  • Seasonal variations of dominant algae species and the effects of these algae on coagulation and filteration of water treatment were investigated at Chilseo water treatment plant in downstream of Nakdong river from January in 1995 to Desember of 1998. The water quality of Nakdong river was found to be a hyper eutrophic state during the investigation periods. In the measurement, Chlorophyll-a contents ranged $20.7{\sim}180.9{\mu}g/l$ and total nitrogen contents(T-N) and total phosphorus contents(T-P) exceeded more than 3.4mg/l and 0.1mg/l, respectively. The changes in dominant algae species was in the order of Stepanodiscus sp., Asterionella sp., Melosira sp., Microcystis sp. and Synedra sp. from spring to winter. Microcystis sp. especially, was blooming during summer and Synedra sp. and Stepanodiscus sp. during winter. Although most diatomous algae appeared in the water treatment process caused filter clogging and reduced efficiency of coagulation and sedimentation, Synedra sp. and Stepanodiscus sp were revealed as the main trouble algae. Malfunction of water treatment process caused by Synedra sp. and Stepanodiscus sp. started at the algae concentrations of 800cells/ml and 1,820cells/ml, respectively. When chlorophyll-a content was $18.9{\mu}g/l$, the optimum amounts of coagulant were found to be 40mg/l of Alum and 16mg/l of PACS. Under condition of chlorophyll-a content of $154.1{\mu}g/l$, addition of Alum at the level of 75mg/l and PACS at the level of 35mg/l showed the lowest turibidity. The result indicates that increased amounts of the coagulants should be added for a better water treatment as chlorophyll-a contents increased. Addition of Alum at the amount of 60mg/l and 30mg/l of PACS removed Stepanodiscus sp. algae at the rate of 85% and 83%, respectively. In case of Synedra sp., 50mg/l of Alum and 25mg/l of PACS showed removal rates of 79% and 81%, respectively. Synedra sp. algae at the standing crops of 1,500cells/ml started filter clogging and a filtering process was completely inhibited after 8 hours. At this situation the filter clogging by Synedra sp. algae occurred at the depth of 5cm from the top anthracite layer. On the other, other algae did filter clogging at the depth of 10cm.

  • Purpose: Gastric cancer (GC) has high morbidity and mortality, the cure rate of surgical treatment and drug chemotherapy is not ideal. Therefore, development of new treatment strategies is necessary. We aimed to identify the mechanism underlying Sp1 regulation of GC progression. Methods and Methods: The levels of Sp1, β-catenin, SET domain bifurcated 1 (SETDB1), and 15-hydroxyprostaglandin dehydrogenase (HPGD) were detected by quantitative reverse transcription polymerase chain reaction and western blot analysis. The targets of SETDB1 were predicted by AnimalTFDB, and dual-luciferase reporter assay was used for confirming the combination of Sp1, β-catenin, and SETDB1. HGC27 or AGS cells (1×106 cells/mouse) were injected into mice via the caudal vein for GC model establishment. The level of Ki67 was detected using immunohistochemistry, and hematoxylin and eosin staining was performed for evaluating tumor metastasis in mice with GC. Results: HPGD was inhibited, while the protein levels of Sp1, β-catenin, and SETDB1 were up-regulated in GC tissues and cell lines. HPGD overexpression or SETDB1 silencing inhibited the proliferation, invasion, and migration of GC cells, and Sp1 regulated the proliferation, invasion, and migration of GC cells in a β-catenin-dependent manner. Furthermore, HPGD served as a target of SETDB1, and it was negatively regulated by SETDB1; additionally, Sp1 and β-catenin bound to the SETDB1 promoter and negatively regulated HPGD expression. We proved that Sp1 regulated GC progression via the SETDB1/HPGD axis. Conclusions: Our findings revealed that Sp1 transcriptionally inhibited HPGD via SETDB1 in a β-catenin-dependent manner and promoted the proliferation and metastasis of GC cells.

  • The beneficial effects of Eisenia fetida on soil properties have been attributed to their interaction with soil microorganisms. The bacterial diversity of the intestinal tract of E. fetida was investigated by culture-dependent and culture-independent methods including denaturing gradient gel electrophoresis (DGGE) and pyrosequencing analyses. In a pure culture, Lysinibacillus fusiformis (51%), Bacillus cereus (30%), Enterobacter aerogenes (21%), and L. sphaericus (15%) were identified as the dominant microorganisms. In the DGGE analyses, B. cereus (15.1%), Enterobacter sp. (13.6%), an uncultured bacterium (13.1%), and B. stearothermophilus (7.8%) were identified as the dominant microorganisms. In the pyrosequencing analyses, Microbacterium soli (26%), B. cereus (10%), M. esteraromaticum (6%), and Frigoribacterium sp. (6%) were identified as the dominant microorganisms. The other strains identified were Aeromonas sp., Pseudomonas sp., Borrelia sp., Cellulosimicrobium sp., Klebsiella sp., and Leifsonia sp. The results illustrate that culture independent methods are better able to detect unculturable microorganisms and a wider range of species, as opposed to isolation by culture dependent methods.

  • In order to elucidate chatracteristics of water quality and seasonal dynamics of phytoplankton community in Kyungpo lake, the weekly variation of algal population and 11 environmental factors were investigated from March 1998 to February 1999. A total 121 species of phytoplankton belonged to 57 genera, 4 genera 7 species of the Euglenophyceae, 1 genera 2 species of the Chrysophyceae, 2 genera 3 species of the Cryptophyceae, 1 genera 1 species of the Dinophyceae, 6 genera 9 species of the Cyanophyceae, 28 genera 78 species of the Bacillario-phyceae and 15 genera 21 species of the Chlorophyceae, was identified. Cryptomonas sp., Chamydomonas sp., Nitzschia frustulum, Oscillatoria sp., Lyngbya sp, and Dictyosphaerium pulchellum occurred successively as dominant phytoplankton species from March 1998 to February 1999. Density of phytoplankton cell number showed the peak in early April when Chlamydomoas sp. was dominated. The diversity index of phytoplankton community showed low values in the dominating period of Cryptomonas sp. and Chlamydomonas sp. The concentration of chlorophyll a fluctuated between 1.09 and 107.7$\mu\textrm{g}$ㆍ1$^{-1}$. Water temperature, salinity, DO, conduc tivity and SS were ranged 0.5-27.8$^{\circ}C$, 2.8-25.2 ppt, 4.62-19.21 mgO$_2$ㆍ1$^{-1}$, 5.33-34.76 mS/cm and 0.017-0.184 mgㆍ1$^{-1}$, respectively. Annual means of Secchi disc transparency was 36cm and thai of pH value was 8.5. The concentrations of PO$_4$-P was ranged 0.0012-0.077 mgㆍ1$^{-1}$ , and showed high value from August to October. The concentrations of NO$_2$-N, NO$_3$-N and NH$_4$-N was ranged 0.0011-0.1 mgㆍ1$^{-1}$, 0.074-3.33mgㆍ1$^{-1}$ and 0.0024-0.259 mgㆍ1$^{-1}$, respectively. Among environmental factors, water temperature was negatively correlated with DO, salinity and conductivity, DO was positively correlated with salinity and conductivity. Chlorophyll a, one of the indicies of eutropication in lake, was negatively correlated with NH$_4$-N. The cell density of Cryptomonas sp., which was the most dominant species in this lake, was negatively correlated with transparency.
