Antibacterial Activity of Some Chrysanthemum spp. (Chrysanthemum속(屬) 식물의 항균성)

  • The yields of methanol extracts of several Chrysanthemum spp.(5 species, 9 parts) and their solvent fractions were investigated. The yields of methanol extracts ranged from 16.9%(for flower of C. indium, Cultivated) to 31.5%(for whole plant of C. indium). In the tests of the antibacterial activity. the methanol extracts from flower of C. zawadskii and C. boreale, whole plant of C. zawadskii, and flower of C. coronarium showed excellent antibacterial activity. Generally, the chloroform fractions exhibited stronger antibacterial activity against all the bacteria tested when compared with other solvent fractions. In case of C. boreale, the chloroform fraction of the whole plant was shown stronger antibacterial activity against all four bacterial strains tested when compared with that of the flower. The chloroform fractions from flower and whole plant of C. boreale and C .zawadskii, and flower of C. coronarium were shown a similar TLC pattern.

Antioxidant Activities of the Solvent Extracts from Tetragonia tetragonioides (번행초 추출물의 항산화 활성)

  • To clarify the antioxidation effect of the various solvent fractionation of Tetragonia tetragoniodes which has been known to superior plants for the traditional prevention and treatment of stomach-related diseases, total polyphenol and flavonoid contents, vitamin E content, the elimination activity of a DPPH free radical and superoxide anion, inhibition activity of the superoxide generation, reducing power and metal chelating effects were estimated. The contents of the polyphenol compounds were highest in the DCM and EA fractionation, and the content of flavonoid was high in the order of HX and EA fractionation. The vitamin E content showed high in the order of the HX and DCM fractionation among solvent fractions. $IC_{50}$ for the elimination effect of a DPPH radical were estimated as 554.25 and $394.96{\mu}g/ml$ in the DCM and EA fractions, respectively. These values were higher than that of BHT $784.7{\mu}g/ml$ widely used in antioxidation effect. The inhibition activity of the superoxide generation using the T. tetragoniodes solvent fractions represented the effects similar to that of ${\alpha}$-tocopherol known to prevent the lipoprotein oxidation, but lower consequences than that of the phenol-resins, BHT and BHA, respectively. In the antioxidation activity derived by the reducing capability, the EA fractionation in a 1.5 mg/ml concentration showed higher than that of ${\alpha}$-tocopherol.

Screening of Functional Materials from Solvent Fractions of Apple Flower Leaf Extract (사과꽃잎 추출물의 용매 분획으로부터 기능성 소재의 탐색)

  • Fractional solvent extraction by organic solvents such as hexane, chloroform, ethylacetate, and butanol was carried out using 70% ethanol extract of apple flower leaves. Biological activities including antioxidant, whitening, antimicrobial and anti-wrinkle activities were investigated and bio-active materials of the extracts were identified using GC/MSD. Among the tested solvent fractions, ethylacetate fraction showed the highest total polyphenol content (1218.94 ${\mu}g/mL$), and flavonoid (140 ${\mu}g/mL$). The DPPH radical scavenging activities was over 80% at a dry matterbased concentration of 200 ${\mu}g/{\mu}L$ and SOD-like activity was over 90% at 50 ${\mu}g/mL$ concentration in ethylacetate fraction that was slightly lower than of ascorbic aicd. Tyrosinase inhibition activity related to skin-whitening was over 60% by ethylacetate fraction of 100 ${\mu}g/mL$. As an anti-aging effect, elastase inhibitory activity was about 45% in ethylacetate fraction. Also, it showed a significantly antimicrobial activity against P. acenes. From GC/MSD analysis, a characteristic peak of high content in ethylacetate fraction was identified as kaempferol, which has been reported as a bioactive compound.

Addictive Effects of Ecklonia cava Methanol and its Solvent-soluble Extract Against Antibiotic-resistant Cutibacterium acnes Causing Acne Vulgaris (항생제 내성 Cutibacterium acnes에 대한 감태(Ecklonia cava) 추출물의 항균 시너지 효과)

  • Acne vulgaris is often associated with acne-related bacteria such as Cutibacterium acnes. In this study, we investigated the antibacterial effects of the methanol extract of the algae Ecklonia cava and its solvent-soluble extract against C. acnes. Among five solvent fractions, the ethyl acetate (EtOAc) fraction exhibited the strongest antibacterial activity against C. acnes. Furthermore, the EtOAc-soluble extract exhibited the highest total phenolic contents among the five solvent fractions tested. The EtOAc subfraction 07 (Fr. 07) extract showed the highest antibacterial effect against C. acnes and isolated C. acnes along with minimum inhibitory concentration (MIC) ranging from 32 to 64 ㎍/mL. Additionally, MICs of antibiotics against antibiotic-resistant C. acnes strains were substantially reduced when antibiotics were combined with Fr. 07, suggesting that Fr. 07 restore the antibacterial activity of the antibiotics. The fractional inhibitory concentration indices clearly revealed an additive synergistic effect of Fr. 07 with antibiotics. The results of the present study suggest a potential role for E. cava in the control of infections related to acne vulgaris.

$S^{35}$(1,2-dichloro vinyl) L-Cysteine의 소에 의(依)한 대사물(代謝物)의 분리(分離)

  • It has been ascertained that S-(1, 2-dichlorovinyl) L-cysteine (DCVC) is probably the toxic component in the trichloro-ethylene extracted soybean oil meal on the bovine. For the study of the metabolites of DCVC, the components with radioactive $S^{35}$-in the urine of the calf, to which $S^{35}$-DCVC was administrated, were separated using of cellulose powder with propanol-water (70:30 V/V) which is easily removable by evaporation. As the results followings were obtained: Peak 1, which shows fractions containing single $S^{35}$-radioactive components, whose Rf is 0.815 Peak 2, which shows fractions containing jingle $S^{35}$-radioactive component, whose Rf is 0.447 Peak 3, which shows fractions containing both $S^{35}$-radioactive components whose Rfs are 0.090 and 0.171 Peak 4, which shows fractions containing single $S^{35}$-radioactive component, whose Rf is 0.142. Peak 5, which shows fractions containing single $S^{35}$-radioactive component, whose Rf is 0.084. Besides these, two of small peak were obtained. Although, the resolving power of the cellulose powder column is not sufficient, it is applicable for the study of the components of metabolytes of DCVC conveniently with the rest of removable solvent easily. Also the components with radioactive $S^{35}$ in the feces of the calf were separated using citrate buffer gradient system of Moore and stein. As the results; three $S^{35}$-radioactive components were separated.

Cytotoxic Activity of the Extracts from Curcuma zedoaria

  • The effect of the hexane extract of Curcuma zedoaria roots and its solvent fractions were investigated on the proliferation of SiHa, SNU-1 and HepG2 cell lines. Among those fractions, final fraction H2-3-1 and H2-3-3 showed cytotoxic effect on SiHa and HepG2 cell lines. The hallmark of apoptosis, DNA fragmentation, also appeared in the final fractions H2-3-1 and H2-3-3 after 24h treatment in SiHa cell line. Furthermore, those fractions were shown to be able to induce cell death in $[^3H]$thymidine incorporation test. These two fractions, H2-3-1 and H2-3-3 were determined as (-)-$\alpha$-curcumene and $\beta$-tumerone respectively by NMR and mass spectrum. From these results, it is speculated that te hexane extract of Curcuma zedoaria is necessary for further studies as a potent inhibitor of the growth of cancer cells.

Isolation and Concentration of Organic Components from a Complex Matrix into Three Fractions of Different Volatilities (복합 유기혼합물체로부터 휘발성이 서로 다른 세 유기화합물 그룹의 분리 농축방법의 연구)

  • A simple micro-sampling system is described which facilitates isolation and concentration of complex organic constituents into three fractions of different volatilities. The method involves the headspace trapping of very volatile components from a complex matrix onto a porous polymer, Tenax GC, followed by the solvent elution of the matrix and the subsequent fractions of the eluate into volatile and less-volatile fractions. The headspace and the volatile fractions are then analyzed by high-resolution capillary gas chromatography. The less-volatile fraction is analyzed by high-performance liquid chromatography. Experimental details and the results obtained using tobacco leaves as a complex organic matrix are presented.

Effect of Saliva miltiorrhiza Bunge on Antimicrobial Activity and Resistant Gene Regulation against Methicillin-Resistant Staphylococcus aureus (MRSA)

  • This study was conducted in an effort to evaluate the antimicrobial activity and antibiotic-resistant gene regulation from Saliva miltiorrhiza Bunge on methicillin-resistant Staphylococcus aureus (MRSA). A variety of solvent fractions and methanol extracts of S. miltiorrhiza Bunge were tested in order to determine its antimicrobial activities against S. aureus and MRSA. As a result, the hexane fraction of S. miltiorrhiza Bunge evidenced the highest levels of antimicrobial activity against S. aureus and MRSA. The MICs of the hexane fraction against various MRSA specimens were $64. The hexane fraction evidenced inhibitory effects superior to those of the chloroform fraction. The results showed inhibition zones of hexane (16 mm) and chloroform (14 mm) fractions against MRSA KCCM 40511 at $1,000{\mu}g/disc$. The hexane and chloroform fractions inhibited the expression of the resistant genes, mecA, mecR1, and femA in mRNA. Moreover, the results of Western blotting assays indicated that the hexane and chloroform fractions inhibited the expression of the resistant protein, PBP2a. These results reveal that the hexane and chloroform fractions of S. miltiorrhiza Bunge may prove to be a valuable choice for studies targeted toward the development of new antimicrobial agents.

Physicochemical characteristics of mackerel and tuna viscera as baits for swimming crab Portunus trituberculatus pots (꽃게 통발용 미끼로서의 고등어와 다랑어 내장의 이화학적 특성)

  • This study was conducted to investigate the attractant substances of different solvent fractions of mackerel and tuna viscera. Mackerel and tuna viscera were extracted with methanol and the resultant were fractionated with n-hexane, chloroform, ethyl acetate, n-butanol and water. The chemical compounds such as free amino acid, fatty acid, nucleotide related compounds contents were estimated for mackerel, tuna viscera and various fractions. These fractions were also subjected to attracting experiments in water tank to estimate attracting effects. The aqueous fractions of mackerel and tuna viscera showed the highest attraction effect on swimming crab. The major chemical compounds of the aqueous fractions were histidine, taurine, cysteine, glutamic acid, inosine monophosphate (IMP) and inosine (HxR) for mackerel and arginine, glutamic acid, aspartic aid, alanine, IMP and hypoxanthin (Hx). Results indicated that higher polarity compounds, such as amino acid and nucleotide related compounds have higher attraction activities than nonpolar substances such as neutral lipid.

The effect of isoflavonoid contents in SH003 and its subfractions on breast cancer (SH003과 분획물의 구성 성분인 이소플라본의 유방암에 대한 효과)

  • Objectives: We investigated the isoflavone contained in SH003 and its fractions, and the effect of these components on the inhibition of breast cancer. Methods: The isoflavones in solvent fractions of SH003 extract were identified by UPLC-MS and its contents were quantified using HPLC analysis. The estrogenic activity of SH003 or fractions was assessed by ERE luciferase assay in estrogen receptor (ER)-positive MCF-7 cells. To test the breast cancer inhibitory effect, the cell viability was measured using an MTT assay. Results: In this study, we demonstrated that SH003 and fractions contain 4 isoflavones which are calycosin-7-β-D-glucoside, formononetin-7-β-D-glucoside, calycosin, and formononetin. Despite containing isoflavones, estrogen-dependent transcription activity was not altered by both SH003 and fractions. On the other hand, SH003 and fractions inhibited the cell viability of breast cancer. In addition, its isoflavone components also showed reduced cell viability in various breast cancer cells. Conclusions: Overall, the phytoestrogen included in SH003 and fractions did not influence the estrogenic activity, emphasizing the safety of SH003 and fractions in breast cancer treatment.
