Some Properties of the Polyphenol Oxidase from Potatoes (Solanum tubersum L.) and Inhibiting Effect of the Polyphenol Oxidase by Sulfites (감자 Polyphenol Oxidase의 효소학적 성질 및 아황산염에 의한 활성억제 효과)

  • This study was aimed at obtaining elementary data on enzymatic browning of potato and potato products and examining the inhibitory method of browning. Therefore, we extracted polyphenol oxidase from potatoes(Solanum tubersum L.), and investigates its general properties and inhibiting effects of its activity with the different concentrations of sulfites($Na_2S_2O_4,\;Na_2SO_3{\cdot}7H_2O,\;NaHSO_3$). The optimum pH and temperature of polyphenol oxidase were observed to be 6.5 and $37^{\circ}C$ respectively. The polyphenol oxidase at PH5 was very stable, and the activity of polyphenol oxidase between pH $5.0{\sim}9.0$ was estimated to be relatively high, showing $72{\sim}75%$ of its activity at pH5. The polyphenol oxidase was very stable when heated at $40^{\circ}C$ for one hour, and almost 50% of enzyme activity was decreased when heated at $70^{\circ}C$ for twelve minutes. At 0.1mM concentrating of sulfites the relative activity of polyphenol oxidase was 98% in all the three cases of sulfites. Thus sulfites at 0.1mM concentration was found to have little inhibiting effect on polyphenol oxidase activity. At 1mM concentration of sulfites $NaHSO_3$ showed the lowest 36% relative activity among the three. At 5mM concentration of sulfites, the relative activity of $Na_2SO_3{\cdot}7H_2O$ was the lowest 14%.

Infestation of Potato Cultivars by Potato Aphid, Macrosiphum euphorbiae Thomas, and its Infestation-related Factors (감자수염진딧물(Macrosiphum euphorbiae Thomas)에 의한 감자 품종별 피해정도와 관련요인 분석)

  • Fifty potato cultivars were screened for potato aphid, Macrosiphum euphorbiae Thomas infestation degree in the field. Aphid honey dew secretion and contents of foliar substances(glycoalkaloids, total nitrogen, Ca, K and Mg) of the cultivars were measured in the laboratory to analyze the correlation between infestation degrees and those factors. The number of aphids ranged from 4.1 to 31.4 per five leaves. The lower density(less than 5 aphids) was found on cvs. Jidose, Recent, and Sebago. The aphid secreted highest(40.3 droplets) and lowest(15.7 droplets) honeydew on cvs. Jopoong and Anco, respectively. Degree of infestation is positively correlated to glycoalkaloids and total nitrogen contents of the potato leaf.

First Report of Potato Stem-End Rot Caused by Fusarium oxysporum in Korea

  • In this study, we identified the causative agent of stem-end rot in potatoes that were grown in Gangwon alpine areas of Korea in 2013. The disease symptoms included appearance of slightly sunken circular lesion with corky rot on the potato surface at the stem-end portion. The fungal species isolated from the infected potatoes were grown on potato dextrose agar and produced white aerial mycelia with dark violet pigments. The conidiophores were branched and monophialidic. The microconidia had ellipsoidal to cylindrical shapes and ranged from $2.6{\sim}11.4{\times}1.9{\sim}3.5{\mu}m$ in size. The macroconidia ranged from $12.7{\sim}24.7{\times}2.7{\sim}3.6{\mu}m$ in size and had slightly curved or fusiform shape with 2 to 5 septate. Chlamydospores ranged from $6.1{\sim}8.1{\times}5.7{\sim}8.3{\mu}m$ in size and were present singly or in pairs. The causal agent of potato stem-end rot was identified as Fusarium oxysporum by morphological characterization and by sequencing the internal transcribed spacer (ITS1 and ITS4) regions of rRNA. Artificial inoculation of the pathogen resulted in development of disease symptoms and the re-isolated pathogen showed characteristics of F. oxysporum. To the best of our knowledge, this is the first study to report that potato stem-end rot is caused by F. oxysporum in Korea.

Potato Basal Stem Rot Caused by Pythium myriotylum in Hydroponic Cultural System (양액 재배시 발생하는 Pythium myriotylum에 의한 감자 줄기기부썩음병)

  • A basal stem rot disease of potato (Solanum tubersum) grown on hydroponic system in greenhouse was found in Jeju-city, Jeju Province of Korea in January 2001. The symptoms occurred on basel stem and root, and than plants eventually were died. The casual fungus isolate from basal stem rot symptom and identified to be Pythium myriotylum on the basis of mycological characteristics; Main hyphae wide were 8.4 ${\mu}{\textrm}{m}$ in size. Oogonia were spherical, smooth, mostly terminal in shape and 26.4∼31.2 ${\mu}{\textrm}{m}$ in size. Antheridia were 9.6∼14,4${\times}$4.8-9.6 ${\mu}{\textrm}{m}$ in size. Oospores were spherical, smooth, aplerotic in shape and 21.6∼26.4 ${\mu}{\textrm}{m}$ in size. This is the first report on the basal stem rot of potato caused by P. myriotylum in Korea.

Effect ofMunicipal Sludge on contents of Cadmium and Zinc in Crop Plants (도시하수(都市下水) Sludge 의 경작지(耕作地) 처리(處理)가 식물체(植物體)중 Cd 및 Zn 함량(含量)에 끼치는 영향(影響))

  • Four crop plants, such as green onion(Allium ascalonicum L.), radish(Raphanus sativus L.), potato(Solanum tubersum L.) and chinese cabbage(Brassica pekinensis Rupr), were grown in greenhouses to determine the accumulations of Cd and Zn absorbed by the plants grown on soils treated with municipal sewage sludge of different levels of 0, 22.5, 45.0 and 90.0 ton $ha^{-1}$ as soil dry weight. The result was analyzed to be comparable to the heavy metal content in the plant tissues among the crop species and the statistical characteristics were investigated to determin a possible relationship between causes of increased contents of Cd and Zn in plants and in soils. The results were as follows : 1. The Cd and Zn content in experimental crop plants increased with increase in quantity of treatment sludge. 2. The Cd and Zn content in roots of all experimental plants increased with sludge treatment amount in soil, in the following order : green onion>potato>chinese cabbage>radish. 3. The Cd content in leaves of crop plants were high in this order : green onion>radish>potato>chinese cabbage, and the Zn content in leaves of crop plants was high in this order. : potato>radish>chinese cabbage>green onion. 4. Concentrations of Cd in potato tubers, radish roots and chinese cabbage leaves ranged from 0.03 to 0.12 ppm. 5. Concentrations of Cd and Zn in plant tissues grown in sludge treated soil of 90.0 ton $ha^{-1}$ were 1.2 to 10 times and 0.4 to 2.3 times higher as compared with those absorbed from sludge-free soil, respectively. 6. The accumulation rates of Cd and Zn in sludge-treated plant tissue were almost constant except for green onion roots.
