Anti-Wrinkling Effect of Noni (Morinda citrifolia) by Antioxidant and Anti-Inflammatory Properties

  • Noni has been used for medicinal purposes for more than 2,000 years in South Pacific Polynesia, China and India, and has been heavily ingested as an extract for its excellent antioxidant and anti-inflammatory effects. However, a recent study found that the noni extract causes digestive disorders, kidney problems, and liver diseases, which made it necessary to use it for other purposes than as an extract. In this study, we want to evaluate the potential of noni as an anti-oxidant, anti-inflammatory and anti-wrinkling agent. Methods: Noni was freeze-dried, extracted in water, and concentrated. Skin cells were treated with the noni extract for 24 hrs and then were exposed to UVB (55 mJ/cm2). After 48 hrs of incubation, pro-inflammatory cytokine, elastase, MMP-1 and type-1 procollagen levels were measured by ELISA. Results: To find out the antioxidant effect of the noni extract, the DPPH and ABTS radical scavenging activity experiments were conducted and the noni extract showed 97.0 % and 92.0 % antioxidant efficacy at 200 ㎍/mL respectively. The noni extract (50 and 100 ㎍/mL) decreased IL-6 and TNF-α in RAW 264.7 cells induced by LPS in a concentration-dependent manner. In the RT-PCR experiment involving NO production, the noni extract (50 and 100 ㎍/mL) inhibited NO production by strongly inhibiting iNOS mRNA expression, and also inhibited the elevation of MMP-1 and elastases caused by UVB irradiation by 25.0 % and 7.0 % respectively. In addition, type-1 procollagen was elevated by 20.0 % by the noni extract treatment in HaCaT cells. Conclusion: The noni extract has photoprotective ability by reducing proinflammatory mediators, elastase and MMP-1 production, and elevation of collagen synthesis. Our findings suggest that the noni extract might be a good natural substance to protect against UVB-induced premature skin aging.

Southeast Asia as Theoretical Laboratory for the World

  • Area studies are sometimes framed as focused on specific localities, rooted in deep linguistic, cultural and historical knowledge, and hence empirically rich but, as a result, as yielding non-transferable/non-translatable findings and hence as theoretically poor. In Europe and North America some social science disciplines like sociology, economics and political science routinely dismiss any reference to local specifics as parochial "noise" interfering with their universalizing pretensions which in reality obscure their own Euro-American parochialism. For more qualitatively oriented disciplines like history, anthropology and cultural studies the inherent non-universality of (geographically constricted) area studies presents a predicament which is increasingly fought out by resorting to philosophical concepts which usually have a Eurocentric pedigree. In this paper, however, I argue that concepts with arguably European pedigree - like religion, culture, identity, heritage and art - travel around the world and are adopted through vernacular discourses that are specific to locally inflected histories and cultural contexts by annexing existing vocabularies as linguistic vehicles. In the process, these vernacularized "universal" concepts acquire different meanings or connotations, and can be used as powerful devices in local discursive fields. The study of these processes offer at once a powerful antidote against simplistic notions of "global"/"universal" and "local," and a potential corrective to localizing parochialism and blindly Eurocentric universalism. I develop this substantive argument with reference to my own professional, disciplinary and theoretical trajectory as an anthropologist and historian focusing on Vietnam, who used that experience - and the empirical puzzles and wonder encountered - in order to develop theoretical interests and questions that became the basis for larger-scale, comparative research projects in Japan, China, India, South Africa, Brazil and Europe. The subsequent challenge is to bring the results of such larger, comparative research "home" to Vietnam in a meaningful way, and thus overcome the limitations of both area studies and Eurocentric disciplines.

Histopathological Study and Expression of Beta-Catenin in Congenital Choledochal Cyst in a Tertiary Care Pediatric Referral Center in South India

  • Purpose: Choledochal cysts are congenital anomalies that occur as localized cystic or fusiform dilatations of the biliary tree. Reflux and stasis of pancreatic enzymes in the biliary duct may relate to the development of intestinal metaplasia which might be an important factor related to the carcinogenesis of choledochal cyst, thus the expression of beta-catenin in the metaplastic epithelium might be associated with malignant transformation of choledochal cyst epithelium. Methods: This study was conducted at a tertiary care pediatric center between October 2014 and March 2017. Forty patients were evaluated for epithelial lining, mural ulceration, fibrosis, inflammation, and metaplasia. Results: Out of 40, 12 cases (30.0%) were the infantile age group and 28 cases (70.0%) were in the classic pediatric group. Ulceration was classified as grade 0 (14 cases, 35.0%), grade 1 (17 cases, 42.5%), or grade 2 (nine cases, 22.5%). Inflammation was classified as grade 0 (2 cases, 5.0%), grade 1 (26 cases, 65.0%), or grade 2 (12 cases, 30.0%). Fibrosis was classified as grade 0 (five cases, 12.5%), grade 1 (11 cases, 27.5%), grade 2 (17 cases, 42.5%), or grade 3 (seven cases, 17.5%). Metaplasia was noted in five (12.5%) out of 40 cases. All choledochal cysts with metaplasia showed beta-catenin nuclear positivity on immunohistochemistry and were followed up. Conclusion: This study emphasizes the importance of detailed histopathological examination and documentation of metaplastic changes. Metaplasia was associated with beta-catenin nuclear positivity. These findings suggest a potential role for beta-catenin as a marker of metaplastic changes in choledochal cysts.

International Microsurgery Club and World Society for Reconstructive Microsurgery Webinar: Career Building in Microsurgery

  • Career building can be challenging for young surgeons, especially when topics such as lifestyle, work-life balance and subspecialization arise. Suggestions and advice from senior colleagues is very valuable but many young surgeons do not have such opportunities or are limited to a few senior surgeons. The International Microsurgery Club (IMC), in collaboration with the World Society of Reconstructive Microsurgery, organized a combined webinar for this topic and invited world renownedmicrosurgery masters polled by the IMCmembers to join, including Prof. Peter Neligan (Emeritus from University of Washington, United States), Prof. Raja Sabapathy (Ganga Hospital, India), Dr. Gregory Buncke (The Buncke Clinic, United States), Prof. Isao Koshima (Hiroshima University Hospital, Japan), Prof. David Chwei-Chin Chuang (Chang Gung Memorial Hospital, Taiwan), and Prof. Eric Santamaria (Hospital General Dr. Manuel Gea Gonzalez, Mexico) on May 1, 2022. Prof. Joon-Pio Hong (Asan Medical Center, South Korea) and Prof. Fu-Chan Wei (Chang Gung Memorial Hospital, Taiwan) were also selected but unfortunately could not make it and were therefore invited to another event in April 2023, summarized in a recently published paper. There is ample literature reporting on different aspects of developing a microsurgical career but the goal of this session was to offer an opportunity for direct exchange with experienced mentors. Moreover, insights from experienced microsurgeons from different part of the world were more likely to offer different perspectives on aspects such as career building, failure management, and team culture. This webinar event was moderated by Dr. Jung-Ju Huang (Taiwan), Dr. Susana Heredero (Spain), and Dr. Wei F. Chen (United States).

SARS-CoV-2 Delta (B.1.617.2) Variant: A Unique T478K Mutation in Receptor Binding Motif (RBM) of Spike Gene

  • Over two hundred twenty-eight million cases of coronavirus disease 2019 (COVID-19) in the world have been reported until the 21st of September 2021 after the first rise in December 2019. The virus caused the disease called severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Over 4 million deaths blame COVID-19 during the last one year and 8 months in the world. Currently, four SARS-CoV-2 variants of concern are mainly focused by pandemic studies with limited experiments to translate the infectivity and pathogenicity of each variant. The SARS-CoV-2 α, β, γ, and δ variant of concern was originated from United Kingdom, South Africa, Brazil/Japan, and India, respectively. The classification of SARS-CoV-2 variant is based on the mutation in spike (S) gene on the envelop of SARS-CoV-2. This review describes four SARS-CoV-2 α, β, γ, and δ variants of concern including SARS-CoV-2 ε, ζ, η, ι, κ, and B.1.617.3 variants of interest and alert. Recently, SARS-CoV-2 δ variant prevails over different countries that have 3 unique mutation sites: E156del/R158G in the N-terminal domain and T478K in a crucial receptor binding domain. A particular mutation in the functional domain of the S gene is probably associated with the infectivity and pathogenesis of the SARS-CoV-2 variant.

아프가니스탄 틸랴 테페의 사르마티아(Sarmathia)식 검집 패용 방식의 전개 과정으로 본 동서교섭 (Thinking in Terms of East-West Contacts through Spreading Process of Sarmathia-Pattened Scabbard on Tillya-Tepe Site in Afghanistan)

  • 최근 북방유목민족인 사르마티아에 관한 고고학적 자료들이 증가하면서 사르마티아인들의 활동 양상과 그 실체들이 조금씩 규명되고 있다. 이 글에서는 사르마티아인들의 동서 교역의 활동 양상을 사르마티아식 검집의 확산 지역 중심으로 살펴보았다. 사르마티아인들의 검은 스키타이나 페르시아인들이 사용한 아키나케스(Akinakes)의 계보를 잇는 것이지만 손잡이나 패용 방식에서 손쉽게 칼을 뺄 수 있도록 변화되었다. 즉 양 측면에 달린 4개 돌기에 끈으로 연결한 검집을 허벅지에 부착시키는 패용 구조로 변한 것이다. 사르마티아식 검은 기원전 5세기 파지리크시기 알타이 지역에서 처음 등장한 이래 사르마티아인들의 활동 중심지인 남시베리아, 박트리아, 파르티아, 로마로 확산되었다. 이 글에서는 동서 교섭의 요충지인 박트리아지역에 속하는 아프가니스탄 틸랴 테페 4호묘에서 출토된 검집에 관심을 두었다. 사르마티아식 검을 패용한 채 매장되었던 틸랴 테페 4호묘의 묘주는 당시 쿠샨왕조의 지배하에 있었던 박트리아에서 상당한 위치의 인물로 파악된다. 묘주가 2자루의 사르마티아식 검을 패용하고 있었던 것은 박트리아에 사르마티아의 영향이 상당이 수용된 사실을 의미한다. 사르마티아인들이 동서교섭에서 중요한 역할을 담당할 수 있었던 배경에는 중국산 물자를 실크로드에 공급하였던 이들의 역할과도 관련이 있다. 이에 사르마티아인들이 활동한 북방 초원 루트에 중국 한대 동경, 메론형 구슬, 수정구슬 등의 장식구슬, 금환연접구슬 등 중국 특히 비단이 생산되었던 남중국의 공예품들이 출토되는 경위에 관심을 가졌다. 모자이크 구슬은 당시 남중국 그리고 사르마티아와의 관계에 의한 것으로 살펴보았다. 거리적으로 먼 사르마티아와 남중국이 교섭할 수 있었던 중간 매체로는 흉노가 주목된다. 이와 관련하여 흉노의 유적에서 남중국에 가장 많은 분포를 보이는 금환연접구슬이 발견되는 점을 참고하였다. 남인도와 동남아시아에서 유행한 금환연접구슬이나 모자이크구슬이 아프카니스탄의 틸랴 테페 유적에는 없는 반면 같은 초원길로 연결되는 사르마티아 유적이나 흉노의 몽골 유적에서 등장하는 점이 흥미로운 것이다. 이는 남중국과 사르마티아, 흉노와의 연결 루트를 상정하게 한다. 한편 사르마티아인들의 중심연대인 기원전 2세기에서 기원후 2세기 사이 초원루트와 남중국을 잇는 교통로로는 운남에서 미안마를 지나 파키스탄, 아프가니스탄을 지나 인도 동부로 들어가는 서남실크로드로 판단된다. 1세기 동서교섭의 루트에는 북방 초원 루트, 남해 루트, 오아시스 루트 등 다양한 루트가 존재한다. 이들 루트들은 운반, 집산되는 물자의 특성이나 당시 정치적 관계에 의해 다양하게 사용되고 형성되었다. 이 글에서는 인도와 중앙아시아, 그리고 남중국을 잇는 최단거리의 길인 서남루트가 북방 유목민들인 사르마티아인들에 의해 사용되고 개척된 상황을 살펴본 것이다.

역내 해양협력 및 신뢰증진을 위한 한국 해군의 기여방안 (ROK Navy's Role for a Confidence Building and Mutual Cooperation on the East Asian Sea)

  • We are witnessing the growing maritime tension on the East Asian sea these days. Each naval powers in the region are competing each other to acquire more advanced naval capabilities. Based upon the rapid economic development, China is actively beefing up its naval capabilities and expand its boundary of naval activities all over the East Asian region. Chinese Navy already unveiled its expansive naval strategy replacing the traditional concept of 'Near-Sea Defense' with the new concept of 'Far-Sea Defense' strategy. In response to potential rival's naval build up, the U.S. is redeploying its naval forces focusing on the Asia-Pacific region. The U.S. enhances its joint naval exercises with the countries in the region, such as Japan, India, Australia and so on. In addition, Washington is devising new naval strategy under the concept of 'Air-Sea Battle' to deter Peking's so-called 'Anti-Access/ Area Denial(A2AD)' strategy. As a close ally of the U.S., Japan also disclosed its clear intention to strengthen the Maritime Self Defense Force(MSDF)'s capabilities by introducing the new concept of 'Dynamic Defense Force' in 2011. Under the new concept, JMSDF is pursuing the additional acquisition of submarines, quasi-aircraft carriers, Aegis-equipped destroyers, etc. Under the new president's strong leadership, Russia is also invigorating the naval build-up. Especially, Russia is fortifying the Pacific Fleet's naval assets by deploying new-type of naval ships such as the Mistral which was imported from France. In the midst of competitive naval build-up among the major naval powers in the region, we are observing the growing maritime conflicts on the East China Sea as well as South China Sea. Those naval conflicts can pose severe threats to our national interests. Maritime conflicts on the East or South China Sea can imperil our sea lanes which will be indispensible for national economic development. Neighboring countries' maritime conflicts also will cast an uncertainty on the path to mobilize international cooperation to resolve the North Korean issues. We should contribute to ease the maritime tension in the region by various ways. First, we should actively galvanize the bilateral maritime dialogue among the major naval powers in the region. Second, we also should take the lead to form a multilateral maritime cooperation mechanism in the region. Above all, we should set the aim to be a peaceful maritime power who can contribute to a building of stable maritime order in the region with a considerable naval power.

Screening for the 3' UTR Polymorphism of the PXR Gene in South Indian Breast Cancer Patients and its Potential role in Pharmacogenomics

  • Background: Breast cancer, the commonest cancer among women in the world, ranks top in India with an incidence rate of 1,45,000 new cases and mortality rate of 70,000 women every year. Chemotherapy outcome for breast cancer is hampered due to poor response and irreversible dose-dependent cardiotoxicity which is determined by genetic variations in drug metabolizing enzymes and transporters. Pregnane X receptor (PXR), a member of the nuclear receptor superfamily, induces expression of drug metabolizing enzymes (DMEs) and transporters leading to regulation of xenobiotic metabolism. Materials and Methods: A genomic region spanning PXR 3' UTR was amplified and sequenced using genomic DNA isolated from 96 South Indian breast cancer patients. Genetic variants observed in our study subjects were queried in miRSNP to establish SNPs that alter miRNA binding sites in PXR 3' UTR. In addition, enrichment analysis was carried out to understand the network of miRNAs and PXR in drug metabolism using DIANA miRpath and miRwalk pathway prediction tools. Results: In this study, we identified SNPs rs3732359, rs3732360, rs1054190, rs1054191 and rs6438550 in the PXR 3; UTR region. The SNPs rs3732360, rs1054190 and rs1054191 were located in the binding site of miR-500a-3p, miR-532-3p and miR-374a-3p resulting in the altered PXR level due to the deregulation of post-transcriptional control and this leads to poor treatment response and toxicity. Conclusions: Genetic variants identified in PXR 3' UTR and their effects on PXR levels through post-transcriptional regulation provide a genetic basis for interindividual variability in treatment response and toxicity associated with chemotherapy.

네팔에서의 인공습지 적용: 최근 개발 및 향후 고려사항 (Status of Constructed Wetlands in Nepal: Recent Developments and Future Concerns)

  • 네팔은 중국과 인도 사이에 위치한 남부 아시아의 내륙국가로 폐수관리는 도시지역에서 가장 중요한 문제 중 하나이다. 네팔의 폐수처리 시스템은 비싼 운영비, 불연속적인 전력 공급, 유지관리 및 기술인력의 부족으로 인해 조성과 운영이 어렵다. 이러한 이유로 인공습지는 폐수처리시설의 대안기술로써 다양한 지역에 적용되고 있다. 일반적으로 인공습지 기술의 효율 지속 가능성은 적절한 운영과 유지관리 및 적극적인 지역사회 참여에 의존한다. 따라서 본 연구에서는 26개의 인공습지에서의 제거 효율, 방류수질, 네팔 수질기준, 운영 및 유지관리 활동 등을 조사하고 문제점을 분석하여 관리방안 등을 도출하였다. 오염물질당 제거효율은 ka-1의 인공습지가 가장 높은 것으로 나타났으며, B-1, L-3, Ka-5, k-1 순으로 높은 것으로 나타났다. 네팔의 인공습지 조성기술은 최근 20년간 기술개발 없이 전통적 방식에 의존해온 결과 비효율적인 성능으로 인해 많은 습지가 가동이 중단되거나 폐쇄되는 것으로 나타났다. 또한 기술개발의 부족, 자연재해, 대중의 인공습지에 대한 낮은 인식 및 예산 부족 등은 인공습지의 지속적인 개발을 저해하고 있는 것으로 조사되었지만, 인공습지는 네팔 도시지역의 폐수문제에 대한 해결방안으로 고려되고 있다.

Maxent 모델을 이용한 반달가슴곰의 서식지 분포변화 예측 (Habitat Distribution Change Prediction of Asiatic Black Bears (Ursus thibetanus) Using Maxent Modeling Approach)

  • 본 연구는 국립공원을 포함한 국내 보호지역에서 반달가슴곰 (Ursus thibetanus)을 효과적으로 보전하고, 종 복원을 성공하기 위해서 종의 재도입에 적합한 지역을 객관적으로 평가하기 위한 기초자료를 제공하는 데 그 목적이 있다. 이를 위해서 Maxent 모델과 기후, 지형, 그리고 도로 및 토지이용과 관련된 환경 변수를 이용하여 반달가슴곰의 출현 기록이 있는 동아시아, 동남아시아, 그리고 인도를 대상으로 잠재 서식지를 예측하고, 이와 관련된 기후 및 환경 변수의 영향을 평가하였다. 또한 미래 기후변화에 따라서 반달가슴곰에게 적합한 서식 범위의 면적과 지리적인 변화를 분석하였다. 생물보전을 위해서 야생생물의 서식지분포연구에 널리 활용되고 있는 Maxent 모델의 판별정확도를 나타내는 AUC 값이 0.893 (sd=0.121)으로 산출되었다. 이는 반달가슴곰의 잠재 서식지를 예측하고 미래 기후변화에 따른 서식지 변화특성을 평가하는 데 유용하였다. IUCN에서 평가한 반달가슴곰의 분포지도와 비교해서, 현존 지역 (Extant)은 Maxent 모델로 예측된 서식 확률이 국가별 지역적으로 다양하고, 멸종 지역 (Extinct)은 상대적으로 낮았다. 이는 반달가슴곰이 서식하는 환경 특성의 차이가 지역적으로 반영된 결과라 할 수 있다. 반달가슴곰의 잠재 서식지 분포에 영향을 주는 환경은 기후, 지형 그리고 인위적 요소인 도로로부터의 거리와 같은 요소보다 토지피복 유형의 영향이 가장 높았는데, 특히 낙엽활엽수림지역이 더욱 선호될 것으로 예측되었다. 또한 기온의 연간범위보다 연평균강수량과 건조시기의 강수량의 영향이 더욱 클 것으로 예측되었고 도로로부터 거리가 멀어질수록 서식가능성이 높은 것으로 나타났다. 이는 반달가슴곰은 먹이자원뿐만 아니라 인간의 간섭이 없는 보다 안정된 지역을 선호할 것으로 추측된다. 미래 기후변화에 따라서 서식적합지역은 점차 확장할 것으로 전망되었고, 남한에서는 전남, 전북 그리고 강원도지역이, 일본에서는 Kyushu, Chugoku, Shikoku, Chubu, Kanto 그리고 Tohoku의 접경 지역이, 중국에서는 Jiangxi, Zhejiang 그리고 Fujian의 접경 지역이 향후 아시아지역에서 반달가슴곰이 서식할 수 있는 핵심지역이 될 것으로 예상된다. 본 연구는 반달가슴곰의 서식지 보전과 효율적인 관리, 인위적으로 도입된 개체의 방사지점 선정, 향후 서식 범위의 확장에 따른 보호지역 설정 그리고 인간과 충돌지역의 관리에 대한 기초자료로서 활용될 것으로 기대된다.
