Sustainable Production of Dihydroxybenzene Glucosides Using Immobilized Amylosucrase from Deinococcus geothermalis

  • The amylosucrase encoding gene from Deinococcus geothermalis DSM 11300 (DgAS) was codon-optimized and expressed in Escherichia coli. The enzyme was employed for biosynthesis of three different dihydroxybenzene glucosides using sucrose as the source of glucose moiety. The reaction parameters, including temperature, pH, and donor (sucrose) and acceptor substrate concentrations, were optimized to increase the production yield. This study demonstrates the highest ever reported molar yield of hydroquinone glucosides 325.6 mM (88.6 g/l), resorcinol glucosides 130.2 mM (35.4 g/l) and catechol glucosides 284.4 mM (77.4 g/l) when 400 mM hydroquinone, 200 mM resorcinol and 300 mM catechol, respectively, were used as an acceptor substrate. Furthermore, the use of commercially available amyloglucosidase at the end of the transglycosylation reaction minimized the gluco-oligosaccharides, thereby enhancing the target productivity of mono-glucosides. Moreover, the immobilized DgAS on Amicogen LKZ118 beads led to a 278.4 mM (75.8 g/l), 108.8 mM (29.6 g/l) and 211.2 mM (57.5 g/l) final concentration of mono-glycosylated product of hydroquinone, catechol and resorcinol at 35 cycles, respectively, when the same substrate concentration was used as mentioned above. The percent yield of the total glycosides of hydroquinone and catechol varied from 85% to 90% during 35 cycles of reactions in an immobilized system, however, in case of resorcinol the yield was in between 65% to 70%. The immobilized DgAS enhanced the efficiency of the glycosylation reaction and is therefore considered effective for industrial application.

  • Many super-resolution reconstruction algorithms have been proposed since it was the first proposed in 1984. The spatial domain approach of the super-resolution reconstruction methods is accomplished by mapping the low resolution image pixels into the high resolution image pixels. Generally, a super-resolution reconstruction algorithm by using the spatial domain approach has the noise problem because the low resolution images have different noise component, different PSF, and distortion, etc. In this paper, we proposed the new super-resolution reconstruction method that uses the L1 norm to minimize noise source and also uses the Huber norm to preserve edges of image. The proposed algorithm obtained the higher image quality of the result high resolution image comparing with other algorithms by experiment.

  • This paper proposes a new method for the inverse problem of the three-dimensional reconstruction of the electrical activity of the brain from electroencephalography (EEG). Compared to conventional direct methods using additional parameters, the proposed approach solves the EEG inverse problem iteratively without any parameter. We describe the Lagrangian corresponding to the minimization problem and suggest the numerical inverse algorithm. The restriction of influence space and the lead field matrix reduce the computational cost in this approach. The reconstructed divergence of primary current converges to a reasonable distribution for three dimensional sphere head model.

Domain Adaptation Image Classification Based on Multi-sparse Representation

  • Generally, research of classical image classification algorithms assume that training data and testing data are derived from the same domain with the same distribution. Unfortunately, in practical applications, this assumption is rarely met. Aiming at the problem, a domain adaption image classification approach based on multi-sparse representation is proposed in this paper. The existences of intermediate domains are hypothesized between the source and target domains. And each intermediate subspace is modeled through online dictionary learning with target data updating. On the one hand, the reconstruction error of the target data is guaranteed, on the other, the transition from the source domain to the target domain is as smooth as possible. An augmented feature representation produced by invariant sparse codes across the source, intermediate and target domain dictionaries is employed for across domain recognition. Experimental results verify the effectiveness of the proposed algorithm.

Application of a deep learning algorithm to Compton imaging of radioactive point sources with a single planar CdTe pixelated detector

  • Compton imaging is the main method for locating radioactive hot spots emitting high-energy gamma-ray photons. In particular, this imaging method is crucial when the photon energy is too high for coded-mask aperture imaging methods to be effective or when a large field of view is required. Reconstruction of the photon source requires advanced Compton event processing algorithms to determine the exact position of the source. In this study, we introduce a novel method based on a Deep Learning algorithm with a Convolutional Neural Network (CNN) to perform Compton imaging. This algorithm is trained on simulated data and tested on real data acquired with Caliste, a single planar CdTe pixelated detector. We show that performance in terms of source location accuracy is equivalent to state-of-the-art algorithms, while computation time is significantly reduced and sensitivity is improved by a factor of ~5 in the Caliste configuration.

  • In order to render 3D model of the bone, the stack of cross-sectional images must be reconstructed from a series of X-ray radiographs, served as the projections. In the case where the distance between x-ray source and detector is not infinite, image reconstruction from projection based on parallel-beam geometry provides an error in the cross-sectional image. In such case, image reconstruction from projection based on conebeam geometry must be exercised instead. This paper is devoted to the determination of detector center for SART conebeam Technique which is critically effect the performance of the resulting 3D modeling.

  • Squalene is a linear triterpene formed via the MVA or MEP biosynthetic pathway and is widely distributed in bacteria, fungi, algae, plants, and animals. Metabolically, squalene is used not only as a precursor in the synthesis of complex secondary metabolites such as sterols, hormones, and vitamins, but also as a carbon source in aerobic and anaerobic fermentation in microorganisms. Owing to the increasing roles of squalene as an antioxidant, anticancer, and anti-inflammatory agent, the demand for this chemical is highly urgent. As a result, with the exception of traditional methods of the isolation of squalene from animals (shark liver oil) and plants, biotechnological methods using microorganisms as producers have afforded increased yield and productivity, but a reduction in progress. In this paper, we first review the biosynthetic routes of squalene and its typical derivatives, particularly the squalene synthase route. Second, typical biotechnological methods for the enhanced production of squalene using microbial cell factories are summarized and classified. Finally, the outline and discussion of the novel trend in the production of squalene with several updated events to 2015 are presented.

It's not only mistreated as a limitation of personal asset but also treated as national source Despite of rather compulsive and constructive method must be noticed by whoever owns for, more positive follows, promotion and directing must be able to guide its understanding and adherence of importance and inevitability.

  • The key to model-based iterative reconstruction (MBIR) algorithms for transmission computed tomography lies in the ability to accurately model the data formation process from the emitted photons produced in the transmission source to the measured photons at the detector. Therefore, accurately modeling the system matrix that accounts for the data formation process is a prerequisite for MBIR-based algorithms. In this work we compared quantitative performance of the three representative ray-driven methods for calculating the system matrix; the ray-tracing method (RTM), the distance-driven method (DDM), and the strip-area based method (SAM). We implemented the ordered-subsets separable surrogates (OS-SPS) algorithm using the three different models and performed simulation studies using a digital phantom. Our experimental results show that, in spite of the more advanced features in the SAM and DDM, the traditional RTM implemented in the OS-SPS algorithm with an edge-preserving regularizer out-performs the SAM and DDM in restoring complex edges in the underlying object. The performance of the RTM in smooth regions was also comparable to that of the SAM or DDM.

  • We are familiar with the holography in these days. For making holography the coherent sources like LASER are used in many fields. But coherent holography has many problems. Coherent holography needs many instrument for practical use like 3-D TV case. In solving the problem we use the non-coherent source. Nowadays many methods like conoscopic holo graphy using anisotropic crystal, shadow casting and interferometric systems are suggested. In this paper we make the hologram using the triangular interferometric systems. [1],[2],[3],[4]. We explain the afocal and double-afocal system which consists of the triangular interferometric system. The holography made in one point and two point cases is imaged on CCD camera and we handle the image data digitally for the reconstruction efficiently. In reconstructing the hologram the Fraunhofer diffraction theory is used. We adopt the rectangular aperture for the convenience of calculation. In the future we must reconstruct the perfect 3-Dimensional object by optical method. For this, we have many problems like resolution problem. We must solve these problem for perfect reconstruction.
