind of malomelo yeast CMY-28 was used for wine fermentation. The wine was succeedingly fermented for 8 days at $30^{\circ}C$ after inoculation of seed vinegar which contained active cells of acid producing bacteria CV1. Inoculum size of seed vinegar was controlled to $10\%$(v/v) of citrus wine. The wine converted into vinegar by the fermentation. Citrus vinegar, the final product of fermentation, was colored with very thin radish-yellow and transparent. It's acidity ranged between $5.8\~6.2\%$ as acetic acid. The vinegar got the best score by sensory test among several natural fruit vinegars. It was clear from the results that citrus vinegar in high quality could be produced from concentrated citrus juice, however fermentation conditions should be improved to reduce the amount of reducing alcohol.

Influence on Composting of Waste Mushroom Bed from Agaricus bisporus by using Mixed Organic Materials (혼용자재 특성이 양송이 폐상배지를 이용한 퇴비제조에 미치는 영향)

  • This study was conducted to select organic materials (OM) and nitrogen sources in composting of waste mushroom bed from Agaricus bisporus. We examined physio-chemical properties of the organic materials and the mixture ratio for preparing the wasted mushroom bed (M) compost. The carbon content of sawdust was higher than those of rice straw (R) as OM source and the nitrogen content was high in the order of fowl manure (F)>> pig manure (P)> cow manure (C). The compost was prepared to maintain the criteria of above 25% organic matter and then the change of their ingredients was estimated during the process of fermentation. The temperature of waste mushroom bed+pig manure+rice straw (MRP) treatment was varied fast throughout fermentation, on the other hand the temperature of waste mushroom bed+pig manure+sawdust (MSP) treatment was steadily elevated to the middle of composting. The pH of the compost was somewhat high to pH 8.5~9.0 at the early stage, but decreased to 7.5 at the end stage of composting. The content of OM after fermentation was decreased to the level of 19~21% in rice straw, but the sawdust treatment maintained 25~27% organic matter. The waste mushroom bed+fowl manure+rice straw (MRF) treatment, which contains 26.2% organic matter and 0.68% nitrogen, was the highest among them. The volume of compost was reduced to 50% by using rice straw as organic matter, but reduced to 30% by using the sawdust. The contents of heavy metal in the compost were suitable within the legal criteria. The number of microorganisms were higher in the rice straw than those in the sawdust. It was high in the order of fowl manure> pig manure> cow manure. The major groups consisted of aerobic bacteria, gram negative bacteria and Bacillus sp. and their populations after fermentation were increased to $1{\times}10^1{\sim}1{\times}10^2\;cfu\;g^{-1}$ rather than those before fermentation. Therefore we concluded that the waste mushroom bed+fowl manure+sawdust (MSF 3:9:1 v/v/v) treatment was suitable combination for high organic matter and nitrogen source, and the periods of composting were 50~60 days.

Expression of CDX2 and Villin in Gastric Cardiac Intestinal Metaplasia and the Relation with Gastric Cardiac Carcinogenesis

  • Objective: To determine whether CDX2 and villin protein expression are associated with intestinal metaplasia (IM) in gastric cardiac mucosa and to explore the relationship with evolution of gastric cardiac adenocarcinoma (GCA). Methods: We studied 143 gastric cardiac biopsy or resection specimens from Henan province China, including 25 cardiac gastritis specimens with IM, 65 dysplasia specimens with IM and 35 gastric cardiac adenocarcinoma specimens and stained them for CDX2 and villin by the immunohistochemical SP method. 15 normal gastric cardiac biopsy specimens were also collected as control. Results: (1) Normal gastric mucosa presented no CDX2 and villin expression. The positive rates of CDX2 protein in cardiac gastritis with IM, dysplasia with IM, and carcinoma tissues were 84.0% (21/25), 66.7% (32/48) and 36.4% (20/55), respectively. While the positive rates of villin protein in cardiac gastritis with IM, dysplasia with IM, and carcinoma tissues were 76.0% (19/25), 70.8% (34/48) and 45.5% (25/55), respectively. There were significant differences among the three groups for both CDX2 and villin (P<0.01). Spearman's rank correlation coefficient(rho) showed a close correlation between the two proteins (r=0.843, P<0.01) and both were positively related with tumor differentiation (both P<0.05), but not associated with age, sex, invasion and metastasis of lymph node (P>0.05). Conclusion: Our results suggest that ectopic expression of CDX2 and villin may be involved in early-stage IM and tumorigenesis in gastric cardia and the expression of villin may be regulated by CDX2.

  • Water samples collected from marine fish culture system in Korea were compared for their capability to reduce the infectivity titers of HRV (rhabdovirus olivaceus), FBV(flounder birnavirus) and RVS(retrovirus of salmonid). In addition, interaction between viruses and microorganisms present in the rearing seawater was examined. The titer of HRV and RVS were reduced at $15^{\circ}C$ to less than detectable limits within 3 to 5 days using untreated samples of seawater. No reduction of infectivity was noted in bacteria-free water treated by filtration or autoclaving. Bacteria (Pseudomonas and Vibrio sp.) isolated from the water collected from a flounder culture system showed the inactivation activity of HRV.

Rhizoctonia Blight of Azolla japonica Caused by Rhizoctonia solani (Rhizoctonia solani에 의한 큰물개구리밥(Azolla japonica) 마름병)

  • Azolla Lam. is a small aquatic fern with deeply bilobed leaves, which are consisted of a thick greenish, with chlorophyll, upper (dorsal) lobe and a thinner, translucent lower (ventral) lobe, without chlorophyll, submerged in the water. Azolla blight was observed at a lotus pond. Mycological characteristics of the fungus associated with Azolla blight was immediately determined as Rhizoctonia sp. by the thickness and branching of hypha at right angles at the point toward the distal end of septa, with branching hypha is constricted. The fungus produced brown mycelia and dark brown sclerotia on PDA. The optimum temperature for mycelial growth and sclerotia formation were $25^{\circ}C$ and $30^{\circ}C$, respectively. The optimum temperature for fungal infection was $30^{\circ}C$, when spray inoculated. Phylogenetic analysis of rDNA-ITS revealed that the fungus was identified as Rhizoctonia solani (AG-1 IA) closest to one causing rice sheath blight disease. This is the first report on the blight disease of Azolla caused by R. solani in Korea.

Isolation and Characterization of Endophytic Bacteria from Rice Root Cultivated in Korea (한반도 중부지방의 벼 뿌리로부터 내생 세균의 분리와 특성 분석)

  • The 44 endophytic bacterial strains were isolated from surface-sterilized root of rice cultivated in seven different locations of Chungcheong province, Korea. Each isolate was introduced into rice seedlings grown gnotobiotically by inoculating scissor-cut first true leaf with cell suspensions, and the colonization capacity of each isolate in root tissue was analyzed at 7 days after inoculation. Sixteen out of 44 isolates were re-isolated from root successfully with the frequency of $10^{3-5}$ CFU/g tissue. Interestingly, seven out of 16 isolates were identified as Burkholderia species. The identity between inoculated strains and re-isolates was confirmed by genomic finger-printing and 16S rDNA sequence analysis. By a confocal laser scanning microscopic observation it was revealed that KJ001 strain, one of the sixteen isolates tagged with gfp colonized in root tissue especially around xylem. Six out of seven Burkholderia strains obtained in this study showed antagonizing activities against seven different fungal pathogens, contain nifH gene, and five of them enhanced growth of cucumber over 30%. The isolates showed no hypersensitive response on tobacco leaves and no pathogenecity in rice. From these results it was found that the endophytic Burkholderia strains will be useful in agriculture to develop a biocontrol agent or a bio-fertilizer.
