Plant Regeneration of Soybean Cultivars via Somatic Embryogenesis

  • Effective plant regeneration from immature cotyledons of soybean [Glycine max (L.) Merr.] cultivars was achieved via somatic embryogenesis. Somatic embryogenesis was performed with the cotyledons of immature embryos 14-20 d after flowering. Immature cotyledons of cv. Whangkeum were placed abaxial or adaxial side down on modified MS medium containing 20mg/L 2,4-D. The greatest number of somatic embryos, 1.2 per cotyledon, was produced from those of 4.0-4.9 mm in length which had been placed abaxial side down. Among cvs. Pecking, Whangkeum and Baekwoon, Pecking had the highest embryo induction efficiency with 4.3 somatic embryos per cotyledon in 20mg/L 2,4-D treatment and with 1.0 embryo per cotyledon in 8mg/L NAA treatment. Germinable globular somatic embryos were induced with the highest efficiency, 27.6%, in 20mg/L 2,4-D and were proliferated efficiently on liquid medium containing 10mg/L 2,4-D. The globular somatic embryos developed into germinable mature somatic embryos on medium containing 10 $\mu$M CoCl2, 9% sucrose, and 0.5% activated charcoal. These mature somatic embryos germinated on hormone-free mediu. After transfer to the soil, regenerated plants with seeds were obtained.

Genetic Diversity and Identification of Korean Elite Soybean Cultivars including Certified Cultivars Based on SSR Markers (한국 콩 보급품종을 포함한 엘리트품종의 SSR마커에 의한 유전적 다양성과 품종판별)

  • A total of 26 Korean elite soybean cultivars including 21 certified cultivars was assessed to evaluate genetic diversity and to analyze relationship among them based on 15 SSR markers. Fifteen SSR markers generated a total of 201 alleles. Average number of alleles per SSR marker was 13.4 with a range from 8 to 19. Genetic diversity of 26 cultivars estimated by PIC value ranged from 0.782 to 0.931 with an average of 0.874. PIC value of Satt197 was the highest with 0.931 and Satt141 was the lowest with 0.782 among 15 SSR markers. Cluster analysis based on genetic distances classified 26 soybean cultivars into 3 clusters. Cluster I, II and III included 2, 7 and 17 cultivars, respectively. Average genetic diversity within clusters was 0.769 with a range from 0.720 to 0.799. Average genetic diversity between clusters was 0.813 with a range from 0.725 to 0.857. Genetic diversity was higher between clusters than within clusters. Genetic relationship among clusters was near between I and II, and I and III and far between II and III cluster. All of 26 Korean elite soybean cultivars could be identified by using any of 5 combinations of 2 SSR markers with higher PIC value, i.e, $Satt197+Sat_088$, Satt197+Satt245, $Sat_088+Sat_-036$, $Sat_088+Satt245$ and Satt185+Satt245.

Difference of Hypocotyl and Growth Characters of Soybean-sprouts by Sowing Date in Sprout-soybean Cultivar (나물용 콩 파종기에 따른 콩나물의 하배축 및 생장특성 차이)

  • This study was conducted to investigate the effects of sowing date on soybean-sprout growth characters and for improving cultural techniques to produce high quality soybean-sprout. Six sprout-soybean cultivars were planted on three sowing dates in 2000 and 2001. The growth of hypocotyl in soybean sprouts was accelerated at higher temperature and the differences among cultivars became greater at later culture days. The longest of hypocotyl and root lengths of soybean sprouts was obtained from July 5 sowing, followed by May 25 and June 15 sowing. Doremikong and Hannamkong had the thickest and the thinness hypocotyls, respectively. Total length of soybean-sprouts was longer in seeds harvested from later sowing, respectively. Eunhakong produced the longest soybean-sprouts, while Tawonkong produced the shortest. Percentage of rotten seed and imperfectly germinated seed harvested at later sowing was lower. Somyeongkong and Tawonkong showed the highest and the lowest percentages of rotten seeds, respectively, while Pungsannamulkong and Hannamkong showed the lowest and the highest percentages of imperfectly germinated seeds. The soybean-sprouts yields were higher in seeds harvested in 2001 and later sowing. Although Eunhakong and Doremikong showed higher soybean-sprout yield, Pungsannamulkong and Somyeongkong showed stable soybean-sprout yield regardless of the sowing dates.

Physicochemical Properties of Soybean Leaf by Cultivar and Development of Soybean Curd Prepared with Soybean Leaf Powder (콩잎 품종에 따른 이화학적 특성 비교 및 콩잎 분말을 첨가한 두부 개발)

  • The objectives of this study were to investigate the physicochemical characteristics of soybean leaves in different cultivars and to develop a soybean curd prepared with soybean leave powder as a functional food. Four cultivars (Daewonkong leaf, Daepungkong leaf, Hwangguemkong leaf, and Seoritae leaf) were selected for this experiment. A significant difference was observed in the proximate composition of soybeans leaves (p<0.05). Soybean leaves had the highest content of carbohydrates. The mineral composition and isoflavone content in soybean leaves were significantly different among the cultivars (p<0.05). In particular, Daepongkong leaf had the highest content of genistein, daidzein, and total isoflavones. This study was also conducted to determine the quality characteristics of a soybean curd developed from daepongkong leaf with various concentrations (0, 0.1, 0.2, and 0.3%) of soybean leaf powder (SLP). The soybean curd yield rate increased according to the level of SLP added, whereas the L and a color values decreased. In the sensory evaluation, intensity scores for color, after taste, leafy taste, and chewiness were highest for the 0.3% soybean curd. The soybean curd with 0.2% SLP soybean milk attained the highest overall acceptability score. These results showed that soybean leaf was preferred over soybean curd containing 0.2% soybean leaf powder.

  • Collected 38 varieties of colored-soybean seed cultivated under a given cultural conditions were tested in the various chemical compositions and rheological properties in seeds. As it were, protein, fat, carbohydrate, ash and water contents as a chemical contents were studied not only on their statistical variations, but also on interrelationships with their collaborated rheological characteristics; hardness, cohesiveness, elasticity, gumminess and chewiness. For the rheological analysis of the materials, the General Foods Texturometer and typical texturometer curve were used.

  • Competitive relationships between soybean cultivars and weeds were determined at different plant spacings and weed competition times. Soybean yields did not differ significantly between plant spacings. Annual weed competition more signficantly reduced yields and yield components of 'Williams' soybeans than those of 'Dongbuktae' and 'Suweon 83'. 'Suweon 83' and 'Dongbuktae' were more competitive to weeds than 'Williams'.

Geographical Shift of Quality Soybean Production Area in Northern Gyeonggi Province by Year 2100 (경기북부지역 콩 생산에 미치는 지구온난화의 영향)

  • Potential impacts of the future climate change on crop production can be inferred by crop simulations at a landscape scale, if the climate data may be provided at appropriate spatial scales. Northern Gyunggi Province is one of the few prospective regions in South Korea for growing quality soybeans. Any geographical shift of production areas under the changing climate may influence the current land planning policy in this region. A soybean growth simulation was performed at 342 land units in northern Gyunggi province to test the potential geographical shift of the current production areas for quality soybeans in the near future (form 2011 to 2100). The land units for soybean cultivation were selected by the land use, the soil characteristics, and the minimum arable land area. Daily maximum and minimum temperature, precipitation, the number of rain days and solar radiation were extracted for each land unit from the future digital climate models (DCM, 2011-2040, 2041-2070, 2071-2100). Daily weather data for 30 years were randomly generated for each land unit for each normal year by using a well-known statistical method. They were used to run CROPGRO-Soybean model to simulate the growth, phonology, and yields of 3 cultivars representing different maturity groups grown at 342 land units. According to the model calculations, the warming trend in this region will accelerate the flowering and physiological maturity of all cultivars, resulting in a 7 to 9 days reduction in overall growing season and a 1 to 15% reduction in grain yield of early to medium maturity cultivars. There was a slight increase in grain yield of the late maturing cultivar under the projected climate by 2070, but a decreasing tend was dominant by the year 2100.
