The Infection Experiment of Pleistophora sp. to eel, Anguilla japonica and the Histopathological Investigation of the Infection Development (뱀장어 Pleistophora 증(症)의 감염실험(感染實驗)과 병리조직학적(病理組織學的) 연구(硏究))

  • Pleistophora disease is well known as microsporidiosis at eel cultural farms in Korea in recent years. Major objects of this study were to undertake the induction of Pleistophora infection experimentally and to carry out histopathological investigation from December 1984 to June 1986. Experimental infection of Pleistophora spores into young eels was carried out by oral and immersed administration. Both methods induced the same symptom successfully as that in naturally occurring diseased fish. Remarkable whitish lesions developed mainly on the body surface around the abdomen when orally administrated. On the other hand, they were scattered over the whole body when administrated through immersion. Histopathological investigation revealed that some cysts in the muscle were observed 21 days after administration. Spores were developed within the cyst. Each sporont has undergone several nuclear divisions to form a pair of multinucleate cells (Sporoblasts) enclosed within a common coat (Pansporoblast). All stages were surrounded by cyst. The cysts were destroyed and mature spores were scattered in the muscle.

Geological Distribution of Aquatic Invertebrates Living in Paddy Fields of South Korea (한국의 논에 서식하는 수서무척추생물의 지리적 군집 분포)

  • The roles of paddy fields are re-evaluated as an artificial wetland. Although 44% of total aquatic invertebrates in South Korea are observed in a rice paddy, information of their distribution and characteristics of habitat use is limited yet. This study was carried out to provide information and characterizing distribution of community of aquatic invertebrates using a rice paddy through the South Korea. Aquatic invertebrates were collected at 284 sites of a rice paddy in South Korea from June to August, 2005, 2006 and 2007. We grouped sampling sites according to its species and population of aquatic invertebrates using a non-metric multidimensional scaling (NMDS). Total 21 orders 60 families 114 species were collected from a rice paddy. Coleoptera (25%), Heteroptera (17%), Diptera (17%) and Odonata (12%) were observed. Aquatic invertebrates were classified into three groups (Group 1: Gangwon, Gyeonggi and Chungbuk; Group 2: Chungnam, Jeonbuk, Jeonnam and Jeju; Group 3: Gyeongbuk and Kyeongnam). In Group 1, Muljarus japonicas distributed mainly Gangwondo and Chungbuk. In Group 2, Sigara nigroventralis and S. substriata were mainly observed. In Group 3, higher density of Daphnia sp. and Chironomidae gen. spp. was found in the southern part of Korea.

Regulation of vascular smooth muscle phenotype by cross-regulation of krüppel-like factors

  • Regulation of vascular smooth muscle cell (VSMC) phenotype plays an essential role in many cardiovascular diseases. In the present study, we provide evidence that $kr{\ddot{u}}ppel$-like factor 8 (KLF8) is essential for tumor necrosis factor ${\alpha}$ ($TNF{\alpha}$)-induced phenotypic conversion of VSMC obtained from thoracic aorta from 4-week-old SD rats. Stimulation of the contractile phenotype of VSMCs with $TNF{\alpha}$ significantly reduced the VSMC marker gene expression and KLF8. The gene expression of KLF8 was blocked by $TNF{\alpha}$ stimulation in an ERK-dependent manner. The promoter region of KLF8 contained putative Sp1, KLF4, and $NF{\kappa}B$ binding sites. Myocardin significantly enhanced the promoter activity of KLF4 and KLF8. The ectopic expression of KLF4 strongly enhanced the promoter activity of KLF8. Moreover, silencing of Akt1 significantly attenuated the promoter activity of KLF8; conversely, the overexpression of Akt1 significantly enhanced the promoter activity of KLF8. The promoter activity of SMA, $SM22{\alpha}$, and KLF8 was significantly elevated in the contractile phenotype of VSMCs. The ectopic expression of KLF8 markedly enhanced the expression of SMA and $SM22{\alpha}$ concomitant with morphological changes. The overexpression of KLF8 stimulated the promoter activity of SMA. Stimulation of VSMCs with $TNF{\alpha}$ enhanced the expression of KLF5, and the promoter activity of KLF5 was markedly suppressed by KLF8 ectopic expression. Finally, the overexpression of KLF5 suppressed the promoter activity of SMA and $SM22{\alpha}$, thereby reduced the contractility in response to the stimulation of angiotensin II. These results suggest that cross-regulation of KLF family of transcription factors plays an essential role in the VSMC phenotype.

A Study of Sulfur Nutrition on the Flavour Components of Garlic (Allium sativum L.) (유황성분(硫黃成分)이 마늘의 향기성분(香氣成分)에 미치는 영향(影響))

  • The relationship between the sulfur fertilization, and the quality and yield of garlic was investigated. The garlic was grown in pot and field at the five fertilization levels of sulfur. The growth characteristics, yield and flavour components of the garlic were studied. The results were summarized as follows; 1. The sulfur applications contributed to a weight and yield of the bulb in SP-10 (10kg/10a) and SF-20 (20kg/10a) significantly, respectively. 2. From the volatile oil obtained by steam distillation of garlic, fourteen organie sulfur components were separated, identified and quantified by GC and GC/MASS and the SP-10 and SF-20 were higher in the concentration of the flavour components in the volatile oil than other treatments. 3. It is considered that the organic sulfur components were synthesized in the top and transported to the bulb of the garlic. 4. In conclusion, SF-20, 20kg/sulfur per 10a, could be recommended to get maximum yield and a good quality of garlic in view of evaluation through the contents of organic sulfur components in the volatile oil and the yield of the bulb.

Effects of Biodiversity and Pests Insects Occurrence on Surface Management Methods in Persimmon Orchards (지표면 관리방법이 감 해충 발생과 생물다양성에 미치는 영향)

  • This study was investigated to evaluate the effects of biodiversity and pest insect occurrence by soil surface management methods such as green manure crop, grass planting and clean-tilled crops on persimmon orchards. Green manure crop pests was investigated 7 species including Hypera postica, Laodephax striatellus, Megoura crassicauda, Aphis craccivora, Nephotettix cincticeps, Liriomyza sp., black spot and the most dangerous pest was H. postica but it did not damaged persimmon tree. Amount of pests and natural enemies dwelling soil covering plant was not difference between organic farming (hairy vetch planting) and conventional farming (natural weed). Homona magnanima occurred three times a year but it rarely damaged leaf of persimmon. Total adult moth catched by pheromone trap was 1,261 organic farm, 1,003 conventional farm and 621 clean-tilled farm. Grapholita molesta occurred four times a year but it also rarely damaged twig of persimmon. Number of collected invertebrate species and individuals on soil sample was follow as : 838 individuals of 22 species at organic farm, 421 individuals of 17 species at conventional farm and 381 individuals of 15 species at clean-tilled farm. The richness index was lined up by organic farm 3.12> conventional farm 2.65> clean-tilled farm 2.36.

A Study on the Transport Mode Choice for Asian Highway No.6 Route (아시안하이웨이 6번 노선의 국제여객 교통수단선택에 관한 연구)

  • By promoting the international transportation and the improvement of advanced exchange between South and North Korea, the Asian Highway plan was expected to make considerable economical effects in the North-East Asian region. This study focuses on a part of AH6 route (i.e., South Korea-North Korea-China-Far East Russia) and aims to analyze the behavioral change of international passenger transport around the Korean Peninsula by assuming the non-existent North Korea land transport network connectivity. The study was performed using two main methods. First, a stated preference survey was performed to derive the actual modal share of each travel mode. Second, a disaggregate analysis was performed to develop possible mode-choice models for international passenger travel and find out the suitable choice. Based on the results of the model estimation, it is found that over 90% of the international passenger transport would be converted to land transport (rail+highway). In addition, international railroad modal share rate would be increased from 62.8% to 66.1% and international bus would be decreased from 29.6% to 21.9% as the distance lengthened (903km~1,631km).

The Community Structure of Macrozoobenthos and Its Spatial Distribution in the Subtidal Region off the Namhaedo Island, South Coast of Korea (남해도 주변 조하대 해역의 대형저서동물 분포)

  • An ecological study on subtidal macrobenthic fauna was conducted off the Namhaedo Island, south coast of Korea from July 2012 to April 2013. A total of 247 species of macrobenthos occurred with a mean density of $1,027ind./m^2$ and a mean biomass of $148.7g/m^2$. Polychaetes was the richest and most abundant faunal group that comprised 37% in both total species number and density whereas echinoderms were biomass-dominant faunal group that accounted for 44% of the mean biomass. There was a seasonal variation in the species richness and abundance of macrobenthos with more species in winter and higher density in spring. Mean faunal density was relatively high at the stations near Namhaedo Island, but gradually decreased toward offshore stations. The most dominant species in terms of density was an amphipod species, Eriopisella sechellensis which occurred as a top ranker during three seasons except spring recorded the fourth rank. E. sechellensis showed its high density at the near shore stations of Namhaedo Island, but this species did not occur around the entrance of Gwangyang and Saryang Bays where Theora fragilis and Lumbrineris longifolia showed high densities. In particular, Tharyx sp., recorded high density between Gwangyang Bay entrance and offshore after Sea Prince oil spill, did not occur in the same area during this study period. The bottom temperature and sorting value of the surface sediments were highly correlated to the spatial distribution of macrobenthic fauna from the Bio-Env analysis. From the cluster analysis, the study area has five station groups with more similar faunal affinities from inner area toward offshore area. Based on the SIMPER analysis T. fragilis, Magelona japonica, E. sechellensis, L. longifolia and Paraprionospio cordifolia were mainly contributed to the classification of station groups. From the BPI, benthic communities in the entrance of Gwangyang Bay and Saryang Bay were considered to be in a slightly polluted condition in contrast to the normal healthy community at the offshore of Namhaedo Island. These results suggested that the benthic community of this area should be regularly monitored to assess the health status of this benthic ecosystem.

Macrobenthic Community at the Subtidal Area Around Taebudo in Kyeonggi Bay, Korea (경기만 대부도 주변 조하대 해역의 저서동물 군집)

  • Macrobenthic community structure was studied at thirteen stations in Taebudo subtidal area, Korea, from July to October 1996. Triple macrobenthos samples were collected using a van Veen grab (0.1 $m^2$) at each station during the study period. A total of 209 species of macrobenthos was sampled with a mean density of 1,093 ind./$m^2$ and biomass of 134,86 g/$m^2$. Of these, there were 72 species of polychaetes ($34.5\%$), 69 crustaceans ($33.0\%$) and 49 molluscs ($23.4\%$). Polyalaetes were represented as a density-dominant faunal group with a mean density of 608 ind./$m^2$, comprising $55.6\%$ of the total benthic animals. It was followed by crustaceans with 307 ind./$m^2$($28.1\%$ of the total density), Echinoderms were represented as a biomass-dominant faunal group with a mean biomass of 54.21 g/$m^2$($40.2\%$ of total biomass). Total number of species and diversity were low in the inner part of the study area with high mud content and high in the offshore stations of mixed sediments. Major dominant species were three polychaetes, Heteromastus filifomis, Scoloplos armiger and Tharyx sp. whose mean densities were 70 ind./$m^2$, 67 ind./$m^2$, and 66 ind./$m^2$, respectively. Cluster analysis showed that the study area could be divided into five stational groups based on the faunal composition, that is, the innermost stations, coastal stations, transitional stations and two offshore station groups. The species diversity of these groups increased from the inner stational group toward the outer groups.

Evaluation of A Revolving Plate-Type Biofilter for Use in Recirculating Eel Culture System and Control of Fish Diseases (파상회전원판을 사용한 뱀장어사육과 질병대책)

  • The main purpose of this study is to evaluate a revolving plate-type biofilter system for mass culture of eels (Anguilla japonica) based on the experimental rearing for 199 days. Water quality, growth efficiency of fish and effects of fish disease control were critically evaluated. The experiment was conducted in two different units, each unit consisting of a cement tank containing $20m^3$ of water. In unit A, a biofilter which includes 400 rotating undulated P. V. C. plates being 70 cm in diameter which rotates at 6 rpm and also 400 undulated P. V. C. plates fixed in the settling chamber of an area of $66{\times}62cm$. Water was continuously passed through the filter at a rate of 260 l/min., and supplemental water was added to the fish tank at a rate of $4m^3$ a day. In unit B, the biofilter has 400 P. V. C. plates being $66{\times}62cm$ each was installed in the settling tank. The results gained from the experimental rearing for 199 days from April 21, 1984 to November 5, 1984 are as follows. In the growth experiment, the weight of fish in unit A increased from 3.0 kg to 815.6 kg, while in unit B, from 3.0kg to 416.0kg. During the period of the experiment, in the both units the fish grew at an acceptable rate at the temperature at which they were held. Observing every aspect of eel culture, including growth rate, disease control and water quality, unit A appears to have adventages over unit B, which makes it particuraly attractive in intensive recirculating fish culture system. It was further observed that certain parasites such as Trichodina sp. and Costia sp. could easily be controled by appling 4 ppm of $KMNO_4$.

A Comparative Study on Limno-biological Aspects of the Dammed Lakes in the Youngsan River in Korea - Centering on Fish Fauna - (영산강 수계 댐호의 육수생물학적 비교연구 ( 1 ) - 어류상을 중심으로 -)

  • Some aspects of limno-biology of the five dammed lakes such as Tamyang, Changsong, Kwangju, Naju, and Youngsan Lake along the Yongsan River were investigated from June 1986 to July 1988 for the comparison on the lentic ecosystems in relation to the fish fauna. Sixty four fish species representing 48 genera and 21 families of fishes were collected in these dammed areas, which included 29 species of the family Cyprinidea and 40 speaies of the primary fresh-water fishes. Among, these, 12 species such as Rhodeus Acheilognothus yamatsutae, A canathorhodeus gracilis, Sarcocheilichthys nigripinis morii, S. variegatus wakiyae, Gnathopogon strigatus, Squalidus gracilis majimae, Microphysogobio yaluensis, Cobitis longicorpus sp, Liobagrus mediadiposalis and Odontobutis platycephala are enddmic to Korea. Leiocassis nitidus and Gasterosteus aculeatus which occur in these water areas take note of the zooge rographic study of Korea. The fish species in each of the dammed lakes is in proportion of the scale of the lakes such as 32 species in Thamyang, 40 species in Changsong, 24 species in Kwangju, 35 species in Jaju, and 52 species in Yongsan Lake. The dominant fish species of the 4 upper dammed lakes of Thamyang, Changsong, Kwangju and Naju Lake are Zacco platypus and Rhodeus uyekii, while those of Youngsan Lake are Hemiculter eigenmanni and Carassius auratus. This difference of fish species of those areas suggests that the upper damed lake differ from the lower dammed lakes in the habitable condition of fishes. The migratory fish species have significantly declined and will continue to decline in both population size and number of species in the dammed lakes in near future as a result of the dam construction barriers in the river estuary. The major migratory fishes of this water area are as follows; Anguilla japonica, Plecoglossus altivelis, Hypomesus olidus, Gasterosteus aculeatus, Konosirus Punctatus, Coilia ectens, Hemirhampus sajori, Mugil cephalus. Acanthogobius flavimanus, and Takifugu ocellatus.
