Characterictics and Stability of Anthocyanin Pigment Extracted from Purple-Fleshed Potato (자색감자 Anthocyanin 색소의 특성 및 안정성)

  • Effects of pH, sugars, organic acids, metal ions, ascorbic acid and light on the stability of anthocyanin pigment extracted from purple-fleshed potato (Solanum tuberosum L.) were studied. The pH had marked influences on the color of the potato anthocyanin pigment: i.e., the lower the pH of the anthocyanin solution was, the more stable and intenser of the pigment was. It showed characteristic bathochromic shift as the pH of the solution increased. Generally, the addition of sugars into the purple-fleshed potato anthocyanin solution caused decrease in color stability of the pigment. Among the sugars tested, maltose was the most deleterious followed by sucrose, galactose, fructose and glucose. The addition of organic acids greatly increased the stability of the pigment. Malic acid was found to be the most effective in stabilizing the pigment followed by tartaric, citric and succinic acids, while malonic acid was found to be deteriorative to the stability of the pigment. Most metal ions except $Cu^{2+}$ increased stability of the pigment-especially, $Cd^{2+}\;and\;Al^{3+}$ were more effective than the others. Ascorbic acid degraded the pigment considerably, but the adverse effect was diminished by adding thiourea. Light gave an adverse effect to the stability of the purple-fleshed potato anthocyanin pigment, which could be minimized by shielding the light from the pigment.

Quality Changes of Fresh-Cut Potatoes during Storage Depending on the Packaging Treatments (신선한 감자절편의 포장방법에 따른 저장 중 품질변화)

  • To investigate pertinent packaging treatment of fresh-cut potatoes (Solanum tuberosum L.), potato cubes were subjected to passive, gas exchange, and vacuum packaging conditions. Low density polyethylene film (LDPE), polypropylene film (PP), anti-fogging film (AP), and perforated film (PF) were used as passive packaging treatments. Mixed gases of 5% $CO_2/5%\;O_2$ (MA1) and 10% $CO_2/5%\;O_2$ (MA2) were applied as gas exchange packaging. Packs filled with cubes were kept at $5^{\circ}C$, and changes in weight loss, surface color, vitamin C content and sensory quality of cubes were analyzed during storage. Respiration rate of cubes was 2.11 times higher than that of intact raw potatoes at $5^{\circ}C$. Gas concentrations in passive packaging was maintained at 1-2% $O_2$ and 4-14% $CO_2$ after 7 days. Gas levels changed depending on films used. Cubes packed in PP and MA2 showed lowest weight loss and browning during storage. Firmness of cubes was not affected by packaging treatment. Vitamin C content was highest in cubes packed with AF. Cubes packed in MA2 showed highest quality upto 10days storage, followed by those packed in PP and AF.

Selection of bactericides for control of potato Blackleg disease in Korea (감자흑각병 (Potato Blackleg Disease) 방제를 위한 살균제 선발)

  • Potato blackleg disease caused by Erwinia carotovora subsp. atroseptica has been a serious problem in Korea. It was previously reported that four mixtures [streptomycin (9.3 ppm) + copper oxide (171.6 ppm)/copper hydroxide (146.3 ppm), streptomycin sulfate (7.0 ppm)+copper oxide (171.6ppm)/copper hydroxide (146.3 ppm)] were effective for the control of E. carotovora subsp. atroseptica. in in vitro test. Using those four mixtures and two antibiotics [streptomycin (81.4 ppm) and streptomycin sulfate (61.3 ppm)], the effectiveness of control for E. carotovora subsp. atroseptica. was conducted in the field. Two antibiotics showed over 60% of control efficacy under different soil conditions, while mixtures of two antibiotics with copper compounds did not show any control effects on the infected seed potato. Two mixtures [streptomycin (27.9 ppm)+copper hydroxide (438.9 ppm), streptomycin sulfate (21.0 ppm) + copper oxide (514.8 ppm)] were effective in the control of potato blackleg disease on the infected potato plants under different climate conditions.

Molecular Cloning, Sequence Analysis, and in Vitro Expression of Flavanone 3β-Hydroxylase from Gypsophila paniculata (안개초(Gyposphila paniculata)로부터 Flavanone 3β-Hydroxylase 유전자의 분리 및 분석)

  • Flavanone 3$\beta$-hydroxylase (FHT) is an enzyme acting in the central part of the flavonoid biosynthesis pathway. FHT catalyses the hydroxylation of flavanone to dihydroflavonols in the anthocyanin pathway. In this paper we describe the cloning and expression of the genes encoding the flavonoid-biosynthetic enzyme FHT in Gypsophila paniculata L. A heterologous cDHA probe from Dianthus cavophyllus was used to isolate FHT-encoding cDHA clones from Gypsophila paniculata L.. Inspection of the 1471 bp long sequence revealed an open reading frame 1047 bp, including a 190 bp 5' leader region and 288 bp 3' untranslated region. Comparison of the coding region of this FHT cDHA sequence including the sequences of Arabidopsis thaliana, Citrus sinensis, Dianthus caryophyllus, Ipomoea batatas, Matthiola incana, Nierembergia sp, Petunia hybrida, Solanum tuberosum, Vitis vinifera reveals a identity higher than 69% at the nucleotide level. The function of this nucleotide sequences were verified by comparison with amino acid sequences of the amino-terminus and tryptic peptides from purified plant enzyme, by northern blotting with mRHA from wild type and mutant plants, by in vitro expression yielding and enzymatically active hydroxylase, as indicated by the small dihydrokaempferol peak. Genomic southern blot analysis showed the presence of only one gene for FHT in Gypsophila paniculata.

Gui Valley: A High Yielding Potential and Good Processing Potato Cultivar

  • The main objective of this breeding program is to develop high yielding, disease resistance and good processing potato cultivar. 'Gui Valley' is a clonal selection resulting from a cross between 'ND2471-8' and 'Cona'. It has medium plant height and light green foliage. 'Gui Valley' has medium flowering habit and light pink flowers. 'Gui Valley' is a medium maturing potato cultivar and tubers are smooth, yellow skin, light yellow flesh, long-oval tuber shape, distinct red eyes with medium depth and medium dormancy. It has high level of tuber uniformity and good keeping quality. 'Gui Valley' demonstrates resistance to potato virus Y (PVY), soft rot, but moderately susceptible to late blight and common scab. It is also resistant to most of the internal and external physiological disorders particularly dehiscence, hollow heart and internal brown spot. The specific gravity of 'Gui Valley' is significantly higher (1.097) than that of 'Shepody' (1.078). 'Gui Valley' has suitable for processing mainly French fries and chips. This cultivar has high level of tuber uniformity and capable of yielding 37.6 $t{\cdot}ha^{-1}$, which is 18.2% higher than the control potato cultivar 'Shepody' under optimum agronomical practices.

Tuber Yield and Size Distribution of Potato 'Dejima' (Solanum tuberosum L.) Affected by Stem Cutting Ages and Harvest Time in Aeroponics (경삽묘 연령과 수확시기가 분무경재배 씨감자 '대지'의 생육과 수량에 미치는 영향)

  • This study was carried out to investigate the effects of stem cutting ages and harvest time on the growth and yield of potato 'Dejima' in aeroponics. The stem cuttings were produced from in vitro plantlets and transplanted into an aeroponic system with 20, 30, 40 and 50 day-old stem cuttings (DOS). Tubers were harvested 60, 70, 80 and 90 days after transplanting (DAT) and sorted into following categories: $1{\sim}5$, $5{\sim}10$, $10{\sim}20$, $20{\sim}30$, $30{\sim}40$ and over 40 g. Plant height from the 40 DOS was the highest during the growing periods but no significant difference was observed on 75 DAT. The tuber weight increased until 90 DAT with the greatest weight of tubers in the 20 and 40 DOS. However, there was no significant difference among 20, 30 and 40 DOS in the number and weight of tubers. Harvesting at 80 and 90 DAT increased the number of tubers over 5 g, which are usually considered as appropriate for direct field planting.

  • This study was carried out to discriminate potato cultivars and breeding lines by specific molecular markers using random amplified polymorphic DNA (RAPD) analysis. The genotypes of potatoes used for analysis were eight cultivars and five breeding lines. Some of those show much phenotypic resemblances among them because 'Jopung', 'Daekwan70', 'Gawon', and 'Daekwan72' have immediate parental relationship with 'Superior', 'Irish Cobbler', 'Namsuh', and 'Atlantic', respectively. So, there are many difficulties to distinguish the varieties by the morphological characteristics. Three URP primers, URP2, URP4, and URP8 were selected for promising primers to discriminate potato genotypes or cultivars. The three URP primers were shown very high reproducibility because of the relatively high annealing temperature and long primer size. Although the results of similarity analyses did not always reflect the genetic relationship between potato varieties, the reproducible pattern of amplified DNA bands by URP primers showed possibility for molecular markers for discrimination of potato genotype or cultivar.

Glycoalkaloid content in potato tubers by various cooking methods and potato products (감자의 가공제품 종류와 조리방법에 따른 Glycoalkaloid의 함량)

  • The potato tubers of Solanum tuberosum L. contain potato glycoalkaloids (PGA), a class of naturally occurring toxicants. The primary constituents of this class of compounds in potatoes are $\alpha$-chaconine and $\alpha$-solanine. The purpose of this study was to examine the effects of various cooking methods on the PGA contents in the common Korean potato Irish Cobbler variety and those in commercially produced potato products consumed in Korea. After cooking, most potatoes showed big decreases in their PGA contents compared to the uncooked samples, with the exception of baked potatoes, which maintained 90.86% of their PGA contents. The PGA levels in boiled and blanched potatoes were reduced by 91.68 and 84.17%, respectively. However, the levels in samples boiled in 1% salted water were only reduced by 52.10%. Potatoes fried and sauteed in oil had their PGA contents reduced to 49.42 and 49.51% of their original levels. The reductions in the relative PGA contents by cooking method were in the order boiling〉blanching〉boiling with 1% NaCl〉steaming〉sauteing〉frying〉microwaving〉baking. The highest remaining PGA content of the potato products in Korea was in potato chips, at 69.57mg/100g, but made with imported potatoes.

Quality Changes in Fresh-Cut Potato (Solanum tuberosum var. Romano) after Low-Temperature Blanching and Treatment with Anti-Browning Agents (저온 blanching 및 갈변저해물질의 처리에 따른 최소가공 감자의 저장 중 품질변화)

  • Quality changes in fresh-cut potatoes during storage at $4^{\circ}C$ after treatment with low-temperature blanching and antibrowning agents were studied. Fresh-cut potatoes were treated by dipping for 1.5 min in a browning inhibitor solution containing 0.5% (w/v) ascorbic acid, 0.5% (w/v) citric acid, 0.5% (w/v) sodium chloride, 0.1% (w/v) trehalose, and 0.005% (w/v) biotin, at $60^{\circ}C$, with subsequent cooling for 1.5 min and storage at $4^{\circ}C$. The browning properties of fresh-cut potatoes were examined by measurement of polyphenol oxidase (PPO) activity and total phenolic content. Changes in quality attributes over a 14-day period were assessed in terms of titratable acidity, pH, water-soluble solid level, and gas analysis at $4^{\circ}C$. During storage, PPO activity increased, with the lowest activity seen after about 7 days of storage. Treatment with antibrowning solution at $4^{\circ}C$ increased visual sensory attributes during storage. Low-temperature blanching in distilled water more effectively inhibited browning compared with exposure to browning inhibitor solution, as assessed after 7 days of storage. Fresh-cut potatoes respired aerobically after different treatments during storage at $4^{\circ}C$.

Effect of Irrigation and Fertigation Treatments on Growth and Yield in Spring Potato Cultivation (봄 감자 재배 시 관수 및 관비 처리가 생육 및 수량에 미치는 영향)

  • This study was conducted to determine the suitable amount of irrigation and fertigation for the growth and yield of spring potatoes (Solanum tuberosum L.). To investigate the effect of soil moisture content on the growth and yield of potato, five irrigation treatments were performed based on soil moisture tension: -10 kPa, -20 kPa, -30 kPa, -40 kPa, and non-irrigation. The growth and yield among the irrigation treatments was the best in the -10 kPa treatment, where the yield increased by 47% compared to the non-irrigation treatment. To determine the standard of additional fertilizer for fertigation cultivation of spring potatoes, seven treatments were performed, including a control (basic fertilization without additional fertilizer), compound fertilizer (NPK) based on 0.5 times the standard amount of fertilizer (NPK 0.5; N-P2O5-K2O: 2.5-2.2-3.25 kg/10a), NPK 0.8 (N-P2O5-K2O: 4-3.5-5 kg/10a), and NPK 1.2 (N-P2O5-K2O: 6-5.3-7.8 kg/10a), and potassium fertilizer of 2 kg (K 2), 5 kg (K 5), and 7 kg (K 7). The growth and yield among the fertilization treatments was highest in the K 5 treatment, where the yield increased by approximately 27% compared to the control. Therefore, this study suggests that irrigation with -10 kPa or fertigation with K 5 during the tuber initiation and tuber filling stage could increase the yield of spring potatoes.
