Karyotypic Analysis and Physical Mapping of rRNA Gene Loci in Persicaria tinctoria (쪽의 핵형분석과 rRNA 유전자의 염색체상 위치)

  • Karyotypic analysis and FISH (fluorescence in situ hybridization) with 45S and 5S rRNA genes were carried out in Persicaria tinctoria H Gross. The somatic metaphase chromosomes were ranged from 2.25 ${\mu}m$ to 1.50 ${\mu}m$ in length. Chromosome number was 2n = 4x = 40 with the basic number of x = 10. The chromosome complement of the species consisted of 16 pairs of metacentrics (chromososomes 1,2,3,4,6,7,8,9, 10, 11, 12, 13, 15, 18, 19 and 20) and 4 pairs of submetacentrics (chromosome 5, 14, 16 and 17). The karyotype formula was K(2n) = 4x = 32 m + 8 sm. In FISH analysis, three pairs of 45S rRNA gene loci on the terminal region of submetacentrics (chromosomes 5, 16 and 17) and two pairs of 5S rRNA gene loci on the centromeric region of metacentrics (chromosomes 9 and 11) were detected, respectively.

  • The karyotypes of Biomphalaria tenagophila collected from Rio de Janeiro, Brazil were studied using the air-drying method. Somatic cells of this species had 2n=36. The 18 chromosome pairs were identified and classified into 3 groups. The diploid cell has 7 pairs of metacentric, 8 pairs of submetacentric, and 3 pairs of subtelocentric chromosomes. Observed chromosomes ranged from 2.4 to $6.4{\mu}m$, and the total length was $122.3{\mu}m$. This is the first report on the chromosome of B. tenagophila.

  • To elucidate cytogenetic differences, karyotype analysis and FISH (fluorescence in situ hybridization) with 45S and 5S rDNAs were carried out in the three Astragalas species: Astragalas membranaceus Bunge, A. membranaceus var. alpinus Nakai and A. mongholicus Bunge. The somatic metaphase chromosome numbers of all three species were 2n=2x=16 and the size of chromosomes ranged $2.19{\sim} 5.73\;{\mu}m$. The chromosome complement of A. membranaceus consisted of each four pairs of metacentrics (chromosomes 3,4,6 and 7) and submetacentrics (chromosomes 1,2,4 and 8). In A. membranaceus var. alpinus, the chromosome complement consisted of two pairs of metacentrics (chromosomes 4 and 8) and six pairs of submetacentrics (chromosomes 1,2,3,5,6 and 7). A. mongholicus had three pairs of metacentrics (chromosomes 6,7 and 8) and five pairs of submetacentrics (chromosomes 1,2,3,4 and 5). Using bicolor-FISH, one pair of 45S and 5S rDNA signals could be detected on the centromeric regions of chromosomes 8 and 7 of A. membranaceus and A. mongholicus, respectively. In contrast, A, membranaceus var. alpinus had one pair of 45S signals on the centromeric region of chromosome 8 and two pairs of 5S rDNA signals on the short arms of chromosomes 7 and 8.

Hypoxis aurea Lour. (Hypoxidaceae): a Rare Species from Jeju Island which is Rediscovered Seventy Years after its First Collection in Korea

  • We described and illustrated a rare species in Korea, Hypoxis aurea Lour. (Hypoxidaceae) which was rediscovered about 70 years after its first collection from Jeju island in Korea. The members of the family Hypoxidaceae R. Br. are distinguished from the plants of Amaryllidaceae J. St-Hill. by having grass-like leaves, an invisible stem which is modified into a corm or a rhizome, trimerous, and radially symmetric flowers with an inferior ovary developing into a capsule on scapes. Hypoxis aurea Lour. is readily distinguishable from Curculigo orchinoides Gsertn. in Japan by beakless ovary and capsular fruit. The number of somatic chromosome is 2n=54.

  • Leaf mesophyll protoplasts of Nicotiana tabacum (nitrate reductase deficient mutant) were fused with cell suspension protoplasts of albino Petunia inflata in an electric field. Hybrid cell colonies were selected for nitrate reductase proficiency and chlorophyll synthesis. Five hybrid plant lines, regenerated from the selected calli lines, were analysed by electrophoresis, number of chromosomes and morphological characters. Somtic hybrid plants showed both parent patterns in the isozymesof isoleucine aminopeptidase and esterase. The hybrids had the expected chromosome number of 62 and exhibited an intermediate floral morphology when compared with the parents, but plant height and leaf arrangement were similar to N. tabacum.

Karyotypes of Korean Endemic Land Snail, Koreanohadra koreana (Gastropoda: Bradybaenidae)

  • The karyotypes of Korean endemic land snail, Koreanohadra koreana, using air-drying method wereinvestigated. Somatic cells of this species had 2n = 58. Karyotypes were also analysed with 16 metacentric, 12 submetacentric and one subtelocentric chromosome pairs. Observed chromosomes ranged from 2.6 to $8.9{\mu}m$ and the total length was $122.3{\mu}m$. This is the second report on the chromosome numbers and the karyotype of K. koreana.

  • We report somatic chromosome numbers 2n = 12 for three Carex sect. Siderostictae Franch. ex Ohwi (Cyperaceae) from Korean populations: Carex ciliatomarginata Nakai, C. okamotoi Ohwi, and C. siderosticta Hance. This study is the first chromosome number report for the species C. ciliatomarginata from Korean populations. As found in other Carex species, all the chromosomes examined in the section exhibit nonlocalized centromere (polycentric or holocentric) and large (more than ca. $1{\mu}m$ long) chromosomes. Considering the basal phylogenetic position of the section in tribe Cariceae Pax, small numbers of large chromosomes have been hypothesized as primitive characters in Cariceae, and our observation supports the hypothesis. Further investigations of chromosomes in Carex are needed for a better understanding of species richness in the genus.

Centromere Repeat DNA Originated from Brassica rapa is Detected in the Centromere Region of Raphanus sativus Chromosomes

  • Fluorescence in situ hybridization (FISH) is a powerful tool for the detection of DNA sequences in the specific region of the chromosomes. As well as for the integrated physical mapping, FISH karyotype analysis has to be preceded. Karyotype of Raphanus sativus 'Wonkyo 10039' was analyzed by a dual-color FISH technique; using various repetitive DNA probes, including 5S rDNA, 45S rDNA, and centromere retrotransposon. The length of the somatic metaphase chromosome ranged from 1.35 to $2.06{\mu}m$ with a total length of $15.29{\mu}m$. The chromosome complements comprised of eight pairs of metacentrics and one pair of submetacentric. Bleached DAPI Band analysis revealed a heterochromatin region, covering 28.6% to 50.4% each chromosomes. 5S and 45S rDNA sequences were located on two and three pairs of chromosomes, respectively. The centromere retrotransposon of Brassica (CRB) is a major component in Brassica related species that has been maintained as a common centromere component. CRB signals were detected on the centromere and pericentromeric region of R. sativus 'Wonkyo 10039' and three basic Brassica species (B. rapa, B. nigra, and B. oleracea). These results will provide a valuable background for physical mapping and elucidation of the evolutionary relationship among the Brassica related species.

  • Karyotype analysis and chromosomal localization of 5S and 45S rDNAs using FISH (fluorescence in situ hybridization) technique were carried out in Nicotiana plumbaginifolia. Somatic chromosome number of N. plumbaginifolia was 2n=20 and kyarotype was composed of three pairs of metacentric chromosomes (1,2 and 7) and seven pairs of submetacentric chromosomes (3, 4, 5, 6, 8, 9 and 10). Length of chromosomes was ranged from 2.29 to 4.50 ${\mu}{\textrm}{m}$. Chromosome 1 and 2 were identified as satellite chromosomes. In FISH experiment, one pair of 5S rDNA signals was detected on the centromeric region of chromosome 2 and one pair of 45S rDNA signals was detected on the terminal region of chromosome 1.

  • This study was carried out to determine the chromosomal localization of the 5S and 18S-26S ribosomal DNA(rDNA) genes by means of fluorescence in situ hybridization(FISH) techniques, and the constitutive heterochromatin detected by means of Gimsa C-banding technique in rye(Secale cereale L.). The somatic chromosomes number was 2n=14. The karyotype consists of four pairs of metacentrics(chromosomes 1, 2, 3, and 7) and three pairs of submetacentrics(chromosomes 4, 5, and 6). Secondary constrictions appeared in the short arm of chromosome 1. The 5S rDNA genes have been located on two pairs of chromosomes 1 and 5, and 18S-26S rDNAs genes have been located on one pair of chromosome 1. 5S rDNA genes were detected on the distal region of the secondary constrictions in nucleolus organizer regions(NOR) in chromosome 1, and other detected on the intercalary region in the short arm of chromosome 5.
