Soluble Expression of Recombinant Human Smp30 for Detecting Serum Smp30 Antibody Levels in Hepatocellular Carcinoma Patients

  • Senescence marker protein 30 (SMP30), a hepatocellular carcinoma (HCC) associated antigen, was earlier shown by our research group to be highly expressed in HCC paracancerous tissues, but have low levels in HCC tissues. In order to detect anti-SMP30 antibody in serum of HCC patients, we established pET30a-SMP30 and pColdIII-SMP30 expression systems in Escherichia coli. However, the expression product was mainly in the form of inclusion bodies. In this research, we used several combinations of chaperones, four molecular chaperone plasmids with pET30a-SMP30 and five molecular chaperone plasmids with pColdIII-SMP30 to increase the amount of soluble protein. Results showed that co-expression of HIS-SMP30 with pTf16, combined with the addition of osmosis-regulator, and a two-step expression resulted in the highest enhancement of solubility. A total of 175 cases of HCC serum were studied by ELISA to detect anti-SMP30 antibody with recombinant SMP30 protein. Some 22 were positive and x2 two-sided tests all showed P>0.05, although it remained unclear whether there was a relationship between positive cases and clinical diagnostic data.

Effect of Solvent on the Grafting Polymerization of Polyethylene Wax with Maleic anhydride (폴리에틸렌 왁스와 무수 말레인산의 그라프팅 중합 반응에서 용매가 미치는 영향)

  • In this study, we have investigated the influence of the nature of solvent on the grafting reaction of maleic anhydride onto polyethylene wax obtained as a by-product in a high density polyethylene plant. The results show that the grafting ratio in xylene as a solvent was higher than toluene. This is because xylene has excellent monomer solubility, swelling property and miscibility. It has been also observed that grafting degree shows an initial jump in percentage of grafting with increasing amount of solvent, from 0% v/w to 200% v/w giving maximum grafting in 200% v/w and then slightly decreases on further increase in the amount of solvent and becomes almost constant. It can be also seen that gel content was not formed under the use of solvent. It means that solvent prevented cross-linking reaction due to chain transfer reactions to solvent molecules. Studies of melt viscosity at $140^{\circ}C$ showed that viscosity increased after grafting of maleic anhydride onto polyethylene wax.

A Study on the Separation of Cerium from Rare Earth Precipitates Recovered from Waste NiMH Battery (폐니켈수소전지에서 회수된 희토류복합 침전분말로부터 세륨 회수에 대한 연구)

  • In order to recover the cerium contained in the spent nickel metal hydride batteries (NiMH battery), the recovered rare earth complex precipitates from NIMH were converted into rare earth hydroxides through ion exchange reaction to react with NaOH aqueous solution at a reaction temperature of 70 ℃, for 4 hours. Rare earth hydroxides were oxidized by injecting air at 80 ℃ for 4 hours to oxidize Ce3+ to Ce4+. The oxidation rate of cerium was confirmed to be about 25 % through XPS, and the oxidized powder was separated from the rest of the rare earth using the difference in solubility in dilute sulfuric acid. The finally recovered powder has a crystal phase of cerium hydroxide (Ce(OH)4). The cerium purity of the final product was about 94.6 %, and the recovery rate was 97.3 %.

Extraction of nitrate salts from de-NOx waste scrubbing solution (탈질 스크러버의 폐세정액으로부터 질산염 추출 연구)

  • Recovery of low level nitric acid and sulfuric acid ions, which were contained in wastewater of a wet scrubber for de-NOx and de-SOx from ship engines, was attempted as fertilizing materials. This study utilized a selective extraction method using four organic solvents to precipitate the solid salts of ammonium nitrate and ammonium sulfate. The IR analysis showed almost same composition of the extracted ammonium salts with a commercial product, and recovery rate of nitrogen and sulfuric ions was 89% and 80% respectively. It was found that the selectivity and solubility consequently could be the crucial factors to recover the low level ions from the waste scrubbing water.

  • Fructans are polyfructose molecules that function as nonstructural storage carbohydrates in several plants. In addition, it has been suggested that, due to their solubility, they can play an important role in helping plants survive periods of osmotic stress. In order to study the effect of levan synthesis on plant growth, the coding region of the levansucrase gene, which was isolated from Zymomonas mobilis, was introduced into tobacco plants using Agrobacterium tumefaciens-mediated transformation. The presence of the levansucrase gene in transgenic plants was verified by genomic DNA gel blot analysis. RNA gel blot and immunoblot analyses showed an accumulation of the corresponding transcript and protein product of the bacterial levansucrase gene in transgenic plants. Furthermore, a thin layer chromatography analysis revealed that fructans were synthesized and deposited in transgenic tobacco plants. When $T_1$ seeds were germinated and grown under polyethylene glycol-mediated drought stress or cold stress, the transgenic seedlings displayed a substantially higher level of growth than that of untransformed plants. These results suggest that fructans may playa significant role in the tolerance of plants under osmotic stress.

Quality Characteristics of Instant Coffee with Probiotics and Microground Coffee (Probiotics와 미세분쇄 원두커피가 첨가된 인스턴트 커피의 품질적 특성)

  • Bacillus coagulans was added as probiotics in instant coffee with microground roasted coffee which is recently rising premium coffee and the instant coffee (ProBio coffee) was compared with 3 commercial instant coffees with microground roasted coffee in quality characteristics to understand the competitiveness of ProBio coffee. In sensory evaluation, ProBio coffee had inferior aroma intensity and overall acceptance compared with control group (brewed coffee) (p<0.05) but it had equal quality or more compared with 3 commercials. Total polyphenol content, chlorogenic acid content, DPPH free radical scavenging activity and caffeine content were $110.72{\pm}1.99mg/g$, $2,700{\pm}20mg/g$, $146.22{\pm}3.62TEAC\;mg/g$, $28.1{\pm}3.2ppm$ respectively. In general quality characteristics, water content, solubility, particle size and particle strength of ProBio coffee were similar to general instant coffee but had darker color than it. In safety assessment, acrylamide content was $502{\pm}10ppb$, and residual pesticides and ochratoxin A were not detected. Over 70% of B. coagulans were maintained in 4 months of storage and 16 months of shelf life was predicted in ProBio coffee by $Q_{10}$ model. Therefore, ProBio coffee was confirmed to have a sufficient product competiveness compared with 3 commercials.

Characteristics of the Shells and Calcined Powders from the Butter Clam Saxidomus purpuratus and Littleneck Clam Ruditapes philippinarum as a Natural Calcium Resource (천연칼슘소재로서 개조개(Saxidomus purpuratus)와 바지락(Ruditapes philippinarum) 패각 및 소성분말의 특성)

  • Shell waste from the butter clam Saxidomus purpuratus and littleneck clam Ruditapes philippinarum is a large by-product of shellfish aquaculture, and it is desirable to convert it into value-added products for industrial applications. In this study, calcium carbonate (CaC) polymorphs from butter clam (BCSP) and littleneck clam (LCSP) shell powders and commercial CaC were characterized using Fourier transmission infrared spectroscopy (FT-IR), X-ray diffraction (XRD), and scanning electron microscopy (SEM). The results revealed that the optimal calcination conditions to eliminate organic substances and improve solubility for both BCSP and LCSP were $800^{\circ}C$ for 8 h in an electrical furnace. Calcination improved the white index of the butter clam (BCCP) and littleneck clam (LCCP) calcined powder compared with shell powders. The calcium content in BCCP (51.1%) was higher than that of LCCP (44.9%) or commercial calcium oxide (CaO, 44.7%). The XRD patterns of BCCP and LCCP were similar to that of CaO. Cubic-like crystals of CaC and irregular crystals of BCCP and LCCP were observed by SEM. The FT-IR and XRD analyses revealed the presence of calcite and aragonite in the BCSP and aragonite in the LCSP, whereas the CaC contained calcite. These results indicate that butter and littleneck clam shells are potential biomass resources for calcium carbonate and calcium oxide.

Synthesis of Subphthalocyanine Compound Derivatives and Their Properties (Subphthalocyanine 화합물 유도체들의 합성 및 특성 비교)

  • Subphthalocyanine (SubPc) derivatives consisting of the different kinds and positions of substitute group were successfully synthesized using various types of the precursor as the intermediate compound. The chemical structure of precursor and product was determined by the means of $^{1}H-NMR$ and FT-IR spectrometer. Also, spectroscopic and crystalline properties were determined by means of UV-Vis spectrometer and XRD, respectively. It was found that SubPc derivatives absorbed selectively visible light ranging from 560 to 600 nm, their solubility was enhanced by introducing substitute groups, and their maximum absorbing wavelength was shifted by changing the kind and position of substitute groups.

Influences of Proteolytic Ability of Lactic Acid Bacteria on Acid Production and Precipitates Occurrence in Liquid Yogurt Preparation (액상발효유 제조시 유산균 Starter의 단백질 분해능이 산생성 및 침전발생에 미치는 영향)

  • In making liquid yoghurt, the influences of proteolytic ability of lactic acid bacteria on acid production and on protein stability were investigated. L. bulgaricus CH-2, L. helviticus IAM 1042 and L. jugurti 3048 showed a comparatively high proteolytic activity in milk, while L. casei YIT 9018 did not show any marked proteolysis. Starter organisms having high proteolytic ability showed more rapid growth and acid production than those having low ability in milk. The most active proteolysis occurred during logarithmic growth phase of yogurt organisms, and most of the proteolysis took place in the first 24-48 hrs of incubation. Highly proteolysed yogurts made by L. bulgaricus CH-2, L. jugurti 3048, L. helviticus IAM 1042, L. acidophilus L-54 and L. casei 3012 had low protein solubility at pH 3.5 and had much protein precipitates during storage of product, but those having little protein hydrolysates made by L. casei YIT 9018 or artificial acidification showed no precipitation during keeping.

Characterization of Acid- and Pepsin-soluble Collagens from Rockfish Sebastes schlegeli Skin

  • Biochemical and functional properties of acid-soluble collagen (ASC) and pepsin-soluble collagen (PSC) from rockfish skin were characterized. Yield of PSC (90.0%) was higher than that of ASC (63.2%). Both ASC and the PSC consisted of ${\alpha}1$ and ${\alpha}2$ chains, and $\alpha$-cross-linked components. According to the results of hydroxylation of proline and lysine, and FT-IR, no difference between the helical structure of ASC and PSC was identified. Thermal denaturation temperature (TDT) of ASC from rockfish skin was $22.8^{\circ}C$, the same as exhibited in PSC. Both ASC and PSC were higher in water absorption capacity (WAC) and oil absorption capacity (OAC) than other vegetable proteins. According to the results of emulsifying activity (EA) and cooking stability (CS), both ASC and PSC from rockfish skin were inferior compared to the commercial emulsifier (Tween-80). The results of FT-IR suggested that the structure of PSC was slightly different when compared to that of ASC. No differences in solubility were established between ASC and PSC from rockfish skin at various pH and NaCl concentrations.
