충남 대둔산의 토양으로부터 분리한 점액세균을 ARJ라 명명하고 그 특성을 알아보았다. CT 배지에서 swarming을 하며 성장한 이 균주는 slime을 형성하였고, 액체 및 고체배지 상에서 황록색의 색소를 방출하였으며 swarming 부분을 파장이 366 nm인 자외선으로 조사하였을 때 초록색의 형광을 띄었다. 또한 이 균주는 영양분이 고갈되었을 때 fruiting body를 형성하는 것으로 보아 Myxobacteria라고 생각되었으며 주사전자 현미경 관찰 결과 이 균주가 형성한 fruiting body는 stalk이 없었고 naked myxospore를 가지고 있었다. Myxobacteria의 분류에 있어서 가장 중요한 이러한 형태학적 특성들로 볼때 이 균주는 Myxococcus virescens와 가장 유사한 것으로 밝혀졌다. 한편 이 균주는 특히 gram 양성 세균에 대해 antimicrobial activity가 있는 것으로 나타났는데, 여과한 배양 여액에서도 이렇ㄴ activity가 있었지만 비변성 polyacrylamide 전기영동을 하여 본 결과 이 물질이 단백질은 아닌 것으로 밝혀졌다. 또한 이 배양 여액은 상당한 proteolytic activity가 있었으며 최소한 2개의 proteolytic enzyme이 있는 것으로 비변성 polyacrylamide 전기영동을 통하여 밝혀졌다.
The strain S4-14 which produced alkaline lipase and had resistance against linear alkylbenzene sulfonate was isolated from soil or water samples. The isolated strain S4-14 was identified a species belong to Pseudomonas. Alkalin lipase secreted by Pseudomonas sp. S4-14 was purified by ammonium sulfate precipitation procedure follwed by DEAE-Cellulose, DEAE-Sepharose and gel filtration chromatohraphies with 995.15 U/mg protein and 16.1% yield. The molecular weight of the enzyme was estimated to be 65,000 dalton by SDS-PAGE. The optimum pH and temperature of the purified enzyme was 10.5 and 45$\circ $C, respectively. The emzyme was stable at 45$\circ $C for 1 hr and in a pH range from 8.0 to 12.0 for 24 hr at 4$\circ $C. The activity of lipase was enhanced by Ca$^{2+}$ while inhibited strongly by Pb$^{2+}$, Zn$^{2+}$ or Fe$^{3+}$. The activity of lipase was inactivated about 50~60% in the presence of 50 mg/l linear alkylbenzene sulfonate, $\alpha $-olefin sulfonate, alcohol ethoxylate or perborate.
The seed paper was used in farm field recently for a sound young plant. The most of seed paper are made of synthetic non-woven sheet. Therefore, it is very difficult to bio-degrade in soil and is very hard to have some special function, for example keeping herbicide and/or insecticide activity because of its lack of chemical acceptability. The purpose of this research is manufacture of seedling paper which have a function of herbicide activity from waste paper. The fiber properties from waste paper were remarkably improved by fine removal with washing and/or flotation process. The paper-making ability for seed paper was enhanced with enzyme treatment of secondary fibers. The paper for seedling must have a good bio-degradation ability in soils. The absorption amount of chemical like as dithiopyr was increased remarkably in enzyme treated base paper. The embossing treatment of base paper was very effective for seed attachment and chemicals retention. And also, the developed seed paper showed a good penetration property of young root through embossed paper.
A bacterial strain No. HB-5, which was capable of producing a pretense in the culture conditions, was isolated from the soil . The pretense was purified from cultural filtrate of Bacillus sp. HB-5 by membrane ultrafiltration and DEAE- cellulose chromatography, gel filtration on Sephadex G-100. The molecular weight was estimated to be 60k4a. The optimal pH and temperature for the activity of the purified pretense pH were 11 and 5$0^{\circ}C$ , respectively. The enzyme was stable within a pH range 8-12 and up to 6$0^{\circ}C$ . The enzyme activity was highly inhibited by PMSF at 1mM. The proteolytic actions of pretense and papain on human epidermis keratins which are major protein impurities on the skin, were compared. The bacterial pretense degraded more effectively than papain. Product containing 2% protease exhibited 21% increase on the skin coloration index. These results suggest that cosmetic product containing pretense produced by Bacillus sp. HB-5 could remove the adherent keratin layer and then make a softer skin.

A bacterial strain NS-83 isolated from soil was able to produce an extracellular thermostable protease. The strain was identified as Pseudomonas aeruginosa based on its morphological and physiological characteristics. A thermostable protease from this strain has been purified to homogeneity as judged by SDS-PAGE and isoelectric focusing. The purification procedures included hydrophobic interaction, ion exchange, and gel filtration chromatography. The $M_r$ and the pl of the enzyme were 32,000 and 5.9, respectively. The optimal pH at 55$^{\circ}C$ and the optimal temperature at pH 7.0 were 8.0 and 60$^{\circ}C$, respectively. The D-values of the enzyme at 60, 65, and 70$^{\circ}C$ were 22, 2.1, and 0.75 hrs, respectively. The enzyme activity was significantly inhibited in the presence of 1 mM o-phenanthroline or EDTA, suggesting that the enzyme is metalloprotease. The $K_m$, and $V_{max}$ for Hammarsten casein were found to be 3.2 mg/ml and 0.918 unit/ml, respectively. These enzymatic properties were similar to those of elastase produced from P. aeruginosa IFO 3455, but the enzyme was clearly different from the reported elastase, in respect to $Ca^{++}$ effects on enzyme-thermostability. This property, together with amino acid composition analysis, confirmed that the enzyme differs from the known P. aeruginosa elastase.

A novel deoC gene was identified from Paenibacillus sp. EA001 isolated from soil. The gene had an open reading frame (ORF) of 663 base pairs encoding a protein of 220 amino acids with a molecular mass of 24.5 kDa. The amino acid sequence was 79% identical to that of deoxyribose 5-phosphate aldolase (DERA) from Geobacillus sp. Y412MC10. The deoC gene encoding DERA was cloned into an expression vector and the protein was expressed in Escherichia coli. The recombinant DERA was purified using Ni-NTA affinity chromatography and then characterized. The optimum temperature and pH of the enzyme were $50^{\circ}C$ and 6.0, respectively. The specific activity for the substrate deoxyribose 5-phosphate (DR5P) was $62\;{\mu}mol/min/mg$. The $K_m$ value for DR5P was determined to be 145 mM with the $k_{cat}$ value of $3.2{\times}10^2/s$ from Lineweaver-Burk plots. The EA001 DERA showed stability toward a high concentration of acetaldehyde (100 mM).
토성 및 토양수분에 따른 시호근의 생육특성과 saikosaponin함량 및 항산화효소 활성 변화를 조사해 본 결과, 수분과다 토양에서 는 삼도시 호는 장수시호보다 생육 억제정도가 매우 높았고, 건물중은 과습, 적습, 과건 재배 순으로 높았고, 수분조건에 따른 토성별로는 과건은 식양토, 사양토, 사토 순으로, 적습과 과습시는 사토, 사양토, 식양토 순으로 높았다. 토양수분조건별 saikosapon a, d, c함량은 과건, 적습, 과습 순으로 높았고, 토성간에는 사토, 사양토, 식양토 순으로 높았다. POD 활성은 지상부는 장수시호가 삼도에 비하여 매우 높았고, 토양 수분함량간에는 장수는 과건, 과습, 적습 조건 순으로 높았으나, 삼도시 호는 과건, 적습, 과습 순으로 높았다. SOD활성은 지상부와 지하부 공히 장수시호가 높은 경향이나, 수분조건별로는 과건, 과습, 적습순으로 높았으며, 건조상태의 경우 두 품종 모두 사토에서 높았다. 시호에서 뿌리의 Saikosaponin함량과 POD 및 SOD 함량과는 정의상관을 나타내는 경향이었고 줄기 및 뿌리길이와 POD함량과는 부의 상관을 나타내었다.
폐단백질을 활용하는 방도의 하나로 폐단백질 자원으로 부터 불용성 단백질의 분리 효율성을 높이고 기능성을 개선하기 위하여 protease를 생산하는 Aspergillus sp. MS-18 균주를 토양으로 부터 분리하고 이 균주가 생산하는 효소를 정제하여 특성을 살펴보았다. 효소 생산을 위한 최적 배양조건은 3% arabinose, 0.5% polypepton, 0.1% ammonium sulfate, 0.1% magnesium chloride 첨가로 3 일 배양이었다. 효소는 ion exchange chromatography, gel filtration 등으로 16.9 배 정제할 수 있었으며 비활성역가는 340.4 unit/mg이었다. 정제효소는 polyacryl amide gel 전기영동상 단일 밴드로 나타났으며, 분자량은 30,000 정도로 추정되었고 결정구조는 모서리가 둥그스럼한 막대 모양이었다. 정제 효소의 최적작용 pH와 온도는 9.0, $60^{\circ}C$였으며, pH 7.0-12.0까지 $50^{\circ}C$에서 안정하였다. 금속이온중 $Na^+$, $Mg^{2+}$, $Mn^{2+}$등에 의해 활성이 증대 되었으나, $Hg^{2+}$, $Cu^{2+}$, $Zn^{2+}$, $Pb^{2+}$에 의해 효소 활성이 저해되었고 저해제중 ethylenediaminetetra acetic acid와 phenyl methanesulfonyl fluoride에 의한 활성 저해가 관찰되어 금속 이온이 효소 활성에 관여하는 serine protease로 추정되었으며 정제효소의 Km, Vmax는 $29.33\;{\mu}mole/L$, $5.13\;{\mu}g/min$이었다.
The process of biodegradation in lingo-cellulosic materials is critically relevant to biospheric carbon. The study of this natural process has largely involved laboratory investigations, focused primarily on the biodegradation and recycling of agricultural by-products, generally using basidiomycetes species. In order to collect super white rot fungi and evaluate its ability to degrade lingo-cellulosic material, 35 fungal strains, collected from forests, humus soil, livestock manure, and dead trees, were screened for enzyme activities and their potential to decolorize the commercially used Poly-R 478 dye. In the laccase enzymatic analysis chemical test, 33 white rot fungi and 2 brown rot fungi were identified. The degradation ability of polycyclic aromatic hydrocarbons (PAHs) according to the utilized environmental conditions was higher in the mushrooms grown in dead trees and fallen leaves than in the mushrooms grown in humus soil and livestock manure. Using Poly-R 478 dye to assess the PAH-degradation activity of the identified strains, four strains, including Agrocybe pediades, were selected. The activities of laccase, MnP, and Lip of the four strains with PAH-degrading ability were highest in Pleurotus incarnates. 87 fungal strains, collected from forests, humus soil, livestock manure, and dead trees, were screened for enzyme activities and their potential to decolorize the commercially used Poly-R 478 dye on solid media. Using Poly-R 478 dye to assess the PAHdegrading activity of the identified strains, it was determined that MKACC 51632 and 52492 strains evidenced superior activity in static and shaken liquid cultures. Subsequent screening on plates containing the polymeric dye poly R-478, the decolorization of which is correlated with lignin degradation, resulted in the selection of a strain of Coriolus versicolor, MKACC52492, for further study, primarily due to its rapid growth rate and profound ability to decolorize poly R-478 on solid media. Considering our findings using Poly-R 478 dye to evaluate the PAH-degrading activity of the identified strains, Coriolus versicolor, MKACC 52492 was selected as a favorable strain. Coriolus versicolor, which was collected from Mt. Yeogi in Suwon, was studied for the production of the lignin-modifying enzymes laccase, manganese-dependent peroxidase (MnP), and lignin peroxidase (LiP).
