Anti-inflammatory and anti-allergic effect of soybean extracts produced by organic cultivation (유기농 대두 추출물의 항염증 및 항알레르기 효과)

  • This present study was carried out to investigate the biological effects of soybean extracts comparing organic and conventional cultivation. Cellular and molecular analysis was performed to determine anti-oxidative, anti-inflammatory, anti-apoptotic, and anti-inflammatory effects of both soybean extracts. First, we obtained various solvent extracts of soybeans such as water, ethanol, and methanol. Molecular and cellular analysis were performed with 0.1 mg/ml concentration of each solvent extracts. The results of anti-oxidative, anti-inflammatory and anti-apoptotic effects of organic cultivated soybean extracts were prominent than conventional cultivated soybean extracts. However, discrepancy between organic and conventional cultivated soybean extracts was not observed in anti-allergic effects determined by releasing histamine from rat mast cell line, RBL-2H3. Conclusively, organic cultivated soybeans have stronger effects than conventional cultivated soybeans in suppression of inflammation. In addition, organic soybeans could be applied as a functional food ingredient for treatment of chronic inflammation, asthma, and atopic dermatitis with enhanced anti-inflammatory activities.

Analysis of Propylen Chlorohydrin in Hydroxypropyl Distarch Phosphate by Accelerated Solvent Extractor (ASE) (가속화용매추출기를 이용한 히드록시프로필인산이전분의 프로필렌클로로히드린 분석방법에 관한 연구)

  • Accelerated solvent extraction (ASE) was used for a rapid and simple extraction of propylenchlorohydrin (PCH) residue in hydroxylpropyl modified starch. The effects of temperature, pressure and extraction solvent on the extraction efficiency were investigated to find the optimal condition of ASE. The optimal conditions for PCH extraction in hydroxylpropyl modified starch were static time of 50 min, purge time of 300 sec, heating time of 5min, temperature of 12$0^{\circ}C$, pressure of 2500 psi, flush (%) with 100 volumes, and ethylacetate as an extraction solvent. The recovery (96.1%) of this method was higher than that (76.4%) of Code of Food Additive. Therefore, the ASE was a good method in both aspects of efficiency and effectiveness.

The Oxidative Stability of Solvent Extracts of Sea Tangle Powder(STP)and Maejakgwa Made with STP (다시마 용매추출물과 다시마 분말 첨가에 의한 매작과의 산화안정성)

  • The purpose of this study was to investigate the antioxidative effects of Sea tangle powder(STP) solvent extracts as well as Maejakgwa made with STP. The STP solvent extracts were added to soybean oil at a quantity of 0.05%. The solvents used for extraction were methanol, ethanol, ethyl acetate, and petroleum ether. Soybean oil without added STP was used as a negative control, and soybean oil samples containing 0.02% butylated hydroxy toluene(BHT) and $\alpha$-tocopherol were used as positive control, respectively. Each sample was stored at $50^{\circ}C$ for 30 days. The oxidation levels of these samples were determined by measuring their acid values, peroxide values, and thiobarbituric acid(TBA) values. The soybean oil samples containing the STP extracts had lower oxidation levels than both the negative control and $\alpha$-tocopherol positive control, and the sample containing the 0.05% methanol extracts had the lowest oxidation. According to the Rancimat method, the methanol extract(320 min) and ethanol extract(316 min) demonstrated longer induction periods as, compared to the control(253 min), $\alpha$-tocopherol(255 min), and BHT(309 min) samples. For the Maejakgwa, acid values increased over the storage period, however, the samples made with STP had lower values than the control group. Peroxide values increased rapidly for 30 days and then decreased. The TBA values of the Maejakgwa samples made with 3% and 9% STP were lower than those of the 15% STP sample and the control. In conclusion, the oxidative stability of soybean oil containing solvent extracts of STP and Maejakgwa made with STP were increased.

Anti-invasive Effect of Artemisia scoparia Halophyte Extract and its Solvent-partitioned Fractions in Human Fibrosarcoma Cells (인간 섬유육종세포에서 비쑥 추출물과 유기용매 분획물의 암전이 억제 효과)

  • The halophyte Artemisia scoparia is an edible medicinal plant, with insecticidal, anti-inflammatory, anticholesterol, antipyretic, and antibacterial effects. The aim of this study was to assess the inhibitory effect of crude extract and solvent-partitioned fractions obtained from A. scoparia on MMP-2 and MMP-9 activity in phorbol-12-myristate-13-acetate (PMA)-stimulated human fibrosarcoma HT-1080 cells using four different activity tests: gelatin zymography, MMP enzyme-linked immunosorbent assay (ELISA), wound healing assay, reverse transcription-polymerase chain reaction (RT-PCR), and Western blot assay. A. scoparia samples were extracted twice with methylene chloride (MC) and twice with methanol (MeOH). After the MC and MeOH crude extracts were combined, the combined crude extracts showed a significant inhibitory effect against MMP-2 and MMP-9 enzymes. They were then fractionated into n-hexane, 85% (v/v) aqueous methanol (85% (v/v) aq.MeOH), n-butanol, and water according to solvent polarity. Among the four solvent-partitioned fractions, n-hexane and 85% (v/v) aq. MeOH fractions significantly inhibited MMP-2 and MMP-9 activity and cell mobility. In addition, the n-hexane and 85% (v/v) aq.MeOH fractions effectively inhibited MMP-2 and -9 activity in the gelatin zymography and MMP ELISA assay. In the wound healing assay, RT-PCR, and Western blot assay, all solvent-partitioned fractions, except the H2O fraction, significantly suppressed cell migration, as well as the expression levels of MMP-2 and -9 mRNA and proteins.

Effect of Exposure Chlorpyrifos (CPF) on Survival, Hatching rate, Reproductivity and Histological Changes of Japanese Medaka, Oryzias latipes (Chlorpyrifos(CPF)가 송사리, Oryzias latipe의 생존, 부화율, 번식력에 미치는 영향 및 조직학적 변화)

  • Chlorpyrifos (CPF) is a widely used organophosphate insecticide. Japanese medaka, Oryzias latipes was selected to investigate the effects of chlorpyrifos on survival rate, hatching rate, reproductivity and histopathological changes. Adult fish were exposed to CPF at concentrations of 20, 40, 80 and $160{\mu}g/L$, and the test duration was 4 weeks. An experiment with eggs was also performed under CPF exposure continued for 7 days covering from the fecundation to end-hatching stages. Survival rate of the adult medaka decreased in CPF $80{\mu}g/L$ and $160{\mu}g/L$ treatment groups compared with control, solvent control, $20{\mu}g/L$ or $40{\mu}g/L$L treatment group. Spontaneous start of feeding significantly decreased compared with the control (p < 0.01) when exposed to CPF at above 40 $\mu g/L$. Total spawning decreased by the 20 or $40{\mu}g/L$ CPF treatment compared with the control or solvent control in the test with eggs obtained from untreated adult medaka, and abnormal eggs increased in those CPF-treated groups. There were certain adverse effects at above $40{\mu}g/L$ determined from liver and gonad histological examinations. The results indicate that CPF has insignificant toxic effects in Japanese medaka at less than $20{\mu}g/L$ in long term exposure.

Anti-inflammatory and Anti-Atopic Effects of Crude Extracts and Solvent Fractions of Phormium tenax leaf (신서란(Phormium tenax) 잎 조추출물 및 용매 분획물의 항염증 및 항아토피 효과)

  • This study describes a preliminary evaluation of the anti-inflammatory activity and anti-atopic activity of Phormium tenax leaf extracts. P. tenax leaf was extracted using 70% ethanol and then fractionated sequentially with n-hexane, methylene chloride, ethyl acetate, n-butanol. In order to effectively screen for anti-inflammatory agents, we first investigated the inhibitory effects of P. tenax leaf crude extracts and solvent fractions on production of pro-inflammatory factors[nitric oxide(NO), prostaglandin $E_2(PGE_2)$, inducible nitric oxide synthase(iNOS) and cyclooxygenase-2(COX-2)] and pro-inflammatory cytokines [tumor necrosis $factor-{\alpha}(TNF-{\alpha})$, interleukin-6(IL-6) and $interleukin-1{\beta}(IL-1{\beta})$] in lipopolysaccharide(LPS)-stimulated RAW 264.7 cells. In addition, we also evaluated of their inhibitory effect on the atopic dermatitis-like inflammatory markers such as macrophage-derived chemokine(MDC) and thymus and activation-regulated chemokine(TARC) in HaCaT cells. Among the five solvent fractions of P. tenax, methylene chloride and ethyl acetate fractions inhibited production of pro-inflammatory factors and pro-inflammatory cytokines in a dose dependent manner, respectively. These fractions were also showed inhibitory activity for MDC and TARC expression levels in $IFN-{\gamma}-stimulated$ HaCaT cells, respectively. These results suggest that P. tenax have significantly effects of anti-inflammatory activity and anti-atopic activity that might be beneficial for the topical treatment of inflammatory skin disorders.

Theoretical Study of the N-(2,5-Methylphenyl)salicylaldimine Schiff Base Ligand: Atomic Charges, Molecular Electrostatic Potential, Nonlinear Optical (NLO) Effects and Thermodynamic Properties

  • Optimized geometrical structure, atomic charges, molecular electrostatic potential, nonlinear optical (NLO) effects and thermodynamic properties of the title compound N-(2,5-methylphenyl)salicylaldimine (I) have been investigated by using ab initio quantum chemical computational studies. Calculated results showed that the enol form of (I) is more stable than keto form. The solvent effect was investigated for obtained molecular energies, hardneses and the atomic charge distributions of (I). Natural bond orbital and frontier molecular orbital analysis of the title compound were also performed. The total molecular dipole moment (${\mu}$), linear polarizability (${\alpha}$), and first-order hyperpolarizability (${\beta}$) were calculated by B3LYP method with 6-31G(d), 6-31+G(d,p), 6-31++G(d,p), 6-311+G(d) and 6-311++G(d,p) basis sets to investigate the NLO properties of the compound (I). The standard thermodynamic functions were obtained for the title compound with the temperature ranging from 200 to 450 K.

Anti-lipid Peroxidation and Liver Protective Effects of Polygonum aviculare L. (마디풀(Polygonum aviculare L.)의 전초가 지질과산화(脂質過酸化) 및 간기능(肝機能)에 미치는 영향)

  • Polygonum aviculare L. was extracted by methanol and then fractionated systematically with solvents and column chromatographic method in order to isolate ingredients with anti-lipid peroxidation and liver protectective effects. Among these fractions, ethylacetate soluble part(MWE) showed the strongest in the anti-lipid peroxidation. Furthermore, the crude subtraction(MWE 4) with Rf value near 0.42, which is separated from MWE by column chromatography using a solvent system, inhibited the lipid peroxidation of rat liver in vitro. Moreover, MWE 4 decreased GOT, GPT and TBA value compared with control and suggested high protective effects against hepatotoxicity induced by carbon tetrachloride in mice. The active compounds in MWE 4 were assumed to be flavonoid glucosides.

Temperature, Medium and Structural Effects on the Acid Dissociation Constants of Certain Schiff Bases Derived from Isatin with Some Amino Acids and Aroylhydrazines

  • The acid dissociation constants of certain Schiff bases derived from isatin with p-substituted benzoyl hydrazines and somee amino acids of glycine, $\beta$-and $\alpha$-alanine, valine, $\beta$-phenyl-$\alpha$-alanine and anthranilic acid have been detemined potentiometrically at different temperatures in different aquo-organic solvent mixtures (ethnol, dioxane, dimethyl formamide, methanol, acetone and tetrahydrofurance). The pKa values were demonstrated on the light of the different electornic and steric effects of the substituents and the sovlent characteristics. In all the mixed media used, ionization of the compounds decreased by increasing the mole fraction of the organic cosolvent. Theemodynamic parameters $(\Delta{H}^\circ,\;\Delta{G}^\circ,\;\Delta{S}^\circ)$ were evaluated. The structural effects of the investigated compounds on these parameters were reported and discussed.
