Studies on the Grassland Development in the Forest Ⅷ. Effect of shading degrees on the quality, digestibility and nitrate nitrogen concentration of main grasses (林間草地 開發에 關한 硏究 Ⅷ. 遮光程度가 主要 牧草의 品質, 消化率 및 窒酸態窒素含量에 미치는 影響)

  • A field experiment was conduted to determine the effects of shading degrees (O: full sunlight, 25, 50 and 75%) on the proximate components, cell wall constituents (CWC), digestibility, water soluble carbohydrates (WSC) and nitrate nitrogen ($NO_3$-N) concentration of grasses grown in forest. For the test different artificial shading houses were established and pasutre species used were orchardgrass, timothy, perennial ryegrass and ladino clover. the experiment was performed at LES in Suwon, 1985 1. Considering proximate components, CWC and digestibility of grasses, ladino clover showed the best quality, and then perennial ryegrass. 2. The contents of crude protein, crude ash, and digestibility of grasses were increased with shadking, regardless of pasture species. As the shading degrees are increased, the contents of crude fibe in orchardgrass, perennial ryegrass and timothy were decreased, while that in ladino clover was increased with shading. 3. Grasses grwon in spring showed higher digestibility than those grwon in summer season. 4. The content of WSC was the highest in perennial ryegrass, and then ladino clover, orchardgrass, and timothy, in that order. Also WSC was decreased as the shading degrees are in creased. 5. The content of $NO_3$-N was the highest in perennial ryegrass, and then orchardgrass, ladino clover and timothy, in that order. Also the $NO_3$-N was significantly increased with higher shading level. In the regression equation between shading degrees and $NO_3$-N ($r^2=0.90^{**},\;r^2=0.95^{**}$), shading degree of 43 to 44% was critical level, causing nitrate poisoning to animal. 6. Considering grass quality, dry matter yield and $NO_3$-N, less than 40% of shading degree (over 60% of full sunlight) was desirable for better grassland improvement, management and utilization in the forest.

  • Contents of proline and chloride in halophytes were $80-1700\;{\mu}g/gfw$ and 0.13-0.45 mM/gfw respectively. The content of proline was inversely proportional to that of chloride. Rhizomes of Phargmites communis Trin, a halophyte, were grown in non-saline medium and then taken to saline treatment for one or two weeks. Growth of P. communis was inhibited when salinized with 0.25M NaCl. Total nitrogen decreased and alcohol soluble nitrogen and proline increased when growth was retarded. The quantity of Fraction 1 protein decreased at 0.25M NaCl treatment. The accumulation of proline at high concentration in P. communis suggested that it might play a role in osmotic adjustment.

Screening of yeast for alcoholic fermentation of no sugar-added tomato concentrate (토마토 농축액을 이용한 무가당 알코올 발효를 위한 효모의 선발)

  • The purpose of this study was to screen desirable yeast strains for alcoholic fermentation of tomato paste without sugar addition. The moisture, crude protein, crude fat, ash, and soluble nitrogen contents of the tomato paste ($25^{\circ}Brix$) were found to be 67.33%, 1.90%, 0.03%, 0.02%, and 30.72%, respectively. Free sugars found in the paste were fructose and glucose. Most abundant free amino acids of the paste were glutamic, aspartic, and ${\gamma}$-aminobutyric acids. Total seven yeast strains (Saccharomyces cerevisiae KDH (TWA), S. cerevisiae Lalvin ICVD-47 (TWB), S. cerevisiae Lalvin RC-212 (TWC), S. cerevisiae Lalvin K1-V1116 (TWD), S. bayanus Lalvin EC-1118 (TWE), S. cerevisiae Enoferm (TWF), and S. cerevisiae DJ97(KCTC8842P) (TWG)) were tasted for alcohol fermentation of the tomato paste. The highest alcohol content (8.2%) and the lowest residual sugar content ($13.25^{\circ}Brix$) were observed in the tomato paste fermented using the S. cerevisiae Lalvin ICVD-47 strain (TWB) after 3 day and 4 day of fermentation, respectively. Sugar and reducing sugar contents, and pH of the tomato paste were not remarkably affected by the difference in yeast strains used, showing $13.25{\sim}13.45^{\circ}Brix$, 28.37~28.48 mg/mL, and 4.43~4.54, respectively, after 4 day of fermentation. Color and total acid content were significantly affected by the types of yeast strains and fermentation time, but the numerical changes were negligible. These results indicate that TWB would be the suitable strain for alcoholic fermentatiom of tomato paste based on its highest alcohol production and the lowest residual sugar content produced during fermentation.

Physicochemical Properties of Ripe and Dry Jujube ($Ziziphus$ $jujuba$ Miller) Fruits (성숙과 건조 대추의 이화학적 특성)

  • This study was carried out to investigate the physicochemical characteristics of maturating and dried jujube. The moisture contents of the unripe and ripe jujube were found to be 84.66 and 66.45%, respectively, but that of the dried jujube was 31.48%. The crude-fat and crude-ash contents of the dried jujube were much higher than those of the maturing jujube. The carbohydrate contents of the unripe, ripe, and dried jujube were found to be 13.51, 27.94, and 61.70%, respectively, and the soluble-protein contents were 0.88, 1.73, and 3.71%. The reducing-sugar content of the dried jujube was 18.82%, higher than that of the ripe jujube. The sucrose contents of the unripe and ripe jujube were 10.15 and 16.66 g/100 g, respectively, and that of the dried jujube was 33.46 g/100 g. The major fatty acids that were found to compose jujube were palmitic, oleic, pamitoleic, linoleic, and linolenic acid. The myristoleic and arachidonic acid contents were higher in the dried jujube than in the ripe jujube, but the palmitoleic acid content was lower in the dried jujube than in the ripe jujube. The major minerals of jujube were found to be Ca, K, Mg, and P, and the total mineral contents of the unripe, ripe, and dried jujube in this study were 343.38, 584.94, and 331.56 mg/100 g, respectively. The polyphenol contents of the unripe, ripe, and dried jujube were 309.12, 248.80, and 23.34 mg/100 g, respectively, while the vitamin C contents were 610.04, 310.22, and 423.27 mg/100 g.

Comparison of Muscle Color, Taste and Nutrition Components Between Red Seabreams Cultured by Feeding and Starving (급이 및 비급이 참돔의 색, 맛 및 영양성분 비교)

  • For the effective use of cultured red seabream, the muscle color, taste and nutrition components between red seabreams cultured by feeding and starving were compared. The proximate composition of red seabream muscle cultured by starving (RCS) was 72.7% moisture, 21.1% protein, 3.7% lipid and 1.4% crude ash. In comparison to red seabream muscle cultured by feeding (RCF), moisture and crude lipid of RCS were each 3% higher and 3% lower. No difference was, however, found in the other proximate compositions. The Hunter color value of RCS was 37.52 for L value, -1.47 for a value, 0.71 for b value and 59.33 for ${\Delta}E$ value, which was slightly higher in the L value than that of red seabream muscle cultured feeding (RCF); however, no differences were found in the other Hunter color values. TCA soluble-N content of RCS was 403.8 mg/100 g, which was higher than that of RCF (314.7 mg/100 g). In taste values, the major free amino acids of both RCS and RCF were glutamic acid, alanine, lysine and histidine. Total amino acid content of RCS was 21.2 g/100 g, which was higher than that of RCF. The mineral content of RCS was slightly higher in potassium than that of RCF, while lower in magnesium. According to the result of sensory evaluation, RCS was superior in taste and texture to RCF, while similar in color and flavor.

Food Component Characterization of Muscle around Pectoral Fin, Salmon Fillet Processing By-products (연어 Fillet 부산물인 가슴지느러미 부근 근육의 식품성분 특성)

  • For the effective use of salmon processing by-products, the food components of muscle around pectoral fin (MAPF) were investigated and compared with those of salmon fillet muscle (SFM). The proximate composition of MAPF was 64.4% for the moisture, 17.2% for the crude protein, 16.2% for the crude lipid and 1.4% for the ash. The pH, volatile basic nitrogen (VBN) content and yield of MAPF were 6.73, 7.8 mg/100 g and 7.4 (g/100 g SFM), respectively. The trichloroacetic acid (TCA) soluble-N content of MAPF was 479 mg/100 g, which was lower than that (612 mg/100 g) of SFM. The Hunter value of MAPF was 54.86 for L value, 22.08 for a value, 22.41 for b value and 52.35 for ${\Delta}$E value, which were similar to those of SFM. The total content in amino acids of MAPF was 17.1 g/100 g, its major amino acids were aspartic acid, glutamic acid, leucine and lysine. The major fatty acids of MAPF were 16:0, 18:1n-9 and 18:2n-6, 20:5n-3 and 22:6n-3. No differences were found in the major fatty acids and the major amino acids between MAPF and SFM. The calcium and phosphorus contents of MAPF were 19.1 mg/100 g and 211.7 g/100 g, respectively. These results suggested that MAPF could be used as seafood resources.

Physicochemical and Physiological Activities of Garlic from Different Area (산지별 마늘의 이화학적 특성 및 생리활성 작용)

  • The physicochemical and physiological activities of domestic garlic from 3 different areas (Namhae, Jeju and Uiseong) were analyzed. The contents of moisture, ash, crude protein and crude fiber in garlic were little different in 3 kinds of area. Total sugar and water soluble phenolic compounds were higher in garlic from Namhae. The free sugars found in garlic were fructose, sucrose and lactose. Five kinds of organic acids were determined. Malonic acid and citric acid contents in garlic from Namhae were 23.7${\pm}$1.16 mg% and 22.1${\pm}$0.82 mg%, respectively. Total mineral content of garlic samples were in a range of 7112.6~9067.3 mg%, the potassium content showed the highest concentration (4117.3${\pm}$7.19~5175.3${\pm}$9.61 mg%). The electron donating abilities in 0.2% of garlic from Namhae and Uiseong showed 46.2${\pm}$1.25% and 37.0${\pm}$1.l6%, respectively. The nitrite scavenging effect was measured at different conditions (pH 1.2 and 4.2). The nitrite scavenging effects were higher at pH 1.2, and reached more than 95% by adding 0.2% and 0.1 % of garlic juice at pH 1.2. Addition 0.02~0.001 % garlic juice in showed the SOD-like activities. Its activity of garlic from Namhae. was a range of 6.0${\pm}$0.37~14.4~0.69%. It was found that 0.2% and 0.1% garlic showed strong antimicrobial action against growth of all the tested bacteria. Antimicrobial action. was showed 74.7${\pm}$0.70% and 51.7${\pm}$1.l1% on Sal. typhimurium in 0.2% of garlic from Namhae and Uiseong and 28.6${\pm}$0.90% on B. subtilis in garlic from Jeju.

Biological Activities and Quality Characteristics of Rice Germ after Microbial Fermentation (미생물 발효 쌀 배아의 품질특성 및 생리활성)

  • This study investigated the quality characteristics and biological activities of rice germ fermented by Bacillus spp. During the milling process, the contents of rice germ in the rice bran (Control) were adjusted to 30% (RG30) and 70% (RG70). The approximate composition, pH, total acidity, total soluble solid, total sugar, polyphenol and flavonoid contents were measured. DPPH radical scavenging activity, xanthine oxidase (XO) and angiotensin converting enzyme (ACE) activities were also determined. We observed that the moisture content decreased after fermentation, while the crude protein was significantly increased. Fermentation remarkably lowered the pH from 5.83~6.26 to 4.77~4.93, thereby elevating the total acidity. Fermentation also increased the total solid contents, from $0.40{\sim}0.87^{\circ}Bx$ to $1.63{\sim}2.20^{\circ}Bx$. The total sugar decreased to 136.81~151.53 mg/mL from 377.56~450.64 mg/mL. Polyphenol contents were the highest in control (0.59 and 0.73 mg/mL before and after fermentation, respectively). Fermentation significantly affected the increase of the polyphenols in both rice germ 30% and 70% samples, from 0.26 and 0.28 mg GAE/g before fermentation, to 0.52 and 0.70 mg GAE/g after fermentation, respectively. There was a slight increase in the flavonoid contents after fermentation. The $IC_{50}$ value of the electron donating ability, as evaluated by the DPPH method, was the lowest in control (3.77 and 3.36 mg/mL before and after fermentation, respectively). Fermentation increased the XO inhibition activity up to 63.69% in control, 49.81% in rice germ 30%, and 59.32% in rice germ 70%. The ACE inhibition activities were also increased in the fermented control, rice germ 30% and 70%, to 40.51%, 22.69% and 33.91%, respectively.

Quality Properties of Appenzeller Cheese Added with Chlorella (클로렐라 첨가 아펜젤러 치즈의 품질 특성)

  • Effects of Chlorella powder on the growth of lactic acid bacteria, ripening velocity and organoleptic properties in Appenzeller cheese were investigated. Added levels of Chlorella powder were 0, 0.5, 1.0 and 2.0%. The lactic acid bacteria count was higher in cheese added with Chlorella than those in the control cheese. The pH of cheese increased gradually after 3 weeks, reaching pH $5.4{\sim}6.2$ at 15 weeks of maturation, and the pH was slightly lower in Chlorella added cheese than in control cheese. The soluble nitrogen compounds, non casein nitrogen (NCN) and non protein nitrogen (NPN), in Appenzeller cheese increased during 15 weeks of ripening, and they were higher in Chlorella added cheese than in control cheese. Electrophoresis of cheese proteins revealed that caseins were degraded more rapidly in Chlorella cheese as the level of Chlorella increased so that the cheese with 2% Chlorella could have developed a bitter taste and a stench by an excessive degradation of proteins. Sensory scores of the cheese ripened for 15 weeks were diminished as the level of Chlorella increased especially the cheese added with 2% Chlorella obtained significantly lower values of sensory scores than control cheese. Among the Chlorella cheeses, 0.5% Chlorella added cheese showed the highest score in overall sensory preference. From the results, the adequate level of Chlorella powder being added to produce an Appenzeller cheese product with acceptable quality was suggested to be 0.5%.

Effect of Supercritical Carbon Dioxide on Physicochemical Properties and Microbial Reduction of Freeze-Dried Bovine Liver (초임계 이산화탄소 처리가 동결 건조된 소간의 이화학적 특성 및 미생물 저감화에 미치는 영향)

  • Supercritical carbon dioxide ($SC-CO_2$) treatment has been becoming an important method for substituting the use of organic solvents for samples extraction prior to analysis due to its low toxicity, ease of handling, low cost of disposal etc. Freeze-dried bovine liver was treated with $SC-CO_2$ under different pressures (200, 300, and 450 bar) in order to investigate effects on physicochemical properties and reduction of microbial load. The yield of lipid extraction from bovine liver by $SC-CO_2$ treatment increased with increasing pressure, with values of 84, 86, and 90% in response to 200, 300, and 450 bar, respectively. Results of high performance liquid chromatography analysis showed that vitamin A and coenzyme $Q_{10}$ ($CoQ_{10}$), which is soluble in lipid, were almost removed from bovine liver by $SC-CO_2$ treatment. Saturated fatty acids ratio of bovine liver decreased with increasing pressure, whereas polyunsaturated fatty acids increased with increasing pressure. Total content of amino acids in bovine liver treated by $SC-CO_2$ was less than that of the control sample without treatment. The number of aerobic bacteria in bovine liver, which was stored at $5^{\circ}C$ for 5 days and freeze-dried, decreased from 6.2 to 4.2 log CFU/g by $SC-CO_2$ treatment at 100 bar for 3 h. Interestingly, coliform bacteria were not found in the bovine liver sample by $SC-CO_2$ at 100 bar for 3 h under all storage conditions. This indicates that $SC-CO_2$ treatment can effectively reduce coliform bacteria in the food matrix even at low moisture. In conclusion, freeze-dried bovine liver by proper $SC-CO_2$ treatment may be used as a potential high protein source, with increasing microbial safety and stability of lipid oxidation.
